Tainting of outdoor pond-reared barramundi Lates calcarifer by muddy-earthy off-flavours is frequently reported across tropical Australia. To investigate the possible causes and effects of off-flavour tainting, we analysed water samples from outdoor rearing ponds for the presence of geosmin (GSM) and 2-methylisoborneol (2-MIB), 2 microbial metabolites often associated with tainting episodes. We then conducted controlled dose-effect experiments which measured the accumulation of tainting metabolites in the flesh, and the impact tainting had on taste and flavour attributes. GSM was deemed to be the compound most likely responsible for off-flavour tainting, persisting at moderate (similar to 1.00 mu g l(-1)) to extreme levels (similar to 14.36 mu g l(-1)), while 2-MIB was never detected during the study. Controlled experiments revealed that the accumulation of GSM in the flesh of market-sized barramundi was directly related to GSM levels of the holding water (0 to similar to 4 mu g l(-1)), with higher levels resulting in significant increases in undesirable taste and flavour attributes, particularly muddy-earthy flavour and weedy aftertaste. We identified the sensory detection threshold for GSM in farmed barramundi to be <= 0.74 mu g kg(-1), similar to estimates for GSM detection in rainbow trout Oncorhynchus mykiss (similar to 0.9 mu g kg(-1)) and for 2-MIB in channel catfish Ictalurus punctatus (0.7 mu g kg(-1)). Quantitative estimation of flesh-bound GSM using gas chromatography-mass spectrometry (GC-MS) agreed well with human sensory assessment scores and highlights the reliability of chemical analysis of GSM in barramundi flesh while also indicating the value of GC-MS analysis in predicting the impact of GSM on the sensory properties of farmed barramundi.

The effects of temperature and light on the growth and geosmin production of Lyngbya kuetzingii were determined. Of the three temperatures tested, 10, 25 and 35A degrees C, the maximal geosmin concentration and geosmin productivity were yielded at 10A degrees C, while the highest chl a production was observed at 25A degrees C. In the studies on light intensity, the maximal geosmin concentration and geosmin productivity were observed at 10 mu mol m(-2) s(-1), while the highest chl a production was at 20 mu mol m(-2) s(-1). It was suggested that more geosmin was synthesized with lower chl a demand. Meanwhile, the relative amounts of extra- and intracellular geosmin were investigated. Under optimum growth conditions (20 mu mol m(-2) s(-1), 25A degrees C; BG-11 medium), the amounts of extracellular geosmin increased as the growth progressed and reached the maximum in the stationary phase, while the intracellular geosmin reached its maximum value in the late exponential phase, and then began to decline. However, under the low temperature (10A degrees C) or light (10 mu mol m(-2) s(-1)) conditions, more intracellular geosmin was synthesized and mainly accumulated in the cells. The proportions of extracellular geosmin were high, to 33.33 and 32.27%, respectively, during the stationary phase at 35A degrees C and 20 mu mol m(-2) s(-1). It was indicated that low temperature or light could stimulate geosmin production and favor the accumulation of geosmin in cells, while more intracellular geosmin may be released into the medium at higher temperatures or optimum light intensity.

We have previously reported the effectiveness of TiO2 photocatalysis in the destruction of species generated by cyanobacteria, specifically geosmin and microcystin-LR. In this paper we report an investigation of factors which influence the rate of the toxin destruction at the catalyst surface. A primary kinetic solvent isotope effect of approximately 1.5 was observed when the destruction was performed in a heavy water solvent. This is in contrast to previous reports of a solvent isotope effect of approximately 3, however, these studies were undertaken with a different photocatalyst material. The solvent isotope effect therefore appears to be dependent on the photocatalyst material used. The results of the study support the theory that the photocatalytic decomposition occurs on the catalyst surface rather than in the bulk of the solution. Furthermore it appears that the rate determining step is not oxygen reduction as previously reported.

