The main goal of this work was to develop a simple analytical method for quantification of glycerol based on the electrocatalytic oxidation of glycerol on the copper surface adapted in a flow injection system. Under optimal experimental conditions, the peak current response increases linearly with glycerol concentration over the range 60-3200 mg kg(-1) (equivalent to 3-160 mg L(-1) in solution). The repeatability of the electrode response in the flow injection analysis (FIA) configuration was evaluated as 5% (n = 10), and the detection limit of the method was estimated to be 5 mg kg(-1) in biodiesel (equivalent to 250 mu g L(-1) in solution) (S/N = 3). The sample throughput under optimised conditions was estimated to be 90 h(-1). Different types of biodiesel samples (B100), as in the types of vegetable oils or animal fats used to produce the fuels, were analysed (seven samples). The only sample pre-treatment used was an extraction of glycerol from the biodiesel sample containing a ratio of 5 mL of water to 250 mg of biodiesel. The proposed method improves the analytical parameters obtained by other electroanalytical methods for quantification of glycerol in biodiesel samples, and its accuracy was evaluated using a spike-and-recovery assay, where all the biodiesel samples used obtained admissible values according to the Association of Official Analytical Chemists. No presente trabalho estudou-se a produção de ésteres etílicos de ácido graxo de Ricinus communis L. através da tranesterificação alcalina do óleo de mamona com etanol. Esta metodologia foi adotada para determinar as melhores condições para a produção de biodiesel a partir de óleo de mamona usando o mínimo de operações unitárias com benefícios do ponto de vista econômico e de produção de efluentes. Para a obtenção dos ésteres etílicos através do processo de transesterificação (etapa 1) utilizou-se como catalisador 1% de NaOH com etanol em uma razão molar de 6:1 seguido da adição de ácido sulfúrico. Após, a reação de esterificação (etapa 2) dos ácidos graxos contidos no biodiesel foi realizada visando reduzir o índice de acidez da amostra, ficando em torno de 2 mg de KOH/g. A quebra in situ dos sabões (provenientes da reação paralela de saponificação do triglicerídeo) pela adição de ácido sulfúrico ao meio reacional foi bem sucedida melhorando a separação dos FAEEs do glicerol. O processo em duas etapas transesterificação/esterificação apresentou boa conversão para os ésteres etílicos, diminuindo o índice de acidez e atingindo as especificações para glicerina total e livre. O biodiesel proveniente do óleo de mamona foi composto de 90,6% ácido ricinoléico (C18:1, OH), 3,2% ácido oléico (C18:1), 4,5% ácido linoléico (C18:2), 0,7% ácido esteárico (C18:0), 1,0% ácido palmítico (C16:0), triacilgliceróis (TGs, 0%), diacilgliceróis (DGs, 0,37%) monoacilgliceróis (MGs, 0,46%) e glicerol livre (0,25%) após o processo em duas etapas transesterificação/esterificação. O processo em duas etapas foi muito importante para determinar a integralidade da reação no rendimento do produto. Os resultados demonstram que o procedimento desenvolvido para a produção de FAEEs em escala de laboratório pode ser escalonado para uma planta piloto.

A utilização do óleo de mamona como matéria-prima para produção de biodiesel mostra-se inviável na combustão interna do motor devido sua natureza química incomum que lhe confere especificações acima do permitido nas especificações técnicas da legislação nacional brasileira. Desta forma, a blenda com óleo de arroz refinado (OM:OA) qualifica o biodiesel atendendo a legislação nacional, além de corrigir a elevada acidez do óleo bruto de mamona prejudicial ao processo de transesterificação homogênea básica. No presente estudo realizou-se a produção de biodiesel etílico em escala piloto a partir de blendas de óleo bruto de mamona e óleo refinado de arroz em dois processos: o primeiro processo adotando a mistura direta dos dois óleos e o segundo processo pela esterificação antecipada do óleo bruto de mamona. Ambos os processos foram aplicados visando tanto definir os critérios de processo (acidez inicial) quanto o cumprimento das especificações técnicas (viscosidade e densidade). A produção em escala piloto (200 litros/batelada) foi realizada na Usina Demonstrativa para Produção de Biodiesel – BIOSUL (Edital FINEP, 2005) da Universidade Federal do Rio Grande - FURG utilizando em ambos os processos transesterificação, com hidróxido de sódio, e esterificação com ácido sulfúrico. Os processos apresentaram resultados satisfatórios, sendo o processo de mistura direta (Processo A) o que obteve melhores rendimentos (94,04%, blenda 20:80) enquanto que o processo de pré- esterificação (Processo B) foi aquele que proporcionou a maior fração de óleo de mamona na blenda (80,36%, 33:67). Os resultados para o Processo A de glicerol livre, monoacilgliceróis, diacilgliceróis, triacilgliceróis e de glicerol total foram, respectivamente, de 1,322 %, 6,092 %, 1,000 %, 0,884 e 3,152%. Neste estudo foi comprovada a viabilidade do processamento, em batelada, de blendas dos óleos de mamona e arroz. O óleo de mamona bruto pode ser utilizado em até 30% produzindo biodiesel dentro da legislação, verificando-se assim a viabilidade do uso da mamona na produção de biocombustíveis.

