The use of sustainability indicators for evaluating sanitation systems is applied to the Erdos Eco- Town Project (EETP) in China for illustration. The EETP is the largest urban settlement in the world employing ecological sanitation, which incorporates separation of waste streams, dry toilets, and resource recovery. The EETP’s dry sanitation system is compared against the Dongsheng District’s conventional sewer and centralised STP. The two systems are compared based on technological, environmental, economic, and societal indicators. Overall, the two systems perform reasonably well from a technological perspective. The conventional system performs significantly better than the dry system with regards to land and energy requirements, and global warming potential; it also performs better based on freshwater aquatic and terrestrial ecotoxicity potentials, but by a smaller margin. The dry system has superior environmental performance based on water consumption, eutrophication potential, and nutrient and organic matter recovery. The dry system is a more costly system as it requires greater infrastructure and higher operational costs, and does not benefit from economies of scale. The waterborne system performs better based on the societal indicators largely because it is a well-established system.

This doctoral thesis describes the extension of the resonance ionization laser ion source RILIS at CERN/ISOLDE by the addition of an all-solid state tunable titanium:sapphire (Ti:Sa) laser system to complement the well-established system of dye lasers. Synchronous operation of the so called Dual RILIS system of Ti:Sa and dye lasers was investigated and the potential for increased ion beam intensity, reliability, and reduced setup time has been demonstrated. In-source resonance ionization spectroscopy was performed at ISOLDE/CERN and at ISAC/TRIUMF radioactive ion beam facilities to develop an efficient and selective three-colour ionization scheme for the purely radioactive element astatine. A LabVIEW based monitoring, control and measurement system was conceived which enabled, in conjunction with Dual RILIS operation, the spectroscopy of high lying Rydberg states, from which the ionization potential of the astatine atom was determined for the first time experimentally.

Hematopoiesis is a well-established system used to study developmental choices amongst cells with multiple lineage potentials, as well as the transcription factor network interactions that drive these developmental paths. Multipotent progenitors travel from the bone marrow to the thymus where T-cell development is initiated and these early T-cell precursors retain lineage plasticity even after initiating a T-cell program. The development of these early cells is driven by Notch signaling and the combinatorial expression of many transcription factors, several of which are also involved in the development of other cell lineages. The ETS family transcription factor PU.1 is involved in the development of progenitor, myeloid, and lymphoid cells, and can divert progenitor T-cells from the T-lineage to a myeloid lineage. This diversion of early T-cells by PU.1 can be blocked by Notch signaling. The PU.1 and Notch interaction creates a switch wherein PU.1 in the presence of Notch promotes T-cell identity and PU.1 in the absence of Notch signaling promotes a myeloid identity. Here we characterized an early T-cell cell line, Scid.adh.2c2, as a good model system for studying the myeloid vs. lymphoid developmental choice dependent on PU.1 and Notch signaling. We then used the Scid.adh.2c2 system to identify mechanisms mediating PU.1 and Notch signaling interactions during early T-cell development. We show that the mechanism by which Notch signaling is protecting pro-T cells is neither degradation nor modification of the PU.1 protein. Instead we give evidence that Notch signaling is blocking the PU.1-driven inhibition of a key set of T-regulatory genes including Myb, Tcf7, and Gata3. We show that the protection of Gata3 from PU.1-mediated inhibition, by Notch signaling and Myb, is important for retaining a T-lineage identity. We also discuss a PU.1-driven mechanism involving E-protein inhibition that leads to the inhibition of Notch target genes. This is mechanism may be used as a lockdown mechanism in pro-T-cells that have made the decision to divert to the myeloid pathway.

