Culinary herbs and spices have long been considered essentially as flavor enhancers or preservatives, with little attention given to their potential health-promoting properties. Nevertheless, recent research has shown them to be significant dietary sources of bioactive phenolic compounds. Despite noteworthy efforts performed in recent years to improve our knowledge of their chemical composition, a detailed phenolic profile of these plant-based products is still lacking. In the present work, antioxidant activities and phenolic composition of five herbs and spices, namely caraway, turmeric, dill, marjoram and nutmeg, have been studied. The use of liquid chromatography coupled to LTQ-Orbitrap mass spectrometry enabled the identification of up to 42 phenolic compounds. To the best of our knowledge, two of them, apigenin-C-hexoside-C-pentoside and apigenin-C-hexoside-C-hexoside have not been previously reported in turmeric. Qualitative and quantitative differences were observed in polyphenol profiles, with the highest phenolic content found in caraway. Multivariate statistical treatment of the results allowed the detection of distinctive features among the studied herbs and spices.

In the present study a fast, sensitive and robust validated method to quantify chlorpheniramine in human plasma using brompheniramine as internal standard (IS) is described. The analyte and the IS were extracted from plasma by LLE (diethyl ether-dichloromethane, 80:20, v/v) and analyzed by HPLC-ESI-MS/MS. Chromatographic separation was performed using a gradient of methanol from 35 to 90% with 2.5 mm NH(4)OH on a Gemini Phenomenex C(8) 5 mu m column (50 x 4.6 mm i.d.) in 5.0 min/run. The method fitted to a linear calibration curve (0.05-10 ng/mL, R > 0.9991). The precision (%CV) and accuracy ranged, respectively: intra-batch from 1.5 to 6.8% and 99.1 to 106.6%, and inter-batch from 2.4 to 9.0%, and 99.9 to 103.1%. The validated bioanalytical procedure was used to assess the comparative bioavailability in healthy volunteers of two dexchlorpheniramine 2.0 mg tablet formulations (test dexchlorpheniramine, Eurofarma, and reference Celestamine (R), Schering-Plough). The study was conducted using an open, randomized, two-period crossover design with a 2 week washout interval. Since the 90% confidence interval for C(max) and AUC ratios were all within the 80-125% interval proposed by ANVISA and FDA, it was concluded that test and reference formulations are bioequivalent concerning the rate and the extent of absorption. This work describes the formation of transformation products (TPs) by the enzymatic degradation at laboratory scale of two highly consumed antibiotics: tetracycline (Tc) and erythromycin (ERY). The analysis of the samples was carried out by a fast and simple method based on the novel configuration of the on-line turbulent flow system coupled to a hybrid linear ion trap – high resolution mass spectrometer. The method was optimized and validated for the complete analysis of ERY, Tc and their transformation products within 10 min without any other sample manipulation. Furthermore, the applicability of the on-line procedure was evaluated for 25 additional antibiotics, covering a wide range of chemical classes in different environmental waters with satisfactory quality parameters. Degradation rates obtained for Tc by laccase enzyme and ERY by EreB esterase enzyme without the presence of mediators were ∼78% and ∼50%, respectively. Concerning the identification of TPs, three suspected compounds for Tc and five of ERY have been proposed. In the case of Tc, the tentative molecular formulas with errors mass within 2 ppm have been based on the hypothesis of dehydroxylation, (bi)demethylation and oxidation of the rings A and C as major reactions. In contrast, the major TP detected for ERY has been identified as the “dehydration ERY-A”, with the same molecular formula of its parent compound. In addition, the evaluation of the antibiotic activity of the samples along the enzymatic treatments showed a decrease around 100% in both cases

This paper reports an HPLC-ESI-MS/MS investigation on the oxidation of 3,5- and 4,5- dicaffeoylquinic acid using iron(III) tetraphenylporphyrin chloride as catalyst. Two major mono-oxidised products of the quinic acid moiety have been identified for both compounds. However, only the 4,5-derivative afforded two different tri-oxo products. Thus, it seems that the oxidation pattern depends on the number and positions of the caffeic acid moieties present in caffeoylquinic acid molecules.

A HPLC-ESI-IT-MSn method, based on high-performance liquid chromatography coupled to electrospray negative ionization multistage ion trap mass spectrometry, was developed for rapid identification of 24 flavonoid and naphthopyranone compounds. The methanol extracts of the capitulae and scapes of P. chiquitensis exhibited mutagenic activity in the Salmonella/microsome assay, against strain TA97a. © 2013 by the authors.

