Lead toxicity was studied in rats exposed from conception until weaning and assessed by monitoring offspring behavior in both the open field and elevated plus maze and by determining tissue lead in an assessment schedule extended to first (F1) and second (F2) generations. Dams utilized for the F1 generation were submitted to 750 ppm of lead (acetate) in drinking water during pregnancy and lactation. For F1 pups, behavioral alterations were not detected in the elevated plus maze, while in the open field, spontaneous locomotor activity as well as time of both grooming and rearing increased, while freezing time decreased in 30- and 90-day-old rats. Lead content was higher in tissues of 1- and 30-day-old pups. However, in 90-day-old rats, lead was detected only in the femur. F2 generation was lead-free but still presented alterations in both locomotor activity and grooming in 30- and 90-day-old pups. It appears that developmental lead exposure may cause behavioral effects during the developmental stage of the F1 generation, which remains throughout the animal's adult life as a sequel, regardless of lead accumulation, and is extended to the F2 generation of rats. A range of vectors were made in which the EYFP gene or the Cre gene were inserted in the start codon of the NG2 gene. The NG2-EYFP vectors were used to generate NG2-EYFP “knockin” mice by homologous recombination. The F1 generation showed lack of EYFP expression, due to NeoR cassette interference. Excision of the NeoR, by breeding the F1 generation to ELLA-Cre mice allowed proper expression of EYFP. NG2-EYFP heterozygous mice were characterized in detail for astrocytic, neurogenic and oligodendrocytic properties through antibody labeling. NG2-EYFP+ cells did not label for the astrocyte marker GFAP, but some cells did express S100 Beta. The cells did not label with any neuronal markers like Beta III tubulin, Neun, and double cortin, but many of the NG2-EYFP+ cells made intimate contacts to the neurons. These contacts are widespread throughout the grey and white matter of the brain. The NG2-EYFP+ cells did label for oligodendrocyte markers like PDGFα-R, NG2, Olig2, O4, and Sox 10. There were a few cells termed phantom cells that did label for NG2, but had no EYFP expression. This could have been caused by improper excision of the NeoR cassette in the F2 generation. The heterozygous mouse is a tool to allow the characterization of the in vivo properties of the NG2+ cells. Breeding of these mice to homozygosity yielded an NG2-knockout mouse, which was also subjected to initial characterization. The NG2-EYFP homozygous showed equivalent cell labeling results to the NG2-EYFP heterozygous mouse, but the phantom cells disappeared in the knockout. The results show that the NG2 cells are a heterogenous population that does not express astrocytic or neuronal markers. The homozygous mouse is an ideal tool to further dissect the properties of the cells, lacking NG2 in vivo.

‘KP4’ is based on selected F4 progeny of 8 plants showing a low, creeping, tight-matted, late flowering growth habit. The original parental breeding population was selected from among 1600 diploid Rhodes grass seedlings grown as spaced plants; seven of the selected parental plants were from ‘Katambora’ and the eighth (which did not contribute as a maternal parent beyond the F1 generation) was a seedling from an unreleased accession. Four (4) cycles of mass selection were conducted, in which the selected plants from the previous generation were allowed to inter-cross in isolation in the field, and the resultant progeny later grown as spaced plants in the field for the next cycle of selection. Selection was for the following attributes: prostrate creeping early growth habit with short stolon internodes resulting in a dense stolon mat; leafy appearance; fine leaf and stem; and late flowering (i.e. a long period of vegetative growth before flowering). ‘KP4’ is a synthetic Rhodes grass cultivar multiplied from the selected fourth-generation plants produced by this line of breeding. Objetivo: Avaliar os efeitos da obesidade materna sobre a estrutura do pâncreas e metabolismo de carboidratos no início da vida adulta, com foco na prole da primeira geração (F1) e segunda geração (F2) de progenitoras F0 alimentadas com dieta rica em gordura nos períodos pré-gestacional, durante a gestação e lactação. Métodos: Camundongos C57BL/6 do gênero feminino (F0) foram alimentadas com dieta padrão (SC) ou dieta rica em gordura (HF) durante as oito semanas que antecederam o acasalamento, durante a gestação e lactação para gerar a F1 (F1-SC e F1-HF). As proles geradas receberam dieta SC até o terceiro mês de vida. Aos três meses de idade, fêmeas F1 foram acasaladas para gerar a F2 (F2-SC e F2-HF). Apenas os machos das geração F1 e F2 foram avaliados aos 3 meses de idade. As características metabólicas foram avaliadas pela massa corporal (MC), glicemia de jejum, área sob a curva no teste oral de tolerância a glicose; análise plasmática de insulina, leptina e adiponectina; distribuição e análise morfológica do pâncreas através da imunohistoquímica e imunofluorescência. Resultados: A prole F1-HF teve aumento significativo na massa corporal e glicemia quando comparado à F1-SC. Por outro lado, as proles F2-HF e F2-SC tiveram massas corporais semelhantes. A prole F1-HF e F2-HF mostrou aumento da ingestão de energia, intolerância à glicose, hiperinsulinemia, hiperleptinemia, hipoadiponectinemia, resistência à insulina, aumento da massa pancreática, o aumento da densidade de volume de ilhotas com elevada massa de células alfa e beta, ilhotas hipertrofiadas caracterizadas por uma distribuição alterada de células alfa e beta, e fraca imunoreatividade do pâncreas homeobox duodeno-1 (PDx1). Conclusões: Uma dieta HF materna consumida durante o período pré-concepção e durante toda a gestação e lactação em camundongos promoveu programação metabólica do pâncreas endócrino com pré-disposição ao diabetes mellitus tipo 2 precoce nas proles F1 e F2 do gênero masculino, sugerindo que essas mudanças são intergeracionais.

