A pre-column derivatization method for the sensitive determination of aliphatic amines using the labeling reagent 1,2-benzo-3,4-dihydrocarbazole-9-ethyl chloroformate (BCEOC) followed by HPLC with fluorescence detection and APCI/NIS identification in positive-ion mode has been developed. The chromophore of 2-(9-carbazole)-ethyl chloroformate (CEOC) reagent was replaced by the 1,2-benzo-3,4-dihydrocarbazole functional group, which resulted in a sensitive fluorescence derivatizing reagent, BCEOC, that could easily and quickly label amines. Derivatives were stable enough to be efficiently analyzed by HPLC and showed an intense protonated molecular ion corresponding m/z [M + H](+) with APCI/MS in positive-ion mode. The collision induced dissociation of the protonated molecular ion formed characteristic fragment ions at m/z 264.1, m/z 246.0 and m/z 218.1, corresponding to the cleavages of CH2CH2O-CO, CH2CH2-OCO, and N-CH2CH2O bonds. Studies on derivatization conditions demonstrated that excellent derivatization yields close to 100% were observed with a 3 to 4-fold molar reagent excess in acetonitrile solvent, in the presence of borate buffer (pH 9.0) at 40 degrees C for 10 min. In addition, the detection responses for BCEOC derivatives were compared with those obtained with CEOC and FMOC as labeling reagents. The ratios I-BCEOC/I-CEOC and I-BCEOC/I-FMOC were, respectively, 1.40-2.76 and 1.36-2.92 for fluorescence responses (here, I was the relative fluorescence intensity). Separation of the amine derivatives had been optimized on an Eclipse XDB-C-8 column. Detection limits calculated from an 0.10 pmol injection, at a signal-to-noise ratio of 3, were 18.65-38.82 fmol (injection volume 10 mu L for fluorescence detection. The relative standard deviations for intraday determination (n = 6) of standard amine derivatives (50 pmol) were 0.0063-0.037% for retention times and 3.36-6.93% for peak areas. The mean intra-and inter-assay precision for all amines were <5.4% and 5.8%, respectively. The recoveries of amines ranged from 96 to 113%. Excellent linear responses were observed with correlation coefficients of >0.9994. The established method provided a simple and highly sensitive technique for the quantitative analysis of trace amounts of aliphatic amines from biological and natural environmental samples.

A sensitive method for the determination of 30 kinds of free fatty acids (FFAs, C-1-C-30) with 1-[2-(p-toluenesulfonate)-ethyl]-2-phenylimidazole-[4,5-f] 9,10-phenan- threne (TSPP) as labeling reagent and using high performance liquid chromatography with fluorescence detection and identification by online postcolumn mass spectrometry with atmospheric pressure chemical ionization (APCI) source in positive-ion mode (HPLC/MS/APCI) has been developed. TSPP could easily and quickly label FFAs in the presence of K2CO3 catalyst at 90 degrees C for 30 min in N,N-dimethylformamide (DMF) solvent, and maximal labeling yields close to 100% were observed with a 5-fold excess of molar reagent. Derivatives were stable enough to be efficiently analyzed by high performance liquid chromatography. TSPP was introduced into fatty acid molecules and effectively augmented MS ionization of fatty acid derivatives and led to regular MS and MS/MS information. The collision induced cleavage of protonated molecular ions formed specific fragment ions at m/z [MH](+)(molecular ion), m/z [M'+CH2CH2](+)(M' was molecular mass of the corresponding FFA) and m/z 295.0 (the, mass of protonated molecular core structure of TSPP). Fatty acid derivatives were separated on a reversed-phase Eclipse XDB-C-8 column (4.6 x 150 mm, 5 mu m, Agilent) with a good baseline resolution in combination with a gradient elution. Linear ranges of 30 FFAs are 2.441 x 10(-3) to 20 mu mol/L, detection limits are 3.24 similar to 36.97 fmol (injection volume 10 mu L, at a signal-to-noise ratio of 3, S/N 3:1). The mean interday precision ranged from 93.4 to 106.2% with the largest mean coefficients of variation (R.S.D.) < 7,5%. The mean intraday precision for all standards was < 6.4% of the expected concentration. Excellent linear responses were observed with correlation coefficients of > 0.9991. Good compositional data could be obtained from the analysis of extracted fatty acids from as little as 200 mg of bryophyte plant samples.Therefore, the facile TSPP derivatization coupled with HPLC/MS/APCI analysis allowed the development of a highly sensitive method for the quantitation of trace levels of short and long chain fatty acids from biological and natural environmental samples.

