第一部分:青蒿开花与青蒿素生物合成相关性的研究 青蒿素是从中药青蒿中分离出的倍半萜内酯化合物,目前是世界上唯一有效的治疗脑型疟疾和抗氯喹恶性疟疾的药物。青蒿植株中青蒿素含量在开花期最高,但是目前尚不清楚开花与青蒿素生物合成的关系。为此,我们用光周期(短日照)诱导青蒿提前开花,不仅同时获得了开花与不开花的青蒿植株,而且还成功地在同一植株上诱导部分分枝开花,另一部分分枝保持营养生长状态。这一实验体系为研究青蒿开花与青蒿素生物合成的相关性奠定了基础。实验结果表明,开花与不开花青蒿植株青蒿素含量有明显差异。开花植株的青蒿素含量在前2周内逐渐提高,第三周(开花期)达到最高,并保持一周左右,在随后的2周内下降。青蒿植株开花后,叶片便开始老化变黄,逐渐死亡。未开花青蒿植株的青蒿素含量动态在前三周内与开花植株类似,但是这种高青蒿素含量状态能保持较长时间,至少在随后的2周内没有下降。未开花植株的叶片依然保持绿色。这一结果表明,开花不是导致青蒿素含量提高的直接原因。 扫描电镜观察结果表明,幼嫩叶片上的毛状腺体( trichrome)结构是完整的,而在老化的叶片上,则观察到了相当比例(40-50%)破损的腺体。这可能是导致青蒿素含量下降的直接原因。 不同生态型青蒿对光周期的反应是不同的。在北京地区,本地青蒿在8月初便开始开花,而来自四川武陵的青蒿则要到9月份才能开花。根据这一特性,采用“南蒿北栽”的方法,能够使青蒿保持较长时间的营养生长状态,延长适于采收的时间。 第二部分:金丝桃和百金花二苯甲酮合酶基因的克隆,异源表达及功能分析 植物次生代谢物山屯酮( Xanthones)仅存在于龙胆科和藤黄科植物中。它们具有抑制单胺氧化酶,细胞毒素及抗肿瘤活性。 含有1 3个碳原子的二苯甲酮是山屯酮生物合成的中间产物,是由二苯甲酮合酶催化合成的,这一反应是山屯酮生物合成的关键步骤。二苯甲酮合酶已经在金丝桃和百金花细胞悬浮培养系统中检测到,并进行了细致的生化水平上的研究。本研究是在上述研究的基础上,进一步克隆该酶的基因,并进行异源表达及功能分析工作,以便更好地了解和调控山屯酮的生物合成。 用PCR和RT-PCR技术,从金丝桃cDNA文库和逆转录产物中分别克隆到一个基因HBPS1和HBPS2,从百金花cDNA文库中克隆到一个基因CBPS1。HBPS1含有1402个碱基,其开放阅读框架编码390个氨基酸,分子量为42.7 kDa,等电点为6.55。HBPS2含有1398个碱基,其开放阅读框架编码395个氨基酸,分子量为42.8 kDa,等电点为5.78。CBPS1含有1383个碱基,其开放阅读框架编码389个氨基酸,分子量为42.7 kDa,等电点为7.88。与GenBank中序列同源性比较结果表明:在氨基酸水平上,HBPS1与茶(Camellia sinensis)查尔酮合酶的同源性高达92%,HBPS2与萝卜(Raphanus sativus)查尔酮合酶的同源性为64%,CBPS1与茶(Camellia sinensis)查尔酮合酶的同源性为71%。HBPS1与HBPS2的同源性仅为62%。 将三个新克隆的基因的ORF整合到载体pGEX-G上的谷胱甘肽还原酶S基因下游,构建成转化质粒,并在大肠杆菌中诱导表达。结果表明,这三个基因的ORF片段均能被表达成约68 kDa的产物,这与期望的结果一致。 活性检测结果表明,HBPS1是查尔酮合成酶,其底物为香豆酰辅酶A和丙二酸单酰辅酶A,对这两种底物的亲和性KM分别为:香豆酰辅酶A 2.8μM,丙二酸单酰辅酶A,11.2μM。最适反应条件是350C,pH7.0,DTT浓度10 μM。 HBPS2是二苯甲酮合酶,其底物是苯甲丙氨酰辅酶A,和丙二酸单酰辅酶A,对这两种底物的亲和性KM分别为:苯甲丙氨酰辅酶A 2.4 μM,丙二酸单酰辅酶A 9.6μM。最适反应条件是350C,pH 6.5,DTT浓度50 μM。而CBPS1则没有检测到任何活性。从同一种植物中同时获得了查尔酮合酶和二苯甲酮合酶,对研究这两种十分相近的酶的差异表达,酶促反应机制等问题将非常有利。

