The main analytical variables of a modified Folin-Ciocalteu method were studied by UV-Vis and gradient HPLC-PDA methods, using purified (PC) and technical grade (TGC) casein. Rutin and an aqueous extract of Psidium guajava L. leaves were used as models. The best results were ascribed to TGC. Certainly PC bonds the polyphenols of the P. guajava extract better than TGC, but TGC afforded better precision. A lack of specificity occurred when rutin was analyzed together with the P guajava extract. Additional analyses performed with the flavonoid fraction of P. guajava extract by HPLC-PDA had confirmed that casein was able to bind catechin, gallic acid and P. guajava flavonoids in a non-specific way.

The infusion of aerial parts of Ilex paraguariensis is widely consumed. Its antioxidant activity suggests an important role of this plant in the treatment/prevention of oxidative stress related diseases. Plant extract active compounds are frequently found in esterified form that may be poorly absorbed. Hydrolysis of the extract is a possible approach to increase its bioavailability. The aim of this study was to perform a phytochemical analysis and evaluate in rats the plasma concentration and tissue distribution of antioxidant compounds in the hydroethanolic extract of Ilex paraguariensis, before and after enzymatic hydrolysis. Both extracts presented high antioxidant activity and phenolic content. Rats given single or repeated doses of the hydrolyzed extract showed increased plasma antioxidant activity and higher plasma levels of caffeic acid. However, no changes of endogenous antioxidants were observed. In conclusion, hydrolysis of the extract of Ilex paraguariensis is a strategy to improve its bioavailability and in vivo antioxidant activity.

The total phenolic and flavonoid content, color, and antioxidant activity were evaluated from ten honey samples from Apis mellifera L. collected from Roraima State, Brazil. The total phenolic content was determined using the Folin-Ciocalteu reagent and ranged from 250 to 548 mg gallic acid kg-1 of honey. The total flavonoid content was obtained using two methods: total flavones/flavonols were determined with aluminum chloride, and flavonones/dihydroflavonols were determined with 2,4-dinitrophenylhydrazine. The results ranged from 9 to 48.6 mg of quercetine kg-1 of honey and 1805 to 2606 mg of pinocembrin kg-1 of honey, respectively. Antioxidant activity was measured with 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavengers, and the results of the IC50 ranged from 3.17 to 8.79 mg/mL. Statistical analysis demonstrated positive correlations between color intensity, flavone and flavonol content, and phenolic content and negative correlations with antioxidant capacity.

A new method is presented for spectrophotometric determination of total polyphenols content in wine. The procedure is a modified CUPRAC method based on the reduction of Cu(II), in hydroethanolic medium (pH 7.0) in the presence of neocuproine (2,9-dimethyl-1,10-phenanthroline), by polyphenols, yielding a Cu(I) complexes with maximum absorption peak at 450 nm. The absorbance values are linear (r = 0.998, n = 6) with tannic acid concentrations from 0.4 to 3.6 mu mol L(-1). The limit of detection obtained was 0.41 mu mol L(-1) and relative standard deviation 1.2% (1 mu mol L(-1); n = 8). Recoveries between 80% and 110% (mean value of 95%) were calculated for total polyphenols determination in 14 commercials and 2 synthetic wine samples (with and without sulphite). The proposed procedure is about 1.5 more sensitive than the official Folin-Ciocalteu method. The sensitivities of both methods were compared by the analytical responses of several polyphenols tested in each method. (C) 2010 Elsevier Ltd. All rights reserved.

The antioxidant activity of aqueous extracts of five edible tropical fruits (Spondias lutea, Hancornia speciosa, Spondias purpurea, Manilkara zapota and Averrhoa carambola) was investigated using different methods. The amount of phenolic compounds was determined by the Folin-Ciocalteu reagent. The M. zapota had Total Antioxidant Capacity (TAC) higher than the other fruits. Extracts showed neither reducing power nor iron chelation (between 0.01 and 2.0 mg/mL). H. speciosa exhibited the highest superoxide scavenging activity (80%, 0.5 mg/mL). However, at high concentrations (8.0 mg/mL) only A. carambola, S. purpurea and S. lutea scavenging 100% of radicals formed. M. zapota and S. purpurea had higher phenolic compound levels and greater OH radical scavenging activity (92 %, 2.0 mg/mL). Antiproliferative activity was assessed with 3T3 fibroblasts and cervical tumor cells (HeLa). The most potent extract was S. purpurea (0.5 mg/mL), which inhibited HeLa cell proliferation by 52%. The most fruits showed antioxidant and antiproliferative properties, characterizing them as functional foods.

