The Identification and Classification of Bacteria (ICB) database (http:/www.mbio.co.jp/icb) contains currently available information about the DNA gyrase subunit B (gyrB) gene in bacteria. The database is designed to provide the scientific community with a reference point for using gyrB as an evolutionary and taxonomic marker. Nucleic and amino acid sequence data are currently available for over 850 strains, along with alignments at several different taxonomic levels and an exhaustive review of primer selection and background information.

It is known that Aeromonas spp. possess different chromosomal beta-lactamase genes. Presence and phenotypic expression of bla(TEM), bla(SHV), and bla(CTX-M) ESBL-encoding genes were investigated in environmental water isolates of Aeromonas hydrophila and Aeromonas jandaei. Presence of blaSHV and blaCTX-M genes was not observed, and blaTEM gene was verified in 91% of the isolates. Sequencing of 10 fragments showed the occurrence of bla(TEM-116)

Using two mouse strains with different abilities to generate interferon (IFN)-gamma production after Mycobacterium tuberculosis infection, we tested the hypothesis that the frequency and activity of regulatory T (Treg) cells are influenced by genetic background. Our results demonstrated that the suppressive activity of spleen Treg cells from infected or uninfected BALB/c mice was enhanced, inhibiting IFN-gamma and interleukin (IL)-2 production. Infected C57BL/6 mice exhibited a decrease in the frequency of lung Treg cells and an increased ratio CD4(+):CD4(+)Foxp3(+) cells compared with infected BALB/c mice and uninfected C57BL/6 mice. Moreover, infected C57BL/6 mice also had a decrease in the immunosuppressive capacity of spleen Treg cells, higher lung IFN-gamma and IL-17 production, and restricted the infection better than BALB/c mice. Adoptive transfer of BALB/c Treg cells into BALB/c mice induced an increase in bacterial colony-forming unit (CFU) counts. Furthermore, BALB/c mice treated with anti-CD25 antibody exhibited lung CFU counts significantly lower than mice treated with irrelevant antibody. Our results show that in BALB/c mice, the Treg cells have a stronger influence than that in C57BL/6 mice. These data suggest that BALB/c and C57BL/6 mice may use some different mechanisms to control M. tuberculosis infection. Therefore, the role of Treg cells should be explored during the development of immune modulators, both from the perspective of the pathogen and the host. Neospora caninum is an apicomplexan parasite responsible for major economic losses due to abortions in cattle. Toll-like receptors (TLRs) sense specific microbial products and direct downstream signaling pathways in immune cells, linking innate, and adaptive immunity. Here, we analyze the role of TLR2 on innate and adaptive immune responses during N. caninum infection. Inflammatory peritoneal macrophages and bone marrow-derived dendritic cells exposed to N. caninum-soluble antigens presented an upregulated expression of TLR2. Increased receptor expression was correlated to TLR2/MyD88-dependent antigen-presenting cell maturation and pro-inflammatory cytokine production after stimulation by antigens. Impaired innate responses observed after infection of mice genetically deficient for TLR2((-/-)) was followed by downregulation of adaptive T helper 1 (Th1) immunity, represented by diminished parasite-specific CD4(+) and CD8(+) T-cell proliferation, IFN-gamma:interleukin (IL)-10 ratio, and IgG subclass synthesis. In parallel, TLR2(-/-) mice presented higher parasite burden than wild-type (WT) mice at acute and chronic stages of infection. These results show that initial recognition of N. caninum by TLR2 participates in the generation of effector immune responses against N. caninum and imply that the receptor may be a target for future prophylactic strategies against neosporosis. Trophozoites from cultures of Entamoeba histolytica strains isolated and grown axenically in Brazil (ICB-CSP, ICB-462 and ICB-32) were used for immune sera production and for characterization of their antigens by using electrophoretic and glycoproteic profiles, in parallel with a standard strain isolated and kept under axenic conditions in USA (HK-9). Hyperimmune sera, presenting high antibody titers with homologous and heterologous antigens, were obtained. The four strains in study revealed similar and complex electrophoretic and glycoproteic profiles showing polypep-tides with molecular weights ranging from 200 to less than 29 kDa. No significant differences were detected between the pathogenic and non-pathogenic strains.