Sofrito is a key component of the Mediterranean diet, a diet that is strongly associated with a reduced risk of cardiovascular events. In this study, different Mediterranean sofritos were analysed for their content of polyphenols and carotenoids after a suitable work-up extraction procedure using liquid chromatography/electrospray ionisation-linear ion trap quadrupole-Orbitrap-mass spectrometry (LC/ESI-LTQ-Orbitrap- MS) and liquid chromatography/electrospray ionisation tandem triple quadrupole mass spectrometry (LC/ESI-MS-MS). In this way, 40 polyphenols (simple phenolic and hydroxycinnamoylquinic acids, and flavone, flavonol and dihydrochalcone derivatives) were identified with very good mass accuracy (<2 mDa), and confirmed by accurate mass measurements in MS and MS2 modes. The high-resolution MS analyses revealed the presence of polyphenols never previously reported in Mediterranean sofrito. The quantification levels of phenolic and carotenoid compounds led to the distinction of features among different Mediterranean sofritos according to the type of vegetables (garlic and onions) or olive oil added for their production. © 2013 Elsevier Ltd. Sekundäres organisches Aerosol (SOA) ist ein wichtiger Bestandteil von atmosphärischen Aerosolpartikeln. Atmosphärische Aerosole sind bedeutsam, da sie das Klima über direkte (Streuung und Absorption von Strahlung) und indirekte (Wolken-Kondensationskeime) Effekte beeinflussen. Nach bisherigen Schätzungen ist die SOA-Bildung aus biogenen Kohlenwasserstoffen global weit wichtiger als die SOA-Bildung aus anthropogenen Kohlenwasserstoffen. Reaktive Kohlenwasserstoffe, die in großen Mengen von der Vegetation emittiert werden und als die wichtigsten Vorläufersubstanzen für biogenes SOA gelten, sind die Terpene. In der vorliegenden Arbeit wurde eine Methode entwickelt, welche die Quantifizierung von aciden Produkten der Terpen-Oxidation ermöglicht. Die Abscheidung des größenselektierten Aerosols (PM 2.5) erfolgte auf Quarzfilter, die unter Zuhilfenahme von Ultraschall mittels Methanol extrahiert wurden. Nach Aufkonzentrierung und Lösungsmittelwechsel auf Wasser sowie Standardaddition wurden die Proben mit einer Kapillar-HPLC-ESI-MSn-Methode analysiert. Das verwendete Ionenfallen-Massenspektrometer (LCQ-DECA) bietet die Möglichkeit, Strukturaufklärung durch selektive Fragmentierung der Qasimolekülionen zu betreiben. Die Quantifizierung erfolgte teilweise im MS/MS-Modus, wodurch Selektivität und Nachweisgrenze verbessert werden konnten. Um Produkte der Terpen-Oxidation zu identifizieren, die nicht als Standards erhältlich waren, wurden Ozonolysexperimente durchgeführt. Dadurch gelang die Identifizierung einer Reihe von Oxidationsprodukten in Realproben. Neben schon bekannten Produkten der Terpen-Oxidation konnten einige Produkte erstmals in Realproben eindeutig als Produkte des α Pinens nachgewiesen werden. In den Proben der Ozonolyseexperimente konnten auch Produkte mit hohem Molekulargewicht (>300 u) nachgewiesen werden, die Ähnlichkeit zeigen zu den als Dimeren oder Polymeren in der Literatur bezeichneten Substanzen. Sie konnten jedoch nicht in Feldproben gefunden werden. Im Rahmen von 5 Messkampagnen in Deutschland und Finnland wurden Proben der atmosphärischen Partikelphase genommen. Die Quantifizierung von Produkten der Oxidation von α-Pinen, β-Pinen, 3-Caren, Sabinen und Limonen in diesen Proben ergab eine große zeitliche und örtliche Variationsbreite der Konzentrationen. Die Konzentration von Pinsäure bewegte sich beispielsweise zwischen etwa 0,4 und 21 ng/m³ während aller Messkampagnen. Es konnten stets Produkte verschiedener Terpene nachgewiesen werden. Produkte einiger Terpene eignen sich sogar als Markersubstanzen für verschiedene Pflanzenarten. Sabinen-Produkte wie Sabinsäure können als Marker für die Emissionen von Laubbäumen wie Buchen oder Birken verwendet werden, während Caren-Produkte wie Caronsäure als Marker für Nadelbäume, speziell Kiefern, verwendet werden können. Mit den quantifizierten Substanzen als Marker wurde unter zu Hilfenahme von Messungen des Gehaltes an organischem und elementarem Kohlenstoff im Aerosol der Anteil des sekundären organischen Aerosols (SOA) errechnet, der von der Ozonolyse der Terpene stammt. Erstaunlicherweise konnten nur 1% bis 8% des SOA auf die Ozonolyse der Terpene zurückgeführt werden. Dies steht im Gegensatz zu der bisherigen Meinung, dass die Ozonolyse der Terpene die wichtigste Quelle für biogenes SOA darstellt. Gründe für diese Diskrepanz werden in der Arbeit diskutiert. Um die atmosphärischen Prozesse der Bildung von SOA vollständig zu verstehen, müssen jedoch noch weitere Anstrengungen unternommen werden.

