无类囊体蓝藻Gloeobacter violaceus的细胞有厚胶质鞘包裹并形成群体,难以进行遗传操作.用注射器反复吸打的方法将胶质鞘剥落,得到大量单个细胞,通过接合转移将外源广宿主质粒pKT210转入G.violaceus.用斑点杂交方法以及对质粒进行酶切图谱分析,均证实外源质粒pKT210确已导入G.violaceus并稳定复制,未发生结构上的变化.从G.violaceus提取的pKT210可转化mcr和mrr基因缺陷的大肠杆菌DH10B却不能转化mcr+mrr+菌株DH5α,表明G.vio-lace

Gloeobacter violaceus, a cyanobacterium lack of thylakoids, is refractory to genetic manipulations because its cells are enveloped by a thick gelatinous sheath and in colonial form. In this study, a large number of single cells were obtained by repeated pumping with a syringe with the gelatinous sheath removed. And an exogenous broad host range plasmid pKT210 was conjugatively transferred into G. violaceus. Analyses with dot-blot hybridization and restriction mapping showed that the exogenous plasmid pKT210 had been introduced into G. violaceus and stably maintained with no alteration in its structure. pKT210 extracted from G. violaceus exconjugants could be transformed into the mcr - mrr - E. coli strain DH10B but not the mcr(+) mrr(+) strain DH5alpha, which suggests that a methylase system may be present in G. violaceus.

Fatty acid desaturases are enzymes that introduce double bonds into the hydrocarbon chains of fatty acids. The fatty acid desaturases from 37 cyanobacterial genomes were identified and classified based upon their conserved histidine-rich motifs and phylogenetic analysis, which help to determine the amounts and distributions of desaturases in cyanobacterial species. The filamentous or N-2-fixing cyanobacteria usually possess more types of fatty acid desaturases than that of unicellular species. The pathway of acyl-lipid desaturation for unicellular marine cyanobacteria Synechococcus and Prochlorococcus differs from that of other cyanobacteria, indicating different phylogenetic histories of the two genera from other cyanobacteria isolated from freshwater, soil, or symbiont. Strain Gloeobacter violaceus PCC 7421 was isolated from calcareous rock and lacks thylakoid membranes. The types and amounts of desaturases of this strain are distinct to those of other cyanobacteria, reflecting the earliest divergence of it from the cyanobacterial line. Three thermophilic unicellular strains, Thermosynechococcus elongatus BP-1 and two Synechococcus Yellowstone species, lack highly unsaturated fatty acids in lipids and contain only one Delta 9 desaturase in contrast with mesophilic strains, which is probably due to their thermic habitats. Thus, the amounts and types of fatty acid desaturases are various among different cyanobacterial species, which may result from the adaption to environments in evolution. 类胡萝卜素在生物体内具有重要的生理功能,其中虾青素是一类高附加值值的类胡萝卜素。本文通过比较基因组学和实验手段,探索了类胡萝卜素相关基因的转录调控,为类胡萝卜素的代谢工程奠定的了基础。 1、对已测序的18种蓝藻的类胡萝卜素合成基因进行了比较基因组学研究,发现除了Gloeobacter violaceus PCC 7421的类胡萝卜素合成途径是细菌型外,其余的蓝藻类胡萝卜合成途径均属于植物型。序列比对发现蓝藻中参与合成途径上游的酶基因在进化中保守性较高。研究还发现,一些类胡萝卜素合成酶在结构和功能上存在趋同或趋异进化,揭示它们进化上的多样性。 2、 通过比较基因组学分析,发现在集胞藻Synechocystis sp.PCC6803等几种蓝藻中,没有典型的番茄红素环化酶基因的同源基因,而存在着与绿硫细菌中的γ-carotene环化酶基因相似的基因,例如集胞藻中的sll0147和sll0659。但是研究结果显示sll0147和sll0659的突变对番茄红素环化过程影响不明显,提示在这些蓝藻中可能有其他基因参与环化过程,而sll0659基因可能参与了细胞的分裂过程。 3、 从经济绿藻雨生红球藻中克隆了参与类胡萝卜素合成途径的八氢番茄红素合成酶基因(psy)和八氢番茄红素脱氢酶基因(pds)的cDNA和基因组序列。通过基因组步移的方法克隆了这两个基因的5’侧翼序列,以本实验室建立的lacZ为报告基因的瞬间表达体系研究了这两个基因上游区域的启动子活性。 4、 通过ABA、 N饥饿和高光诱导雨生红球藻积累虾青素,利用半定量RT-PCR方法分析了在相同环境因子诱导下雨生红球藻中类胡萝卜素合成基因的表达调控模式,结果显示在虾青素积累过程中,除了番茄红素环化酶基因变化不明显,其他一些基因均存在明显的转录上调。 本研究首次利用比较基因组学的方法分析了类胡萝卜素基因的进化,发现大部分蓝藻的类胡萝卜合成途径属于植物型,一些类胡萝卜合成酶存在结构和功能的多样性。同时分离了雨生红球藻中类胡萝卜素相关基因八氢番茄红素合成酶基因(psy)和八氢番茄红素脱氢酶基因(pds)的5’上游侧翼序列,验证了它们的启动子功能,研究了雨生红球藻虾青素合成酶基因在相同环境因子诱导下的表达调控模式。本文将基础研究与实验验证相结合,为下一步研究类胡萝卜素合成的代谢工程提供了线索。