Five kinds of Schiff bases of chitosan and carboxymethyl chitosan (CMCTS) have been prepared according to a previous method and the antioxidant activity was studied using an established system, such as superoxide and hydroxyl radical scavenging. Obvious differences between the Schiff bases of chitosan and CMCTS were observed, which might be related to contents of the active hydroxyl and amino groups in the molecular chains. Three new kinds of 1,3,5-thiadiazine-2-thi one derivatives of chitosan with two different molecular weight (SATTCS1, SATTCS2, TITTCS1, TITTCS2, CITTCS1 and CITTCS2) have been prepared. Their structures were characterized by IR spectroscopy. The substitution degree of derivatives calculated by elemental analyses was 0.47, 0.42, 0.41, 0.38, 0.41 and 0.36, respectively. The result shows that substitution degree of derivatives was higher with lower molecular weight. The antioxidant activity was studied using an established system, such as bydroxyl radical scavenging, superoxide radical scavenging and reducing power. Antioxidant activity of the 1,3,5-thiadiazine-2-thione derivatives of chitosan were stronger than that of chitosans and antioxiclant activity of low molecular weight derivatives were stronger than that of high molecular weight derivatives. Le sous-continent latino-américain vit dans une pauvreté endémique où la lutte pour la survie est au quotidien. Les peuples deviennent de plus en plus pauvres et la vie de la personne humaine est méprisée. Est-il question d’une carence de biens matériels ou d’une mauvaise répartition de la richesse? Nous affirmons qu’il s’agit d’une situation de domination, d’oppression et d’injustice institutionnalisée. Ce mémoire tend à faire une lecture de l’éthique théologique de la libération d’Enrique Dussel dans une perspective émancipatoire. Nous partons du principe que cette éthique matérielle de la vie peut donner une réponse à la morale dominatrice établie et peut contribuer à la transformation de la situation injuste. L’éthique théologique de la libération dusselienne analyse l’être latino-américain considéré comme autre par le système en vigueur et cherche à changer qualitativement les conditions réelles de vie de chaque personne en communauté. Pour ce faire, elle repense de façon critique la totalité des principes de la morale dominatrice à partir de la négation de la vie des victimes et propose une alternative qui doit se réaliser ici maintenant dans l’histoire : la libération des victimes dans n’importe quel système de domination. Ce projet tient compte de la dimension spirituelle du peuple latino-américain. Un sous-continent dit majoritairement chrétien se questionne sur la présence de Dieu et de son agir dans la lutte quotidienne pour la libération. Il se demande comment vivre la foi en Dieu dans une situation d’oppression et de lutte pour la libération. C’est à ce parcours que ce mémoire nous convie en nous rappelant constamment qu’un monde meilleur est possible et que le Dieu de la vie n’abandonne pas sa création.

Nach 35 Jahren Entwicklungszeit wurde im Jahr 2004 in Shanghai die erste kommerzielle Anwendung des innovativen Magnetbahnsystems Transrapid in Betrieb genommen; in Deutschland konnte bislang keine Transrapid-Strecke realisiert werden, obwohl dieses System entsprechend den Ergebnissen einer vom damaligen Bundesverkehrsminister beauftragten Studie aus dem Jahr 1972 für den Einsatz in Deutschland entwickelt wurde. Beim Transrapid handelt es sich um eine echte Produkt-Innovation im Bahnverkehr und nicht um eine Weiterentwicklung oder Optimierung wie beim ICE, und ist somit als innovativer Verkehrsträger der Zukunft in die langfristige Entwicklung der Verkehrssysteme einzufügen. Die modernen HGV Bahnsysteme (Shinkansen/TGV/ICE) hingegen sind, ähnlich der Clipper in der Phase der Segelschifffahrt im Übergang zum Dampfschiff, letzte Abwehrentwicklungen eines am Zenit angekommenen Schienen-Verkehrssystems. Die Einführung von Innovationen in einen geschlossenen Markt stellt sich als schwierig dar, da sie zu einem Bruch innerhalb eines etablierten Systems führen. Somit wird in der vorliegenden Arbeit im ersten Teil der Themenkomplex Innovation und die Einordnung der Magnet-Schwebe-Technologie in diese langfristig strukturierten Abläufe untersucht und dargestellt. Das Transrapid-Projekt ist demzufolge in eine zeitstrukturelle Zyklizität einzuordnen, die dafür spricht, die Realisierung des Gesamtprojektes in eine Zeitspanne von 20 bis 30 Jahre zu verlagern. Im zweiten Teil wird auf der Basis einer regionalstrukturellen Analyse der Bundesrepublik Deutschland ein mögliches Transrapidnetz entworfen und die in diesem Netz möglichen Reisezeiten simuliert. Weiterhin werden die Veränderungen in den Erreichbarkeiten der einzelnen Regionen aufgrund ihrer Erschließung durch das Transrapidnetz simuliert und grafisch dargestellt. Die vorliegende Analyse der zeitlichen Feinstruktur eines perspektiven Transrapidnetzes ist ein modellhafter Orientierungsrahmen für die Objektivierung von Zeitvorteilen einer abgestimmten Infrastruktur im Vergleich zu real möglichen Reisezeiten innerhalb Deutschlands mit den gegebenen Verkehrsträgern Schiene, Straße, Luft. So würde der Einsatz des Transrapid auf einem entsprechenden eigenständigen Netz die dezentrale Konzentration von Agglomerationen in Deutschland fördern und im Durchschnitt annähernd 1 h kürzere Reisezeiten als mit den aktuellen Verkehrsträgern ermöglichen. Zusätzlich wird noch ein Ausblick über mögliche Realisierungsschritte eines Gesamtnetzes gegeben und die aufgetretenen Schwierigkeiten bei der Einführung des innovativen Verkehrssystems Transrapid dargestellt.