A vitamina D, atualmente, é relacionada também ao metabolismo da glicose e o desenvolvimento de órgãos. Fêmeas de camundongos suíços (F0) foram alimentadas por uma das dietas experimentais: SC (dieta padrão) ou VitD- (dieta sem vitamina D). A prole de machos foi estudada nas idades: nascimento, 10 dias, desmame e seis meses, nas gerações F1 e F2. Avaliou-se a biometria [Massa Corporal (MC), Comprimento nasoanal (CNA) e Pressão Arterial (PA)], urina de 24 horas, glicemia e Teste Oral de Tolerância à Glicose (TOTG). Durante a eutanásia, o sangue foi coletado para análise bioquímica e os tecidos foram removidos para análise estereológica, morfométrica e Western blotting (WB). Não houve diferença de MC ao nascimento. Ao desmame, o grupo F2-VitD- teve maior MC que F2-SC (P=0,03) e aos seis meses, os grupos F1 e F2-VitD- tiveram MC mais elevada (P<0,05 vs SC). A PA foi crescente na prole VitD-, sendo maior em F1-VitD- (P=0,001). A glicemia e TOTG foram alterados somente na F1-VitD-, seguida de esteatose hepática (+99%), hipertrofia da ilhota pancreática (+40%) e elevação do triglicerídeo sanguíneo (P<0,01). O WB de fígado mostrou elevação de FAS (+18%, P<0,01), no grupo com esteatose. Curiosamente, embora a F2-VitD- tenha apresentado elevação de MC, somente o colesterol total fora alterado (P<0,05). Quanto à nefrogênese, houve 50% mais glomérulos imaturos em F1-VitD- que F1-SC (P<0,0001). Porém, na F2 houve aumento somente de 20% (P<0,001). Aos 10 dias, F1-VitD- teve 150% mais glomérulos imaturos e 25% mais glomérulos maduros que SC-F1 (P<0,0001). O WB de rim mostrou que a prole F1-VitD- apresentou maior expressão de renina, ao desmame e aos seis meses, enquanto que a expressão de podocina foi reduzida (P=0,0004). Não houve diferença na análise de WT1. A restrição materna em vitamina D altera a morfologia do pâncreas e fígado, com resistência à insulina, altera a expressão renal de importantes fatores, assim como retarda a maturação glomerular estendendo o período da nefrogênese, principalmente na geração F1.

Studies were undertaken to produce genetic clones derived from all homozygous mitotic gynogenetic individuals in rohu, Labeo rohita Ham. ln view of this, attempts were made to interfere with the normal functioning of the spindle apparatus during the first mitotic cell division of developing eggs using heat shocks, there by leading to the induction of mitotic gynogenetic diploids in the F1 generation. Afterwards, viable mitotic gynogenetic alevins were reared and a selected mature female fish was used to obtain ovulated eggs which were fertilized later with UV-irradiated milt. Milt was diluted with Cortland’s solution and the sperm concentration was maintained at 10⁸/ml. The UV-irradiation was carried out for 2 minutes at the intensity of 200 to 250 µW/cm² at 28± 1°C. The optimal heat shock of 40°C for 2 minutes applied at 25 to 30 minutes a.f. was used to induce mitotic gynogenesis in first (F1) generation and at 3 to 5 minutes a.f. to induce meiotic gynogenesis in the second (F2) generation. The results obtained are presented and the light they shed on the timing of the mitotic and meiotic cell division in this species is discussed.

