Literary Meals in Canada étudie Pig Tails ’n Breadfruit d'Austin Clarke, Chorus of Mushrooms et The Kappa Child de Hiromi Goto, This Body de Tessa McWatt, ainsi que Diamond Grill de Fred Wah. Cette thèse entreprend d’établir la signification de la nourriture dans ces récits, ce qu'elle permet aux auteur(e)s d'exprimer par rapport à divers thématiques—les structures sociales, la culture, le langage, ou encore la subjectivité—et comment ils/elles établissent des connexions entre elles, et quelles conclusions ils/elles en tirent. En d'autres termes, cette thèse s'interroge sur les stratégies utilisées par ces auteur(e)s lorsqu'ils écrivent de la “nourritéra-ture.” Ma lecture de ces oeuvres est aussi ancrée au sein d'une conversation sur la nourriture au sens large: que ce soit dans les cercles académiques, dans les supermarchés, par l'intermédiaire des étiquettes, ou dans les médias. J'examine comment mon corpus littéraire répond, infirme, ou confirme les discours actuels sur la nourriture. Divisé en quatre chapitres—Production, Approvisionnement, Préparation, et Consommation—ce mémoire précise la signification du “literary supermarket” de Rachel Bowlby, en s'appuyant sur les travaux de Michael Pollan et Hiromi Goto; compare la haute cuisine d'Escoffier à la “hot-cuisine” d'Austin Clarke; recherche les connections entre l’acte de faire la cuisine et celui de l’écrire chez Luce Giard, Austin Clarke, et Fred Wah; confronte les préceptes d'Emily Post concernant les bonnes manières de la table à la cacophonie et aux bruits de mastication chez Hiromi Goto; et relie Tessa McWatt et Elspeth Probyn qui partagent, toutes deux, un intérêt et une approche à la sustentation des corps. Les textes qui composent ce corpus sont des “foodbooks” (“aliment-textes”). La nourriture, et les différentes activités qui y sont associées, y est transcrite. C’est pourquoi cette thèse accorde une grande importance aux particularités de ce moyen d'expression.

Problème: Ma thèse porte sur l’identité individuelle comme interrogation sur les enjeux personnels et sur ce qui constitue l’identification hybride à l’intérieur des notions concurrentielles en ce qui a trait à l’authenticité. Plus précisément, j’aborde le concept des identifications hybrides en tant que zones intermédiaires pour ce qui est de l’alternance de codes linguistiques et comme négociation des espaces continuels dans leur mouvement entre les cultures et les langues. Une telle négociation engendre des tensions et/ou apporte le lien créatif. Les tensions sont inhérentes à n’importe quelle construction d’identité où les lignes qui définissent des personnes ne sont pas spécifiques à une culture ou à une langue, où des notions de l’identité pure sont contestées et des codes communs de l’appartenance sont compromis. Le lien créatif se produit dans les exemples où l’alternance de code linguistique ou la négociation des espaces produit le mouvement ouvert et fluide entre les codes de concurrence des références et les différences à travers les discriminations raciales, la sexualité, la culture et la langue. Les travaux que j’ai sélectionnés représentent une section transversale de quelques auteurs migrants provenant de la minorité en Amérique du Nord qui alternent les codes linguistiques de cette manière. Les travaux détaillent le temps et l’espace dans leur traitement de l’identité et dans la façon dont ils cernent l’hybridité dans les textes suivants : The Woman Warrior de Maxine Hong Kingston (1975-76), Hunger of Memory de Richard Rodriguez (1982), Comment faire l’amour avec un nègre sans se fatiguer de Dany Laferrière (1985), Borderlands/La Frontera de Gloria Anzalduá (1987), Lost in Translation de Eva Hoffman (1989), Avril ou l’anti-passion de Antonio D’Alfonso (1990) et Chorus of Mushrooms de Hiromi Goto (1994). Enjeux/Questions La notion de l’identification hybride est provocante comme sujet. Elle met en question l’identité pure. C’est un sujet qui a suscité beaucoup de discussions tant en ce qui a trait à la littérature, à la politique, à la société, à la linguistique, aux communications, qu’au sein même des cercles philosophiques. Ce sujet est compliqué parce qu’il secoue la base des espaces fixes et structurés de l’identité dans sa signification culturelle et linguistique. Par exemple, la notion de patrie n’a pas les représentations exclusives du pays d’origine ou du pays d’accueil. De même, les notions de race, d’appartenance ethnique, et d’espaces sexuels sont parfois négativement acceptées si elles proviennent des codes socialement admis et normalisés de l’extérieur. De tels codes de la signification sont souvent définis par l’étiquette d’identification hétérosexuelle et blanche. Dans l’environnement généralisé d’aujourd’hui, plus que jamais, une personne doit négocier qui elle est, au sens de son appartenance à soi, en tant qu’individu et ce, face aux modèles locaux, régionaux, nationaux, voire même globaux de la subjectivité. Nous pouvons interpréter ce mouvement comme une série de couches superposées de