The neurodegeneration and amyloid deposition of sporadic Alzheimer disease (AD) also occur in familial AD and in all trisomy-21 Down syndrome (DS) patients, suggesting a common pathogenetic mechanism. We investigated whether defective processing of damaged DNA might be that mechanism, as postulated for the neurodegeneration in xeroderma pigmentosum, a disease with defective repair not only of UV radiation-induced, but also of some oxygen free radical-induced, DNA lesions. We irradiated AD and DS skin fibroblasts or blood lymphocytes with fluorescent light, which is known to cause free radical-induced DNA damage. The cells were then treated with either beta-cytosine arabinoside (araC) or caffeine, and chromatid breaks were quantified. At least 28 of 31 normal donors and 10 of 11 donors with nonamyloid neurodegenerations gave normal test results. All 12 DS, 11 sporadic AD, and 16 familial AD patients tested had abnormal araC and caffeine tests, as did XP-A cells. In one of our four AD families, an abnormal caffeine test was found in all 10 afflicted individuals (including 3 asymptomatic when their skin biopsies were obtained) and in 8 of 11 offspring at a 50% risk for AD. Our tests could prove useful in predicting inheritance of familial AD and in supporting, or rendering unlikely, the diagnosis of sporadic AD in patients suspected of having the disease.

Nitric oxide (NO) in NTS plays an important role in regulating autonomic function to the cardiovascular system. Using the fluorescent dye DAF-2 DA, we evaluated the NO concentration in NTS. Brainstem slices of rats were loaded with DAF-2 DA, washed, fixed in paraformaldehyde and examined under fluorescent light. In different experimental groups, NTS slices were pre-incubated with 1 mM L-NAME (a non-selective NOS inhibitor), 1 MM D-NAME (an inactive enantiomere of L-NAME), 1 mM kynurenic acid (a nonselective ionotropic receptors antagonist) or 20 mu M bicuculline (a selective GABA(A) receptors antagonist) before and during DAF-2 DA loading. Images were acquired using a confocal microscope and the intensity of fluorescence was quantified in three antero-posterior NTS regions. In addition, slices previously loaded with DAF-2 DA were incubated with NeuN or GFAP antibody. A semi-quantitative analysis of the fluorescence intensity showed that the basal NO concentration was similar in all antero-posterior aspects of the NTS (rostral intermediate, 15.5 +/- 0.8 AU: caudal intermediate, 13.2 +/- 1.4 AU; caudal commissural, 13.8 +/- 1.4 AU, n = 10). In addition, the inhibition of NOS and the antagonism of glutamatergic receptors decreased the NO fluorescence in the NTS. On the other hand, D-NAME did not affect the NO fluorescence and the antagonism of GABAA receptors increased the NO fluorescence in the NTS. It is important to note that the fluorescence for NO was detected mainly in neurons. These data show that the fluorescence observed after NTS loading with DAF-2 DA is a result of NO present in the NTS and support the concept that NTS neurons have basal NO production which is modulated by L-glutamate and GABA. New emerging contaminants could represent a danger to the environment and Humanity with repercussions not yet known. One of the major worldwide pharmaceutical and personal care productions are antimicrobials products, triclosan, is an antimicrobial agent present in most products. Despite the high removal rate of triclosan present in wastewater treatments, triclosan levels are on the rise in the environment through disposal of wastewater effluent and use of sewage sludge in land application. Regulated in the EC/1272/2008 (annex VI, table 3.1), this compound is considered very toxic to aquatic life and it has been reported that photochemical transformation of triclosan produces dioxins. In the current work it was defined three objectives; determination of the most efficient process in triclosan degradation, recurring to photochemical degradation methods comparing different sources of light; identification of the main by-products formed during the degradation and the study of the influence of the Fenton and photo-Fenton reaction. Photochemical degradation methods such as: photocatalysis under florescent light (UV), photocatalysis under visible light (sunlight), photocatalysis under LEDs, photo-Fenton and Fenton reaction have been compared in this work. The degradation of triclosan was visualized through gas chromatography/mass spectrometry (GC/MS). In this study photo-Fenton reaction has successfully oxidized triclosan to H2O and CO2 without any by-products within 2 hours. Photocatalysis by titanium dioxide (TiO2) under LEDs was possible, having a degradation rate of 53% in an 8 hours essay. The degradation rate of the Fenton reaction, UV light and sunlight showed degradation between 90% and 95%. The results are reported to the data observed without statistic support, since this was not possible during the work period. Hydroquinone specie and 2,4-dichlorophenol by-products were identified in the first hour of photocatalysis by UV. A common compound, possibly identified has C7O4H , was present at the degradation by UV, sunlight and LEDs and was concluded to be a contaminant. In the future more studies in the use of LEDs should be undertaken given the advantages of long durability and low consumption of energy of these lamps and that due to their negative impact on the environment fluorescent lamps are being progressively made unavailable by governments, requiring new solutions to be found. Fenton and photo-Fenton reactions can also be costly processes given the expensive reagents used.