In this study, the development of a new sensitive method for the analysis of alpha-dicarbonyls glyoxal (G) and methylglyoxal (MG) in environmental ice and snow is presented. Stir bar sorptive extraction with in situ derivatization and liquid desorption (SBSE-LD) was used for sample extraction, enrichment, and derivatization. Measurements were carried out using high-performance liquid chromatography coupled to electrospray ionization tandem mass spectrometry (HPLC-ESI-MS/MS). As part of the method development, SBSE-LD parameters such as extraction time, derivatization reagent, desorption time and solvent, and the effect of NaCl addition on the SBSE efficiency as well as measurement parameters of HPLC-ESI-MS/MS were evaluated. Calibration was performed in the range of 1–60 ng/mL using spiked ultrapure water samples, thus incorporating the complete SBSE and derivatization process. 4-Fluorobenzaldehyde was applied as internal standard. Inter-batch precision was <12 % RSD. Recoveries were determined by means of spiked snow samples and were 78.9 ± 5.6 % for G and 82.7 ± 7.5 % for MG, respectively. Instrumental detection limits of 0.242 and 0.213 ng/mL for G and MG were achieved using the multiple reaction monitoring mode. Relative detection limits referred to a sample volume of 15 mL were 0.016 ng/mL for G and 0.014 ng/mL for MG. The optimized method was applied for the analysis of snow samples from Mount Hohenpeissenberg (close to the Meteorological Observatory Hohenpeissenberg, Germany) and samples from an ice core from Upper Grenzgletscher (Monte Rosa massif, Switzerland). Resulting concentrations were 0.085–16.3 ng/mL for G and 0.126–3.6 ng/mL for MG. Concentrations of G and MG in snow were 1–2 orders of magnitude higher than in ice core samples. The described method represents a simple, green, and sensitive analytical approach to measure G and MG in aqueous environmental samples.

The complex nature of venom from spider species offers a unique natural source of potential pharmacological tools and therapeutic leads. The increased interest in spider venom molecules requires reproducible and precise identification methods. The current taxonomy of the Australian Funnel-web spiders is incomplete, and therefore, accurate identification of these spiders is difficult. Here, we present a study of venom from numerous morphologically similar specimens of the Hadronyche infensa species group collected from a variety of geographic locations in southeast Queensland. Analysis of the crude venoms using online reversed-phase high performance liquid chromatography/electrospray ionisation mass spectrometry (rp-HPLC/ESI-MS) revealed that the venom profiles provide a useful means of specimen identification, from the species level to species variants. Tables defining the descriptor molecules for each group of specimens were constructed and provided a quick reference of the relationship between one specimen and another. The study revealed that the morphologically similar specimens from the southeast Queensland region are a number of different species/species variants. Furthermore, the study supports aspects of the current taxonomy with respect to the H. infensa species group. Analysis of Australian Funnel-web spider venom by rp-HPLC/ESI-MS provides a rapid and accurate method of species/species variant identification. (c) 2006 Elsevier Ltd. All rights reserved.