To study the influence of Hypericum perforatum extract (HPE) on piglets infected with porcine respiratory and reproductive syndrome virus (PRRSV), enzyme-labeled immunosorbent assay (ELISA) and cytopathic effect (CPE) were used to determine in vitro whether HPE could induce swine pulmonary alveolar macrophages (PAMs) to secrete IFN-gamma, and whether PRRSV titers in PAMs were affected by the levels of HPE-induced IFN-gamma. HPE (200 mg kg(-1)) was administrated by oral gavage to piglets infected with the PRRSV in vivo to observe whether HPE affected the viremia, lung viral titers, and weight gain of piglets infected with PRRSV. The results showed that HPE was capable of inducing PAMs to produce IFN-gamma in a dose dependent manner and HPE pretreatment was capable of significantly reducing PRRSV viral titers in PAMs (P<0.01). Administration of HPE to the PRRSV-infected animals significantly (P<0.05) reduced viremia over time as compared with the PRRSV-infected animals. But there was not significant decrease in lung viral titers at day 21 post-infection between the HPE-treated animals and the PRRSV-infected control piglets. There were no significant differences in weight gain over time among the HPE-treatment animals, the normal control, and the HPE control animals. The PRRSV-infected animals caused significant (P<0.01) growth retardation as compared with the HPE controls and the normal piglets. It suggested that HPE might be an effective novel therapeutic approach to diminish the PRRSV-induced disease in swine.

Objectives: This study aimed to investigate the efficacy of St. John's wort extract (SJW) as a treatment for premenstrual symptoms. Design: The study was a randomized, double-blinded, placebo-controlled trial, with two parallel treatment groups. After a no-treatment baseline cycle, volunteers were randomized to either SJW or placebo for a further two menstrual cycles. Settings/location: A postal trial conducted from The University of Reading, Berkshire, England. Subjects: One hundred and sixty-nine (169) normally menstruating women who experienced recurrent premenstrual symptoms were recruited onto the study. One hundred and twenty-five (125) completed the protocol and were included in the analysis. Interventions: Six hundred milligrams (600) mg of SJW (standardized to contain 1800 mug of hypericin) or placebo (containing lactose and cellulose). Outcome measure: A menstrual diary was used to assess changes in premenstrual symptoms. The anxiety-related subgroup of symptoms of this instrument was used as the primary outcome measure. Results: After averaging the effects of treatment over both treatment cycles it was found that there was a trend for SJW to be superior to placebo. However, this finding was not statistically significant. Conclusion: The possibility that this nonsignificant finding resulted from insufficient statistical power in the study, rather than a lack of efficacy of SJW, is discussed. Following this discussion the recommendation is made that, in future, similar studies should be powered to detect a minimum clinically relevant difference between treatments.

There is an association between smoking and depression, yet the herbal antidepressant St John's wort (Hypericum perforatum L.: SJW) herb extract has not previously been investigated as an aid in smoking cessation. In this open, uncontrolled, pilot study, 28 smokers of 10 or more cigarettes per day for at least one year were randomised to receive SJW herb extract (LI-160) 300mg once or twice daily taken for one week before and continued for 3 months after a target quit date. In addition, all participants received motivational/behavioural support from a trained pharmacist. At 3 months, the point prevalence and continuous abstinence rates were both 18%, and at 12 months were 0%. Fifteen participants (54%) reported 23 adverse events up to the end of the 3-month follow-up period. There was no statistically significant difference in the frequency of adverse events for participants taking SJW once or twice daily (p > 0.05). Most adverse events were mild, transient and non-serious. This preliminary study has not provided convincing evidence that a SJW herb extract plus individual motivational/behavioural support is likely to be effective as an aid in smoking cessation. However, it may be premature to rule out a possible effect on the basis of a single, uncontrolled pilot study, and other approaches involving SJW extract may warrant investigation.

The volatile Constituents of the fresh materials of Hypericum cordatum were isolated by hydrodistillation kind analyzed by CC and GC/MS. The leaves produced 0.04% of a yellowish essential oil and the flowers did not. The main components of the oil were myrcene (40.18%), alpha-pinene (16.40%), and limonene (12%). The antibacterial activities of the oil against Saccharomyces aureus and Escherichia coli and the anti-fungal activities of the oil against the fungi Cladosporium cladosporioides and C. sphaerospemum were evaluated. The oil showed an antibacterial activity against the bacteria S. aureus and anti-fungal activity against the two fungi.