Gli istoni sono proteine basiche che possono essere classificate in varie classi: H1, H2A, H2B, H3 e H4. Queste proteine formano l’ottamero proteico attorno al quale si avvolge il DNA per formare il nucleosoma che è l’unità fondamentale della cromatina. A livello delle code N-terminali, gli istoni possono essere soggetti a numerose modifiche posttraduzionali quali acetilazioni, metilazioni, fosforilazioni, ADP-ribosilazioni e ubiquitinazioni. Queste modifiche portano alla formazione di diversi siti di riconoscimento per diversi complessi enzimatici coinvolti in importanti processi come la riparazione e la replicazione del DNA e l’assemblaggio della cromatina. La più importante e la più studiata di queste modifiche è l’acetilazione che avviene a livello dei residui amminici della catena laterale dell’amminoacido lisina. I livelli corretti di acetilazione delle proteine istoniche sono mantenuti dall’attività combinata di due enzimi: istone acetil transferasi (HAT) e istone deacetilasi (HDAC). Gli enzimi appartenenti a questa famiglia possono essere suddivisi in varie classi a seconda delle loro diverse caratteristiche, quali la localizzazione cellulare, la dimensione, l’omologia strutturale e il meccanismo d’azione. Recentemente è stato osservato che livelli aberranti di HDAC sono coinvolti nella carcinogenesi; per questo motivo numerosi gruppi di ricerca sono interessati alla progettazione e alla sintesi di composti che siano in grado di inibire questa classe enzimatica. L’inibizione delle HDAC può infatti provocare arresto della crescita cellulare, apoptosi o morte cellulare. Per questo motivo la ricerca farmaceutica in campo antitumorale è mirata alla sintesi di inibitori selettivi verso le diverse classi di HDAC per sviluppare farmaci meno tossici e per cercare di comprendere con maggiore chiarezza il ruolo biologico di questi enzimi. Il potenziale antitumorale degli inibitori delle HDAC deriva infatti dalla loro capacità di interferire con diversi processi cellulari, generalmente non più controllati nelle cellule neoplastiche. Nella maggior parte dei casi l’attività antitumorale risiede nella capacità di attivare programmi di differenziamento, di inibire la progressione del ciclo cellulare e di indurre apoptosi. Inoltre sembra essere molto importante anche la capacità di attivare la risposta immunitaria e l’inibizione dell’angiogenesi. Gli inibitori delle HDAC possono essere a loro volta classificati in base alla struttura chimica, alla loro origine (naturale o sintetica), e alla loro capacità di inibire selettivamente le HDAC appartenenti a classi diverse. Non è ancora chiaro se la selettività di queste molecole verso una specifica classe di HDAC sia importante per ottenere un effetto antitumorale, ma sicuramente inibitori selettivi possono essere molto utili per investigare e chiarire il ruolo delle HDAC nei processi cellulari che portano all’insorgenza del tumore. Nel primo capitolo di questa tesi quindi è riportata un’introduzione sull’importanza delle proteine istoniche non solo da un punto di vista strutturale ma anche funzionale per il destino cellulare. Nel secondo capitolo è riportato lo stato dell’arte dell’analisi delle proteine istoniche che comprende sia i metodi tradizionali come il microsequenziamento e l’utilizzo di anticorpi, sia metodi più innovativi (RP-LC, HILIC, HPCE) ideati per poter essere accoppiati ad analisi mediante spettrometria di massa. Questa tecnica consente infatti di ottenere importanti e precise informazioni che possono aiutare sia a identificare gli istoni come proteine che a