la signification. Quand nous rencontrons une personne pour la première fois, nous ne voyons que la couche supérieure. D’ailleurs, son soi intérieur est caché par de nombreuses couches superposées (voir Joseph D. Straubhaar). Toutefois, sous cette couche supérieure, on retrouve beaucoup d’autres couches et tout comme pour un oignon, on doit les enlever une par une pour que l’individualité complète d’une personne soit révélée et comprise. Le noyau d’une personne représente un point de départ crucial pour opposer qui elle était à la façon dont elle se transforme sans cesse. Sa base, ou son noyau, dépend du moment, et comprend, mais ne s’y limite pas, ses origines, son environnement et ses expériences d’enfance, son éducation, sa notion de famille, et ses amitiés. De plus, les notions d’amour-propre et d’amour pour les autres, d’altruisme, sont aussi des points importants. Il y a une relation réciproque entre le soi et l’autre qui établit notre degré d’estime de soi. En raison de la mondialisation, notre façon de comprendre la culture, en fait, comment on consomme et définit la culture, devient rapidement un phénomène de déplacement. À l’intérieur de cette arène de culture généralisée, la façon dont les personnes sont à l’origine chinoises, mexicaines, italiennes, ou autres, et poursuivent leur évolution culturelle, se définit plus aussi facilement qu’avant. Approche Ainsi, ma thèse explore la subjectivité hybride comme position des tensions et/ou des relations créatrices entre les cultures et les langues. Quoique je ne souhaite aucunement simplifier ni le processus, ni les questions de l’auto-identification, il m’apparaît que la subjectivité hybride est aujourd’hui une réalité croissante dans l’arène généralisée de la culture. Ce processus d’échange est particulièrement complexe chez les populations migrantes en conflit avec leur désir de s’intégrer dans les nouveaux espaces adoptés, c’est-à-dire leur pays d’accueil. Ce réel désir d’appartenance peut entrer en conflit avec celui de garder les espaces originels de la culture définie par son pays d’origine. Ainsi, les références antérieures de l’identification d’une personne, les fondements de son individualité, son noyau, peuvent toujours ne pas correspondre à, ou bien fonctionner harmonieusement avec, les références extérieures et les couches d’identification changeantes, celles qu’elle s’approprie du pays d’accueil. Puisque nos politiques, nos religions et nos établissements d’enseignement proviennent des représentations nationales de la culture et de la communauté, le processus d’identification et la création de son individualité extérieure sont formées par le contact avec ces établissements. La façon dont une personne va chercher l’identification entre les espaces personnels et les espaces publics détermine ainsi le degré de conflit et/ou de lien créatif éprouvé entre les modes et les codes des espaces culturels et linguistiques. Par conséquent, l’identification des populations migrantes suggère que la « community and culture will represent both a hybridization of home and host cultures » (Straubhaar 27). Il y a beaucoup d’écrits au sujet de l’hybridité et des questions de l’identité et de la patrie, toutefois cette thèse aborde la valeur créative de l’alternance de codes culturels et linguistiques. Ce que la littérature indiquera Par conséquent, la plate-forme à partir de laquelle j’explore mon sujet de l’hybridité flotte entre l’interprétation postcoloniale de Homi Bhabha concernant le troisième espace hybride; le modèle d’hétéroglossie de Mikhail Bakhtine qui englobent plusieurs de mes exemples; la représentation de Roland Barthes sur l’identité comme espace transgressif qui est un modèle de référence et la contribution de Chantal Zabus sur le palimpseste et l’alternance de codes africains. J’utilise aussi le modèle de Sherry Simon portant sur l’espace urbain hybride de Montréal qui établit un lien important avec la valeur des échanges culturels et linguistiques, et les analyses de Janet Paterson. En effet, la façon dont elle traite la figure de l’Autre dans les modèles littéraires au Québec fournisse un aperçu régional et national de l’identification hybride. Enfin, l’exploration du bilinguisme de Doris Sommer comme espace esthétique et même humoristique d’identification situe l’hybridité dans une espace de rencontre créative. Conséquence Mon approche dans cette thèse ne prétend pas résoudre les problèmes qui peuvent résulter des plates-formes de la subjectivité hybride. Pour cette raison, j’évite d’aborder toute approche politique ou nationaliste de l’identité qui réfute l’identification hybride. De la même façon, je n’amène pas de discussion approfondie sur les questions postcoloniales. Le but de cette thèse est de démontrer à quel point la subjectivité hybride peut être une zone de relation créatrice lorsque l’alternance de codes permet des échanges de communication plus intimes entre les cultures et les langues. C’est un espace qui devient créateur parce qu’il favorise une attitude plus