Broiler production is highly dependent on the use of artificial light. The light source may affect the effectiveness of housing conditions due to increasing ambient temperature and concentration of noxious gases. This research aimed to evaluate the effects of different bulb types on the thermal, aerial, and acoustic environment of broiler aviaries. The experiment was carried out at a commercial broiler farm in Dourados, Mato Grosso do Sul State, Brazil. Three aviaries were used, and two flocks of male broilers from Cobb® genetic strain were reared from the first day to slaughter. Each aviary was equipped with a different light source, comprising the adopted treatments (A1 - incandescent light bulb, control; A2 - sodium vapor light bulb; A3 - fluorescent light bulb). The aviaries were divided into nine quadrants, and the environmental data (ambient dry bulb temperature and relative humidity), litter surface temperature, CO2 and NH3 concentrations, and bird sound pressure behavior were recorded in each quadrant. The aviary with incandescent light presented higher air and litter temperatures, and concentration of gases than the other tested alternatives. It also presented higher level of sound pressure in the second week of the growing period; however, from this period up to slaughter, there was no effect of the light source on the results of broiler sound pressure level.

Cloudy apple juice has been found to develop off-flavors during storage in daylight. The development of off-flavors and volatile compounds was monitored in reconstituted juice prepared from 'Golden Delicious' and 'Fuji' apple concentrates stored in glass bottles under fluorescent light (3000 Ix, 8 degrees C). A strong metallic off-flavor was formed by photooxidation. A major contributor to the off-flavor was identified as 1-octen-3-one by gas chromatography-olfactometry. In addition, six volatile compounds, pentanal, 2-methyl-1-penten-3-one, hexanal, (E)-2-heptenal, 6-methyl-5-hepten-2-one, and (E)-2octenal, increased significantly after light exposure and could contribute to the off-flavor. Except for pentanal and hexanal, these volatiles were found only after light exposure. Higher levels of volatiles were observed in juice from 'Golden Delicious' apples than in juice from 'Fuji' apples, and this difference was consistent with higher levels of suspended solids. When the suspended solids were removed by centrifugation, the development of volatiles on exposure to light was reduced significantly.

The purpose of this study was to investigate lymphatic clearance of the human skin in patients with acute deep thrombosis of the femoral vein. In 13 patients with deep vein thrombosis and no other cause for swelling of the limbs, lymphatic clearance of the skin at the foot was measured. Ten microliters of fluorescein isothiocyanatedextran 150,000 were injected intradermally and the fluorescent light intensity of the deposit measured 10 min and 24 hours after injection by window densitometry. In addition, intralymphatic pressure was measured by the servo-nulling system. The results were compared with a sex- and age-matched control group. Fluorescent light intensity decreased by 23.8 +/- 12.3 arbitrary units or by a factor of 1.8 +/- 0.5 in patients with DVT after 24 hours, which was significantly less than in healthy controls (33.7 +/- 8.9 arbitrary units or by factor 5.0 +/- 4.1, p < 0.013). Intralymphatic pressure was not different between the two groups. These results indicate that lymphatic clearance is significantly reduced in the acute phase of deep venous thrombosis.