individuare i siti coinvolti nelle modifiche post-traduzionali. Nel capitolo 3 è riportata la prima parte del lavoro sperimentale di questa tesi volto alla caratterizzazione delle proteine istoniche mediante tecniche cromatografiche accoppiate alla spettrometria di massa. Nella prima fase del lavoro è stato messo a punto un nuovo metodo cromatografico HPLC che ha consentito di ottenere una buona separazione, alla linea di base, delle otto classi istoniche (H1-1, H1-2, H2A-1, H2A-2, H2B, H3-1, H3-2 e H4). La separazione HPLC delle proteine istoniche ha permesso di poter eseguire analisi accurate di spettrometria di massa mediante accoppiamento con un analizzatore a trappola ionica tramite la sorgente electrospray (ESI). E’ stato così possibile identificare e quantificare tutte le isoforme istoniche, che differiscono per il tipo e il numero di modifiche post-traduzionali alle quali sono soggette, previa estrazione da colture cellulari di HT29 (cancro del colon). Un’analisi così dettagliata delle isoforme non può essere ottenuta con i metodi immunologici e permette di eseguire un’indagine molto accurata delle modifiche delle proteine istoniche correlandole ai diversi stadi della progressione del ciclo e alla morte cellulare. Il metodo messo a punto è stato convalidato mediante analisi comparative che prevedono la stessa separazione cromatografica ma accoppiata a uno spettrometro di massa avente sorgente ESI e analizzatore Q-TOF, dotato di maggiore sensibilità e risoluzione. Successivamente, per identificare quali sono gli specifici amminoacidi coinvolti nelle diverse modifiche post-traduzionali, l’istone H4 è stato sottoposto a digestione enzimatica e successiva analisi mediante tecniche MALDI-TOF e LC-ESI-MSMS. Queste analisi hanno permesso di identificare le specifiche lisine acetilate della coda N-terminale e la sequenza temporale di acetilazione delle lisine stesse. Nel quarto capitolo sono invece riportati gli studi di inibizione, mirati a caratterizzare le modifiche a carico delle proteine istoniche indotte da inibitori delle HDAC, dotati di diverso profilo di potenza e selettività. Dapprima Il metodo messo a punto per l’analisi delle proteine istoniche è stato applicato all’analisi di istoni estratti da cellule HT29 trattate con due noti inibitori delle HDAC, valproato e butirrato, somministrati alle cellule a dosi diverse, che corrispondono alle dosi con cui sono stati testati in vivo, per convalidare il metodo per studi di inibizione di composti incogniti. Successivamente, lo studio è proseguito con lo scopo di evidenziare effetti legati alla diversa potenza e selettività degli inibitori. Le cellule sono state trattate con due inibitori più potenti, SAHA e MS275, alla stessa concentrazione. In entrambi i casi il metodo messo a punto ha permesso di evidenziare l’aumento dei livelli di acetilazione indotto dal trattamento con gli inibitori; ha inoltre messo in luce differenti livelli di acetilazione. Ad esempio il SAHA, potente inibitore di tutte le classi di HDAC, ha prodotto un’estesa iperacetilazione di tutte le proteine istoniche, mentre MS275 selettivo per la classe I di HDAC, ha prodotto modifiche molto più blande. E’ stato quindi deciso di applicare questo metodo per studiare la dose e la tempo-dipendenza dell’effetto di quattro diversi inibitori delle HDAC (SAHA, MS275, MC1855 e MC1568) sulle modifiche post-traduzionali di istoni estratti da cellule HT29. Questi inibitori differiscono oltre che per la struttura chimica anche per il profilo di selettività nei confronti delle HDAC appartenenti alle diverse classi. Sono