This study describes the pedagogical impact of real-world experimental projects undertaken as part of an advanced undergraduate Fluid Mechanics subject at an Australian university. The projects have been organised to complement traditional lectures and introduce students to the challenges of professional design, physical modelling, data collection and analysis. The physical model studies combine experimental, analytical and numerical work in order to develop students’ abilities to tackle real-world problems. A first study illustrates the differences between ideal and real fluid flow force predictions based upon model tests of buildings in a large size wind tunnel used for research and professional testing. A second study introduces the complexity arising from unsteady non-uniform wave loading on a sheltered pile. The teaching initiative is supported by feedback from undergraduate students. The pedagogy of the course and projects is discussed with reference to experiential, project-based and collaborative learning. The practical work complements traditional lectures and tutorials, and provides opportunities which cannot be learnt in the classroom, real or virtual. Student feedback demonstrates a strong interest for the project phases of the course. This was associated with greater motivation for the course, leading in turn to lower failure rates. In terms of learning outcomes, the primary aim is to enable students to deliver a professional report as the final product, where physical model data are compared to ideal-fluid flow calculations and real-fluid flow analyses. Thus the students are exposed to a professional design approach involving a high level of expertise in fluid mechanics, with sufficient academic guidance to achieve carefully defined learning goals, while retaining sufficient flexibility for students to construct there own learning goals. The overall pedagogy is a blend of problem-based and project-based learning, which reflects academic research and professional practice. The assessment is a mix of peer-assessed oral presentations and written reports that aims to maximise student reflection and development. Student feedback indicated a strong motivation for courses that include a well-designed project component.

Linkage disequilibrium (LD) is defined as the nonrandom association of alleles at two or more loci in a population and may be a useful tool in a diverse array of applications including disease gene mapping, elucidating the demographic history of populations, and testing hypotheses of human evolution. However, the successful application of LD-based approaches to pertinent genetic questions is hampered by a lack of understanding about the forces that mediate the genome-wide distribution of LD within and between human populations. Delineating the genomic patterns of LD is a complex task that will require interdisciplinary research that transcends traditional scientific boundaries. The research presented in this dissertation is predicated upon the need for interdisciplinary studies and both theoretical and experimental projects were pursued. In the theoretical studies, I have investigated the effect of genotyping errors and SNP identification strategies on estimates of LD. The primary importance of these two chapters is that they provide important insights and guidance for the design of future empirical LD studies. Furthermore, I analyzed the allele frequency distribution of 26,530 single nucleotide polymorphisms (SNPs) in three populations and generated the first-generation natural selection map of the human genome, which will be an important resource for explaining and understanding genomic patterns of LD. Finally, in the experimental study, I describe a novel and simple, low-cost, and high-throughput SNP genotyping method. The theoretical analyses and experimental tools developed in this dissertation will facilitate a more complete understanding of patterns of LD in human populations. ^

O Grupo Poéticas Digitais foi criado em 2002, no Departamento de Artes Plásticas da ECA-USP, com a intenção de gerar um núcleo multidisciplinar, promovendo o desenvolvimento de projetos experimentais e a reflexão sobre o impacto das novas tecnologias no campo das artes. O Grupo é um desdobramento do projeto wAwRwT, iniciado em 1995 por Gilbertto Prado e tem como participantes professores, artistas, pesquisadores e estudantes. O objetivo deste texto é apresentar algumas experimentações recentes de projetos poéticos como Desluz, de 2009/2010, e Amoreiras, de 2010.