A dissertação de mestrado “O Impacto das Novas Biotecnologias no Pensamento Político – A problemática das células estaminais embrionárias” partiu do pressuposto basilar de que a Humanidade se depara com uma ruptura de modelo de pensamento sem paralelo na História. O Homem detém hoje um conhecimento científico sem precedentes e vê-se perante o potencial das novas biotecnologias que, pela primeira, vez podem alterar a forma de olhar sobre si próprio, não apenas enquanto ser social mas sobretudo como entidade biológica. Todo o enquadramento da dissertação tem em consideração os diferentes momentos da História em que certos homens levados pela inevitabilidade do progresso intelectual e científico contribuíram decisivamente para alterar profundamente os modelos de pensamento. Modelos que, surgidos em determinado contextos históricos, foram considerados de ruptura e revolucionários. Em sentido contrário, numa espécie de reacção conservadora, foram surgindo forças de autoridade e de poder, rejeitando novos modelos e paradigmas que, de uma maneira ou de outra, pudessem pôr em causa o sistema de sociedade instituído. As grandes rupturas na História da Humanidade resultaram desse confronto de ideias, entre um modelo de pensamento vigente e um novo paradigma proposto. Ao longo da dissertação apresentada são analisados vários períodos de ruptura, com particular enfoque para o advento da genética no século XIX e posterior revolução biotecnológica nos Estados Unidos que, num futuro próximo, poderá vir a curar doenças congénitas e degenerativas, funcionando como uma espécie de “kit de reparação do corpo humano, e, num horizonte mais alargado, poderá potenciar a possibilidade da criação de um “outro eu”, produto do Homem e não do livre arbítrio. Pela primeira vez, o Homem tem conhecimento e técnica para criar um mundo pós-humano, onde cada um é resultado da vontade individual dos seus progenitores, dando-se, assim, início a uma nova História. Mas, tudo isto levanta uma série de questões morais, éticas e políticas. Dilemas quanto aos processos de investigação e quanto às consequências que a sua aplicação poderá trazer para a própria Humanidade. Como trabalho de Ciência Política não cabe no propósito deste tecer cenários filosóficos quanto ao futuro do Homem face aos avanços da investigação genética, mas sim tentar analisar e procurar encontrar um padrão de comportamento na forma como os legisladores e governantes, mediante a sua base doutrinária, têm abordado uma matéria cujas implicações terão eventualmente impacto na concepção da própria Humanidade.

O presente trabalho discute o tema dos dirigentes públicos, atores recentemente debatidos pela literatura como um grupo específico que atua orientado a resultados e à criação de valor público. Em geral, são escassos os estudos que discutem os dirigentes públicos empiricamente; assim, mostraremos os papéis que desempenham argumentando que são atores fundamentais para a formulação e implementação das políticas públicas na medida em que são capazes de dialogar tanto com a esfera política como com a da gestão. Para isso, analisaremos o caso Chileno, experiência bastante consolidada que reconhece o dirigente público como ator distinto dos demais, regido por regras específicas de recrutamento, seleção, nomeação e gestão segundo acordos de resultados. Apresentaremos uma análise dos dirigentes chilenos baseada em uma perspectiva histórica tanto sobre o surgimento e consolidação do Sistema de Alta Direção Pública como em relação às tensões e complementaridades entre tecnocratas e políticos que marcaram os governos chilenos no período que compreende desde a redemocratização até os dias atuais. Mostraremos que este Sistema nasce no contexto de um acordo político entre governo e oposição em busca da modernização e profissionalização do Estado e que as dimensões da política e da gestão são pilares centrais do modelo, presentes em todas as suas esferas, tais como seu desenho institucional e regras de gestão de pessoas e do desempenho.

