What are the ethical and political implications when the very foundations of life —things of awe and spiritual significance — are translated into products accessible to few people? This book critically analyses this historic recontextualisation. Through mediation — when meaning moves ‘from one text to another, from one discourse to another’ — biotechnology is transformed into analysable data and into public discourses. The unique book links biotechnology with media and citizenship. As with any ‘commodity’, biological products have been commodified. Because enormous speculative investment rests on this, risk will be understated and benefit will be overstated. Benefits will be unfairly distributed. Already, the bioprospecting of Southern megadiverse nations, legally sanctioned by U.S. property rights conventions, has led to wealth and health benefits in the North. Crucial to this development are biotechnological discourses that shift meanings from a “language of life” into technocratic discourses, infused with neo-liberal economic assumptions that promise progress and benefits for all. Crucial in this is the mass media’s representation of biotechnology for an audience with poor scientific literacy. Yet, even apparently benign biotechnology spawned by the Human Genome Project such as prenatal screening has eugenic possibilities, and genetic codes for illness are eagerly sought by insurance companies seeking to exclude certain people. These issues raise important questions about a citizenship that is founded on moral responsibility for the wellbeing of society now and into the future. After all, biotechnology is very much concerned with the essence of life itself. This book provides a space for alternative and dissident voices beyond the hype that surrounds biotechnology.

This plea for changes in the way epidemiology is practiced presumes a progressive spirit among many readers, sharing a desire and in a position to serve humanity. The criteria used to begin this study of our role in the development of epidemiology is not simply a matter of technocratic formulae, but is based, rather on a committed point of view about the human dimension in the development of epidemiology. The aim is to generate propositions that will eliminate processes that destroy and threaten human life, promoting, instead, supportive measures and processes to protect society, the family and the individual.

Humane Nierenzellkarzinom-(NZK)-Zelllinien wurden etabliert, um sie zur Generierung von autologen zytotoxischen T-Zelllinien einzusetzen. Erst nach Modifikation mit dem kostimulierenden B7-1-Molekül wurden mit der NZK-Zelllinie MZ1257RC autologe, tumorspezifische T-Zelllinien generiert und charakterisiert. Die Aufklärung eines T-Zell-definierten TAA eines autologen, zytotoxischen T Zellklons wurde mittels Expressionsklonierung einer hergestellten cDNS-Expressionsbank begonnen. Nach in vitro-Sensibilisierung von peripheren Blutmonozyten mit der autologen NZK-Zelllinie MZ2733RC wurde die HLA-Klasse I-restringierte T Zelllinie XIE6 generiert, die die autologe und verschiedene allogene NZK- sowie Zervixkarzinom-Zelllinien, jedoch nicht autologe Nierenzellen lysiert. Die T Zellen exprimieren TZR Vβ13.6-Ketten und sezernieren GM-CSF und IL-10 nach Antigenstimulation. Jedoch ist die NZK-Zelllinie MZ2733RC wenig sensitiv gegenüber autologen und allogenen Effektorzellen. Erst die Blockade ihrer HLA Klasse I-Moleküle auf der Zelloberfläche erhöht ihre Sensitivität gegenüber allogenen lymphokin-aktivierten Killer-Zellen. Verantwortlich dafür können nicht-klassische HLA Klasse Ib-Moleküle, insbesondere HLA-G sein, dessen Transkripte in der RNS der NZK-Zellen, jedoch nicht in Nierenzellen detektiert wurden. In einer detaillierten Studie wurden HLA-G-Transkripte in NZK-Zelllinien (58%), in NZK-Biopsien (80%), und nur in wenigen Nierenepithelbiopsien (10%) nachgewiesen. In der NZK-Zelllinie MZ2733RC wurde eine konstitutive HLA-G1-Proteinexpression beobachtet, die durch eine IFN-γ-Behandlung induzierbar ist.

