Plant extracts are some of the most attractive sources of new drugs and have shown promising results for the treatment of gastric ulcers. Several folk medicinal plants and herbs have been used to treat gastrointestinal disorders, including gastric ulcers. Mammea americana L. (Guttiferae) fruit is very common in the diet of the population of northern South America. Our research interest in this plant arose because of its potential medicinal value as a tonic and against stomachache, as used in folk medicine. In this paper we evaluated three different extracts (ethanolic/EtOH, methanolic/MeOH and dichloromethane/DCM) obtained from M. americana L., for their ability to protect the gastric mucosa against injuries caused by necrotizing agents (0.3 M HCI/60% EtOH), hypothermic restraint stress, nonsteroidal anti-inflammatory drugs (NSAID, indomethacin) and pylorus ligation. In the HCI/EtOH-induced gastriculcer model, EtOH and DCM extracts demonstrated significant inhibition of the ulcerative lesion index by 54% (12.0 +/- 2.6 mm) and 86% (3.7 +/- 1.8 mm), respectively, in relation to the control value (26.0 +/- 1.4 mm) (p < 0.000 1). In the NSAID/cholinomimetic-induced lesion model, both EtOH and DCM extracts showed antiulcerogenic effects with significant reduction in the damage to these gastric lesions of 36% (8.3 +/- 2.0 mm) and 42% (7.5 +/- 1.4 mm), respectively, as compared to the control group (13.0 +/- 0.9 mm) (p < 0.0001). In the gastric ulcer induced by hypothermic-restraint stress, both extracts also showed significant activity, and inhibited the gastric lesion index by 58% and 75%, respectively. The EtOH and DCM extracts also changed gastric juice parameters as well as those of cimetidine, decreased gastric acid secretion significantly (p < 0.0001), increased pH values and promoted reduced acid output (p < 0.0001). In all gastric-ulcer-induced models, MeOH extract did not show any significant antiulcerogenic activity, nor did it change gastric-juice parameters (p > 0.05). The results suggest that EtOH and DCM extracts obtained from M. americana possess excellent antisecretory and/or gastrotective effect in all gastric ulcer models. These results suggest that the antiulcerogenic compound(s) present in M. americana may be clustered in the apolar fraction, which will be investigated by our group for the probable mechanisms of action. Plantas da família Guttiferae apresentam diversas atividades biológicas sendo Hypericum o gênero mais importante devido às atividades antidepressiva, antibacteriana e antiviral de algumas espécies. Hypericum connatum, utilizado no sul do Brasil para o tratamento de feridas de boca, demonstrou atividade contra o lentivírus, responsável pela imunodeficiência felina. Objetivo: isolar e identificar as substâncias majoritárias de H. connatum e testar a atividade de extratos obtidos das partes aéreas e raízes e das substâncias isoladas frente a duas cepas do herpesvírus simples tipo 1 (HSV-1). Método: foram obtidos frações nhexano, diclorometano e metanol e extratos bruto, aquosos a diferentes temperaturas e hidro-etanólico das partes aéreas e raízes. As frações n-hexano e metanólica das partes aéreas foram submetidas à coluna cromatográfica para o isolamento de substâncias. Os extratos e substâncias isoladas foram testados frente ao HSV-1, cepas KOS e ATCC-VR733. Determinou-se a concentração máxima não tóxica (CMNT) à célula e a concentração que provoca alteração morfológica em 50% das células (CC50) pela técnica da alteração morfológica celular, utilizando-se células VERO, linhagem ATCC CCL-81. A avaliação da atividade antiviral foi realizada em placas de microtitulação e medida pela inibição do efeito citopático (ECP) provocado pelo vírus. Resultados e Conclusões: da fração n-hexano foi isolado hiperbrasilol B, da fração metanólica foram isolados amentoflavona, hiperosídeo e guaijaveriana, além de um flavonol de estrutura ainda não definida (HCN3). A fração n-hexano e o extrato bruto das raízes inibiram o ECP das cepas KOS e ATCC-VR733. Os demais extratos testados não apresentaram atividade antiviral. Dentre as substâncias analisadas, hiperbrasilol B, amentoflavona e HCN3 foram ativos frente às duas cepas. Os flavonóides hiperosídeo e guaijaverina não apresentaram atividade anti-HSV-1.

O bacuri (Platonia insignis, Mart.) é um dos mais importantes entre os frutos da Amazônia. Todavia, pouco se sabe sobre sua fisiologia pós-colheita, bem como, estádio de maturação, mudanças durante o armazenamento sob condições ambientais e padrão respiratório. Os frutos foram colhidos em três estádios de maturação baseados na coloração do epicarpo: verde-escuro, verde-claro e de vez (50% amarelo), com o objetivo de se determinar as modificações de coloração e padrão respiratório durante armazenamento sob condições de ambiente (25,2 ºC, 75,1 % UR). Os frutos de todos os estádios de maturação apresentaram, a partir do terceiro dia de colheita, comportamento respiratório não climatérico, com os frutos de vez apresentando maior taxa de produção de CO2 até o quarto dia de armazenamento (177,63 mg CO2.kg-1.h-1). O amarelecimento aumentou durante o armazenamento e foi relacionado com a diminuição da luminosidade (L), cromaticidade e ângulo hue. Os frutos de vez podem ser armazenados sob condições de ambiente por até 10 dias sem nenhuma perda da qualidade comercial.