No presente trabalho foi investigada a transesterificação de blendas dos óleos de soja e de tungue com metanol ou etanol empregando catalisador alcalino (NaOH ou KOH). Foi investigado o tempo reacional, a proporção da blenda, a concentração e o tipo de catalisador, tipo de álcool e razão molar, temperatura e metodologia empregada no tratamento da reação. Nas reações com metanol obtiveram-se melhores conversões com tempo reacional de 1,5h; temperatura de 60°C; proporção blenda dos óleos de soja e de tungue de 90:10 (m/m); concentração de NaOH de 0,5% em relação a massa da blenda e razão molar metanol:blenda de 6:1. O tratamento dos ésteres metílicos produzidos na reação foi realizado por lavagem com água a 60°C após o processo de decantação das fases, metodologia C. O rendimento de ésteres metílicos foi superior a 96% e, o teor de mono-, di- e triacilglicerídeos, glicerol livre e total ficou abaixo dos limites estabelecidos pela ANP, indicando boa conversão (> 96,5%). Nas reações com etanol verificou-se que as melhores condições reacionais foram com uma concentração de catalisador de 0,8% de NaOH em relação a massa da blenda, razão molar etanol:blenda de 9:1, tempo de 1,5h e temperatura de 60°C. O tratamento dos produtos da reação foi realizado por lavagem com água a 60°C após o processo de remoção do etanol e decantação das fases, metodologia D. A concentração do catalisador foi um fator determinante na separação das fases. Uma maior concentração de catalisador favorece a saponificação, dificultando a separação das fases e afetando o rendimento do biodiesel sintetizado, tanto para o metílico quanto o etílico. O índice de acidez, tanto para o biodiesel metílico como o etílico, para qualquer proporção da blenda dos óleos de soja e tungue, ficaram dentro das normas da ANP, com valores abaixo de 0,5 mg.g-1 de KOH.

Glass transition and relaxation of the glycerol-water (G-W) binary mixture system have been studied over the glycerol concentration range of 5-85 mol% by using the highly sensitive technique of electron spin resonance (ESR). For the water rich mixture the glass transition,sensed by the dissolved spin probe, arises from the vitrified mesoscopic portion of the binary system. The concentration dependence of the glass transition temperature manifests a closely related molecular level cooperativity in the system. A drastic change in the mesoscopic structure of the system at the critical concentration of 40 mol is confirmed by an estimation of the spin probe effective volume in a temperature range where the tracer reorientation is strongly coupled to the system dynamics.

We report an efficient one-pot conversion of glycerol (GLY) to methyl lactate (MLACT) in methanol in good yields (73 % at 95 % GLY conversion) by using Au nanoparticles on commercially available ultra-stable zeolite-Y (USY) as the catalyst (160 °C, air, 47 bar pressure, 0.25 M GLY, GLY-to-Au mol ratio of 1407, 10 h). The best results were obtained with zeolite USY-600, a catalyst that has both Lewis and Brønsted sites. This methodology provides a direct chemo-catalytic route for the synthesis of MLACT from GLY. MLACT is stable under the reaction conditions, and the Au/USY catalyst was recycled without a decrease in the activity and selectivity. From glycerol to green building blocks and solvents! An efficient, base-free conversion of glycerol to methyl lactate in methanol is reported, achieving good yields (73 % at 95 % glycerol conversion) using Au/ultra-stable zeolite-Y (USY) as the catalyst and environmentally benign oxygen as the oxidant by combining two separate reaction steps efficiently in a one pot procedure. The base free oxidation of glycerol with molecular oxygen in water using bimetallic Au-Pt catalysts on three different acidic zeolite supports (H-mordenite, H-β and H-USY) was explored in a batch setup. At temperatures between 140 and 180 °C, lactic acid formation was significant and highest selectivity (60 % lactic acid at 80 % glycerol conversion) was obtained using Au-Pt/USY-600 (180 °C). A selectivity switch to glyceric acid (GLYA) was observed when the reactions were performed at 100 °C. Highest conversion and selectivity towards GLYA were obtained with Au-Pt/H-β as the catalyst (68 % selectivity at 68 % conversion).

