Dogs and cats are the animals that owners most frequently seek assistance for potential poisonings, and these species are frequently involved with toxicoses due to ingestion of poisonous food. Feeding human foodstuff to pets may prove itself dangerous for their health, similarly to what is observed in Allium species toxicosis. Allium species toxicosis is reported worldwide in several animal species, and the toxic principles present in them causes the transformation of hemoglobin into methemoglobin, consequently resulting in hemolytic anemia with Heinz body formation. The aim of this review is to analyze the clinicopathologic aspects and therapeutic approach of this serious toxicosis of dogs and cats in order to give knowledge to veterinarians about Allium species toxicosis, and subsequently allow them to correctly diagnose this disease when facing it; and to educate pet owners to not feed their animals with Alliumcontaing food in order to better control this particular life-threatening toxicosis.

A doença falciforme é um termo genérico usado para determinar um grupo de alterações genéticas caracterizadas pelo predomínio de hemoglobina (Hb) S. Os principais genótipos que compõem o grupo de doença falciforme são os seguintes: SS, SF [S/beta0 talassemia e S/persistência hereditária de Hb fetal (PHHF)], SFA (S/beta+ talassemia), SC, SD e SH (S/alfa talassemia). O presente trabalho analisa os resultados das avaliações de produtos provenientes da oxidação da Hb S, identificados pela concentração da metemoglobina e de eritrócitos com corpos de Heinz em dois genótipos da doença falciforme (SS e S/beta0 talassemia) e no traço falcêmico (AS), em comparação com o genótipo normal (AA). A análise dos produtos da degradação oxidativa da hemoglobina, evidenciados pelo aumento dos valores das médias referentes à concentração de metemoglobina e do número de eritrócitos com corpos de Heinz, está diretamente relacionada com o aumento da concentração da Hb S. Assim, a degradação oxidativa da hemoglobina decresce entre os genótipos estudados da seguinte forma: SS>SF>AS>AA. É importante destacar que as análises indicaram que a simples presença de Hb S no eritrócito, como é o caso do genótipo AS, é capaz de causar elevação da concentração de metemoglobina em 52,62% das amostras analisadas e de induzir a precipitação de corpos de Heinz em 73,68% dos casos estudados. Explicações referentes aos processos oxidativos e redutores das hemoglobinas estudadas são apresentados no texto. Destaca-se, entre os resultados apresentados, a identificação por meio de eletroforese em agarose alcalina da fração de globina alfa-livre em todas as amostras do genótipo SF provenientes de pessoas com Hb S/beta0 talassemia. É proposto um esquema para explicar a origem da globina alfa-livre, especialmente para o genótipo S/beta0 talassemia, e a importância da sua identificação no diagnóstico laboratorial de Hb S/beta0 talassemia.

Foram analisados 496 indivíduos de Cubatão, SP (Brasil), cidade com alto teor de poluição industrial, com o objetivo de verificar alterações hematológicas induzidas por poluentes industriais. Dos estudos citológicos dos eritrócitos dessa população estudada, foram observadas 188 (38%) com alterações, isoladas ou combinadas em um único indivíduo, das quais 26% apresentaram policromatofilia, 24% com pontilhados basófilos, 15% com corpos de Heinz, e 8% com reticulocitose. As freqüências de metahemoglobinemia e sulfohemoglobinemia foram,respectivamente, de 35% e 32% em moradores da vila Parisi - um bairro cercado pela maioria das indústrias de Cubatão - 15% e 5% em operários das indústrias, e 12% e 4% em habitantes de áreas distantes entre 3 e 8 km do polo industrial. Esses resultados indicam que as alterações são causadas por poluentes tóxico-oxidantes e que as conseqüências fisio-patológicas no sangue dos moradores de Cubatão parecem indicar que estão relacionadas com o tempo de exposição e com a proximidade dos focos emissores de poluentes.

Dapsone (DDS) is useful in the treatment of a number of inflammatory conditions which are characterized by neutrophil infiltration. It is the drug of choice for the treatment of leprosy and prophylaxis of malaria. Haematological side effects of methaemoglobinaemia and haemolysis have been long recognized. However, the frequency and severity of these side effects in patients already treated with DDS as a single drug or as part of a multidrug therapy (MDT) have not been well documented. We report herein an investigation of the effect of dapsone long-term treatment on the haematological and biochemical alterations in leprosy patients undergoing dapsone as a single drug (DDS group) or as part of a multidrug therapy in combination with rifampin and clofazimine (MDT group). Methaemoglobinaemia and haemolytic anaemia were the principal side effects observed. Reticulocytes were found to be elevated (> 1.5%) in 90% of the patients. Heinz bodies were also detected (6.6% of the patients). The osmotic fragility test showed a reduction in cell resistance and in the evaluation of white cells a severe eosinophilia was found. Hepatic, pancreatic and renal evaluation by the determination of biochemical parameters showed rare and occasional changes of no apparent clinical significance. We conclude that haematological side effects of dapsone are significant even at doses currently used to treat leprosy (100 mg/day) and that rifampin and clofazimine do not increase the incidence of these effects during long-term treatment.