ouverte vis-à-vis les différents champs qui passent par la culture, aussi bien la langue, que la sexualité, la politique ou la religion. Les zones hybrides de l’identification nous permettent de contester les traditions dépassées, les coutumes, les modes de communication et la non-acceptation, toutes choses dépassées qui emprisonnent le désir et empêchent d’explorer et d’adopter des codes en dehors des normes et des modèles de la culture contenus dans le discours blanc, dominant, de l’appartenance culturelle et linguistique mondialisée. Ainsi, il appert que ces zones des relations multi-ethniques exigent plus d’attention des cercles scolaires puisque la population des centres urbains à travers l’Amérique du Nord devient de plus en plus nourrie par d’autres types de populations. Donc, il existe un besoin réel d’établir une communication sincère qui permettrait à la population de bien comprendre les populations adoptées. C’est une invitation à stimuler une relation plus intime de l’un avec l’autre. Toutefois, il est évident qu’une communication efficace à travers les frontières des codes linguistiques, culturels, sexuels, religieux et politiques exige une négociation continuelle. Mais une telle négociation peut stimuler la compréhension plus juste des différences (culturelle ou linguistique) si des institutions académiques offrent des programmes d’études intégrant davantage les littératures migrantes. Ma thèse vise à illustrer (par son choix littéraire) l’identification hybride comme une réalité importante dans les cultures généralisées qui croissent toujours aujourd’hui. Les espaces géographiques nous gardent éloignés les uns des autres, mais notre consommation de produits exotiques, qu’ils soient culturels ou non, et même notre consommation de l’autre, s’est rétrécie sensiblement depuis les deux dernières décennies et les indicateurs suggèrent que ce processus n’est pas une tendance, mais plutôt une nouvelle manière d’éprouver la vie et de connaître les autres. Ainsi les marqueurs qui forment nos frontières externes, aussi bien que ces marqueurs qui nous définissent de l’intérieur, exigent un examen minutieux de ces enjeux inter(trans)culturels, surtout si nous souhaitons nous en tenir avec succès à des langues et des codes culturels présents, tout en favorisant la diversité culturelle et linguistique. Recent releases from the International Federation of Accountants (IFAC) highlight the importance of ethics education. Academic institutions employ varying methods of teaching ethics and place varying levels of emphasis on ethics teaching during a business/accounting degree. This paper attempts to evaluate whether teaching ethics to final year accountancy students is beneficial. At the commencement of a semester 85 final year accounting students were given five ethical scenarios on which to make an ethical decision. During the semester they were subject to two different methods of teaching ethics, a traditional lecture/tutorial process and a group assignment. ----- After a significant gap, students were re-presented with the ethical scenarios and asked what action they now considered appropriate. In all five instances students offered a more ethical response the second time. When asked to evaluate the methodologies the students considered both training methods to have a positive effect on their ethical thinking. The results suggest it is beneficial to include ethics teaching in accountancy courses, if the profession’s goal of ethical practitioners is to be achieved.

The expression of transgenes in plant genomes can be inhibited by either transcriptional gene silencing or posttranscriptional gene silencing (PTGS). Overexpression of the chalcone synthase-A (CHS-A) transgene triggers PTGS of CHS-A and thus results in loss of flower pigmentation in petunia. We previously demonstrated that epigenetic inactivation of CHS-A transgene transcription leads to a reversion of the PTGS phenotype. Although neomycin phosphotransferase II (nptII), a marker gene co-introduced into the genome with the CHS-A transgene, is not normally silenced in petunia, even when CHS-A is silenced, here we found that nptII was silenced in a petunia line in which CHS-A PTGS was induced, but not in the revertant plants that had no PTGS of CHS-A. Transcriptional activity, accumulation of short interfering RNAs, and restoration of mRNA level after infection with viruses that had suppressor proteins of gene silencing indicated that the mechanism for nptII silencing was posttranscriptional. Read-through transcripts of the CHS-A gene toward the nptII gene were detected. Deep-sequencing analysis revealed a striking difference between the predominant size class of small RNAs produced from the read-through transcripts (22 nt) and that from the CHS-A RNAs (21 nt). These results implicate the involvement of read-through transcription and distinct phases of RNA degradation in the coincident PTGS of linked transgenes and provide new insights into the destabilization of transgene expression.