An Arthrobacter species (tentatively identified as A. citreus), isolated by the enrichment culture method with glycerol as the sole source of carbon, was studied with a view to elucidate its pathway of glycerol breakdown. Evidence has been obtained against the functioning of the phosphorylative pathway by the study of (1) oxygen uptake with phosphorylated intermediates, (2) uptake of inorganic phosphorus by intact resting cells, (3) action of inhibitors like sodium fluoride, sodium azide, sodium arsenite, sodium iodoacetate, and parachloromercurybenzoate on oxygen uptake with resting cell suspensions and cell-free extracts in some cases. Evidence presented for the functioning of a non-phosphorylative pathway includes studies on the oxidation of glycerol, D-glyceraldehyde, glycerate, glycolic aldehyde, glycolic acid, glyoxylic acid, and formic acid to carbon dioxide and water. Further, the possibility of glyoxylate metabolism through the tricarboxylic acid cycle by its formation of malate was shown. The significance of the above pathway is that it has pointed to an alternative route of carbohydrate metabolism and entry into the tricaboxylic acid cycle without the intervention of pyruvate or the condensing enzyme.

Major facilitators represent the largest superfamily of secondary active transporter proteins and catalyze the transport of an enormous variety of small solute molecules across biological membranes. However, individual superfamily members, although they may be architecturally similar, exhibit strict specificity toward the substrates they transport. The structural basis of this specificity is poorly understood. A member of the major facilitator superfamily is the glycerol-3-phosphate (G3P) transporter (GlpT) from the Escherichia coli inner membrane. GlpT is an antiporter that transports G3P into the cell in exchange for inorganic phosphate (Pi). By combining large-scale molecular-dynamics simulations, mutagenesis, substrate-binding affinity, and transport activity assays on GlpT, we were able to identify key amino acid residues that confer substrate specificity upon this protein. Our studies suggest that only a few amino acid residues that line the transporter lumen act as specificity determinants. Whereas R45, K80, H165, and, to a lesser extent Y38, Y42, and Y76 contribute to recognition of both free Pi and the phosphate moiety of G3P, the residues N162, Y266, and Y393 function in recognition of only the glycerol moiety of G3P. It is the latter interactions that give the transporter a higher affinity to G3P over Pi.

This study was developed with the purpose to investigate the effect of polysaccharide/plasticiser concentration on the microstructure and molecular dynamics of polymeric film systems, using transmission electron microscope imaging (TEM) and nuclear magnetic resonance (NMR) techniques. Experiments were carried out in chitosan/glycerol films prepared with solutions of different composition. The films obtained after drying and equilibration were characterised in terms of composition, thickness and water activity. Results show that glycerol quantities used in film forming solutions were responsible for films composition; while polymer/total plasticiser ratio in the solution determined the thickness (and thus structure) of the films. These results were confirmed by TEM. NMR allowed understanding the films molecular rearrangement. Two different behaviours for the two components analysed, water and glycerol were observed: the first is predominantly moving free in the matrix, while glycerol is mainly bounded to the chitosan chain. The aim of the present study was to investigate the participation of the sympathetic nervous system (SNS) in the control of glycerol-3-P (G3P) generating pathways in white adipose tissue (WAT) of rats in three situations in which the plasma insulin levels are low. WAT from 48 h fasted animals, 3 day-streptozotocin diabetic animals and high-protein, carbohydrate-free (HP) diet-fed rats was surgical denervated and the G3P generation pathways were evaluated. Food deprivation, diabetes and the HP diet provoke a marked decrease in the rate of glucose uptake and glycerokinase (GyK) activity, but a significant increase in the glyceroneogenesis, estimated by the phosphoenolpyruvate carboxykinase (PEPCK) activity and the incorporation of 1-[C-14]-pyruvate into glycerol-TAG. The denervation provokes a reduction (similar to 70%) in the NE content of WAT in fasted, diabetic and HP diet-fed rats. The denervation induced an increase in WAT glucose uptake of fed, fasted, diabetic and HP diet-fed rats (40%, 60%, 3.2 fold and 35%, respectively). TAG-glycerol synthesis from pyruvate was reduced by denervation in adipocytes of fed (58%) and fasted (36%), saline-treated (58%) and diabetic (23%), and HP diet-fed rats (11%). In these same groups the denervation reduced the PEPCK mRNA expression (75%-95%) and the PEPCK activity (35%-60%). The denervation caused a similar to 35% decrease in GyK activity of control rats and a further similar to 35% reduction in the already low enzyme activity of fasted, diabetic and HP diet-fed rats. These data suggest that the SNS plays an important role in modulating G3P generating pathways in WAT, in situations where insulin levels are low. (C) 2012 Elsevier Inc. All rights reserved.