Analysis of the products of oxidative degradation of Hb S was made by methahemoglobin measurement and a count of red blood cells with Heinz bodies. Free radicals originating from oxidation cause extensive injury to erythrocytes, decreasing their useful survival period especially in Hb S carriers. The Superoxide ion (O 2) is the most responsible for the oxidation process of Hb forming membrane-bound haemachromes which afterwards evolve to Heinz bodies, damaging the membrane and provoking erythrocytes hemolysis. The results from this work showed that the SS genotype is more susceptible to the action of the free radicals than the S/Tal genotype. The β genotype has a lower oxidative susceptibility than the SS because it has only one β s mutation. The results allowed us to conclude that: a) the simple presence of Hb S, independent of its genotype and its concentration, is sufficient to produce methaemoglobin from this Hb; b) there is not a direct relationship between methaetnoglobin concentration and the Heinz bodies count; c) the intensity of Heinz bodies in the sickled erythrocytes seems to be independent of the Hb Fetal concentration; d) The genotype SS is more susceptible to Hb oxidation with the release of products of oxidative degradation; e) methaemoglobin formation in blood of people with Hb AA and Hb AS, assessed over 24, 48, 72 and 163 hourly-periods, showed greater oxidative intensity in the Hb AS compared with Hb AA.

O processo inflamatório decorrente da infecção por Mycobacterium leprae e a administração de fármacos com propriedades oxidativas, como a dapsona, são fatores de riscos ao estresse oxidativo ocasionado em pacientes com hanseníase. Este trabalho visa determinar as concentrações plasmáticas de dapsona em pacientes com hanseníase em uso de poliquimioterapia (PQT), correlacionando ao desenvolvimento do estresse oxidativo. Para o estudo, foram selecionados indivíduos saudáveis e pacientes com hanseníase, acompanhados antes (D0) e após a terceira dose supervisionada de PQT (D3). As concentrações plasmáticas de dapsona dos pacientes sob tratamento foram mesuradas por cromatografia liquida de alta eficiência. A avaliação do estresse oxidativo foi realizada através da determinação de metemoglobina (MetHb) e das concentrações de glutationa reduzida (GSH), óxido nítrico (NO), malondialdeído (MDA), capacidade antioxidante total (TEAC) e avaliação das atividades das enzimas catalase (CAT), superóxido dismutase (SOD) e da presença de corpúsculo de Heinz em esfregaços sanguíneos. No período de estudo foram obtidas 23 amostras de pacientes com hanseníase D0, 13 pacientes em D3 e 20 de indivíduos saudáveis e sem a doença. As alterações no pacientes antes do tratamento estavam associadas ao aumento de NO (D0=18.91± 2.39; controle= 6.86±1.79mM) e a redução significativa na enzima SOD (D0=69.88±12.26;controle= 138.42±14.99nmol/mL). Com relação aos pacientes sob tratamento, a concentração de dapsona no plasma foi 0.552± 0.037 μg/mL e as principais alterações observadas foram o aumento significativo no percentual de MetHb (D3= 3.29±0.74;controle=0.66±0.051%) e presença de corpúsculo de Heinz. Nos pacientes em tratamento também se observou aumento nos níveis de GSH (7.01±1.09μg/mL) quando comparados ao controle (3.33±1.09g/mL) e diminuição da atividade de CAT (D3= 10.29± 2.02; controle= 19.52±2.48 U/g de proteínas). Os níveis de MDA antes e durante o PQT não mostraram alteração, enquanto os níveis de TEAC aumentaram significativamente neste pacientes (D0=2.90 ± 0.42; D3=3.04±0.52; controle= 1.42±0.18μmol/mL). Estes dados sugerem que a PQT é a principal responsável pelo desenvolvimento de estresse oxidativo, através da geração de danos oxidativos identificados pela presença de corpúsculo de Heinz e aumento no percentual de MetHb.