Background The Global Burden of Disease Study 2013 (GBD 2013) aims to bring together all available epidemiological data using a coherent measurement framework, standardised estimation methods, and transparent data sources to enable comparisons of health loss over time and across causes, age–sex groups, and countries. The GBD can be used to generate summary measures such as disability-adjusted life-years (DALYs) and healthy life expectancy (HALE) that make possible comparative assessments of broad epidemiological patterns across countries and time. These summary measures can also be used to quantify the component of variation in epidemiology that is related to sociodemographic development. Methods We used the published GBD 2013 data for age-specific mortality, years of life lost due to premature mortality (YLLs), and years lived with disability (YLDs) to calculate DALYs and HALE for 1990, 1995, 2000, 2005, 2010, and 2013 for 188 countries. We calculated HALE using the Sullivan method; 95% uncertainty intervals (UIs) represent uncertainty in age-specific death rates and YLDs per person for each country, age, sex, and year. We estimated DALYs for 306 causes for each country as the sum of YLLs and YLDs; 95% UIs represent uncertainty in YLL and YLD rates. We quantified patterns of the epidemiological transition with a composite indicator of sociodemographic status, which we constructed from income per person, average years of schooling after age 15 years, and the total fertility rate and mean age of the population. We applied hierarchical regression to DALY rates by cause across countries to decompose variance related to the sociodemographic status variable, country, and time. Findings Worldwide, from 1990 to 2013, life expectancy at birth rose by 6·2 years (95% UI 5·6–6·6), from 65·3 years (65·0–65·6) in 1990 to 71·5 years (71·0–71·9) in 2013, HALE at birth rose by 5·4 years (4·9–5·8), from 56·9 years (54·5–59·1) to 62·3 years (59·7–64·8), total DALYs fell by 3·6% (0·3–7·4), and age-standardised DALY rates per 100 000 people fell by 26·7% (24·6–29·1). For communicable, maternal, neonatal, and nutritional disorders, global DALY numbers, crude rates, and age-standardised rates have all declined between 1990 and 2013, whereas for non–communicable diseases, global DALYs have been increasing, DALY rates have remained nearly constant, and age-standardised DALY rates declined during the same period. From 2005 to 2013, the number of DALYs increased for most specific non-communicable diseases, including cardiovascular diseases and neoplasms, in addition to dengue, food-borne trematodes, and leishmaniasis; DALYs decreased for nearly all other causes. By 2013, the five leading causes of DALYs were ischaemic heart disease, lower respiratory infections, cerebrovascular disease, low back and neck pain, and road injuries. Sociodemographic status explained more than 50% of the variance between countries and over time for diarrhoea, lower respiratory infections, and other common infectious diseases; maternal disorders; neonatal disorders; nutritional deficiencies; other communicable, maternal, neonatal, and nutritional diseases; musculoskeletal disorders; and other non-communicable diseases. However, sociodemographic status explained less than 10% of the variance in DALY rates for cardiovascular diseases; chronic respiratory diseases; cirrhosis; diabetes, urogenital, blood, and endocrine diseases; unintentional injuries; and self-harm and interpersonal violence. Predictably, increased sociodemographic status was associated with a shift in burden from YLLs to YLDs, driven by declines in YLLs and increases in YLDs from musculoskeletal disorders, neurological disorders, and mental and substance use disorders. In most country-specific estimates, the increase in life expectancy was greater than that in HALE. Leading causes of DALYs are highly variable across countries. Interpretation Global health is improving. Population growth and ageing have driven up numbers of DALYs, but crude rates have remained relatively constant, showing that progress in health does not mean fewer demands on health systems. The notion of an epidemiological transition—in which increasing sociodemographic status brings structured change in disease burden—is useful, but there is tremendous variation in burden of disease that is not associated with sociodemographic status. This further underscores the need for country-specific assessments of DALYs and HALE to appropriately inform health policy decisions and attendant actions.

