Histoarchitecture, seasonal variation and reproductive function of the neuroendocrine structure, brain of freshwater prawn Macrobrachium gangeticum were studied. Three types of NSCs - 'B', 'C' and 'D' were found to be concentrated in four groups in brain. These cells showed larger diameters and higher activity during breeding season. In case of females, the 'C' cells were more active during vitellogenic period. Brain extracts were found to induce gonadal maturation of both males and females.

In our pursuit to develop new potential anticancer leads, we designed a combination of structural units of indole and substituted triazole; and a library of 1-{1-methyl-2-4-phenyl-5-(propan-2-ylsulfanyl)-4H-1,2,4-triazol-3-yl ]-1H-indol-3-yl}methanamine derivatives was synthesized and characterized. Cytotoxic evaluations of these molecules over a panel of three human cancer cell lines were carried out. Few molecules exhibited potent growth inhibitory action against the treated cancer cell lines at lower micro molar concentration. An in vitro assay investigation of these active compounds using recombinant human SIRT1 enzyme showed that one of the compounds (IT-14) inhibited the deacetylation activity of the enzyme. The in vivo study of IT-14 exemplified its promising action by reducing the prostate weight to the body weight ratio in prostate hyperplasia animal models. A remarkable decrease in the disruption of histoarchitecture of the prostate tissues isolated from IT-14 treated animal compared to that of the positive control was observed. The molecular interactions with SIRT1 enzyme were also supported by molecular docking simulations. Hence this compound can act as a lead molecule to treat prostatic hyperplasia. Morphological investigations of the epithelial cell types that line the ductuli efferentes (DE) of black isogenic mice confirm absorption of the luminal fluid phase by endocytosis as the main function of ductuli efferentes (DE) in this species. :Furthermore, all the histochemical and ultrastructural observations on the DE epithelial histoarchitecture indicate other cellular functions such as exocytosis and probably secretion: including an aprocrine secretory process.

The Picrosirius-polarization method has been indicated as a selective histochemical stain for collagen detection in tissue sections. This method can also be of value for studying collagen degradation given that, under polarized light, collagen displays birefringence due to its molecular order. The aim of this study is to highlight this staining method as an additional instrument for a rapid and excellent confirmatory diagnosis of the presence of collagenolysis in connective tissue in the vaginal wall with vesical prolapse lesion, in tissue sections. Dramatic changes in collagen morphology were found in vaginal mucosa in vesical prolapse disorder: they were weakly stained by Sirius red and under polarized light appeared as thin, pale (weakly birefringent), greenish, and with fibers more scattered, while the histoarchitecture of the organ showed a disrupted appearance. Thus, in the present study, we showed in vaginal mucosa in the vesicle prolapse that corroded collagenous framework appears as fragmentary and irregularly separated collagenous structures, that are weakly birefringent, corresponding to a molecular disorganization of these fibers caused by collagenolysis. Características estruturais da parede das artérias renal e femoral de coelho (Oryctolagus cuniculus)

In the renal and femoral arteries of rabbit was verified that both vessels had walls structured by myostromal components, despite of their different distributive of blood, being the renal artery a visceral blood vessel and the femoral artery a parietal vessel. This wall pattern in these vessels concerned to presence of connective stromal elements (collagen and elastic fibres and lamellae) and smooth muscle cells coexisting with some equilibrium in the wall structure of the renal and femoral arteries, mainly in the medial layer architecture. An intimal folding pattern was verified around the vascular lumen, possibly related to capacitance of the both arteries regarding to variability of pressure levels in cardiac cycle. Furthermore, myostromal relations of connective elements and smooth muscle cells verified in the medial layer and the network formed by connective elements in the adventitial layer of these arteries contributed to maintenance of wall viscoelasticity properties of the vessels.