A espécie Hypericum caprifoliatum é nativa do sul do Brasil. Devido a suas propriedades fitoterápicas é importante estabelecer seu cultivo e evitar sua exploração de forma extrativista. Os objetivos do trabalho foi caracterizar as sementes, avaliar a germinação, comparada à Hypericum perforatum e a sua propagação sexuada. Os experimentos foram conduzidos nos Departamentos de Plantas Forrageiras e Agrometeorologia e no de Horticultura e Silvicultura da Faculdade de Agronomia da UFRGS. Os estudos foram feitos com um lote de sementes de H. caprifoliatum obtido em Teutônia-RS, e um lote comercial de H. perforatum e incluíram: caracterização das sementes de H. caprifoliatum (peso de mil sementes - PMS, cor, tamanho); comparação da germinação com H. perforatum, em BOD e sobre papel, aos 34 dias, quanto à necessidade de luz (sem luz - 7 e 21 dias); tratamentos para superação de dormência (KNO3 - 0,2%, ácido giberélico - 0,5g/l, imersão em água à 70°C - 15 minutos); temperatura para teste de germinação em meio ágar-água - 6g/l (20°C, 25°C, 30°C e 20 - 30°C); emergência de plântulas aos 30 e 52 dias sob cultivo protegido e BOD, utilizando substrato comercial, sob diferentes temperaturas (20 - 30°C, 25°C,30°C); comparação do cultivo protegido e condições de campo para obtenção de mudas. Foram obtidos os seguintes resultados: As sementes apresentam PMS de 0,0205g, cor parda e 0,4 mm de comprimento. A germinação aos 34 dias com luz, foi 18%, e em H. perforatum 57 %; sem luz até 21 dias, foi 15% e, em H. perforatum, 55% com plântulas estioladas. A superação de dormência resultou em: testemunha 9%, KNO3 (0,2%) - 1%, ácido giberélico - 26%, água (70°C - 15 minutos) - 0%. A germinação em meio ágar-água, aos 60 dias: 20°C 64%, 20-30°C - 86%, 25°C - 18% e 30°C17%. A emergência (%) de plântulas em cultivo protegido, aos 30 dias, foi 33% e, em BOD: 20-30°C - 18%, 25°C - 36% e 30°C - 5%. A alteração da temperatura 30°C para 20°C, mantendo os outros tratamentos resultou, na contagem aos 52 dias: 20-30°C - 18%, 25°C - 42 % e 20°C - 29%. Mudas mais vigorosas foram obtidas em condições de campo. Os resultados indicam necessidade de luz, superação de dormência e temperatura inferior à 25°C para germinação de H. caprifoliatum, sendo possível sua propagação sexuada.

Plantas da família Guttiferae apresentam diversas atividades biológicas sendo Hypericum o gênero mais importante devido às atividades antidepressiva, antibacteriana e antiviral de algumas espécies. Hypericum connatum, utilizado no sul do Brasil para o tratamento de feridas de boca, demonstrou atividade contra o lentivírus, responsável pela imunodeficiência felina. Objetivo: isolar e identificar as substâncias majoritárias de H. connatum e testar a atividade de extratos obtidos das partes aéreas e raízes e das substâncias isoladas frente a duas cepas do herpesvírus simples tipo 1 (HSV-1). Método: foram obtidos frações nhexano, diclorometano e metanol e extratos bruto, aquosos a diferentes temperaturas e hidro-etanólico das partes aéreas e raízes. As frações n-hexano e metanólica das partes aéreas foram submetidas à coluna cromatográfica para o isolamento de substâncias. Os extratos e substâncias isoladas foram testados frente ao HSV-1, cepas KOS e ATCC-VR733. Determinou-se a concentração máxima não tóxica (CMNT) à célula e a concentração que provoca alteração morfológica em 50% das células (CC50) pela técnica da alteração morfológica celular, utilizando-se células VERO, linhagem ATCC CCL-81. A avaliação da atividade antiviral foi realizada em placas de microtitulação e medida pela inibição do efeito citopático (ECP) provocado pelo vírus. Resultados e Conclusões: da fração n-hexano foi isolado hiperbrasilol B, da fração metanólica foram isolados amentoflavona, hiperosídeo e guaijaveriana, além de um flavonol de estrutura ainda não definida (HCN3). A fração n-hexano e o extrato bruto das raízes inibiram o ECP das cepas KOS e ATCC-VR733. Os demais extratos testados não apresentaram atividade antiviral. Dentre as substâncias analisadas, hiperbrasilol B, amentoflavona e HCN3 foram ativos frente às duas cepas. Os flavonóides hiperosídeo e guaijaverina não apresentaram atividade anti-HSV-1.