Background The Global Burden of Disease, Injuries, and Risk Factor study 2013 (GBD 2013) is the first of a series of annual updates of the GBD. Risk factor quantification, particularly of modifiable risk factors, can help to identify emerging threats to population health and opportunities for prevention. The GBD 2013 provides a timely opportunity to update the comparative risk assessment with new data for exposure, relative risks, and evidence on the appropriate counterfactual risk distribution. Methods Attributable deaths, years of life lost, years lived with disability, and disability-adjusted life-years (DALYs) have been estimated for 79 risks or clusters of risks using the GBD 2010 methods. Risk–outcome pairs meeting explicit evidence criteria were assessed for 188 countries for the period 1990–2013 by age and sex using three inputs: risk exposure, relative risks, and the theoretical minimum risk exposure level (TMREL). Risks are organised into a hierarchy with blocks of behavioural, environmental and occupational, and metabolic risks at the first level of the hierarchy. The next level in the hierarchy includes nine clusters of related risks and two individual risks, with more detail provided at levels 3 and 4 of the hierarchy. Compared with GBD 2010, six new risk factors have been added: handwashing practices, occupational exposure to trichloroethylene, childhood wasting, childhood stunting, unsafe sex, and low glomerular filtration rate. For most risks, data for exposure were synthesised with a Bayesian meta-regression method, DisMod-MR 2.0, or spatial-temporal Gaussian process regression. Relative risks were based on meta-regressions of published cohort and intervention studies. Attributable burden for clusters of risks and all risks combined took into account evidence on the mediation of some risks such as high body-mass index (BMI) through other risks such as high systolic blood pressure and high cholesterol. Findings All risks combined account for 57·2% (95% uncertainty interval [UI] 55·8–58·5) of deaths and 41·6% (40·1–43·0) of DALYs. Risks quantified account for 87·9% (86·5–89·3) of cardiovascular disease DALYs, ranging to a low of 0% for neonatal disorders and neglected tropical diseases and malaria. In terms of global DALYs in 2013, six risks or clusters of risks each caused more than 5% of DALYs: dietary risks accounting for 11·3 million deaths and 241·4 million DALYs, high systolic blood pressure for 10·4 million deaths and 208·1 million DALYs, child and maternal malnutrition for 1·7 million deaths and 176·9 million DALYs, tobacco smoke for 6·1 million deaths and 143·5 million DALYs, air pollution for 5·5 million deaths and 141·5 million DALYs, and high BMI for 4·4 million deaths and 134·0 million DALYs. Risk factor patterns vary across regions and countries and with time. In sub-Saharan Africa, the leading risk factors are child and maternal malnutrition, unsafe sex, and unsafe water, sanitation, and handwashing. In women, in nearly all countries in the Americas, north Africa, and the Middle East, and in many other high-income countries, high BMI is the leading risk factor, with high systolic blood pressure as the leading risk in most of Central and Eastern Europe and south and east Asia. For men, high systolic blood pressure or tobacco use are the leading risks in nearly all high-income countries, in north Africa and the Middle East, Europe, and Asia. For men and women, unsafe sex is the leading risk in a corridor from Kenya to South Africa. Interpretation Behavioural, environmental and occupational, and metabolic risks can explain half of global mortality and more than one-third of global DALYs providing many opportunities for prevention. Of the larger risks, the attributable burden of high BMI has increased in the past 23 years. In view of the prominence of behavioural risk factors, behavioural and social science research on interventions for these risks should be strengthened. Many prevention and primary care policy options are available now to act on key risks.