The female prostate is a differentiated organ found in several mammal species, including humans and rodents. This gland has been related to important functions on female reproductive biology. Although the factors, which regulate prostate's development and activity are not well known, its functionality has been related to steroid hormones. It is well established that cyclic changes of estradiol and progesterone levels promote histophysiological adaptations of the whole female body. In contrast, only a few is found about those adaptations in female prostate. Thus, this study aimed to evaluate the effect of estradiol and estradiol+testosterone association on gerbil female prostate in order to verify, which hormonal associations are necessary to its homeostasis. For this, adult females had the ovaries surgically removed. After recovering, they received estradiol and estradiol+testosterone doses through 30 days, each 48 h. The prostatic tissue underwent morphological and morphometric-estereological analysis. Hormonal restriction caused great gland involution and decreased secretory activity, aspects that were reverted by exposure to estradiol and estradiol+testosterone. However, these hormones were not able to re-establish the normal prostate histoarchitecture. The immunoreaction of steroid receptors (ER-α, ER-β, and AR) responded differently among the experimental and control groups, and PCNA assay showed a decrease in epithelial cell proliferation within groups that had hormone privation. Therefore, we conclude that estradiol and testosterone are able to influence prostate morphophysiology and the maintenance of gland homeostasis depends on a balance among these and other hormones. © 2013 Wiley Periodicals, Inc.

Background. Clay is often used in cosmetic treatments, although little is known about its action. Aim. To evaluate the effect of topical clay application on the histoarchitecture of collagen fibres in rat skin. Methods. Animals received a daily application of clay and retinoic acid (RA) 0.025% to the dorsal skin over 7 and 14 days, under vaporization at 37 degrees C for 40 min. Control skin was not vaporized. Samples from each region were excised, and stained with picrosirius red for collagen evaluation. Results. Seven days after clay treatment, an increase in the number of collagen fibres was observed in treated skin compared with control skin (51.74 +/- 1.28 vs. 43.39 +/- 1.79%, respectively, P < 0.01), whereas RA did not alter the collagen level (45.66 +/- 1.10%). Clay application over 14 days did not induce a further increase in skin collagen, whereas treatment with RA did (58.07 +/- 1.59%; P = 0.001 vs. control). Conclusion. Clay application promotes an increase in the number of collagen fibres, which may account for its beneficial effects.

Zusammenfassung Humane Mundschleimhaut wurde mit Hilfe eines speziellen dreidimensionalen Zellkultivierungssystems in vitro in großen, zusammenhängenden Arealen gezüchtet. Hierbei handelte es sich um eine methodische Eigenentwicklung, welche die gewebespezifische Mikroumgebung der humanen Mundhöhle optimal nachbildet. Die Beurteilung der physiologischen Integrität des gezüchteten Humangewebes erfolgte durch eine Vielzahl von Kenngrößen. Zu diesen Beurteilungskriterien zählten unter anderem die Expression von Cytokeratinen, die Ausprägung von Komponenten des Cytoskeletts und der extrazellulären Matrix sowie die Detektion von genetischen Mutationen. Mit zunehmender Kultivierungsdauer konnten in den organotypischen Co-Kulturen, neben der Synthese der mesenchymalen Marker Fibronectin und Tenascin, eine gesteigerte Expression des Universalmarkers Cytokeratin 14 sowie der differenzierungsspezifischen Cytokeratine 4 und 13 beobachtet werden. Die Cytokeratinproduktion war ebenfalls, wie das vermehrte Auftreten der interzellulären Bindungselemente Desmoglein 2 und Desmoplakine 1 bzw. 2, ausschließlich auf die epithelialen Zellschichten beschränkt. Nach 14tägiger Kultivierung zeigte sich in der PAS-Färbung die Ausbildung der Basalmembran, die immuncytochemisch mit einer gesteigerten Expression der Hauptkomponenten Collagen Typ IV und Laminin korrelierte. Die Beurteilung des morphologischen Gesamtbilds in der H/E-Darstellung zeigte große strukturelle Gemeinsamkeiten zwischen dem in vitro gezüchteten und dem physiologischen Nativgewebe. Wie die abschließenden Mutationsuntersuchungen bezüglich des Tumorsuppressorgens p53 ergaben, konnten bei den isolierten Epithel- und Bindegewebszellen keine genetischen Alterationen im Sinne einer neoplastischen Entartung gefunden werden.