stati condotti quindi studi di dose-dipendenza che hanno consentito di ottenere i valori di IC50 (concentrazione capace di ridurre della metà la quantità relativa dell’istone meno acetilato) caratteristici per ogni inibitore nei confronti di tutte le classi istoniche. E’ stata inoltre calcolata la percentuale massima di inibizione per ogni inibitore. Infine sono stati eseguiti studi di tempo-dipendenza. I risultati ottenuti da questi studi hanno permesso di correlare i livelli di acetilazione delle varie classi istoniche con la selettività d’azione e la struttura chimica degli inibitori somministrati alle cellule. In particolare, SAHA e MC1855, inibitori delle HDAC di classi I e II a struttura idrossamica, hanno causato l’iperacetilazione di tutte le proteine istoniche, mentre MC1568 (inibitore selettivo per HDAC di classe II) ha prodotto l’iperacetilazione solo di H4. Inoltre la potenza e la selettività degli inibitori nel provocare un aumento dei livelli di acetilazione a livello delle distinte classi istoniche è stata correlata al destino biologico della cellula, tramite studi di vitalità cellulare. E’ stato osservato che il SAHA e MC1855, inibitori potenti e non selettivi, somministrati alla coltura HT29 a dose 50 μM producono morte cellulare, mentre MS275 alla stessa dose produce accumulo citostatico in G1/G0. MC1568, invece, non produce effetti significatici sul ciclo cellulare. Questo studio ha perciò dimostrato che l’analisi tramite HPLC-ESI-MS delle proteine istoniche permette di caratterizzare finemente la potenza e la selettività di nuovi composti inibitori delle HDAC, prevedendone l’effetto sul ciclo cellulare. In maggiore dettaglio è risultato che l’iperacetilazione di H4 non è in grado di provocare modifiche significative sul ciclo cellulare. Questo metodo, insieme alle analisi MALDI-TOF e LC-ESI-MSMS che permettono di individuare l’ordine di acetilazione delle lisine della coda N-terminale, potrà fornire importanti informazioni sugli inibitori delle HDAC e potrà essere applicato per delineare la potenza, la selettività e il meccanismo di azione di nuovi potenziali inibitori di questa classe enzimatica in colture cellulari tumorali.

Wasserlösliche organische Verbindungen (WSOCs) sind Hauptbestandteile atmosphärischer Aerosole, die bis zu ~ 50% und mehr der organischen Aerosolfraktion ausmachen. Sie können die optischen Eigenschaften sowie die Hygroskopizität von Aerosolpartikeln und damit deren Auswirkungen auf das Klima beeinflussen. Darüber hinaus können sie zur Toxizität und Allergenität atmosphärischer Aerosole beitragen.In dieser Studie wurde Hochleistungsflüssigchromatographie gekoppelt mit optischen Diodenarraydetektion und Massenspektrometrie (HPLC-DAD-MS und HPLC-MS/MS) angewandt, um WSOCs zu analysieren, die für verschiedene Aerosolquellen und -prozesse charakteristisch sind. Niedermolekulare Carbonsäuren und Nitrophenole wurden als Indikatoren für die Verbrennung fossiler Brennstoffe und die Entstehung sowie Alterung sekundärer organischer Aerosole (SOA) aus biogenen Vorläufern untersucht. Protein-Makromoleküle wurden mit Blick auf den Einfluss von Luftverschmutzung und Nitrierungsreaktionen auf die Allergenität primärer biologischer Aerosolpartikel – wie Pollen und Pilzsporen – untersucht.rnFilterproben von Grob- und Feinstaubwurden über ein Jahr hinweg gesammelt und auf folgende WSOCs untersucht: die Pinen-Oxidationsprodukte Pinsäure, Pinonsäure und 3-Methyl-1,2,3-Butantricarbonsäure (3-MBTCA) sowie eine Vielzahl anderer Dicarbonsäuren und Nitrophenole. Saisonale Schwankungen und andere