Antifungal activity of natural products has been tested by adapting methods designed for synthetic drugs. In this study, two methods for the determination of antifungal activity of natural products, agar diffusion and broth microdilution, the CLSI reference methods for synthetic drugs, are compared and discussed. The microdilution method was more sensitive. The minimal inhibitory concentrations (MIC) of crude extracts, fractions and pure substances from different species of the plant families Piperaceae, Rubiaceae, Clusiaceae, Fabaceae and Lauraceae, from the Biota project, were determined. Antifungal activities against Candida albicans, C.krusei, C.parapsilosis and Cryptococcus neoformans were produced by several samples.

As atividades antiinflamatória e antinociceptiva do extrato padronizado de Hypericum brasiliense (HBSE) (Guttiferae) foi avaliada em modelos animais. Ratos Wistar machos foram tratados com extrato de H. brasiliense (50, 250 e 500 mg/kg, v.o.) em solução 3% Tween 80 0,9% NaCl. O tratamento com HBSE (500 mg/kg) mostrou inibição significativa sobre o edema induzido por carragenina comparado ao grupo controle. Nessa dose, o edema foi reduzido em 31,25% na terceira hora (pico do edema) após o tratamento, mas na dose de 50 mg/kg, o edema apresentou redução de 53,13% (p < 0,05). Ainda com a dose de 50 mg/kg, a diminuição do edema induzido por dextrana foi similar ao controle positivo, ciproeptadina. Houve diminuição na formação do tecido granulomatoso (6,6%) comparável ao grupo controle. O HBSE também inibiu o número de contorções abdominais em 46,4%, estatisticamente igual ao controle positivo, tratado com indometacina (42,9%). Na dose de 250 mg/kg, houve inibição do número de contorções em 70,7% quando comparado ao grupo controle (p < 0,001). No teste da placa-quente, foi verificado aumento no tempo de latência com a dose de 50 mg/kg. Os resultados demonstram que o HBSE possui atividade antiinflamatória sobre processos agudos, principalmente quando sua gênese está relacionada à síntese dos derivados do ácido araquidônico, e seu efeito analgésico provavelmente envolve ação sobre o Sistema Nervoso Central.

Investigation of carbon isotope fractionation by plants was carried out at two sub-areas located in Reserva Ducke, central Amazonia: open reserve (virgin forest with low density of plant species); and closed reserve (virgin forest with high density of plant species). Preliminary results (δ‰ 13C: 12C values, PDB) of leaf analysis at different plant heights indicate the following: Eschweilera matamata Hub. (Lecythidaceae), common name 'matamata', -31.55±0.61; Protium heptaplyllum March. (Burseraceae), common name 'breu branco', -32.34±1.39; Calophyllum brasiliense Camb. (Guttiferae), common name 'jacareúba', -30.72±0.23; Scleronema micrantthum Ducke. (Bombacaceae), common name 'cardeiro'. -28.81±0.68; and Carapa guianensis Aubl. (Meliaceae), common name 'andiroba', -31.07±0.51. It is possible that the plant species analysed belong to the C3 photosynthetic cycle. In general, the species in the open reserve show differences of the order of 1.66±0.34‰ (greater in 13C) as compared with the same species in the closed reserve. The old leaves show differences in the relative isotopic enrichment (δ) of the order of 1‰, being smaller in new leaves in both reserves. The probable occurrence of an isotopic gradient from the lower (2-5 m) to the upper part (15-20 m) of the plant, of the order of 1.3‰, smaller in 13C, in species from the dense forest was noted. However, only two plants from each species were analysed during a two-year period, data obtained to far are still preliminary, and results should, therefore, be revised. Moreover, according to the literature, the natural carbon isotope fractionation by plants shows metabolic, physiological and environmental dependence. © 1991.

Southern China, especially Yunnan, has undergone high tectonic activity caused by the uplift of Himalayan Mountains during the Neogene, which led to a fast changing palaeogeography. Previous study shows that Southern China has been influenced by the Asian Monsoon since at least the Early Miocene. However, it is yet not well understood how intense the Miocene monsoon system was. In the present study, 63 fossil floras of 16 localities from Southern China are compiled and evaluated for obtaining available information concerning floristic composition, stratigraphic age, sedimentology, etc. Based on such reliable information, selected mega- and micro-floras have been analysed with the coexistence approach to obtain quantitative palaeoclimate data. Visualization of climate results in maps shows a distinct spatial differentiation in Southern China during the Miocene. Higher seasonalities of temperature and precipitation occur in the north and south parts of Southern China, respectively. During the Miocene, most regions of Southern China and Europe were both warm and humid. Central Eurasia was likely to be an arid center, which gradually spread westward and eastward. Our data provide information about Miocene climate patterns in Southern China and about the evolution of these patterns throughout the Miocene, and is also crucial to unravel and understand the climatic signals of global cooling and tectonic uplift.

The Longling Coal Mine (W. Yunnan) is situated in an area of substantial geotectonic activity. Its Late Pliocene palynoflora is of considerable interest, since the area represents a centre of biodiversity. Eighty-two palynomorphs belonging to 61 families were recovered from the lignite. The palynoflora is dominated by angiosperms (68.3%), with ferns (24.4%), gymnosperms (4.9%) and algae (2.4%). Comparisons indicate that most of the palynoflora was derived from the Montane Humid Evergreen Broad-leaved Forest, with lesser contributions from the Tsuga dumosa Forest and Evergreen Coniferous Broad-leaved Mixed Forest, as well as the Montane Mossy Evergreen Broad-leaved Forest. This indicates that the Late Pliocene climate was cooler than that of the present. In the course of the accumulation of the lignite, the climate underwent five major phases of warming and cooling.
