Upon overexpression of integrin αvβ3 and its engagement by vitronectin, we previously showed enhanced adhesion, proliferation, and motility of human ovarian cancer cells. By studying differential expression of genes possibly related to these tumor biological events, we identified the epidermal growth-factor receptor (EGF-R) to be under control of αvβ3 expression levels. Thus in the present study we characterized αvβ3-dependent changes of EGF-R and found significant upregulation of its expression and activity which was reflected by prominent changes of EGF-R promoter activity. Upon disruption of DNA-binding motifs for the transcription factors p53, ETF, the repressor ETR, p50, and c-rel, respectively, we sought to identify DNA elements contributing to αvβ3-mediated EGF-R promoter induction. Both, the p53- and ETF-mutant, while exhibiting considerably lower EGF-R promoter activity than the wild type promoter, retained inducibility by αvβ3. Mutation of the repressor motif ETR, as expected, enhanced EGF-R promoter activity with a further moderate increase upon αvβ3 elevation. The p50-mutant displayed EGF-R promoter activity almost comparable to that of the wild type promoter with no impairment of induction by αvβ3. However, the activity of an EGF-R promoter mutant displaying a disrupted c-rel-binding motif did not only prominently decline, but, moreover, was not longer responsive to enhanced αvβ3, involving this DNA element in αvβ3-dependent EGF-R upregulation. Moreover, αvβ3 did not only increase the EGF-R but, moreover, also led to obvious co-clustering on the cancer cell surface. By studying αvβ3/EGF-R-effects on the focal adhesion kinase (FAK) and the mitogen activated protein kinases (MAPK) p44/42 (erk−1/erk−2), having important functions in synergistic crosstalk between integrins and growth-factor receptors, we found for both significant enhancement of expression and activity upon αvβ3/VN interaction and cell stimulation by EGF. Upregulation of the EGF-R by integrin αvβ3, both receptor molecules with a well-defined role as targets for cancer treatment, might represent an additional mechanism to adapt synergistic receptor signaling and crosstalk in response to an altered tumor cell microenvironment during ovarian cancer progression.

The global efforts to reduce carbon emissions from power generation have favoured renewable energy resources such as wind and solar in recent years. The generation of power from the renewable energy resources has become attractive because of various incentives provided by government policies supporting green power. Among the various available renewable energy resources, the power generation from wind has seen tremendous growth in the last decade. This article discusses various advantages of the upcoming offshore wind technology and associated considerations related to their construction. The conventional configuration of the offshore wind farm is based on the alternative current internal links. With the recent advances of improved commercialised converters, voltage source converters based high voltage direct current link for offshore wind farms is gaining popularity. The planning and construction phases of offshore wind farms, including related environmental issues, are discussed here.

In coastal waters and estuaries, seagrass meadows are often subject to light deprivation over short time scales (days to weeks) in response to increased turbidity from anthropogenic disturbances. Seagrasses may exhibit negative physiological responses to light deprivation and suffer stress, or tolerate such stresses through photo-adaptation of physiological processes allowing more efficient use of low light. Pulse Amplitude Modulated (PAM) fluorometery has been used to rapidly assess changes in photosynthetic responses along in situ gradients in light. In this study, however, light is experimentally manipulated in the field to examine the photosynthesis of Halophila ovalis and Zostera capricorni. We aimed to evaluate the tolerance of these seagrasses to short-term light reductions. The seagrasses were subject to four light treatments, 0, 5, 60, and 90% shading, for a period of 14 days. In both species, as shading increased the photosynthetic variables significantly (P < 0.05) decreased by up to 40% for maximum electron transport rates (ETRmax) and 70% for saturating irradiances (Ek). Photosynthetic efficiencies (a) and effective quantum yields (ΔF/Fm′ ) increased significantly (P < 0.05), in both species, for 90% shaded plants compared with 0% shaded plants. H. ovalis was more sensitive to 90% shading than Z. capricorni, showing greater reductions in ETR max, indicative of a reduced photosynthetic capacity. An increase in Ek, Fm′ and ΔF/Fm′ for H. ovalis and Z. capricorni under 90% shading suggested an increase in photochemical efficiency and a more efficient use of low-photon flux, consistent with photo-acclimation to shading. Similar responses were found along a depth gradient from 0 to10 m, where depth related changes in ETRmax and Ek in H. ovalis implied a strong difference of irradiance history between depths of 0 and 5-10 m. The results suggest that H. ovalis is more vulnerable to light deprivation than Z. capricorni and that H. ovalis, at depths of 5-10 m, would be more vulnerable to light deprivation than intertidal populations. Both species showed a strong degree of photo-adaptation to light manipulation that may enable them to tolerate and adapt to short-term reductions in light. These consistent responses to changes in light suggest that photosynthetic variables can be used to rapidly assess the status of seagrasses when subjected to sudden and prolonged periods of reduced light

