O herpesvírus bovino tipo 1 (BHV-1) está amplamente disseminado no rebanho bovino brasileiro. Visando aprofundar o conhecimento sobre a patogenicidade para o trato respiratório de amostras dos subtipos deste vírus mais freqüentemente encontrados no Brasil (BHV-1.1 e BHV-1.a), isolados autóctones oriundos de casos de doença respiratória foram inoculados em bovinos suscetíveis. As amostras de ambos os subtipos foram capazes de induzir doença respiratória após inoculação pela via intranasal, em intensidade que variou de moderada a grave, independentemente do subtipo de vírus inoculado. Foi ainda caracterizada a resposta imune humoral induzida por tais amostras quanto ao perfil de classes e subclasses de imunoglobulinas, sendo esta igualmente indistinguível com relação aos vírus inoculados. O perfil de classes de imunoglobulinas apresentadas pelos animais permitiu a determinação do status da infecção nos animais inoculados através da análise da resposta sorológica. Na segunda etapa deste trabalho foram testadas as propriedades vacinais de um vírus recombinante, do qual foi deletada a glicoproteína E (gE; 265gE-), gerado a partir de uma amostra brasileira de BHV-1.2a. Experimentos de inoculação do recombinante 265gE- em animais suscetíveis e o desafio destes animais com a amostra parental virulenta demonstraram a segurança e eficácia desta amostra na prevenção de sinais clínicos da infecção pelo BHV-1. Posteriormente, o recombinante 265gE- foi inoculado em vacas em diferentes estágios da gestação. Não foram observadas quaisquer anormalidades nestas gestações e as 22 vacas vacinadas deram à luz a animais saudáveis. Conclui-se que o recombinante é imunogênico e capaz de conferir significativa proteção frente ao desafio com a amostra parental virulenta do vírus. O recombinante também não causou enfermidade nas vacas inoculadas nem tampouco aos fetos quando inoculado em diferentes estágios da gestação.

The bovine Herpesvirus type 1 and type 5 (BoHV-1 and BoHV-5), causing diseases and significant economic losses in farms of worldwide. Both affect the nervous system of cattle, although BoHV-5 has been the most associated with this type of pathogenesis. Given the death of animals with nervous symptoms and negative diagnoses for rabies virus in the area of study, this research focused on the detection of positive reactors to bovine herpes virus serum neutralization. We collected 518 blood samples from animals without Herpesvirus vaccine, in the municipalities of Caparrapi, Cimitarra, Honda and Victoria, in the Middle Magdalena River Region. In addition, epidemiological information useful to discuss neurological disease was collected through primary and secondary sources. For the analysis of data was used chi-square test by identification of relationship between evidence of viral infection and the variables recorded. The results revealed that 286 cases were positive for Herpesvirus infection, corresponding to a prevalence of 55.5%, however, there was no statistical relationship (p < 0.05) between the presence of antibodies and the variables analyzed. In conclusion, some cases of neurological disease in cattle in this region could be due to infection with herpes viruses. We discussed about the presence of BoHV-1 and BoHV-5 in the ambient, diagnosis and monitoring plans, as well as economic losses, which may cause in herds in this area.

The aim of this work was draw an endemic level of reproductive losses and determine positivity for four infectious agents related to reproductive problems in dairy cattle on a property in São Carlos city. Blood serum samples were collected of 142 breeding animals more than two years old, from which 21.1% showed history of abortions or stillbirths in at least one pregnancy. Immunofluorescent antibody technique, tamponated acidified antigen test, serum neutralization technique and microscopic agglutination test, were used for detection of antibody anti-Neospora caninum, anti- Brucella abortus, anti-Bovine Herpesvirus Type-1 (BoHV-1) and anti-Leptospira spp , respectively. The serological tests carried out showed that 28.9% of the animals had titers greater than or equal to 100 of anti-Neospora caninum. Viral neutralization tests demonstrated that 26.8% of the animals had titers greater or equal to 256 for antibody anti-BoHV-1.Only 7.7% of the animals studied had titers equal to or greater than 100 in the microscopic agglutination test for Leptospira spp. There weren’t observed positive reactions to the tamponated acidified antigen test for diagnosis of bovine brucellosis. The study suggests the presence of BoHV-1, N. caninum and Leptospira spp in contact with dairy cattle property and could be involved in the occurrence of abortions and stillbirths in cattle. Reproductive losses are endemic and implies a possible behavior of chronic infections caused by these microorganisms.

Twelve cases of viral meningoencephalitis in Brazilian cattle were submitted to immunohistochemical analysis for inflammatory response description. All the cases showed severe neurological signs followed by death. Mild to moderate histological inflammatory changes in the brain and cerebellum characterized the neurological infection showing meningitis, mononuclear perivascular cuffing, gliosis, haemorrhage, and macrophages (gitter cells) accompanying great areas of malacia. None of the cases showed intranuclear inclusion bodies. However, in five of them it was possible to isolate the BoHV-5. In order to collect data to allow the description of the inflammatory response in these cases, brain samples from all of the cases were analyzed by immunohistochemistry using polyclonal antibodies against CD3 to detect T cells, and against GFAP to detect astrocytes. On the other hand, monoclonal antibodies were used against BLA to detect B cells and, against MAC 387 to detect macrophages. The results indicate different degrees of prominent astrocytic response, and at the same time, T lymphocytes constituted a high percentage of the mononuclear cells which characterized the inflammatory response.