Geosmin is produced by cyanobacteria and actinomycetes in surface waters. It causes undesirable earthy off-flavours in freshwater fish and is a major concern for the drinking water industry. This paper presents the first published study on the use of the novel pelleted Ti02 photocatalyst, Hombikat K01/C, for the removal of geosmin from water. Ti02 in pelleted form eliminates the requirement for the separation of the catalyst from the water following treatment which is normally the case with the widely used powdered catalysts. A laboratory reactor was designed to limit system loss since the compound adsorbs to a wide range of surfaces. Initial concentration, aeration rate and irradiation were evaluated. It was found that degradation of geosmin followed the Langmuir-Hinshelwood model. Elevated aeration had no effect on the photocatalytic removal of geosmin, but increasing irradiation was found to increase degradation rates. The catalyst proved effective within 10 min under optimum conditions.

Geosmin (GSM) and 2-methylisoborneol (MIB) are semi-volatile compounds produced by cyanobacteria in surface waters. These compounds present problems to the drinking water industry and in aquaculture because they can taint water and fish producing an earthy-musty flavour. This paper presents an initial study on the use of TiO2 photocatalysis for the destruction of these compounds in water. The process proved effective with the complete destruction of MIB and GSM being achieved within 60 min. These results suggest that TiO2 photocatalysis will be a successful method for removal of taint compounds from potable water supplies and fish farms.

Taste and odour compounds, especially geosmin (GSM) and 2-methylisoborneol (2-MIB), cause major problems in both drinking water and aquaculture industries world-wide. Aquaculture in particular has experienced significant financial losses due to the accumulation of taint compounds prior to harvest resulting in consumer rejection. UV-TiO2 photocatalysis has been demonstrated to remove GSM and 2-MIB at laboratory scale but the development of a continuous flow reactor suitable for use in water treatment has not been investigated. In this study, a pilot packed bed photocatalytic reactor was developed and evaluated for water treatment with both laboratory and naturally tainted samples. A significant reduction of both 2-MIB and GSM was achieved in both trials using the packed bed reactor unit. With the laboratory spiked water (100ngL-1 of each compound added prior to treatment), detectable levels were reduced by up to 97% after a single pass through the unit. When the reactor was used to treat water in a fish farm where both compounds were being produced in situ (2-MIB: 19ngL-1 and GSM: 14ngL-1) a reduction of almost 90% in taint compounds was achieved. These very encouraging promising results demonstrate the potential of this UV-TiO2 photocatalytic reactor for water treatment in fish rearing systems and other applications.

