Introduction: The aim of this study was to assess the occurrence of apical root transportation after the use of Pro Taper Universal rotary files sizes 3 (F3) and 4 (F4). Methods: Instruments were worked to the apex of the original canal, always by the same operator. Digital subtraction radiography images were produced in buccolingual and mesiodistal projections. A total of 25 radiographs were taken from root canals of human maxillary first molars with curvatures varying from 23-31 degrees. Quantitative data were analyzed by intraclass correlation coefficient and Wilcoxon nonparametric test (P = .05). Results: Buccolingual images revealed a significantly higher degree of apical transportation associated with F4 instruments when compared with F3 instruments in relation to the original canal (Wilcoxon test, P = .007). No significant difference was observed in mesiodistal images (P = .492). Conclusions: F3 instruments should be used with care in curved canals, and F4 instruments should be avoided in apical third preparation of curved canals. (J Endod 2010;36:1052-1055)

Com o objetivo de selecionar progênies de cafeeiros produtivos e resistentes à ferrugem foi instalado e conduzido um experimento em Machado,MG. Foram avaliadas 29 progênies selecionadas pelo programa de melhoramento genético do cafeeiro, coordenado pela EPAMIG, obtidas a partir do cruzamento Icatu e Catimor. O cultivar Rubi MG 1192 foi utilizado como testemunha. O delineamento experimental adotado foi o de blocos casualizados, com três repetições. Foram analisados a produção de café beneficiado em sc.ha-1 de seis colheitas, 2001/2002 a 2006/2007; o vigor vegetativo avaliado após a colheita 2006/2007; e a incidência e severidade da ferrugem no primeiro semestre de 2006. Observouse variabilidade para produção e resistência à ferrugem, resultados confirmados pelas estimativas da herdabilidade de boa magnitude, variando de 58,5% para a produção a 92,6% para a característica incidência da ferrugem. Esses resultados, aliados às boas médias de produção, à baixa incidência e severidade da ferrugem ao bom vigor vegetativo de algumas progênies, demonstram que é possível selecionar progênies superiores com base na produção e resistência à ferrugem.

A espécie Coffea arabica L é cultivada em todas as regiões cafeeiras do Estado de Minas Gerais, com predominância dos cultivares Catuaí e Mundo Novo, por seu vigor vegetativo, alto potencial produtivo e boa qualidade de bebida. Visando a identificar e selecionar progênies de cafeeiro com boas características agronômicas, foi instalado e conduzido um experimento na Fazenda Experimental da EPAMIG, no município de Três Pontas, MG. Foram avaliadas 39 progênies na 4ª geração de autofecundação, após o 2º retrocruzamento entre 'Catuaí' e 'Mundo Novo' (genitor recorrente), desenvolvidas pelo programa de melhoramento genético da Epamig. Os cultivares Catuaí Vermelho MG 99, Rubi MG 1192 e Acaiá Cerrado MG 1474 foram utilizados como testemunhas. Foram analisadas as características: produção de café beneficiado em sc.ha-1 de oito safras (1998/1999 a 2005/2006), vigor vegetativo, percentagem de frutos chochos e classificação quanto à peneira. Ficou evidente que existe variabilidade genética dentro do grupo de progênies estudadas. As maiores produtividades foram encontradas na sétima e oitava colheita. As progênies 1189-9-80-1 e 1189-9-80-3 apresentaram melhor desempenho, em todas as características avaliadas e serão selecionadas para futuros trabalhos de melhoramento genético, sendo lançadas como cultivar ou utilizadas em hibridações.