charakteristische Merkmale werden mit Blick auf Aerosolquellen und -senken im Vergleich zu Daten anderen Studien und Regionen diskutiert. Die Verhätlnisse von Adipinsäure und Phthalsäure zu Azelainsäure deuten darauf hin, dass die untersuchten Aerosolproben hauptsächlich durch biogene Quellen beeinflusst werden. Eine ausgeprägte Arrhenius-artige Korrelation wurde zwischen der 3-MBTCA-¬Konzentration und der inversen Temperatur beobachtet (R2 = 0.79, Ea = 126±10 kJ mol-1, Temperaturbereich 275–300 K). Modellrechnungen zeigen, dass die Temperaturabhängigkeit auf eine Steigerung der photochemischen Produktionsraten von 3-MBTCA durch erhöhte OH-Radikal-Konzentrationen bei erhöhten Temperaturen zurückgeführt werden kann. Im Vergleich zur chemischen Reaktionskinetik scheint der Einfluss von Gas-Partikel-Partitionierungseffekten nur eine untergeordnete Rolle zu spielen. Die Ergebnisse zeigen, dass die OH-initiierte Oxidation von Pinosäure der geschwindigkeitsbestimmende Schritt der Bildung von 3-MBTCA ist. 3-MBTCA erscheint somit als Indikator für die chemische Alterung von biogener sekundärer organischer Aerosole (SOA) durch OH-Radikale geeignet. Eine Arrhenius-artige Temperaturabhängigkeit wurde auch für Pinäure beobachtet und kann durch die Temperaturabhängigkeit der biogenen Pinen-Emissionen als geschwindigkeitsbestimmender Schritt der Pinsäure-Bildung erklärt werden (R2 = 0.60, Ea = 84±9 kJ mol-1).rn rnFür die Untersuchung von Proteinnitrierungreaktionen wurde nitrierte Protein¬standards durch Flüssigphasenreaktion von Rinderserumalbumin (BSA) und Ovalbumin (OVA) mit Tetranitromethan (TNM) synthetisiert.Proteinnitrierung erfolgt vorrangig an den Resten der aromatischen Aminosäure Tyrosin auf, und mittels UV-Vis-Photometrie wurde der Proteinnnitrierungsgrad (ND) bestimmt. Dieser ist definiert als Verhältnis der mittleren Anzahl von Nitrotyrosinresten zur Tyrosinrest-Gesamtzahl in den Proteinmolekülen. BSA und OVA zeigten verschiedene Relationen zwischen ND und TNM/Tyrosin-Verhältnis im Reaktionsgemisch, was vermutlich auf Unterschiede in den Löslichkeiten und den molekularen Strukturen der beiden Proteine zurück zu führen ist.rnDie Nitrierung von BSA und OVA durch Exposition mit einem Gasgemisch aus Stickstoffdioxid (NO2) und Ozon (O3) wurde mit einer neu entwickelten HPLC-DAD-¬Analysemethode untersucht. Diese einfache und robuste Methode erlaubt die Bestimmung des ND ohne Hydrolyse oder Verdau der untersuchten Proteine und ernöglicht somit eine effiziente Untersuchung der Kinetik von Protein¬nitrierungs-Reaktionen. Für eine detaillierte Produktstudien wurden die nitrierten Proteine enzymatisch verdaut, und die erhaltenen Oligopeptide wurden mittels HPLC-MS/MS und Datenbankabgleich mit hoher Sequenzübereinstimmung analysiert. Die Nitrierungsgrade individueller Nitrotyrosin-Reste (NDY) korrelierten gut mit dem Gesamt-Proteinnitrierungsgrad (ND), und unterschiedliche Verhältnisse von NDY zu ND geben Aufschluss über die Regioselektivität der Reaktion. Die Nitrierungmuster von BSA und OVA nach Beahndlung mit TNM deuten darauf hin, dass die Nachbarschaft eines negativ geladenen Aminosäurerestes die Tyrosinnitrierung fördert. Die Behandlung von BSA durch NO2 und O3 führte zu anderend Nitrierungemustern als die Behandlung mit TNM, was darauf hindeutet, dass die Regioselektivität der Nitrierung vom Nitrierungsmittel abhängt. Es zeigt sich jedoch, dass Tyrosinreste in Loop-Strukturen bevorzugt und unabhängig vom Reagens nitriert werden.Die Methoden und Ergebnisse dieser Studie bilden eine Grundlage für weitere, detaillierte Untersuchungen der Reaktionskinetik sowie der Produkte und Mechanismen von Proteinnitrierungreaktionen. Sie sollen helfen, die Zusammenhänge zwischen verkehrsbedingten Luftschadstoffen wie Stickoxiden und Ozon und der Allergenität von Luftstaub aufzuklären.rn