2-Methylisoborneol (MIB) and geosmin (GSM) are sub products from algae decomposition and, depending on their concentration, can be toxic: otherwise, they give unpleasant taste and odor to water. For water treatment companies it is important to constantly monitor their presence in the distributed water and avoid further costumer complaints. Lower-cost and easy-to-read instrumentation would be very promising in this regard. In this study, we evaluate the potentiality of an electronic tongue (ET) system based on non-specific polymeric sensors and impedance measurements in monitoring MIB and GSM in water samples. Principal component analysis (PCA) applied to the generated data matrix indicated that this ET was capable to perform with remarkable reproducibility the discrimination of these two contaminants in either distilled or tap water, in concentrations as low as 25 ng L-1. Nonetheless, this analysis methodology was rather qualitative and laborious, and the outputs it provided were greatly subjective. Also, data analysis based on PCA severely restricts automation of the measuring system or its use by non-specialized operators. To circumvent these drawbacks, a fuzzy controller was designed to quantitatively perform sample classification while providing outputs in simpler data charts. For instance, the ET along with the referred fuzzy controller performed with a 100% hit rate the quantification of MIB and GSM samples in distilled and tap water. The hit rate could be read directly from the plot. The lower cost of these polymeric sensors allied to the especial features of the fuzzy controller (easiness on programming and numerical outputs) provided initial requirements for developing an automated ET system to monitor odorant species in water production and distribution. The production of certain odorous metabolites is an undesirable attribute of cyanobacteria (blue-green algae) growth in aquaculture ponds [e.g., channel catfish(Ictalurus punctatus)] and in drinking water reservoirs. The most common odorous compounds encountered in catfish aquaculture are geosmin (trans-1,10-dimethyltrans-9-decalol) and 2-methylisoborneol(exo-1,2,7,7-tetramethylbicyclo[2.2.1]heptan-2-ol). These compounds are also frequently encountered worldwide in reservoirs and aqueducts used for municipal drinking water systems(Schrader et al. 2002). In this study, several algicides were evaluated using a rapid bioassay to determine their effectiveness in controlling the MIB-producing cyanobacterium Oscillatoria perornata from a west Mississippi catfish pond and the MIBproducing Pseudanabaena sp. (strain LW397) from Lake Whitehurst, Virginia, used as a city water supply reservoir. The cyanobacterium Oscillatoria agardhii , not a MIB-producer, and the green alga Selenastrum capricornutum , found in catfish ponds in the southeastern United States, were included in the bioassay to help determine potential broad-spectrum toxicity of the commercial products. The three Biesbosch Reservoirs are pumped storage reservoirs, fed with rather polluted and highly eutrophic water from the River Meuse. Air injection at the bottom of the reservoirs prevents thermal stratification, which would otherwise result in serious water quality deterioration. Reservoir mixing also serves as an economic algal control measure; mixing over sufficient depth causes light to play the role of limiting factor and this, combined with zooplankton grazing, keeps the biomass of phytoplankton at acceptable levels. Special problems are caused by benthic, geosmin-producing Oscillatoria species growing on the inner embankment. Rooting up the bottom with a harrow is used as the method of control, based on underwater observations by biological staff trained as SCUBA-divers. With regard to pollutant behaviour the three reservoirs act as a series of fully mixed reactors. This enables the application of kinetic models to describe their behaviour and allows the use of a selective intake policy, e.g. for suspended solids with associated contaminants, ammonia and polynuclear aromatic hydrocarbons. A combination of selective intake and self- purification processes - enhanced by the compartmentalisation of the storage volume in three reservoirs - leads to a striking improvement for many water-quality parameters.

采用微波蒸馏-顶空固相微萃取-气质联用检测鱼体中常见的两种土霉味化合物,即2-甲基异茨醇(2-MIB)和土腥素(Geosmin)。研究并优化了微波蒸馏萃取过程的关键参数(微波蒸馏时间、载气流量),结果表明微波蒸馏6min、载气流70mL/min为土霉味化合物微波蒸馏萃取的最佳条件。在此优化的条件下,土霉味化合物能够充分地从鱼体中蒸馏出来,再采用顶空固相微萃取的方法使馏分中的土霉味化合物吸附于纤维涂层上,将其在250℃高温下解吸,并用GC-MS分析。基于此测定方法,鱼肉中2-甲基异茨醇和土腥素的检测限均达到

The presence of the odorous compounds, 2-methylisoborneol (MIB) and geosmin, as well as causative microorganisms in brackish intensive cultivation fishponds in Tianjin, China that had a severe earthy-musty odor were evaluated. The results revealed that MIB was the primary odorous compound present in the Tianjin fishponds, with a concentration ranging from 0.53-5302.7 ng.L-1. Furthermore, the concentration of MIB was found to be closely correlated with the gross biomass of actinomycetes in the water, which ranged from 10.67-1528.24 x 10(6) cfu.ml(-1). Therefore, the sequences of the 16 SrRNA and morphological characteristics of the actinomycetes in the brackish fishponds were investigated. The results revealed that the actinomycetes in the brackish fishponds included 9 species of common and dominant actinomycetes belonging to 4 genera. Of these genera, Streptomyces were the dominant species, and Streptomyces, Nocardioides and Micromonospora were the most common species in the fishponds evaluated. Next, the ability of each of the isolated Streptomyces to produce MIB was measured under laboratory culture conditions. Streptomyces Sp2 was found to have a strong ability to produce MIB, which indicates that this strain may be the primary source of the earthy-musty odor reported in brackish intensive cultivation fishponds in Tianjin, China.