AIMS: Experimental autoimmune myocarditis (EAM) model mirrors important mechanisms of inflammatory dilated cardiomyopathy (iDCM). In EAM, inflammatory CD133(+) progenitors are a major cellular source of cardiac myofibroblasts in the post-inflammatory myocardium. We hypothesized that exogenous delivery of macrophage-colony-stimulating factor (M-CSF) can stimulate macrophage lineage differentiation of inflammatory progenitors and, therefore, prevent their naturally occurring myofibroblast fate in EAM. METHODS AND RESULTS: EAM was induced in wild-type (BALB/c) and nitric oxide synthase 2-deficient (Nos2(-/-)) mice and CD133(+) progenitors were isolated from inflamed hearts. In vitro, M-CSF converted inflammatory CD133(+) progenitors into nitric oxide-producing F4/80(+) macrophages and prevented transforming growth factor-β-mediated myofibroblast differentiation. Importantly, only a subset of heart-infiltrating CD133(+) progenitors expresses macrophage-specific antigen F4/80 in EAM. These CD133(+)/F4/80(hi) cells show impaired myofibrogenic potential compared with CD133(+)/F4/80(-) cells. M-CSF treatment of wild-type mice with EAM at the peak of disease markedly increased CD133(+)/F4/80(hi) cells in the myocardium, and CD133(+) progenitors isolated from M-CSF-treated mice failed to differentiate into myofibroblasts. In contrast, M-CSF was not effective in converting CD133(+) progenitors from inflamed hearts of Nos2(-/-) mice into macrophages, and M-CSF treatment did not result in increased CD133(+)/F4/80(hi) cell population in hearts of Nos2(-/-) mice. Accordingly, M-CSF prevented post-inflammatory fibrosis and left ventricular dysfunction in wild-type but not in Nos2(-/-) mice. CONCLUSION: Active and NOS2-dependent induction of macrophage lineage differentiation abrogates the myofibrogenic potential of heart-infiltrating CD133(+) progenitors. Modulating the in vivo differentiation fate of specific progenitors might become a novel approach for the treatment of inflammatory heart diseases.

Rationale: Experimental autoimmune myocarditis (EAM) mirrors important pathogenic aspects of inflammatory cardiomyopathy, a common cause of heart failure. In EAM, TGF-β-dependent conversion of heart-infiltrating prominin-1+ progenitors into myofibroblasts is critical for development of fibrosis and the end-stage heart failure phenotype. Therapeutic strategies modulating the in vivo fate of prominin-1+ progenitors might therefore prevent TGF-β-mediated cardiac fibrosis and pathological remodelling. Methods and Results: EAM was induced in BALB/c mice using alpha-myosin heavy chain (aMyHC) peptide/complete Freund's adjuvant (CFA) immunization. Prominin-1+ cells were isolated from the inflamed hearts at day 21 after immunization, expanded and treated with Macrophage Colony-Stimulating Factor (M-CSF) or Transforming Growth Factor-beta (TGF-β). Herein, we demonstrated that M-CSF turns, ex vivo and in the EAM, heart-infiltrating prominin-1+ progenitors into immunosuppressive F4/80/CD11b/CD16/32/NOS2-expressing, nitric oxide producing and E.coli bacteria phygocyting macrophages, and protect further TGF-β-stimulated differentiation into pathogenic myofibroblasts. Systemic M-CSF treatment during myocarditis completely prevented post-inflammatory fibrosis, T cell relapse and left ventricular dysfunction. Mechanistically, M-CSF-induced macrophage differentiation from prominin-1+ progenitors critically required nitric oxide synthase 2. Accordingly, M-CSF treatment failed to reduce myocardial fibrosis development in Nos2-/- mice. Conclusions: Altering the in vivo fate of inflammatory prominin-1 expressing progenitors from pro-fibrotic into the F4/80 expressing macrophage phenotype protects from myocarditis progression, cardiac fibrosis, and heart failure. These findings offer a modern therapeutic model and challenge former concepts, which attributed macrophages a detrimental role in inflammatory cardiomyopathy progression.

O objetivo deste trabalho foi identificar progênies de feijoeiro (Phaseolus vulgaris) na geração F4 provenientes do cruzamento entre cultivares resistentes ao Bean golden mosaic virus (BGMV). Os parentais foram as cultivares Carioca-MG (suscetível) e IAPAR-57, IAPAR-72 e IAPAR-65 (resistentes). A população de 480 progênies foi obtida pelo modelo dialélico completo. Estas foram semeadas em campo sob inoculação natural e estudada ao nível de plantas individuais para cálculo do índice de doença de BGMV (I.D.) Em relação ao I.D. médio, as progênies foram mais resistentes do que seus parentais (2,62 e 2,87), respectivamente. O processo de seleção foi realizado pelo I.D. (sigmag² = 0,2729 e h a² = 0,3953), onde foram obtidas famílias com grãos do tipo carioca e com I.D. inferior aos parentais resistentes. Segundo o GS% estimado, não serão necessárias muitas progênies ou famílias para se obter progresso com seleção para BGMV. O genótipo IAPAR-72 foi o parental superior na obtenção de progênies de maior resistência (menores I.D.). Possivelmente o mecanismo de resistência é do tipo resistência parcial.