In view of the energy, environmental, and economic advantages of the foamed asphalt process using local aggregates in cold mixes and the promising results from Research Project HR-212, a 4.2-mile section of county road in Muscatine County was built with foamed asphalt and local aggregates during August-September 1983. Extensive laboratory evaluation was carried out on five plant mixes representing foamed mixes used in the nine test sections, a laboratory prepared foamed mix, and a laboratory prepared hot mix similar to Plant Mix 1. The foamed mixes were compacted, cured under 15 curing conditions and tested for bulk specific gravity, Marshall stability at 77° F and at 140° F, cured moisture content, resilient modulus and effects of moisture damage due to freeze-thaw cycles, water soaking, and vacuum saturation. In addition, four sets of 83 core samples were taken at 1 to 15 months and tested for moisture content, specific gravity, Marshall stability, and resilient modulus. In summary, the test road has performed satisfactorily for almost two years. The few early construction problems encountered were to be expected for experimental projects dealing with new materials and technologies. Overall results to date are encouraging and foamed asphalt mixes have proved to have the potential as a viable base material in areas where marginal aggregates are available. It is hoped and expected that performance evaluation of the test sections will be continued and that more foamed asphalt trial projects will be constructed and monitored so that experiences and findings from this project can be verified and mix design criteria can be gradually established. For future foamed asphalt projects it is recommended that anti-stripping additives, such as hydrated lime, be added in view of the potential moisture susceptibility of foamed mixes observed in the laboratory evaluation.

Il temperamento può essere definito come l’attitudine che un cane esprime verso le persone e verso altri animali, la combinazione di tratti fisici e mentali, acquisiti e non, che determinano il comportamento del cane. Tale parametro delinea perciò il carattere di un individuo, inclinazioni e tendenze, eccitabilità, tristezza, rabbia e il modo caratteristico di comportarsi di un individuo, con particolare riferimento alle interazioni sociali. La presente tesi di Dottorato rappresenta uno studio su alcuni tratti del temperamento nel cane domestico elaborato in 3 progetti sperimentali. Nei primi due progetti sono state analizzate le differenze attitudinali tra alcune razze canine attraverso l’applicazione di un test di temperamento in cuccioli di 60 giorni e in cani adulti, per valutare e confrontarne il temperamento, la socialità ed identificare profili tipici di razza. Nel terzo progetto un campione di cani morsicatori di canile e di proprietà è stato confrontato con due rispettivi gruppi di controllo. Analizzando i risultati è stato possibile mettere in evidenza caratteristiche di razza omogenee nell’interazione con stimoli inanimati, nelle interazioni sociali e in relazione alla possessività e sono stati delineati profili di razza sia nei cuccioli sia negli adulti. Si sono tuttavia, osservate variabilità individuali, intra-razza e intra-cucciolata, a testimonianza dell’influenza complessa e multifattoriale delle componenti genetica e ambientale sul comportamento dei cani. Il confronto tra cani morsicatori di canile e di proprietà ha messo in luce interessanti differenze tra i soggetti in termini di reattività, socievolezza, propensione all’interazione con il proprietario o con un estraneo, comportamenti di evitamento e velocità di reazione agli stimoli presentati. Il test applicato è risultato un valido strumento per valutare il temperamento di cani dichiarati aggressivi che sono stati sottoposti a situazioni nuove e a stimoli sconosciuti per poter ottenere una migliore visione d’insieme del temperamento del soggetto.

The encounter of architecture and urbanism with parcelisation is marked with the sign of a historical debt. Parcelisation is a tremendously powerful design tool. Architecture and urban planning attest to its capacity to shape sociospatial relations across a range of scales and contexts. From Renaissance palazzi to modernist slabs, from nation-wide agrarian reforms and Haussmann’s Paris to Christopher Alexander’s new theory of urban design and Michael Webb’s Suitaloon — most of the historical revolutions and experimental projects that have shaken these fields have relied, consciously or not, on a critique and restructuration of inherited divisions of land and space. The layout of parcel patterns has a series of implications including forms of property, the divide between private and public realms, the distribution of spatial contents and uses through zoning, typological decisions and so forth. The behaviour of any urban structure depends on the tissue of plots that configures the urban fabric and many of the challenges urban design faces today —complexity, urban vitality, social equality, etc.— hinge upon the forms of land division and distribution we adopt.

The experimental projects discussed in this thesis are all related to the field of artificial molecular machines, specifically to systems composed of pseudorotaxane and rotaxane architectures. The characterization of the peculiar properties of these mechano-molecules is frequently associated with the analysis and elucidation of complex reaction networks; this latter aspect represents the main focus and central thread tying my thesis work. In each chapter, a specific project is described as summarized below: the focus of the first chapter is the realization and characterization of a prototype model of a photoactivated molecular transporter based on a pseudorotaxane architecture; in the second chapter is reported the design, synthesis, and characterization of a [2]rotaxane endowed with a dibenzylammonium station and a novel photochromic unit that acts as a recognition site for a DB24C8 crown ether macrocycle; in the last chapter is described the synthesis and characterization of a [3]rotaxane in which the relative number of rings and stations can be changed on command.