This work started from an investigation concerning the process of education, moralization and disciplining of the poor in the backlands of the Brazilian Northeast region, in the second half of the 19th century, through the incorporation of the values of productive work. In order to do so, we took as our field of investigation the missions of Father José Antônio Pereira Ibiapina (1806-1883), in which an extensive work was developed, including the construction of twenty two (22) charity houses in the States of Rio Grande do Norte, Paraíba, Pernambuco, Ceará e Piauí, but also of weirs, churches, graveyards, hospitals and so forth. Given the socio-historical character of the research, we adopted qualitative methods of analysis and the following procedures: survey and analysis of documents, statute and internal rules that ordered the workings of the charity houses; biographies on Father Ibiapina and the missions‟ reports, as well as various sorts of publications on the missionary. Our study comprises his 27 year-period of missionary-religious activity (1856-1883), and undertakes an analysis on the topic of poverty, since the medieval Christian take on it until modernity, when it stops representing a value and becomes a problem to be solved through work. The socio-political context in which Ibiapina lived in the Northeast was marked by the consequence of droughts and economic crises in the region. The analysis of the documents indicates that, in the circumstances of political weakness and lack of interest for regional demands by the national political agenda, his missionary actions produced strong effects. Starting from an ethics of valorization of work, which combined modern and traditional elements, his actions set up disciplinary, moralizing and civil educational practices of the poor, founding, simultaneously, a pragmatic religious experience directed at the resolution of the problems caused by poverty. In that sense, we observe that such actions are connected to the broader process of moralization and education of the poor, but the analysis also suggests that their inscription into the social order articulated both forward as well as conservative aspects of the established system

This research argues about the mathematical knowledge built in the tradition of the cassava flour production, seeking to analyse these mathematical knowledge in the perspective of the categories of time and measure, built and practiced in the flour production, located in Serra do Navio and Calçoene, in Amapá - Brazil. The following work discuss the identification and the description of the mathematics during the production activities of the flour, where is presented elements related to generation and transmission of the traditional knowledge, which is the basis for maintenance of the tradition of the flour, characterizing the research as an Ethnomathematic study. The methodological procedures highlight ethnographical techniques and elements that characterize the participating observation. The results obtained showed us that the flour workers articulate some length, area and volume measure due to own and traditionally acquired systems, which is apprehended and countersigned by other kind of culturally established system; thus they relativism the measures systems and the official calendars. And it lifts as one of the main proposal that the academic mathematics and the tradition establish knowledge make conjunction of the both knowledge, that is important for a possible reflection and application in the construction of a pedagogical practice in mathematical education, trying to establish points of socio-economic and cultural mark

Pichia pastoris, a methylotrophic yeast, is an established system for the production of heterologous proteins, particularly biopharmaceuticals and industrial enzymes. To maximise and optimise the production of recombinant products, recent molecular research has focused on numerous issues including the design of expression vectors, optimisation of gene copy number, co-expression of secretory proteins such as chaperones, engineering of glycosylation and secretory pathways, etc. However, the physiological effects of different cultivation strategies are often difficult to separate from the molecular effects of the gene construct (e.g., cellular stress through over-expression or incorrect post-translational processing). Hence, overall system optimisation is difficult, even though it is urgently required in order to describe and understand the behaviour of new molecular constructs. This review focuses on particular aspects of recombinant protein production related to variations in biomass growth and their implications for strain design and screening, as well as on the concept of rational comparisons between cultivation systems for the development of specific production processes in bioreactors. The relationship between specific formation rates of secreted recombinant proteins, qp, and specific growth rates, μ, has been analysed in a conceptual attempt to compare different systems, particularly those based on AOX1/methanol and GAP/glucose, and this has now evolved into a pivotal concept for bioprocess engineering of P. pastoris.