Les diarrhées post-sevrages causées par des infections à Escherichia coli entérotoxinogène positif pour le fimbriae F4 (ETEC F4), entraînent des pertes économiques importantes chez les producteurs de porc. Depuis quelques années, l’utilisation de probiotiques, comme additif alimentaire pour prévenir ce type d’infection entérique et réduire les traitements aux antimicrobiens, suscite un intérêt grandissant en production porcine. Le but du présent travail est de déterminer l’influence de l’administration des probiotiques Pediococcus acidilactici (PA) et Saccharomyces cerevisiae boulardii (SCB) sur la colonisation et l’attachement des ETEC F4, l’accumulation de fluide intestinal et l’expression de cytokines dans l’iléon de porcelets sevrés. Dès la naissance, différentes portées de porcelets ont été affectées aux traitements suivants : PA, SCB, PA + SCB, témoin et témoin avec antibiotiques (ATB). Une dose quotidienne de probiotiques (1 × 109 UFC) a été administrée aux porcelets des groupes probiotiques durant la lactation et après le sevrage. Sept jours après le sevrage, à 28 jours d’âge, des porcelets positifs pour le récepteur intestinal spécifique pour F4 ont été infectés oralement avec une souche ETEC F4. Les porcelets ont été euthanasiés 24 heures après l’infection (jour 29) et différents échantillons intestinaux ont été prélevés. Chez les porcelets recevant des probiotiques, l’attachement des ETEC F4 à la muqueuse iléale était significativement diminué chez les groupes PA ou SCB en comparaison avec le groupe ATB. Finalement, l’expression de cytokines intestinales était plus élevée chez les porcs du groupe PA + SCB en comparaison avec les porcelets témoins. En conclusion, les résultats de cette étude suggèrent que l’administration de probiotiques pourrait être une alternative pour limiter les infections à ETEC F4 chez le porc.

The aim of the present study was to investigate the in vitro gastric stability of colistin sulfate (CS) and its antimicrobial activity against Escherichia coli and to study the impact of ETEC O149: F4 (K88) infection in pigs on CS intestinal absorption. The stability profile of CS was evaluated in a simulated gastric fluid (SGF). Antimicrobial activity of CS and its degradation products were examined in a 96-well polystyrene microplate model. The effect of experimental infection with ETEC O149: F4 on CS intestinal absorption was determined by quantification of CS systemic concentration using a validated LC–MS/MS method. A rapid degradation of CS accompanied by an increase in CS antimicrobial activity by comparison with non-degraded CS (P < 0.0001) was observed in SGF. Additionally, CS levels were not quantifiable in systemic circulation using a highly sensitive method and concurrent oral challenge did not affect CS absorption in an induction model of subclinical post-weaning diarrhea (PWD).

Enteric Escherichia coli infections are a highly relevant cause of disease and death in young pigs. Breeding genetically resistant pigs is an economical and sustainable method of prevention. Resistant pigs are protected against colonization of the intestine through the absence of receptors for the bacterial fimbriae, which mediate adhesion to the intestinal surface. The present work aimed at elucidation of the mode of inheritance of the F4ad receptor which according to former investigations appeared quite confusing. Intestines of 489 pigs of an experimental herd were examined by a microscopic adhesion test modified in such a manner that four small intestinal sites instead of one were tested for adhesion of the fimbrial variant F4ad. Segregation analysis revealed that the mixed inheritance model explained our data best. The heritability of the F4ad phenotype was estimated to be 0.7±0.1. There are no relations to the strong receptors for variants F4ab and F4ac. Targeted matings allowed the discrimination between two F4ad receptors, that is, a fully adhesive receptor (F4adRFA) expressed on all enterocytes and at all small intestinal sites, and a partially adhesive receptor (F4adRPA) variably expressed at different sites and often leading to partial bacterial adhesion. In pigs with both F4ad receptors, the F4adRPA receptor is masked by the F4adRFA. The hypothesis that F4adRFA must be encoded by at least two complementary or epistatic dominant genes is supported by the Hardy-Weinberg equilibrium statistics. The F4adRPA receptor is inherited as a monogenetic dominant trait. A comparable partially adhesive receptor for variant F4ab (F4abRPA) was also observed but the limited data did not allow a prediction of the mode of inheritance. Pigs were therefore classified into one of eight receptor phenotypes: A1 (F4abRFA/F4acR+/F4adRFA); A2 (F4abRFA/F4acR+/F4adRPA); B (F4abRFA/F4acR+/F4adR-); C1 (F4abRPA/F4acR-/F4adRFA); C2 (F4abRPA/F4acR-/F4adRPA); D1 (F4abR-/F4acR-/F4adRFA); D2 (F4abR-/F4acR-/F4adRPA); E (F4abR-/F4acR-/F4adR-).