Cu(II)/neocuproine (2,9-dimethyl-1,10-phenanthroline) complexes were utilized for spectrophotometric determination of total polyphenol content in beers. This procedure is based on the reduction of Cu(II) by polyphenols in hydroethanolic medium (pH 7.0) in the presence of neocuproine, yielding Cu(I)/complexes with maximum absorption at 454 nm. The sensitivity of the proposed method was compared with the AOAC method using tannic and gallic acid as standards. The average apparent molar absorptivity, in L cm(-1) mol(-1), of tannic acid (3.50 +/- 0.20) x 10(5) and gallic acid (5.12 +/- 0.21) x 10(4), was twice as high for the proposed method. A lower limit of detection (LOD) (2.9 x 10(-1) mg L-1) was found when tannic acid was used in the proposed method. Additionally, less interference from the most common additives in beers was noticed. Total content of polyphenols was analyzed in 17 Brazilian samples. Results ranged from 35.5 to 556 mg L-1 of tannic acid, with higher values for recovery rates (45.4-118%, mean 85.0%) than for gallic acid. Although tannic acid is a mixture of polygalloyl glucoses, the total polyphenol content found in the samples suggests that tannic acid should be used as a standard. A flow system designed with solenoid micro-pumps is introduced for spectrophotometric determination of total tannins based on the Folin- Denis reaction. The procedure minimizes the main drawbacks related to the AOAC batch procedure, i.e. interferences from reducing species in the samples, high reagent consumption and waste generation, and low sampling rate. Linear response was observed for tannic acid concentrations in the range 2-100 mg L-1, with a detection limit (99.7% confidence level) of 0.3 mg L-1. The sampling rate and coefficient of variation (n = 10) were estimated as 75 measurements per hour and 1.1%, respectively. Results of determination of total tannin in tea, beer and wine samples were in agreement with those achieved by the batch reference procedure at the 95% confidence level. In comparison to the batch procedure, the reagent consumption and effluent generation were 83 and 60-fold lower, respectively.

The reaction follows Beer's law in the range 0-50 μg of 11-desoxycorticosterone. 3. Determinations made in human urine and adrenal glands of rats and guinea pigs are comparable with results obtained by other methods.

Three simple, sensitive, economical and reproducible spectrophotometric methods (A, B and C) are described for determination of mesalamine in pure drug as well as in tablet dosage forms. Method A is based on the reduction of tungstate and/or molybdate in Folin Ciocalteu's reagent; method B describes the reaction between the diazotized drug and α-naphthol and method C is based on the reaction of the drug with vanillin, in acidic medium. Under optimum conditions, mesalamine could be quantified in the concentration ranges, 1-30, 1-15 and 2-30 µg mL-1 by method A, B and C, respectively. All the methods have been applied to the determination of mesalamine in tablet dosage forms. Results of analysis are validated statistically.

Neste trabalho foi estudado um subproduto derivado da indústria agroalimentar produtora de sumo concentrado de maçã, conhecido por bagaço de maçã, com o objetivo de avaliar condições de extração de compostos fenólicos, o teor de compostos fenólicos totais, flavonóides e proantocianidinas e ainda a atividade antioxidante. Foram efetuadas extrações a partir do bagaço de maçã variando as condições de tempo, temperatura, razão massa:volume e solvente e os extratos obtidos avaliados quanto ao seu teor em compostos fenólicos totais pelo método FolinCiocalteu. O extrato aquoso do bagaço de maçã para uma temperatura de 100 ºC a um tempo de 2x4h e concentração de 50 mg/mL, apresentou o teor de compostos fenólicos mais elevado (9,37 mgEAG/g de bagaço de maçã, na base seca) em relação a todas as outras temperaturas, tempos de extração e solventes utilizados, como etanol (50% e 70%) e metanol. O doseamento de flavonóides totais baseou-se no método espetrofotométrico, usando o reagente cloreto de alumínio e a rutina como padrão. Os melhores resultados foram obtidos usando etanol (70%) como solvente à temperatura ambiente, cerca de 4,35 mgER/g. A amostra extraída com água apresentou valores bastante similares ao etanol, cerca de 4,27 mgER/g, usando uma temperatura de 100 ºC durante 2x4h. O conteúdo em proantocianidinas foi determinado pelo método 4-dimetilamino cinamaldeído (DMAC). O bagaço de maçã estudado demonstrou ser pobre no seu conteúdo de proantocianidinas, obtendo valores de 0,77 mgEEC/g. A atividade antioxidante do bagaço de maçã foi avaliada através de dois métodos distintos: 2,2-difenil-1-picril-hidrazilo (DPPH∙) e método do poder redutor (FRAP). O extrato aquoso obtido a 100 ºC a um tempo de 2x4h, demonstrou ser aquele com maior potencial, com uma capacidade antioxidante mais elevada que os restantes extratos, com valores de IC50 de 0,48 mg/mL e 0,65 mg/mL, para os métodos de DPPH∙ e FRAP, respetivamente.