CD4+CD25+ regulatorische T-Zellen (CD4+CD25+ Tregs) sind essentiell an der Homöostase des Immunsystems beteiligt, indem sie eine antigenspezifische Toleranzinduktion in der Peripherie vermitteln und vor der Entstehung von Autoimmunerkrankungen schützen. Darüber hinaus sind diese Zellen wesentlich an der Kontrolle von Allergien, Infektionen und Tumoren beteiligt. Innerhalb dieser Arbeit konnten zwei bisher unbekannte Subpopulationen humaner CD4+CD25+ Tregs, isoliert aus dem peripheren Blut des Menschen, nachgewiesen werden. Diese Subpopulationen unterscheiden sich in ihrer Oberflächenexpression und exprimieren die Integrine a4b1 bzw. a4b7. Beide Treg-Subpopulationen supprimieren kokultivierte CD4+ T-Helferzellen Zellkontakt-abhängig und konvertieren gleichzeitig einen Teil dieser Zellen in sekundäre Suppressorzellen (iTregs). a4b1+ Tregs induzieren TGF-β-sezernierende iTregs, a4b7+ Tregs führen zur Bildung von IL-10-produzierenden iTregs. Differentielle Proteomanalysen humaner CD4+CD25+ Tregs, im Vergleich zu CD4+CD25- T-Helferzellen, führten zur Identifizierung von Galectin-10 als Markerprotein, das fast ausschließlich von CD4+CD25+ Tregs und nicht von CD4+ T-Helferzellen exprimiert wird. Galectin-10 ist ein intrazelluläres Protein, das essentiell für die funktionellen Eigenschaften humaner CD4+CD25+ Tregs ist. Die Blockade der Galectin-10-Bildung in den CD4+CD25+ Tregs durch RNA-Interferenz führte zu wesentlichen funktionellen Veränderungen der CD4+CD25+ Tregs. In Abwesenheit von Galectin-10 verlieren humane CD4+CD25+ Tregs ihre suppressiven Eigenschaften und ihren anergischen Phänotyp. Somit konnte mit Galectin-10 erstmals ein spezifischer Marker für humane CD4+CD25+ Tregs identifiziert werden, der wesentlich für den funktionellen Phänotyp dieser Regulatoren peripherer T-Zelltoleranz ist.

We validate, extend, and empirically and theoretically criticize the cultural dimension of humane orientation of the project GLOBE (Global Leadership and Organizational Behavior Effectiveness Research Program). Theoretically, humane orientation is not just a one-dimensionally positive concept about being caring, altruistic, and kind to others as discussed by Kabasakal and Bodur (2004), but there is also a certain ambivalence to this concept. We suggest differentiating humane orientation toward in-group members from humane orientation toward out-group members. A multicountry construct validation study used student samples from 25 countries that were either high or low in humane orientation (N = 876) and studied their relation to the traditional GLOBE scale and other cultural-level measures (agreeableness, religiosity, authoritarianism, and welfare state score). Findings revealed a strong correlation between humane orientation and agreeableness, welfare state score, and religiosity. Out-group humane orientation proved to be the more relevant subfacet of the original humane orientation construct, suggesting that future research on humane orientation should make use of this measure instead of the vague original scale. The ambivalent character of out-group humane orientation is displayed in its positive correlation to high authoritarianism. Patriotism was used as a control variable for noncritical acceptance of one’s society but did not change the correlations. Our findings are discussed as an example of how rigid expectations and a lack of tolerance for diversity may help explain the ambivalent nature of humane orientation

The Stokes County Humane Society has the opportunity to implement sustainable design concepts and demonstrate long-term fiscal savings in the design of an animal shelter for Stokes County, North Carolina. The proposed renovated structure takes advantage of passive design strategies that convert a cold, steel structure into one with warmth, light, and functionality. Proposed design techniques include an interior courtyard, geothermal heating and cooling, solar panels, a green roof, rainwater harvesting, sustainable landscaping, and clever materials selection. A cost analysis identifies capital costs and long term operational costs for both traditional and sustainable construction techniques. The results are capital costs of a sustainable design that are comparable to a traditional build, with long-term operational cost savings and generated revenue.