Apiomithrax violaceus (A. Milne Edwards, 1868 ) is a pisid majoid crab occurring in tropical and subtropical coastal waters of the eastern and western South Atlantic. Larval development consists of two zoeal stages and a megalopa. Beginning with the first zoea, the duration of each larval stage at 24degreesC was 3-8 (5+/-1), 3-5 (4+/-0.5) and 9-15 (11+/-2) days, the megalopa and first crab instar appearing 9-11 (10+/-1) and 20-27 (23+/-2) days after hatching, respectively. Larval characters agree with those proposed for the Majoidea, in having nine or more setae on the scaphognathite in the first zoea and well-developed pleopods in the second zoea. However, larvae of A . violaceus do not fit larval pisid features. Zoeal stages differ from most other Pisidae in having lateral spines, a long rostral spine extending beyond the antenna, two spines per telson fork and a dorsolateral process on the third abdominal somite. The megalopa differs in having a spine dorsally on the carapace and on the basial segment of the second pereiopod. Two characters that are potentially unique to Apiomithrax include a zoeal antenna with an exopod that is much longer than the protopod, and a rostral spine that is longer than the dorsal spine. These characters should facilitate the identification of this taxon and could also be useful for phylogenetic studies. A review of larvae of 28 species among 14 genera indicated that there is no apparent single larval character that differentiates the Pisidae, with more limited phylogenetic analyses suggesting that this is a paraphyletic group. Apiomithrax , Eurynolambrus , Pisoides , Rochinia and Scyra have the most divergent morphological characters within the family. The analysis and inclusion of additional taxa is likely to shed more light on the sister-group relationships of the Pisidae. However, based on the extent of morphological interspecific variability of known larvae it is likely that the group, as presently defined by adult morphology, is not monophyletic.

Neste trabalho, os frutos e sementes de P. violaceus foram descritos morfoanatômica e ontogeneticamente, visando a verificar a origem da ala pericárpica e checar a ocorrência de poliembrionia, previamente descrita para outros gêneros da tribo Dalbergieae. Observouse que o fruto é uma sâmara circular, de aspecto glabro e que a semente possui alguns caracteres típicos de Faboideae, especialmente os relacionados ao hilo. Foram caracterizados atomicamente seis estádios de desenvolvimento. A ala pericárpica origina-se da parede ovariana, por extensões dorso-ventral, apical e basal, produzindo uma estrutura achatada. Não foi possível encontrar poliembrionia, mesmo analisando-se grande número de sementes. Pôde-se também concluir que, no que se refere à estrutura do fruto, P. violaceus é filogeneticamente derivada com relação às outras espécies de Dalbergieae já registradas na literatura, devido à presença de fusão dos feixes ventrais do carpelo.

Adult male Satin Bowerbirds build and decorate stick bowers to which they attract females for matings; females choose among males based on these complex bowers, decorations placed at these bowers, and displays consisting of vocalisations and posturing. Male Satin Bowerbirds undergo an extended period of delayed morphological maturation during which they retain female-like plumage and are assumed to learn adult male behavioural traits. Little is known, however, of how immature males acquire the ability to display and build and decorate bowers, except that they observe the displays of adult males at adults' bowers, and practise their own displays at both adults' and 'practice' bowers. We present data on the home ranges and movement patterns of six immature males, acquired through radio-tracking at the Bunya Mountains in south-east Queensland. Home-range size averaged 13.67 +/- 3.38 ha and immature males visited only some of the bowers located in their home ranges. On average, they visited 2.33 +/- 0.52 adults' bowers and 4.00 +/- 2.00 practice bowers.

Geographic variation in the advertisement call of the male Satin Bowerbird, Ptilonorhynchus violaceus, was investigated in three populations in south-eastern Queensland. The call was found to differ significantly among the three geographically distinct populations. A discriminant function analysis using five measurements of call frequency and duration provided 100% classification success of the 25 individuals. The observed geographic variation in this call may result from adaptation to the local acoustic environment in these populations, or from genetic or cultural divergence among populations. Further research involving the acoustic properties of the habitats, population genetics and a larger number of populations is required to fully understand this pattern of call variation.

We isolated 13 polymorphic microsatellite markers from the satin bowerbird, Ptilonorhynchus violaceus from a genomic library enriched in (AAGG)(n) repetitive elements and characterized them in 20 individuals. The number of alleles ranged from two to 18 per locus with the observed heterozygosity ranging from 0.15 to 1.00. These markers will be useful for analysing questions concerning parentage, population genetic structure and models of speciation.