We compared daily net radiation (Rn) estimates from 19 methods with the ASCE-EWRI Rn estimates in two climates: Clay Center, Nebraska (sub-humid) and Davis, California (semi-arid) for the calendar year. The performances of all 20 methods, including the ASCE-EWRI Rn method, were then evaluated against Rn data measured over a non-stressed maize canopy during two growing seasons in 2005 and 2006 at Clay Center. Methods differ in terms of inputs, structure, and equation intricacy. Most methods differ in estimating the cloudiness factor, emissivity (e), and calculating net longwave radiation (Rnl). All methods use albedo (a) of 0.23 for a reference grass/alfalfa surface. When comparing the performance of all 20 Rn methods with measured Rn, we hypothesized that the a values for grass/alfalfa and non-stressed maize canopy were similar enough to only cause minor differences in Rn and grass- and alfalfa-reference evapotranspiration (ETo and ETr) estimates. The measured seasonal average a for the maize canopy was 0.19 in both years. Using a = 0.19 instead of a = 0.23 resulted in 6% overestimation of Rn. Using a = 0.19 instead of a = 0.23 for ETo and ETr estimations, the 6% difference in Rn translated to only 4% and 3% differences in ETo and ETr, respectively, supporting the validity of our hypothesis. Most methods had good correlations with the ASCE-EWRI Rn (r2 > 0.95). The root mean square difference (RMSD) was less than 2 MJ m-2 d-1 between 12 methods and the ASCE-EWRI Rn at Clay Center and between 14 methods and the ASCE-EWRI Rn at Davis. The performance of some methods showed variations between the two climates. In general, r2 values were higher for the semi-arid climate than for the sub-humid climate. Methods that use dynamic e as a function of mean air temperature performed better in both climates than those that calculate e using actual vapor pressure. The ASCE-EWRI-estimated Rn values had one of the best agreements with the measured Rn (r2 = 0.93, RMSD = 1.44 MJ m-2 d-1), and estimates were within 7% of the measured Rn. The Rn estimates from six methods, including the ASCE-EWRI, were not significantly different from measured Rn. Most methods underestimated measured Rn by 6% to 23%. Some of the differences between measured and estimated Rn were attributed to the poor estimation of Rnl. We conducted sensitivity analyses to evaluate the effect of Rnl on Rn, ETo, and ETr. The Rnl effect on Rn was linear and strong, but its effect on ETo and ETr was subsidiary. Results suggest that the Rn data measured over green vegetation (e.g., irrigated maize canopy) can be an alternative Rn data source for ET estimations when measured Rn data over the reference surface are not available. In the absence of measured Rn, another alternative would be using one of the Rn models that we analyzed when all the input variables are not available to solve the ASCE-EWRI Rn equation. Our results can be used to provide practical information on which method to select based on data availability for reliable estimates of daily Rn in climates similar to Clay Center and Davis.

A Suíte Intrusiva Santa Clara está inserida na Província Estanífera de Rondônia, na porção SW do Cráton Amazônico. Essa suíte intrusiva é composta pelos maciços Santa Clara, Oriente Velho, Oriente Novo, Manteiga-Sul, Manteiga-Norte, Jararaca, Carmelo, Primavera e das Antas. Os litotipos que perfazem a Suíte Santa Clara ocorrem hospedados nas rochas do Complexo Jamari, uma associação polideformada composta por gnaisses ortoderivados e paraderivados. Características observadas em campo e em análises petrográficas permitiram subdividir o Maciço Santa Clara em cinco fácies distintas: fácies porfirítica, fácies isotrópica, fácies fina, fácies piterlítica e fácies viborgítica. Os litotipos observados correspondem a hornblenda-biotita granitos e biotita granitos intermediários a ácidos, com composições médias semelhantes àquelas verificadas para sienogranitos e monzogranitos. Geoquimicamente, três magmas podem ser identificados. O magma menos evoluído corresponde às rochas das fácies porfirítica e equigranular, e o mais evoluído compreende as fácies de granulometria fina e piterlítica. A fácies viborgítica representa o terceiro líquido magmático, e aparentemente é diferente de todas as outras fácies em termos de aspectos de campo e geoquímica. A análise litogeoquímica indica que estes granitoides são subalcalinos, bastante empobrecidos em MgO e exibem caráter metaluminoso a fracamente peraluminoso. Os padrões de elementos-traços evidenciam que tais granitóides possuem alto conteúdo em elementos incompatíveis (Rb, Zr, Y, Ta, Ce) e ETR, com exceção do Eu. Além disso, também exibem leve enriquecimento em LILE, forte depleção em elementos como Sr e Ti, e leve empobrecimento de Ba, indicando que o fracionamento de minerais como plagioclásio e titanita foi importante na evolução do líquido magmático analisado. A anomalia negativa de Nb indica envolvimento de material crustal nos processos magmáticos que geraram estes granitoides. Os litotipos analisados possuem características típicas de granitos tipo-A ferroan, e as razões FeOt/MgO entre 4,27 e 26,22 sugerem tratar-se de uma série de granitos félsicos fracionados. Os padrões de ETR observados para os litotipos analisados exibem um considerável enriquecimento em ETRL, e anomalia negativa de Eu, sugerindo fracionamento de feldspato durante o processo de diferenciação do líquido magmático. Diagramas discriminantes de ambientes tectônicos sugerem que os litotipos do Maciço Intrusivo Santa Clara são típicos de ambiente intraplaca, do tipo-A2, isto é, associados a ambientes pós-colisionais/pós-orogênicos. As características isotópicas observadas para os granitoides do Maciço Santa Clara sugerem que os mesmos foram gerados a partir da fusão parcial de uma crosta inferior pré-existente. As idades U-Pb entre 1,07 e 1,06 Ga são compatíveis com um magmatismo ocorrido nos estágios finais da colagem do supercontinente Rodínia (1,2-1,0 Ga) e estágios finais do Ciclo Orogênico Sunsás-Aguapeí (1320-1100 Ma). Sugere-se ainda que na verdade o Maciço Santa Clara seja formado por uma coalescência das três intrusões graníticas que são representadas pelos três magmas anteriormente descritos.