The role of the therapeutic drug monitoring laboratory in support of immunosuppressant drug therapy is well established, and the introduction of sirolimus (SRL) is a new direction in this field. The lack of an immunoassay for several years has restricted the availability of SRL assay services. The recent availability of a CEDIA (R) SRL assay has the potential to improve this situation. The present communication has compared the CEDIA (R) SRL method with 2 established chromatographic methods, HPLC-UV and HPLC-MS/MS. The CEDIA (R) method, run on a Hitachi 917 analyzer, showed acceptable validation criteria with within-assay precision of 9.1% and 3.3%, and bias of 17.1% and 5.8%, at SRL concentrations of 5.0 mu g/L and 20 mu g/L, respectively. The corresponding between-run precision values were 11.5% and 3.3% and bias of 7.1% and 2.9% at 5.0 mu g/L and 20 mu g/L, respectively, The lower limit of quantification was found to be 3.0 mu g/L. A series of 96 EDTA whole-blood samples predominantly from renal transplant recipients were assayed by the 3 methods for comparison. It was found that the CEDIA (R) method showed a Deming regression line of CEDIA = 1.20 X HPLC-MS/MS - 0.07 (r = 0.934, SEE = 1.47), with a mean bias of 20.4%. Serial blood samples from 8 patients included in this evaluation showed that the CEDIA (R) method reflected the clinical fluctuations in the chromatographic methods, albeit with the variable bias noted. The CEDIA (R) method on the H917 analyzer is therefore a useful adjunct to SRL dosage individualization in renal transplant recipients.

Therapeutic monitoring with dosage individualization of sirolimus drug therapy is standard clinical practice for organ transplant recipients. For several years sirolimus monitoring has been restricted as a result of lack of an immunoassay. The recent reintroduction of the microparticle enzyme immunoassay (MEIA (R)) for sirolimus on the IMx (R) analyser has the potential to address this situation. This Study, using patient samples, has compared the MEIA (R) sirolimus method with an established HPLC-tandem mass spectrometry method (HPLC-MS/MS). An established HPLC-UV assay was used for independent cross-validation. For quality control materials (5, 11, 22 mu g/L), the MEIA (R) showed acceptable validation criteria based on intra-and inter-run precision (CV) and accuracy (bias) of < 8% and < 13%, respectively. The lower limit of quantitation was found to be approximately 3 mu g/L. The performance of the immunoassay was compared with HPLC-MS/MS using EDTA whole-blood samples obtained from various types of organ transplant recipients (n = 116). The resultant Deming regression line was: MEIA = 1.3 x HPLC-MS/MS+ 1.3 (r = 0.967, s(y/x) = 1) with a mean bias of 49.2% +/- 23.1 % (range, -2.4% to 128%; P < 0.001). The reason for the large and variable bias was not explored in this study, but the sirolimus-metabolite cross-reactivity with the MEIA (R) antibody could be a substantive contributing factor. Whereas the MEIA (R) sirolimus method may be an adjunct to sirolimus dosage individualization in transplant recipients, users must consider the implications of the substantial and variable bias when interpreting results. In selected patients where difficult clinical issues arise, reference to a specific chromatographic method may be required.