Suun kautta annosteltava kalsiumherkistäjä parantaa sydämen vajaatoimintaan liittyvää pumppausvajetta kokeellisissa sydämen vajaatoimintamalleissa Huolimatta viime vuosikymmenien lääketieteellisestä kehityksestä krooninen sydämen vajaatoiminta on silti edelleen vakava, elämänlaatua voimakkaasti rajoittava sairaus. Kalsiumherkistäjät ovat uusi, sydämen pumppausvoimaa lisäävä lääkeryhmä. Levosimendaani, kotimaista alkuperää oleva kalsiumherkistäjä, on kliinisessä käytössä akuutin vajaatoiminnan hoitoon suonensisäisesti ja lyhytaikaisesti annosteltavana valmisteena. Levosimendaanilla on aktiivinen metaboliitti, OR-1896, jonka oletetaan olevan vuorokauden mittaisen levosimendaani-infuusion jälkeen havaittujen useita päiviä kestävien hyödyllisisten vaikutuksisten takana. Levosimendaanin kroonisen, suun kautta tapahtuvan annostelun vaikutuksista tieto on vähäisempää, mutta sillä näyttää olevan positiivisia vaikutuksia potilaiden raportoimana. FM Marjut Louhelainen on selvittänyt väitöskirjassaan suun kautta annosteltavan levosimendaanin ja sen pitkäkestoisen aktiivisen metaboliitin vaikutuksia kroonisen vajaatoiminnan hoidossa käyttämällä sekä hypertensiivisen sydäntaudin että 2 tyypin diabeteksen komplisoimaan sydäninfarktin kokeellisia malleja. Tutkimuksessa selvitettiin lisäksi vajaatoimintaan johtavia molekyylitason tapahtumia sydänlihaksessa. Tutkimuksessa osoitettiin, että krooninen suun kautta annosteltu hoito sekä kalsiumherkistäjä levosimendaanilla että sen aktiivisella metaboliitilla estää hypertensiiviseen sydämen vajaatoiminnan aikaasaamaa sydämen uudelleenmuovaantumista ja siihen liittyvää kuolleisuutta. Nämä vaikutukset välittyivät vähentyneen sydänlihassoluhypertrofian, solukuolleisuuden ja neurohumaraalisen aktivaation kautta. Levosimendaanin ja OR-1896:n osoitettiin myös parantavan sydämen pumppausfunktiota tyyppi 2 diabeteksen komplisoimassa sydäninfarktissa. Ei-diabeettiseen tilanteeseen verrattuna diabetekseen liittyvä infarktin jälkeinen vajaatoiminnan kehitys oli yhteydessä lisääntyneeseen tulehdukseen, fibroosiin, solukuolemaan, neurohumoraaliseen aktivaatioon ja ennenaikaiseen kudoksen vanhenemiseen. Sekä levosimendaani, että OR-1869 vähensivät tulehduksen, fibroosin ja solukuoleman merkkejä ja vaimensi neurohumoraalista aktivaatiota. OR-1896 myös vähensi solujen vanhenemiseen liittyvien merkkiaineiden ilmentymistä. Väitöskirjassa todettiin, että suun kautta annosteltuna sekä levosimendaani, että sen aktiivinen metaboliitti OR-1896, omaavat terapeuttista potentiaalia sekä hypertensiivisen sydäntaudin hoitoon että sydäninfarktin jälkeisen vajaatoiminnan estoon. FM Marjut Louhelaisen farmakologian alaan kuuluva väitöskirja Effects of oral calcium sensitizers on experimental heart failure tarkastetaan Helsingin yliopiston Lääketieteellisessä tiedekunnassa perjantaina 29.01.2010 klo 12 (Biomedicum Helsinki, luentosali 2, Haartmaninkatu 8, Helsinki). Increased consumption of low-fat milk products is inversely associated with the risk of hypertension. The beneficial effect of milk on blood pressure is attributed to high calcium and potassium content but also to specific peptide sequences, which are cleaved from milk protein during gastrointestinal digestion, fermentation of milk with proteolytic starter cultures or enzymatic hydrolysis. Milk products fermented with Lactobacillus helveticus contain casein-derived tripeptides isoleucine-proline-proline (Ile-Pro-Pro) and valine-proline-proline (Val-Pro-Pro), which have been shown to possess antihypertensive effects in humans and in experimental animals. The aim of the present series of studies was to investigate the effects of tripeptides Ile-Pro-Pro and Val-Pro-Pro or fermented milk products containing them on vascular function and blood pressure and to elucidate the mechanisms behind them by using different experimental models of hypertension. Another aim was to characterize the acute effects of tripeptides on blood pressure and arterial stiffness in mildly hypertensive humans. Ile-Pro-Pro and Val-Pro-Pro or fermented milk products containing them attenuated the development of hypertension in two experimental models of hypertension, spontaneously hypertensive rat (SHR) and type 2 diabetic Goto-Kakizaki (GK) rat fed with high-salt diet. Significant differences in systolic blood pressure (SBP) were seen after 8 weeks treatment with tripeptide-containing products compared to control product. Plant sterols did not enhance this effect. Two differently produced tripeptide powders produced a similar attenuating effect on SBP in SHR. In mildly hypertensive subjects, a single administration of tripeptide- and plant sterol-containing fermented milk product decreased both SBP and diastolic blood pressure (DBP) over a period of 8 hours. Protective effect of tripeptides Ile-Pro-Pro and Val-Pro-Pro and fermented milk products containing them on vascular function was demonstrated in in vitro studies and long-term experimental studies. The effect was shown to be endothelium-dependent and possibly involving endothelium-derived hyperpolarizing factor (EDHF). In the clinical study, single administration of tripeptide-containing fermented milk product did not affect measures of arterial stiffness. Long-term treatment with fermented milk product containing Ile-Pro-Pro and Val-Pro-Pro inhibited angiotensin-converting enzyme (ACE) and decreased aldosterone levels thus showing beneficial effects on the renin-angiotensin system (RAS) in SHR and GK. No changes in the components of RAS were observed by the single administration of the same product in mildly hypertensive subjects. Increased levels of cGMP, NOx and citrulline suggest increased nitric oxide (NO) production by the tripeptides. Taken together, Ile-Pro-Pro and Val-Pro-Pro -containing products attenuate the development of hypertension after long-term treatment in experimental models of hypertension and possess an acute antihypertensive effect in mildly hypertensive subjects. In addition, these tripeptides show endothelium-mediated beneficial effects on vascular function. Attenuation of blood pressure increase by the tripeptides in experimental animals involves RAS, but its role in the antihypertensive effect in humans remains to be elucidated.