Three major odorous compounds are 2-methylisoborneol ( 2-MIB), geosmin and beta-cyclocitral, which in water were determined by coupling headspace solid-phase microextraction ( HS-SPME) with gas chromatography-mass spectrometry (GC-MS). The operating conditions of HS-SPME, such as fibre type, salt concentration, water temperature, stirring, absorption time and desorption time were studied and discussed.The highest absorption of the odorous compounds were obtained under the following operating conditions as the addition of 30% ( m/V) NaCl, stirring at 60 degrees C for 40 min, using 65 mu m polydimethyl siloxane/divinylbenzene coated fibre. After the odorous compounds had been absorbed in the fibre under the optimal conditions of HSSPME, they were desorbed at 250 degrees C and determined by GC-MS. The limits of detection for geosmin, beta-cyclocitral and 2-MIB in water were 1. 0, 1. 3, 1. 7 ng/L, and the relative standard deviations for them were 4. 9%, 8. 4%, 6. 2%,respectively. There were good linear correlation (the calibration coefficients were all above 0. 997) for the three odorous compounds in the range of 5 similar to 1000 ng/ L. Therefore, trace levels of the odorous compounds at ng/L in water could be quantified by the simple method with satisfactory result.

Les proliférations nuisibles de la cyanobactérie filamenteuse benthique Lyngbya wollei qui forme des tapis déposés sur les sédiments ont augmenté en fréquence au cours des 30 dernières années dans les rivières, lacs et sources de l'Amérique du Nord. Lyngbya wollei produit des neurotoxines et des composés organiques volatils (géosmin, 2-méthylisobornéol) qui ont des répercussions sur la santé publique de même que des impacts d'ordre socioéconomiques. Cette cyanobactérie est considérée comme un habitat et une source de nourriture de piètre qualité pour les invertébrés en raison de sa gaine robuste et de sa production de toxines. Les proliférations de L. wollei ont été observées pour la première fois en 2005 dans le fleuve Saint-Laurent (SLR; Québec, Canada). Nous avons jugé important de déterminer sa distribution sur un tronçon de 250 km afin d'élaborer des modèles prédictifs de sa présence et biomasse en se basant sur les caractéristiques chimiques et physiques de l'eau. Lyngbya wollei était généralement observé en aval de la confluence de petits tributaires qui irriguent des terres agricoles. L’écoulement d’eaux enrichies à travers la végétation submergée se traduisait par une diminution de la concentration d’azote inorganique dissous (DIN), alors que les concentrations de carbone organique dissous (DOC) et de phosphore total dissous (TDP) demeuraient élevées, produisant un faible rapport DIN :TDP. Selon nos modèles, DOC (effet positif), TP (effet négatif) et DIN :TDP (effet négatif) sont les variables les plus importantes pour expliquer la répartition de cette cyanobactérie. La probabilité que L. wollei soit présent dans le SLR a été prédite avec exactitude dans 72 % à 92 % des cas pour un ensemble de données indépendantes. Nous avons ensuite examiné si les conditions hydrodynamiques, c'est-à-dire le courant généré par les vagues et l'écoulement du fleuve, contrôlent les variations spatiales et temporelles de biomasse de L. wollei dans un grand système fluvial. Nous avons mesuré la biomasse de L. wollei ainsi que les variables chimiques, physiques et météorologiques durant trois ans à 10 sites le long d'un gradient d'exposition au courant et au vent dans un grand (148 km2) lac fluvial du SLR. L'exposition aux vagues et la vitesse du courant contrôlaient les variations de biomasses spatiales et temporelles. La biomasse augmentait de mai à novembre et persistait durant l'hiver. Les variations interannuelles étaient contrôlées par l'écoulement de la rivière (niveau d'eau) avec la crue printanière qui délogeait les tapis de l'année précédente. Les baisses du niveau d'eau et l'augmentation de l'intensité des tempêtes anticipées par les scénarios de