Processed tea and herbal infusions were Studied for their phenol content, antioxidant activity and main flavonoids. Total phenolics were determined by Folin-Ciocalteu method and ranged from N.D. to 46.46 +/- 0.44 mg/g GAE. Flavonoids were investigated by HPLC, and myricetin, quercetin, kaempferol were identified in black, green, and chamomile tea. Antioxidant activity was evaluated using two methods: DPPH and beta-carotene bleaching test (BCB). Using the BCB, the highest activities were found for infusions of black tea, mate, lemongrass, chamomile, and fennel; whereas fresh herbal infusions presented the lowest activities. Using the DPPH method, fresh herbal infusions presented the highest activities. Processed leaves with the lowest IC50 values were green and black tea (147.63 and 288.60 mu g/mL, respectively). The results of this research show that the infusions studied are good Source of compounds presenting antioxidant activity in vitro.

Samples of 15 second generation soy-based products (n = 3), commercially available, were analyzed for their protein and isoflavone contents and in vitro antioxidant activity, by means of the Folin-Ciocalteu reducing ability, DPPH radical scavenging capacity, and oxygen radical absorbance capacity. Isoflavone identification and quantification were performed by high-performance liquid chromatography. Products containing soy and/or soy-based ingredients represent important sources of protein in addition to the low fat amounts. However, a large variation in isoflavone content and in vitro antioxidant capacity was observed. The isoflavone content varied from 2.4 to 18.1 mg/100 g (FW), and soy kibe and soy sausage presented the highest amounts. Chocolate had the highest antioxidant capacity, but this fact was probably associated with the addition of cocoa liquor, a well-known source of polyphenolics. This study showed that the soy-based foods do not present a significant content of isoflavones when compared with the grain, and their in vitro antioxidant capacity is not related with these compounds but rather to the presence of other phenolics and synthetic antioxidants, such as sodium erythorbate. However, they may represent alternative sources and provide soy protein, isoflavones, and vegetable fat for those who are not ready to eat traditional soy foods.

The antioxidant capacity of propolis from the southern region of Uruguay was evaluated using in vitro as well as cellular assays. Free radical scavenging capacity was assessed by ORAC, obtaining values significantly higher than those of other natural products (8000 mu mol Trolox equiv/g propolis). ORAC values correlated well with total polyphenol content (determined by Folin-Ciocalteu method) and UV absorption. Total polyphenol content (150 mg gallic acid equiv/g propolis) and flavonoids (45 mg quercetin equiv/g propolis) were similar to values reported for southern Brazilian (group 3) and Argentinean propolis. Flavonoid composition determined by RP-HPLC indicates a strong poplar-tree origin. Samples high in polyphenols efficiently inhibit low-density lipoprotein lipoperoxidation and tyrosine nitration. In addition, Uruguayan propolis was found to induce the expression of endothelial nitric oxide synthase and inhibit endothelial NADPH oxidase, suggesting a potential cardiovascular benefit by increasing nitric oxide bioavailability in the endothelium.

RESUMOA batata-doce (Ipomoea batatas L.) é um vegetal muito cultivado nos países tropicais, usado na alimentação humana e na animal, sendo uma boa fonte de energia, sais minerais e vitaminas. Constitui, ainda, fonte de polifenóis antioxidativos, entre eles, antocianinas e ácidos fenólicos. O objetivo deste estudo foi avaliar a atividade antibacteriana de diferentes extratos de folhas frescas de batata-doce, frente a bactérias de interesse alimentar, e correlacioná-la com os teores de polifenóis totais e de antocianinas. Foram avaliados teores de polifenóis totais e de antocianinas, bem como a atividade antibacteriana de extratos alcoólicos, de infusão e de decocção de folhas de dois acessos de batata-doce, em Porto Alegre, RS, frente a três agentes de interesse em alimentos e alimentação, quais sejam, inóculos padronizados de SalmonellaEnteritidis (ATCC 11076), Staphylococcus aureus(ATCC 25923) e Escherichia coli (ATCC 11229). A avaliação da atividade antibacteriana, expressa em intensidade de atividade de inibição bacteriana (IINIB) e em intensidade de atividade de inativação bacteriana (IINAB), foi baseada em testes de diluição em sistema de tubos múltiplos; as antocianinas foram determinadas por meio de pH diferencial e os polifenóis pelo método de Folin & Ciocalteu. Os extratos alcoólicos inibiram e, ou, inativaram todos os inóculos testados, não obstante o Staphylococcus aureus ter sido significativamente mais resistente. Os extratos obtidos por decocção e infusão, além de apresentarem concentrações muito baixas de antocianinas e menores teores de polifenóis do que os seus correspondentes alcoólicos, não apresentaram atividade antibacteriana. A atividade antibacteriana dos extratos testados foi positivamente correlacionada com os compostos fenólicos e, ou, antocianinas.