Two experimental fishing trials were carried out off the coast of Pernambuco, Brazil, in 1999 and 2001, using a small artisanal longliner. In experiment 1, six-hook baskets with three chemical light-sticks on alternating hooks had significantly higher catch rates than those with zero or with a light-stick on every hook, with most swordfish accounted for by hooks with light-sticks. Analysis of the data from experiment 2 showed no significant difference between electralume attractors, consisting of AA lithium batteries protected by a solid cover and light-sticks that produce a fluorescent light when two chemical products are mixed. Significant differences were detected in mean CPUE by size class, with most swordfish belonging to class 'b' (125-170 cm lower jaw to fork length (LJFL)). No differences, however, were found for swordfish catches in classes 'a' (< 125 cm LJFL) and V, and no evidence was found of interaction between the two factors (attractor and size class). Although there was no significant difference between the total length-frequency distributions of swordfish caught with light-sticks and electralume attractors, signibcant differences were found for fish smaller than 125 cm LJFL, with electralume catches consisting of smaller swordfish than those of gear using light-sticks. Tässä työssä arvioidaan kotitalouksien energiansäästöpotentiaalia valaistuksen osalta Suomessa. Euroopan Unioni on ratifioinut päästövähennystavoitteen 20 % koskien kaikkia primäärienergiantuotantomuotoja. Päästövähennystavoite on tarkoitus toteuttaa vuoteen 2020 mennessä. Tässä kandidaatintyössä arvioidaan saavutettavissa olevaa energiansäästöpotentiaalia Suomessa kotitalouksissa laitesähkön osalta. Energiansäästöpotentiaalin arviointi perustuu hehkulampun korvaamismahdollisuuksien vertailuun. Valaistuksessa voidaan saavuttaa jopa 75 % sähkönsäästö, kun korvataan yksi 60 W hehkulamppu vastaavan valomäärän tuottavalla energiansäästölampulla. Kokonaisuudessa kotitalouksien valaistuksessa on tehostamispotentiaalia noin 60 %. Valaistuksen tehostamisella saavutetaan noin 1,5 TWh säästö Suomen kokonaissähkökulutuksessa. Saavutettavat säästöt voivat olla jopa suuremmatkin, jos käytetään lisäksi älykästä valonohjausta. Johtopäätöksenä voidaan todeta, että tehostamispotentiaalia on merkittävästi ja tehostaminen on yksittäiselle kotitaloudelle taloudellisesti kannattavaa. Energiansäästöpotentiaalin toteuttamisen vaikutuksia kansantalouteen ei ole arvioitu.

Guava nectars were formulated for approximately 10, 12, or 14 ºBrix, with 40% guava pulp. Sodium benzoate, 500 mg.kg-1 was used as preservative. The Brix value was adjusted with saturated sucrose syrup. The guava nectar was pasteurized (85 ºC/42 seconds) in tubular heat exchanger and then hot filled in 500 mL white glass bottles. The products were stored either at room temperature (25 ± 5 ºC) or refrigerated (5 ± 2 ºC) under fluorescent light exposure and analyzed on the day after processing (time zero) and also 40, 80, and 120 days of storage. Eight compounds were identified and quantified by Gas Chromatography (GC) -Mass Spectrometry (MS): hexanal, (E)-hex-2-enal, 1-hexenol, (Z)-hex-3-enol, (Z)-hex-3-enyl acetate, phenyl-3-propyl acetate, cinnamyl acetate, and acetic acid. There was no significant effect of thermal treatment on the volatile compound concentrations, except for a significant decrease (p = 0.0001) in hexanal and (Z)-hex-3-enyl acetate (p = 0.0029). As for the storage time, there was a much greater decrease in the esters contents, such as (Z)-hex-3-enyl and phenyl-3-propyl acetates. Cinnamyl acetate had the greatest decrease over storage time. Refrigeration was better than room temperature for guava nectar volatile compounds stability over storage time, mainly for esters compounds, which are important for the product aroma and flavor