Nowadays, application domains such as smart cities, agriculture or intelligent transportation, require communication technologies that combine long transmission ranges and energy efficiency to fulfill a set of capabilities and constraints to rely on. In addition, in recent years, the interest in Unmanned Aerial Vehicles (UAVs) providing wireless connectivity in such scenarios is substantially increased thanks to their flexible deployment. The first chapters of this thesis deal with LoRaWAN and Narrowband-IoT (NB-IoT), which recent trends identify as the most promising Low Power Wide Area Networks technologies. While LoRaWAN is an open protocol that has gained a lot of interest thanks to its simplicity and energy efficiency, NB-IoT has been introduced from 3GPP as a radio access technology for massive machine-type communications inheriting legacy LTE characteristics. This thesis offers an overview of the two, comparing them in terms of selected performance indicators. In particular, LoRaWAN technology is assessed both via simulations and experiments, considering different network architectures and solutions to improve its performance (e.g., a new Adaptive Data Rate algorithm). NB-IoT is then introduced to identify which technology is more suitable depending on the application considered. The second part of the thesis introduces the use of UAVs as flying Base Stations, denoted as Unmanned Aerial Base Stations, (UABSs), which are considered as one of the key pillars of 6G to offer service for a number of applications. To this end, the performance of an NB-IoT network are assessed considering a UABS following predefined trajectories. Then, machine learning algorithms based on reinforcement learning and meta-learning are considered to optimize the trajectory as well as the radio resource management techniques the UABS may rely on in order to provide service considering both static (IoT sensors) and dynamic (vehicles) users. Finally, some experimental projects based on the technologies mentioned so far are presented.

We define and experimentally test a public provision mechanism that meets three basic ethical requirements and allows community members to influence, via monetary bids, which of several projects is implemented. For each project, participants are assigned personal values, which can be positive or negative. We provide either public or private information about personal values. This produces two distinct public provision games, which are experimentally implemented and analyzed for various projects. In spite of the complex experimental task, participants do not rely on bidding their own personal values as an obvious simple heuristic whose general acceptance would result in fair and efficient outcomes. Rather, they rely on strategic underbidding. Although underbidding is affected by projects’ characteristics, the provision mechanism mostly leads to the implementation of the most efficient project.

Pérez-Castrillo and Wettstein (2002) and Veszteg (2004) propose the use of a multibidding mechanism for situations where agents have to choose a common project. Examples are decisions involving public goods (or public "bads"). We report experimental results to test the practical tractability and effectiveness of the multibidding mechanisms in environments where agents hold private information concerning their valuation of the projects. The mechanism performed quite well in the laboratory: it provided the ex post efficient outcome in roughly three quarters of the cases across the treatments; moreover, the largest part of the subject pool formed their bids according to the theoretical bidding behavior.

Accurate prediction of transcription factor binding sites is needed to unravel the function and regulation of genes discovered in genome sequencing projects. To evaluate current computer prediction tools, we have begun a systematic study of the sequence-specific DNA-binding of a transcription factor belonging to the CTF/NFI family. Using a systematic collection of rationally designed oligonucleotides combined with an in vitro DNA binding assay, we found that the sequence specificity of this protein cannot be represented by a simple consensus sequence or weight matrix. For instance, CTF/NFI uses a flexible DNA binding mode that allows for variations of the binding site length. From the experimental data, we derived a novel prediction method using a generalised profile as a binding site predictor. Experimental evaluation of the generalised profile indicated that it accurately predicts the binding affinity of the transcription factor to natural or synthetic DNA sequences. Furthermore, the in vitro measured binding affinities of a subset of oligonucleotides were found to correlate with their transcriptional activities in transfected cells. The combined computational-experimental approach exemplified in this work thus resulted in an accurate prediction method for CTF/NFI binding sites potentially functioning as regulatory regions in vivo.

This document contains a discussion of the reasons why the project did not succeed. A detailed discussion of the steps taken by the Iowa Department of Transportation to make the experiment work are contained in this document, along with recommendations for future projects.