In the interferon-induced antiviral mechanisms, the Mx pathway is one of the most powerful. Mx proteins have direct antiviral activity and inhibit a wide range of viruses by blocking an early stage of the viral genome replication cycle. However, antiviral activity of piscine Mx remains unclear in vivo. In the present study, an Mx-like gene was cloned, characterized and gene-transferred in rare minnow Gobiocypris rarus, and its antiviral activity was confirmed in vivo. The full length of the rare minnow Mx-like cDNA is 2241 bp in length and encodes a polypeptide of 625 amino acids with an estimated molecular mass of 70.928 kDa and a predicted isoelectric point of 7.33. Analysis of the deduced amino acid sequence indicated that the mature peptide contains an amino-terminal tripartite GTP-binding motif, a dynamin family signature sequence, a GTPase effector domain and two carboxy-terminal leucine zipper motifs, and is the most similar to the crucian carp (Carassius auratus) Mx3 sequence with an identity of 89%. Both P0 and F1 generations of Mx-transgenic rare minnow demonstrated very significantly high survival rate to GCRV infection (P < 0.01). The mRNA expression of Mx gene was consistent with survival rate in F1 generation. The virus yield was also concurrent with survival time using electron microscope technology. Rare minnow has Mx gene(s) of its own but introducing more Mx gene improves their resistance to GCRV. Mx-transgenic rare minnow might contribute to control the GCRV diseases. No unequivocal evidence exists of genetically inherited resistance to metals/metalloids in field populations of earthworms. We studied cocoon production in adult Lumbricus rubellus Hoffmeister collected from an abandoned arsenic and copper mine (Devon Great Consols, Devon, UK), and abandoned tungsten mine (Carrock Fell, Cumbria, UK) and an uncontaminated cultured population. The earthworms were kept in uncontaminated soil for nine weeks. From a total of 42 L. rubellus from each site, Devon Great Consols adults produced 301 cocoons, of which 42 were viable; Carrock Fell 60 cocoons, of which 11 were viable; and the reference population 101 cocoons, of which 62 were viable. The hatchlings were collected and stored at 4 degrees C at weekly intervals. After 12 weeks, all hatchlings were transferred to clean soil and maintained at 15 degrees C for 20 weeks until they showed evidence of a clitellum. In toxicity trials, F1 generation L. rubellus were exposed to 2,000 mg As/kg as sodium arsenate or 300 mg Cu/kg as copper chloride for 28 d. The F1 generation L. rubellus from Devon Great Consols mine demonstrated resistance to arsenate but not copper. All L. rubellus from Devon Great Consols kept in soil treated with sodium arsenate remained in good condition over the 28-d period but lost condition rapidly and suffered high mortality in soil treated with copper chloride. The control population suffered high mortality in soil treated with sodium arsenate and copper chloride. Previous work has shown that field-collected adults demonstrate resistance to both arsenate and Cu toxicity under these conditions. Thus, while arsenate resistance may be demonstrated in F1 generation L. rubellus from one of the contaminated sites, Cu resistance is not. The F1 adults and F2 cocoons did not have significantly higher levels of As than the control population, with no residual As tissue burden, suggesting that resistance to As in these populations may be inherited.

Dendrodrilus rubidus were sampled from a mine spoil soil at Coniston Copper Mine, an abandoned Cu mine in Cumbria, UK and a Cu-free control site. Earthworms were maintained for 14 d in both Kettering loam and a Moorland soil amended with Cu nitrate. Mortality, condition index, weight change and tissue concentration were determined. In both soils D. rubidus native to the mine site were able to tolerate significantly higher soil Cu concentrations (MWRT, p <= 0.001), and exhibited significantly less change in weight (t-test, p <= 0.001) and a lower loss in condition (t-test, p <= 0.001) than control earthworms. For a given soil Cu concentration tissue Cu concentrations were greater in the mine site earthworms. Low cocoon production and viability from the mine site population prevented the determination of toxicity parameters on the F1 generation and may be an indicator of the cost of tolerance to the population. Background: Oil palm is the world’s most productive oil-food crop despite yielding well below its theoretical maximum. This maximum could be approached with the introduction of elite F1 varieties. The development of such elite lines has thus far been prevented by difficulties in generating homozygous parental types for F1 generation. Results: Here we present the first high-throughput screen to identify spontaneously-formed haploid (H) and doubled haploid (DH) palms. We secured over 1,000 Hs and one DH from genetically diverse material and derived further DH/mixoploid palms from Hs using colchicine. We demonstrated viability of pollen from H plants and expect to generate 100% homogeneous F1 seed from intercrosses between DH/mixoploids once they develop female inflorescences. Conclusions: This study has generated genetically diverse H/DH palms from which parental clones can be selected in sufficient numbers to enable the commercial-scale breeding of F1 varieties. The anticipated step increase in productivity may help to relieve pressure to extend palm cultivation, and limit further expansion into biodiverse rainforest.