Type 2 diabetes is a risk factor for the development of cardiovascular disease. Recently, the term diabetic cardiomyopathy has been proposed to describe the changes in the heart that occur in response to chronic hyperglycemia and insulin resistance. Ventricular remodelling in diabetic cardiomyopathy includes left ventricular hypertrophy, increased interstitial fibrosis, apoptosis and diastolic dysfunction. Mechanisms behind these changes are increased oxidative stress and renin-angiotensin system activation. The diabetic Goto-Kakizaki rat is a non-obese model of type 2 diabetes that exhibits defective insulin signalling. Recently two interconnected stress response pathways have been discovered that link insulin signalling, longevity, apoptosis and cardiomyocyte hypertrophy. The insulin-receptor PI3K/Ak pathway inhibits proapoptotic FOXO3a in response to insulin signalling and the nuclear Sirt1 deacetylase inhibits proapoptotic p53 and modulates FOXO3a in favour of survival and growth. --- Levosimendan is a calcium sensitizing agent used for the management of acute decompensated heart failure. Levosimendan acts as a positive inotrope by sensitizing cardiac troponin C to calcium and exerts vasodilation by opening mitochondrial and sarcolemmal ATP-sensitive potassium channels. Levosimendan has been described to have beneficial effects in ventricular remodelling after myocardial infarction. The aims of the study were to characterize whether diabetic cardiomyopathy associates with cardiac dysfunction, cardiomyocyte apoptosis, hypertrophy and fibrosis in spontaneously diabetic Goto-Kakizaki (GK) rats, which were used to model type 2 diabetes. Protein expression and activation of the Akt FOXO3a and Sirt1 p53 pathways were examined in the development of ventricular remodelling in GK rats with and without myocardial infarction (MI). The third and fourth studies examined the effects of levosimendan on ventricular remodelling and gene expression in post-MI GK rats. The results demonstrated that diabetic GK rats develop both modest hypertension and features similar to diabetic cardiomyopathy including cardiac dysfunction, LV hypertrophy and fibrosis and increased apoptotic signalling. MI induced a sustained increase in cardiomyocyte apoptosis in GK rats together with aggravated LV hypertrophy and fibrosis. The GK rat myocardium exhibited decreased Akt- FOXO3a phosphorylation and increased nuclear translocation of FOXO3a and overproduction of the Sirt1 protein. Treatment with levosimendan decreased cardiomyocyte apoptosis, senescence and LV hypertrophy and altered the gene expression profile in GK rat myocardium. The findings indicate that impaired cardioprotection via Akt FOXO3a and p38 MAPK is associated with increased apoptosis, whereas Sirt1 functions in counteracting apoptosis and the development of LV hypertrophy in the GK rat myocardium. Overall, levosimendan treatment protects against post-MI ventricular remodelling and alters the gene expression profile in the GK rat myocardium.

We investigate the electronic structure of Ca1-xSrxVO3 using photoemission spectroscopy. Core level spectra establish an electronic phase separation at the surface, leading to a distinctly different surface electronic structure compared to the bulk. Analysis of the photoemission spectra of this system allowed us to separate the surface and bulk contributions. These results help us to understand properties related to two vastly differing energy scales, namely the low-energy scale of thermal excitations ( $\sim\!k_{\rm B}T$) and the high-energy scale related to Coulomb and other electronic interactions.