Several clinic evaluations have been possible with radiobiocomplexes labeled with technetium-99m (99mTc). Some natural and synthetic drugs are capable of to interfere on the labeling of blood constituents with 99mTc, as well as on the biodistribution of radiobiocomplexes. Authors have also reported about the toxicity of several natural products. The aim of this study was to compare the effects of the Mentha crispa (hortelã) and of the Hypericum perforatum (hipérico) in different experimental models. On the labeling of red blood cells (RBC) and plasma and cellular proteins with 99mTc, both extracts were capable of to decrease the radioactivity percentage on the cellular compartment and on the fixation on plasma and cellular proteins. On the morphometry of the RBC, only the hortelã was capable to alter the shape and the perimeter/area ratio of the RBC. On the biodistribution of the radiobiocomplex sodium pertechnetate (Na99mTcO4), the hortelã increased the Na99mTcO4 distribution in the kidney, spleen, liver and thyroid, meanwhile the hipérico decreased the Na99mTcO4 distribution in the bone, stomach, lungs and thyroid, and increased the Na99mTcO4 distribution in the pancreas. On the bacterial cultures survival, the hipérico was capable of to protect the bacteria against the stannous chloride (SnCl2) effect. The hipérico did not alter the topology of plasmidial DNA and did not protect the plasmidial DNA against the SnCl2 action. Probably, the effects presented by both extracts could be due to chemical compounds of the extracts that could alter the morphology of the RBC and the plasma membrane ions transport, and/or by phytocomplexes that could be formed with different effects dependent on the biological system considered

A utilização de produtos naturais na medicina popular é milenar e persiste até os dias atuais. Entretanto, a idéia de que estes produtos são isentos de toxicidade torna o uso de medicamentos fitoterápicos cada vez maior e indiscriminado. Este trabalho trata de uma revisão sobre as interações que podem ocorrer com a utilização concomitante de Hypericum perforatum L. (erva de são joão) e Piper methysticum F. (kava-kava) com fármacos, podendo levar a sérios efeitos tóxicos, incluindo a fatalidade.

As atividades antiinflamatória e antinociceptiva do extrato padronizado de Hypericum brasiliense (HBSE) (Guttiferae) foi avaliada em modelos animais. Ratos Wistar machos foram tratados com extrato de H. brasiliense (50, 250 e 500 mg/kg, v.o.) em solução 3% Tween 80 0,9% NaCl. O tratamento com HBSE (500 mg/kg) mostrou inibição significativa sobre o edema induzido por carragenina comparado ao grupo controle. Nessa dose, o edema foi reduzido em 31,25% na terceira hora (pico do edema) após o tratamento, mas na dose de 50 mg/kg, o edema apresentou redução de 53,13% (p < 0,05). Ainda com a dose de 50 mg/kg, a diminuição do edema induzido por dextrana foi similar ao controle positivo, ciproeptadina. Houve diminuição na formação do tecido granulomatoso (6,6%) comparável ao grupo controle. O HBSE também inibiu o número de contorções abdominais em 46,4%, estatisticamente igual ao controle positivo, tratado com indometacina (42,9%). Na dose de 250 mg/kg, houve inibição do número de contorções em 70,7% quando comparado ao grupo controle (p < 0,001). No teste da placa-quente, foi verificado aumento no tempo de latência com a dose de 50 mg/kg. Os resultados demonstram que o HBSE possui atividade antiinflamatória sobre processos agudos, principalmente quando sua gênese está relacionada à síntese dos derivados do ácido araquidônico, e seu efeito analgésico provavelmente envolve ação sobre o Sistema Nervoso Central.

Five spirocyclic acylphloroglucinol derivatives (1-5) have been isolated from a hexanes extract of the leaves of Hypericum pyramidatum. Pyramidatones A-D (1-3, 5) are new, and chipericumin C (4) has been previously reported. The acylphloroglucinols were characterized based on spectroscopic (NMR, IR, UV-VIS) and mass spectrometric data.