changements climatiques pourraient accroître la superficie colonisée par L. wollei de même que son accumulation sur les berges. Par la suite, nous avons évalué l'importance relative de L. wollei par rapport aux macrophytes et aux épiphytes. Nous avons examiné l'influence structurante de l'échelle spatiale sur les variables environnementales et la biomasse de ces producteurs primaires (PP) benthiques. Nous avons testé si leur biomasse reflétait la nature des agrégats d'habitat basées sur l'écogéomorphologie ou plutôt le continuum fluvial. Pour répondre à ces deux questions, nous avons utilisé un design à 3 échelles spatiales dans le SLR: 1) le long d'un tronçon de 250 km, 2) entre les lacs fluviaux localisés dans ce tronçon, 3) à l'intérieur de chaque lac fluvial. Les facteurs environnementaux (conductivité et TP) et la structure spatiale expliquent 59% de la variation de biomasse des trois PP benthiques. Spécifiquement, les variations de biomasses étaient le mieux expliquées par la conductivité (+) pour les macrophytes, par le ratio DIN:TDP (+) et le coefficient d'extinction lumineuse (+) pour les épiphytes et par le DOC (+) et le NH4+ (-) pour L. wollei. La structure spatiale à l'intérieur des lacs fluviaux était la plus importante composante spatiale pour tous les PP benthiques, suggérant que les effets locaux tels que l'enrichissement par les tributaire plutôt que les gradients amont-aval déterminent la biomasse de PP benthiques. Donc, la dynamique des agrégats d'habitat représente un cadre général adéquat pour expliquer les variations spatiales et la grande variété de conditions environnementales supportant des organismes aquatiques dans les grands fleuves. Enfin, nous avons étudié le rôle écologique des tapis de L. wollei dans les écosystèmes aquatiques, en particulier comme source de nourriture et refuge pour l'amphipode Gammarus fasciatus. Nous avons offert aux amphipodes un choix entre des tapis de L. wollei et soit des chlorophytes filamenteuses ou un tapis artificiel de laine acrylique lors d'expériences en laboratoire. Nous avons aussi reconstitué la diète in situ des amphipodes à l'aide du mixing model (d13C et δ15N). Gammarus fasciatus choisissait le substrat offrant le meilleur refuge face à la lumière (Acrylique>Lyngbya=Rhizoclonium>Spirogyra). La présence de saxitoxines, la composition élémentaire des tissus et l'abondance des épiphytes n'ont eu aucun effet sur le choix de substrat. Lyngbya wollei et ses épiphytes constituaient 36 et 24 % de l'alimentation in situ de G. fasciatus alors que les chlorophytes, les macrophytes et les épiphytes associées représentaient une fraction moins importante de son alimentation. Les tapis de cyanobactéries benthiques devraient être considérés comme un bon refuge et une source de nourriture pour les petits invertébrés omnivores tels que les amphipodes.

La incidència sobre el suro de defectes sensorials amb descriptors de florit-humitat és un dels principals problemes amb què s'enfronta la indústria surera. Els principals compostos relacionats amb aquest defecte son cloroanisoles, metoxipirazines, geosmina i metilisoborneol. Aquesta recerca s'ha fet amb l'objectiu principal d'aprofundir en els mètodes de determinació de compostos volàtils en suro. S'ha desenvolupat una metodologia basada en la microextracció en fase sòlida i la cromatografia de gasos per a l'anàlisi conjunta d'aquests compostos en macerats de suro. El fet que la majoria dels compostos d'interès tingui un origen microbià ha fet necessari completar les anàlisis químiques amb mètodes de detecció i aïllament dels microorganismes susceptibles de produir-los. S'ha utilitzat un mètode per a l'estudi de la diversitat de fongs i bacteris en mostres de suro basat en l'estudi de marcadors genètics mitjançant l'anàlisi en gradients electroforètics desnaturalitzants. Per últim s'ha avaluat la capacitat de producció de defectes sensorials per microorganismes aïllats de suro utilitzant tècniques olfactomètriques.