A wild house mouse (Mus domesticus) population originally trapped near Reading, Berkshire, United Kingdom, and maintained as a colony in the laboratory, was subjected to the discriminating feeding period of the warfarin resistance test, as used by Wallace and MacSwiney (1976) and derived from the work of Rowe and Redfern (1964). Eighty percent of this heterogeneous population survived the resistance-test. A similar proportion of the population was found to survive the normally lethal dose of bromadiolone administered by oral gavage. The majority of this population of mice were classified as "warfarin-resistant" and "bromadiolone-resistant." The dose of 10mg.kg-1 of bromadiolone administered by oral gavage appeared to give good discrimination between susceptible and resistant individuals. The results of breeding tests indicate a single dominant gene that confers both "warfarin-resistance" and "bromadiolone-resistance", with complete expression of the resistance genotype in both males and females. Individual mice were classified as to genotype by back-crossing to a homozygous-susceptible strain, and resistance-testing the F1 generation. Separate strains of homozygous-resistant and homozygous-susceptible house mice are now being established.

Para o estabelecimento de um programa de controle biológico, o conhecimento de alguns aspectos biológicos e de comportamento dos inimigos naturais é de extrema importância. Os aspectos biológicos do desenvolvimento larval e pupal do predador Ceraeochrysa everes (Banks) foram estudados. Larvas oriundas de adultos da geração F1 foram mantidas em laboratório a 25 ± 21°C, 70 ± 10 % UR e fotofase 14 horas, sendo alimentadas com ovos de Sitotroga cerealella (Olivier). A duração e viabilidade do período embrionário, estágios imaturos de desenvolvimento e o período de ovo a adulto foram avaliados. O período embrionário foi em média de 5,0 dias, enquanto que as durações médias para o primeiro, segundo e terceiro instares foram de 5,1 ± 0,03; 4,3 ± 0,05 e 4,5 ± 0,05 dias, respectivamente, com viabilidade superior a 90 %. Os estágios larval, pré-pupal e pupal apresentaram duração média de 13,9 ± 0,07; 5,7 ± 0,07 e 9,6 ± 0,12 dias, respectivamente. A duração do ciclo biológico foi de 34 ± 0,11 dias em média. Ovos de S. cerelella não foram adequados para a manutenção de C. everes em laboratório, por interferir no desenvolvimento do predador.

Devido a grande importância da cultura de Eucalyptus no Brasil, empresas do setor florestal têm buscado através de programas de melhoramento genético, reduzir as perdas de produção e atender a demanda do mercado de papel e celulose. Um exemplo, é a busca por genes de resistência a doenças, principalmente a ferrugem causada por Puccinia psidii Winter, que resulta em redução da produtividade em plantas altamente suscetíveis. No presente trabalho, mudas de Eucalyptus pertencentes a uma geração F1, provenientes do cruzamento controlado entre parentais híbridos E. grandis X E. urophylla, sendo eles resistente e suscetível, foram inoculadas com Puccinia psidii em casa de vegetação e acompanhadas até o aparecimento dos sintomas da ferrugem. Foram classificadas, em dois grupos: resistentes (ausência de sintomas) e suscetíveis (presença de sintomas e esporulação). As amostras de DNA foram comparadas com o uso de marcadores moleculares associado ao método de BSA (Bulked Segregant Analysis). O polimorfismo entre os grupos foi geneticamente relacionado ao loco que determina a característica de resistência ou sucetibilidade. Dentre os 720 primers testados, 19 foram polimórficos, porém, apenas o marcador AK 01 manteve-se presente, quando testado em todos os indivíduos da população, mostrando-se a uma distância genética estimada de 20 cM em repulsão ao gene de resistência.