Australian wet forests have undergone a contraction in range since the mid-Tertiary, resulting in a fragmented distribution along the east Australian coast incorporating several biogeographical barriers. Variation in mitochondrial DNA and morphology within the satin bowerbird was used to examine biogeographical structure throughout almost the entire geographical extent of these wet forest fragments. We used several genetic analysis techniques, nested clade and barrier analyses, that use patterns inherent in the data to describe the spatial structuring. We also examined the validity of the two previously described satin bowerbird subspecies that are separated by well-defined biogeographical barriers and tested existing hypotheses that propose divergence occurs within each subspecies across two other barriers, the Black Mountain corridor and the Hunter Valley. Our data showed that the two subspecies were genetically and morphologically divergent. The northern subspecies, found in the Wet Tropics region of Queensland, showed little divergence across the Black Mountain corridor, a barrier found to be significant in other Wet Tropics species. Biogeographical structure was found through southeastern Australia; three geographically isolated populations showed genetic differentiation, although minimal divergence was found across the proposed Hunter Valley barrier. A novel barrier was found separating inland and coastal populations in southern New South Wales. Little morphological divergence was observed within subspecies, bar a trend for birds to be larger in the more southerly parts of the species' range. The results from both novel and well-established genetic analyses were similar, providing greater confidence in the conclusions about spatial divergence and supporting the validity of these new techniques.

Geographic variation in vocalizations is widespread in passerine birds, but its origins and maintenance remain unclear. One hypothesis to explain this variation is that it is associated with geographic isolation among populations and therefore should follow a vicariant pattern similar to that typically found in neutral genetic markers. Alternatively, if environmental selection strongly influences vocalizations, then genetic divergence and vocal divergence may be disassociated. This study compared genetic divergence derived from 11 microsatellite markers with a metric of phenotypic divergence derived from male bower advertisement calls. Data were obtained from 16 populations throughout the entire distribution of the satin bowerbird, an Australian wet-forest-restricted passerine. There was no relationship between call divergence and genetic divergence, similar to most other studies on birds with learned vocalizations. Genetic divergence followed a vicariant model of evolution, with the differentiation of isolated populations and isolation-by-distance among continuous populations. Previous work on Ptilonorhynchus violaceus has shown that advertisement call structure is strongly influenced by the acoustic environment of different habitats. Divergence in vocalizations among genetically related populations in different habitats indicates that satin bowerbirds match their vocalizations to the environment in which they live, despite the homogenizing influence of gene flow. In combination with convergence of vocalizations among genetically divergent populations occurring in the same habitat, this shows the overriding importance that habitat-related selection can have on the establishment and maintenance of variation in vocalizations.

Satin bowerbirds Ptilonorhynchus violaceus have an elaborate multi-component sexual display, some components of which have been extensively studied. We describe a relatively unstudied component of this display, bower painting, and birds' responses to manipulations of their paint. Males of this species focus their display around a stick bower constructed on the forest floor which they decorate with a variety of objects and paint. Painting involves a male masticating plant material and wiping the plant-saliva mixture onto the inside walls of the bower; during courtship visits to bowers, females nibble at this paint. We found that 93% of 53 males painted their bowers at our study site and the time males spent painting their bowers accounted for 24% of their time at the bower. We experimentally removed and added paint to bowers to test whether males respond to these changes in their paint. Males gave more advertisement calls and spent less time manipulating sticks at the bower when we added fresh wet paint to their bowers compared to older dried paint or a control treatment. They did not respond to the removal of paint from their bowers, perhaps because it was primarily older dried paint that was removed. We also found that males painted more frequently when there was measurable wind in their bowers, which could have degraded the quality of the signal. Our findings indicate that fresh wet paint is more important to males than older dried paint and, together with previous work at this site, suggest that paint may act as a signal to females. Given that females nibble bower sticks during courtship, we suggest that bower paint may function as a chemical sexual signal rather than a visual signal.

Male Satin Bowerbirds (Ptilonorhynchus violaceus) build stick structures known as bowers that serve as the focus for courtships and matings. Males decorate their bowers with numerous coloured decorations and are known to steal these decorations from one another. We investigated the stealing of bower decorations among males at the Bunya Mountains in Queensland, Australia. We aimed to (1) determine which classes of decorations were targets for theft in the studied population, and (2) examine whether the frequency at which individual decorations were stolen related to their intrinsic properties. To address our first aim, all decorations on the bowers of 21 adult males were labelled and their movements tracked throughout one mating season. To address our second aim, decorations stolen at least three times during the season were collected and their morphological and reflectance properties compared to those of decorations that were not stolen. In terms of the classes of decorations, tail feathers of Crimson Rosella (Platycercus elegans) were stolen more than any other class of decoration, but blue plastic bottletops were the most popular decorations relative to their availability on bowers. Frequently stolen individual decorations were similar to non-stolen items in their weights and surface areas, but were darker blue in colour than the decorations never stolen. Both bottletops and feathers reflected higher levels of ultraviolet (UV) light than did all other classes of bower decorations tested, thus suggesting that males may be using UV reflectance in sexual signalling. The darker blue, stolen decorations may increase contrast between the decoration collection and the platform, while the UV-reflecting subset of most frequently stolen decorations (bottletops and feathers) may increase contrast within the decoration collection. This in turn may increase the attractiveness of the display to females.