This study aims to assess the oxidative stress in leprosy patients under multidrug therapy (MDT; dapsone, clofazimine and rifampicin), evaluating the nitric oxide (NO) concentration, catalase (CAT) and superoxide dismutase (SOD) activities, glutathione (GSH) levels, total antioxidant capacity, lipid peroxidation, and methemoglobin formation. For this, we analyzed 23 leprosy patients and 20 healthy individuals from the Amazon region, Brazil, aged between 20 and 45 years. Blood sampling enabled the evaluation of leprosy patients prior to starting multidrug therapy (called MDT 0) and until the third month of multidrug therapy (MDT 3). With regard to dapsone (DDS) plasma levels, we showed that there was no statistical difference in drug plasma levels between multibacillary (0.518±0.029 μg/mL) and paucibacillary (0.662±0.123 μg/mL) patients. The methemoglobin levels and numbers of Heinz bodies were significantly enhanced after the third MDTsupervised dose, but this treatment did not significantly change the lipid peroxidation and NO levels in these leprosy patients. In addition, CAT activity was significantly reduced in MDT-treated leprosy patients, while GSH content was increased in these patients. However, SOD and Trolox equivalent antioxidant capacity levels were similar in patients with and without treatment. These data suggest that MDT can reduce the activity of some antioxidant enzyme and influence ROS accumulation, which may induce hematological changes, such as methemoglobinemia in patients with leprosy. We also explored some redox mechanisms associated with DDS and its main oxidative metabolite DDS-NHOH and we explored the possible binding of DDS to the active site of CYP2C19 with the aid of molecular modeling software. © 2014 Schalcher et al.

This paper is concerned with the surface profiles of a strip after rigid bodies with serrated (saw-teeth) surfaces indent the strip and are subsequently removed. Plane-strain conditions are assumed. This has application in roughness transfer of final metal forming process. The effects of the semi-angle of the teeth, the depth of indentation and the friction on the contact surface on the profile are considered.

Two perceptions of the marginality of home economics are widespread across educational and other contexts. One is that home economics and those who engage in its pedagogy are inevitably marginalised within patriarchal relations in education and culture. This is because home economics is characterised as women's knowledge, for the private domain of the home. The other perception is that only orthodox epistemological frameworks of inquiry should be used to interrogate this state of affairs. These perceptions have prompted leading theorists in the field to call for non-essentialist approaches to research in order to re-think the thinking that has produced this cul-de-sac positioning of home economics as a body of knowledge and a site of teacher practice. This thesis takes up the challenge of working to locate a space outside the frame of modernist research theory and methods, recognising that this shift in epistemology is necessary to unsettle the idea that home economics is inevitably marginalised. The purpose of the study is to reconfigure how we have come to think about home economics teachers and the profession of home economics as a site of cultural practice, in order to think it otherwise (Lather, 1991). This is done by exploring how the culture of home economics is being contested from within. To do so, the thesis uses a 'posthumanist' approach, which rejects the conception of the individual as a unitary and fixed entity, but instead as a subject in process, shaped by desires and language which are not necessarily consciously determined. This posthumanist project focuses attention on pedagogical body subjects as the 'unsaid' of home economics research. It works to transcend the modernist dualism of mind/body, and other binaries central to modernist work, including private/public, male/female,paid/unpaid, and valued/unvalued. In so doing, it refuses the simple margin/centre geometry so characteristic of current perceptions of home economics itself. Three studies make up this work. Studies one and two serve to document the disciplined body of home economics knowledge, the governance of which works towards normalisation of the 'proper' home economics teacher. The analysis of these accounts of home economics teachers by home economics teachers, reveals that home economics teachers are 'skilled' yet they 'suffer' for their profession. Further,home economics knowledge is seen to be complicit in reinforcing the traditional roles of masculinity and femininity, thereby reinforcing heterosexual normativity which is central to patriarchal society. The third study looks to four 'atypical'subjects who defy the category of 'proper' and 'normal' home economics teacher. These 'atypical' bodies are 'skilled' but fiercely reject the label of 'suffering'. The discussion of the studies is a feminist poststructural account, using Russo's (1994) notion of the grotesque body, which is emergent from Bakhtin's (1968) theory of the carnivalesque. It draws on the 'shreds' of home economics pedagogy,scrutinising them for their subversive, transformative potential. In this analysis, the giving and taking of pleasure and fun in the home economics classroom presents moments of surprise and of carnival. Foucault's notion of the construction of the ethical individual shows these 'atypical' bodies to be 'immoderate' yet striving hard to be 'continent' body subjects. This research captures moments of transgression which suggest that transformative moments are already embodied in the pedagogical practices of home economics teachers, and these can be 'seen' when re-looking through postmodemist lenses. Hence, the cultural practices ofhome economics as inevitably marginalised are being contested from within. Until now, home economics as a lived culture has failed to recognise possibilities for reconstructing its own field beyond the confines of modernity. This research is an example of how to think about home economics teachers and the profession as a reconfigured cultural practice. Future research about home economics as a body of knowledge and a site of teacher practice need not retell a simple story of oppression. Using postmodemist epistemologies is one way to provide opportunities for new ways of looking.