Esse trabalho tem como objetivo compreender como se configura a relação entre o saber psicológico que chega ao grande público na sociedade contemporânea e o fenômeno aqui chamado de especialismo psi de reduções cientificistas. Tal fenômeno será aqui entendido como um tipo de especialismo que acredita possuir o domínio absoluto das narrativas sobre o homem, domínio esse pautado muitas vezes em uma aceitação incondicional da concepção de ciência proveniente do campo das ciências exatas. Para investigar como uma redução cientificista estaria chegando ao grande público, o presente trabalho faz uso de alguns flashes do contemporâneo, incluindo aí uma breve análise de fragmentos jornalísticos que retratam questões psi publicadas nos anos 2009 e 2010 em duas revistas de grande circulação no Rio de Janeiro. O método progressivo-regressivo proposto por Jean-Paul Sartre será a estratégia metodológica adotada nesse trabalho, onde a tentativa de não compreender o fenômeno humano como algo puramente individual nem puramente universal se apresenta como um constante desafio. Uma restrição cientificista no campo psi foi observada a partir de tendências como a do uso do método experimental e sua conseqüente tentativa de criação de relações causais, além da busca pela substancialização de experiências subjetivas, usando para isso conhecimentos da neurociência e fenômenos como o da matematização da existência. A busca por universalidades abstratas e o abandono do sentido singular parecem ser, portanto, características importantes desse saber psi analisado. Para reafirmar a possibilidade de lidar com o conhecimento psicológico a partir da abertura de sentidos, e não a partir de uma restrição cientificista, esse trabalho busca principalmente no pensamento de Jean-Paul Sartre elementos para pensar uma proposta de saber que fuja da formatação dos sentidos feita pela cultura dos especialismos.

A proposta do trabalho é analisar como a morte é entendida pela visão da hipermodernidade e pela visão do Budismo. Na contemporaneidade cuja lógica capitalista é embasada na lógica do mercado onde o consumo assume o papel principal, a morte se tornou um tabu, onde ela é evitada, esvaziada de sentido e descaracterizada. A dor e o sofrimento são depreciados e é exigido do homem uma inabalável postura performática e um desempenho cada vez melhor. Há ainda a crença de que o discurso tecnocientífico trará todas as soluções para as mazelas humanas. A felicidade é, portanto, um imperativo da sociedade hipermoderna e sua busca é exteriorizada isentando os indivíduos de um olhar crítico. Assim, a morte e o luto perdem seu lugar para a busca incessante de satisfação e bem-estar. O Budismo tem uma lógica que segue na contramão. Ensina que a existência humana no Samsara é constituída por principalmente quatro sofrimentos básicos: nascimento, envelhecimento, doença e morte. O Budismo ensina que a morte, assim como a vida, é um fenômeno comum a todos os seres vivos e que o exercício budista possibilita compreender o real significado da vida e da morte. A meditação sobre a impermanência, uma das práticas budistas, visa familiarizar o adepto budista a três pensamentos: certamente vou morrer; a hora da minha morte é totalmente incerta e na hora da minha morte e, depois dela, só a prática do Dharma vai me ajudar. Postula que para alcançar a verdadeira felicidade o homem deve provocar uma mudança interior, exercitar a compaixão e se desapegar da crença de que os fenômenos são permanentes e imutáveis. Tais considerações foram possíveis a partir da pesquisa sobre o Budismo da Nova Tradição Kadampa a partir de uma metodologia etnográfica que incluíram visitas ao campo de estudo, a confecção de um diário de campo e a realização de entrevistas com os praticantes budistas da Nova Tradição Kadampa.

Larval and juvenile rockfishes (Sebastes spp.) are difficult to identify using morphological characters. We developed a key based on sizes of restriction endonuclease fragments of the NADH dehydrogenase-3 and -4 (ND3/ND4) and 12S and 16S ribosomal RNA (12S/16S) mitochondrial regions. The key makes use of variation in the ND3/ND4 region. Restriction endonuclease Dde I variation can corroborate identifications, as can 12S/16S variation. The key, based on 71 species, includes most North American taxa, several Asian species, and Sebastolobus alascanus and Helicolenus hilgendorfi that are closely related to rockfishes. Fifty-eight of 71 rockfish species in our database can be distinguished unequivocally, using one to five restriction enzymes; identities of the remaining species are narrowed to small groups: 1) S. polyspinis, S. crameri, and S. ciliatus or variabilis (the two species could not be distinguished and were considered as a single species) ; 2) S. chlorostictus, S. eos, and S. rosenblatti; 3) S. entomelas and S. mystinus; 4)S. emphaeus, S. variegatus, and S. wilsoni; and 5) S. carnatus and S. chrysomelas.