The drugs studied in this work have been reportedly used to commit drug-facilitated sexual assault (DFSA), commonly known as "date rape". Detection of the drugs was performed using high-performance liquid chromatography with ultraviolet detection (HPLC/UV) and identified with high performance-liquid chromatography mass spectrometry (HPLC/MS) using selected ion monitoring (SIM). The objective of this study was to develop a single HPLC method for the simultaneous detection, identification and quantitation of these drugs. The following drugs were simultaneously analyzed: Gamma-hydroxybutyrate (GHB), scopolamine, lysergic acid diethylamide, ketamine, flunitrazepam, and diphenhydramine. The results showed increased sensitivity with electrospray (ES) ionization versus atmospheric pressure chemical ionization (APCI) using HPLC/MS. HPLC/ES/MS was approximately six times more sensitive than HPLC/APCI/MS and about fifty times more sensitive than HPLC/UV. A limit of detection (LOD) of 100 ppb was achieved for drug analysis using this method. The average linear regression coefficient of correlation squared (r2) was 0.933 for HPLC/UV and 0.998 for HPLC/ES/MS. The detection limits achieved by this method allowed for the detection of drug dosages used in beverage tampering. This method can be used to screen beverages suspected of drug tampering. The results of this study demonstrated that solid phase microextraction (SPME) did not improve sensitivity as an extraction technique when compared to direct injections of the drug standards.

目的:通过HPLC、ESI-MS指纹图谱对中药西洋参与北沙参进行鉴别。方法:利用HPLC和ESI-MS技术,优化西洋参、北沙参提取物的色谱和质谱分离分析条件,建立二者HPLC、ESI-MS指纹图谱。结果:确定出西洋参与北沙参HPLC的各色谱峰相对保留时间;根据MS图中各成份的m/z值,确定相应成分的分子量。二者的液相色谱和质谱指纹图谱完全不同。结论:通过HPLC、ESI-MS指纹图谱可以完全鉴别中药西洋参与北沙参,方法简单准确,重现性好,具有实用价值。

2-(2-Phenyl-1H-phenanthro-[9,10-d]imidazole-1-yl)-acetic acid (PPIA) and 2-(9-acridone)-acetic acid (AAA), two novel precolumn fluorescent derivatization reagents, have been developed and compared for analysis of primary aromatic amines by high performance liquid chromatographic fluorescence detection coupled with online mass spectrometric identification. PPIA and AAA react rapidly and smoothly with the aromatic amines on the basis of a condensation reaction using 1-ethyl-3-(3dimethylaminopropyl)-carbodiimide (EDC) as dehydrating catalyst to form stable derivatives with emission wavelengths at 380 and 440 nm, respectively. Taking six primary aromatic amines (aniline, 2-methylaniline, 2-methoxyaniline, 4-methylaniline, 4-chloroaniline, and 4-bromoaniline) as testing compounds, derivatization conditions such as coupling reagent, basic catalyst, reaction temperature and time, reaction solvent, and fluorescent labeling reagent concentration have also been investigated. With the better PPIA method, chromatographic separation of derivatized aromatic amines exhibited a good baseline resolution on an RP column. At the same time, by online mass spectrometric identification with atmospheric pressure chemical ionization (APCI) source in positive ion mode, the PPIA-labeled derivatives were characterized by easy-to-interpret mass spectra due to the prominent protonated molecular ion m/z [M + H](+) and specific fragment ions (MS/MS) m/z 335 and 295. The linear range is 24.41 fmol-200.0 pmol with correlation coefficients in the range of 0.9996-0.9999, and detection limits of PPIA-labeled aromatic amines are 0.12-0.21 nmol/L (S/N = 3). Method repeatability, precision, and recovery were evaluated and the results were excellent for the efficient HPLC analysis. The most important argument, however, was the high sensitivity and ease-of-handling of the PPIA method. Preliminary experiments with wastewater samples collected from the waterspout of a paper mill and its nearby soil where pollution with aromatic amines may be expected show that the method is highly validated with little interference in the chromatogram.