In this study we have tried to verify whether the interaction "in vitro" with bacteria or small pieces of normal hamster liver would modify the pathogenic behavior of axenic strains of E. histolytica: avirulent ones (ICB-32 and ICB-RPS), of attenuated virulence (ICB-CSP and HM1) and of mean virulence (ICB-462). É apresentado cronologicamente o ciclo de pesquisas de citogenética sobre os "barbeiros" no Departamento de Biologia Geral do ICB-UFMG desde 1948 até hoje. Foram, estudados o cariótipo e os cromossomos sexuais, os fenômenos de heteropicnose de autossomos e sua significação evolutiva, os fenômenos da síntese do DtNA ao longo do túbulo testicular, e a variabilidade da morfologia destes túbulos como caráter sistemático. Uma parte das pssquisas foi dedicada ao estudo dos mecanismos citogenéticos da esterilidade dos híbridos inter específicos.

Data on Schistosoma mansoni-Entamoeba histolytica coinfection are scarce in the literature. In the present study, hamsters that had been infected for 70 days with Schistosoma mansoni (LE strain) were inoculated via the portal vein with two strains of trophozoites of Entamoeba histolytica: ICB-EGG (highly virulent) and ICB-RPS (non-virulent). The most evident result of coinfection was increased morbidity and mortality, in comparison with either of the infections alone. Histologically, there were no evident signs of interaction between these two infections. The morphological findings of schistosomal granuloma and amoebic abscesses in the liver were similar to those seen in the respective single-infection controls. However, there was severe wasting of the animals with both infections and greater numbers of amoebic lesions in their livers. The results obtained indicated that schistosomiasis aggravates the course of amoebiasis in hamsters.

Foi analisado o efeito de dois sistemas silvipastoris nos índices de conforto ambiental e alterações nos parâmetros fisiológicos de bezerros bubalinos criados na Embrapa Amazônia Oriental, Belém-PA (clima Afi), no Período 1 (abril a setembro/2007) e Período 2 (outubro/2007 a março/2008). Foram inseridos onze bezerros no Sistema Silvipastoril 1 (SSP1), que apresentava sombreamento útil nas pastagens de 18 a 21%, e oito no Sistema Silvipastoril 2 (SSP2), sem sombreamento, com lago para banho. Foram mensuradas as variáveis fisiológicas: temperatura retal (TR), frequência respiratória (FR), temperatura da pele (TP), e calculados o Índice de Temperatura e Umidade (ITU) e Índice de Conforto de Benezra (ICB), nos dois períodos experimentais, comparados pelo Teste Tukey (P < 0,05). O ITU apresentou diferença estatística entre horários (P < 0,05) e período do ano (P < 0,05), e oscilou de 73,5 ± 1,3 até 82,2 ± 0,8. A TR apresentou diferença estatística entre horários e períodos do ano (P < 0,05), com amplitude de 38,3 ± 0,26 a 39,3 ± 0,38 °C. A FR apresentou diferença significativa entre horários (P < 0,05), com amplitude de 32,2 ± 9,2 a 56,5 ± 19,0 mov min-1, consideradas acima dos níveis normais, enquanto a TP foi diferente estatisticamente entre períodos e horários (P < 0,05) e variou de 23,6 ± 8,3 a 31,7 ± 5,4 °C. Nos Períodos 1 e 2 e nos dois SSP's, os ICBs estiveram acima do valor ideal, variando de 2,46 ± 0,33 a 3,31 ± 0,62 (SSP1) e 2,42 ± 0,30 a 3,45 ± 0,66 (SSP2).

Asbestos exposure can result in serious and frequently lethal diseases, including malignant mesothelioma. The host sensor for asbestos-induced inflammation is the NLRP3 inflammasome and it is widely assumed that this complex is essential for asbestos-induced cancers. Here, we report that acute interleukin-1β production and recruitment of immune cells into peritoneal cavity were significantly decreased in the NLRP3-deficient mice after the administration of asbestos. However, NLRP3-deficient mice displayed a similar incidence of malignant mesothelioma and survival times as wild-type mice. Thus, early inflammatory reactions triggered by asbestos are NLRP3-dependent, but NLRP3 is not critical in the chronic development of asbestos-induced mesothelioma. Notably, in a two-stage carcinogenesis-induced papilloma model, NLRP3-deficient mice showed a resistance phenotype in two different strain backgrounds, suggesting a tumour-promoting role of NLRP3 in certain chemically-induced cancer types.