Il teff (Eragrostis tef) è uno pseudocereale originario dell’Etiopia che recentemente si sta diffondendo in vari Paesi. Infatti presenta numerose caratteristiche positive (buon contenuto di fibra e composti fenolici, indice glicemico ridotto, assenza di glutine) che potrebbero renderlo adatto alla formulazione di alimenti funzionali o arricchiti, destinati a categorie della popolazione con particolari esigenze (diabetici, celiaci, neonati). I dati attualmente disponibili riguardo il profilo fenolico del teff sono estremamente carenti. Pertanto, lo scopo di questa tesi è la determinazione dei composti fenolici liberi e legati in una farina di teff, mediante la combinazione della cromatografia liquida ad alte prestazioni e la spettrometria di massa con analizzatore a tempo di volo (HPLC-ESI-TOF-MS). L’estrazione dei fenoli liberi e legati dal campione di farina è stata condotta seguendo il metodo messo a punto da Verardo et al. (2011). Dall'analisi dei rispettivi base peak chromatogram sono stati individuati 15 composti fenolici nell'estratto solubile e 5 nell'estratto insolubile. I fenoli liberi riscontrati sono esclusivamente flavonoidi: 11 derivati dall'apigenina, 2 derivati dalla luteolina e 2 composti acetilati derivati dalla vitexina, identificati per la prima volta nella farina di teff. L'estratto insolubile risulta composto da acidi fenolici, rappresentati dall'acido ferulico e i suoi derivati. Non è stato possibile effettuare la determinazione dei composti fenolici totali mediante saggio al reattivo di Folin-Ciocalteu a causa dell’intorbidimento dell’estratto in fase di diluizione: questo ha confermato il limite di tale metodologia, sebbene ampiamente utilizzata, nella determinazione dei composti fenolici.

Herbs and spices have long been used to improve the flavour of food without being considered as nutritionally significant ingredients. However, the bioactive phenolic content of these plant-based products is currently attracting interest.In the present work, liquid chromatography coupled to high-resolution/accurate mass measurement LTQ-Orbitrap mass spectrometry was applied for the comprehensive identification of phenolic constituents of six of the most widely used culinary herbs (rosemary, thyme, oregano and bay) and spices (cinnamon and cumin). In this way, up to 52 compounds were identified in these culinary ingredients, some of them, as far as we know, for the first time. In order to establish the phenolic profiles of the different herbs and spices, accurate quantification of the major phenolics was performed by multiple reaction monitoring in a triple quadrupole mass spectrometer. Purpose: To study the in vivo metabolism of kurarinone, a lavandulyl flavanone which is a major constituent of Kushen and a marker compound with many biological activities, using ultra-performance liquid chromatography coupled with linear ion trap Orbitrap mass spectrometry (UPLC-LTQ-Orbitrap- MS). Methods: Six male Sprague-Dawley rats were randomly divided into two groups. First, kurarinone was suspended in 0.5 % carboxymethylcellulose sodium (CMC-Na) aqueous solution, and was given to rats (n = 3, 2 mL for each rat) orally at 50 mg/kg. A 2 mL aliquot of 0.5 % CMC-Na aqueous solution was administered to the rats in the control group. Next, urine samples were collected over 0-24 h after the oral administrations and all urine samples were pretreated by a solid phase extraction (SPE) method. Finally, all samples were analyzed by a UPLC-LTQ-Orbitrap mass spectrometry coupled with an electrospray ionization source (ESI) that was operated in the negative ionization mode. Results: A total of 11 metabolites, including the parent drug and 10 phase II metabolites in rat urine, were first detected and interpreted based on accurate mass measurement, fragment ions, and chromatographic retention times. The results were based on the assumption that kurarinone glucuronidation was the dominant metabolite that was excreted in rat urine. Conclusion: The results from this work indicate that kurarinone in vivo is typically transformed to nontoxic glucuronidation metabolites, and these findings may help to characterize the metabolic profile of kurarinone.

A sensitive, accurate and simple method using HPLC-MS/MS was developed and validated for levodopa quantitation in human plasma. Analysis was achieved on a pursuit® C18 analytical column (5 µm; 150 x 4.6 mm i.d.) using a mobile phase (methanol and water , 90:10, v/v) containing formic acid 0.5% v/v, after extracting the samples using a simple protein plasma precipitation with perchloric acid. The developed method was validated in accordance with ANVISA guidelines and was successfully applied to a bioequivalence study in 60 healthy volunteers demonstrating the feasibility and reliability of the proposed method.