A method for the determination of long and short chain free fatty acids (FFAs), using 1-[2-(ptoluenesulfonate)-ethyll-2-phenylimidazole-[4,5-f-9,10-phenanthrene (TSPP) as labeling reagent, has been developed. Identification of FFA derivatives was carried out by HPLC-MS with atmospheric pressure chemical ionization (APCI) in positive ion mode. Gradient elution on an Agilent Eclipse XDB-C-8 column gave good separation of the derivatives. Excellent linear responses were observed and good compositional data could be obtained from as little as 200 mg of bryophyte plants and soil samples. Facile TSPP derivatization coupled with HPLC-APCI-MS analysis allowed the development of a highly sensitive method for the quantitative analysis of trace level of FFAs from biological and natural environmental samples.

Hoffmann La Roche Ltd., Switzerland) vs. generic (Lipiblock (R), EMS Sigma Pharma, a generic drug) were carried out with ethanol extracts of commercial samples. The generic formulation contained higher levels of common impurities as well as a considerable number of impurities not found in the reference product. The detected impurity profile of Lipiblock (R) revealed that it most likely is based on fermentation. Since the effect of the impurities is unknown, at this point fully synthetic Xenical (R) appears to offer a better safety margin than Lipiblock (R) which, however, compares quite well to other generic formulations.

Interaction of traditional Chinese Herb Rhizoma Chuanxiong and protein was studied by microdialysis coupled with high performance liquid chromatography. Compounds in Rhizoma Chuanxiong, such as ferulic acid, senkyunolide A and 3-butylphthalide, were identified by HPLC, HPLC-MS and UV-vis. Microdialysis recoveries and binding degrees of compounds in Rhizoma Chuanxiong with human serum albumin (HSA) and other human plasma protein were determined: recoveries of microdialysis sampling ranged from 36.7 to 98.4% with R.S.D. below 3.1%; while binding to HSA ranged from 0 to 91.5% (0.3 mM HSA) and from 0 to 93.5% (0.6 mM HSA), respectively. Compared with HSA, most of compounds bound to human blood serum more extensively and the results showed that binding of these compounds in Rhizoma Chuanxiong was influenced by pH. Two compounds were found to bind to HSA and human blood serum. their binding degrees were consistent with ferulic acid and 3-butylphthalide, the active compounds in Rhizoma Chuanoxiong. A monolithic enzymatic microreactor was prepared in a fused-silica capillary by in situ polymerization of acrylamide, glycidyl methacrylate (GMA) and ethylene dimethacrylate (EDMA) in the presence of a binary porogenic mixture of dodecanol and cyclohexanol, followed by ammonia solution treatment, glutaraldehyde activation and trypsin modification. The choice of acrylamide as co-monomer was found useful to improve the efficiency of trypsin modification, thus, to increase the enzyme activity. The optimized microreactor offered very low back pressure, enabling the fast digestion of proteins flowing through the reactor. The performance of the monolithic microreactor was demonstrated with the digestion of cytochrome c at high flow rate. The digests were then characterized by CE and HPLC-MS/MS with the sequence coverage of 57.7%. The digestion efficiency was found over 230 times as high as that of the conventional method. in addition, for the first time, protein digestion carried out in a mixture of water and ACN was compared with the conventional aqueous reaction using MS/MS detection, and the former solution was found more compatible and more efficient for protein digestion.

A novel approach has been developed to determine ranitidine in paediatric samples using dried blood spots (DBS) on Guthrie cards (Whatman 903). A selective and sensitive HPLC-MS/MS assay has been developed and validated using small volumes of blood (30µl). A 6mm disc was punched from each DBS and extracted with methanolic solution of the internal standard (IS) nizatidine. This was further subjected to solid phase extraction (SPE), followed by reversed phase HPLC separation, using a XBridge™ C18 column and mobile phase 10mM ammonium acetate/methanol (98:2 v/v) with a flow rate of 0.3mL/min. This was combined with multiple reaction monitoring (MRM) mass detection using electrospray ionisation (ESI). The calibration curve for ranitidine was found linear over the range 10-500ng/mL (r=0.996). The limit of quantification (LOQ) of the method was validated at 10ng/mL.