O trabalho realizado contempla a configuração de um caminho de pensamento sobre a espacialização do lixo no município do Rio de Janeiro. Apresenta o problema da destinação do lixo os transtornos causados à população e agravados pela industrialização; adoção de técnicas de coleta e destinação, o surgimento de empresas com o propósito de garantir a limpeza e o aperfeiçoamento técnico para confinamento do lixo. Demonstra um detalhamento da geração dos resíduos sólidos urbanos do município do Rio de Janeiro e sua destinação no fim do século XX. Apresentando quantitativo dos resíduos gerados e posteriormente destinados aos lixões. Analisa também o processo de esgotamento dos aterros sanitários existentes e a constituição de uma nova rede geográfica para destinação final dos resíduos. O exemplo da Central de Tratamento de Resíduos (CTR Rio), projeto executado por iniciativa privada, empresa Ciclus, instalada no município de Seropédica, responsável por receber todo o lixo do município do Rio de Janeiro e tratá-lo por meio de novas técnicas e tecnologias. Os resíduos são recebidos através de estações de transferência (ETR) instaladas em pontos geográficos específicos das regiões administrativas do município do Rio de Janeiro e transportado à CTR Rio para o devido confinamento, por meio de uma nova rede geográfica paradestinação final

阿氏浮丝藻(Planktothrixagardhii)是水华蓝藻的重要类群,实验对我国不同省份分离到的5株阿氏浮丝藻生长速率、色素组成、光合活性参数进行了测定.结果表明分离于广州的HAB1128叶绿素a(Chl.a)含量较低,藻蓝蛋白(C-PC)比例和含量较高.电子传递速率(ETR)测定结果表明,HAB1128的ETR曲线较高,最大ETRmax值也显著高于其余4株,高效的光能电子传递链弥补了光合色素Chl.a的不足,以致HAB1128保持了中性的生长速率.分离于武汉东湖的HAB631则与之正好相反,较高的

初步研究了不同浓度硝基苯对莱茵衣藻(Chlamydomonas reinhardtii)生长和光合作用的影响.结果表明:不同浓度的硝基苯对莱茵衣藻的生长和光合生理有明显抑制作用,主要表现在其明显降低光合色素的含量、光能转换效率(Fv/Fm)、电子传递速率(ETR)、净光合速率(Pn)等方面;硝基苯对莱茵衣藻的影响主要是通过影响光合色素合成,降低光合作用电子传递,从而降低藻类的光合作用,引起生长的抑制.

The physiological differences for three bloom-forming cyanobacteria (Cylindrospermopsis raciborskii, Microcystis aeruginosa, and Aphanizomenon flos-aquae) were investigated. In comparison with M. aeruginosa and A. flos-aquae, C. raciborskii exhibited a significantly higher concentration of carotenoids, higher values in maximum photosynthesis rate (P-m), apparent photosynthetic efficieny (a), and maximum electron transport rate (ETRmax) during the growth period. In addition, higher extracellular alkaline phosphatase activities and lower light compensation point (I-c) were also detected in C raciborskii (p < 0.05, ANOVA). Therefore, it is suggested that the higher photosynthetic activities, more effective uptake and utilization to phosphate, and low light requirements might play important roles in the occurrence and invasive behavior of C. raciborskii. A novel multi-cell device made of organic glass was designed to study morphological and physiological characteristics of Microcystis population trapped in simulated sediment conditions. Changes of colonial morphology and antioxidant activities of the population were observed and measured over the range of 31-day incubation. During the incubation, the antioxidant enzyme activities fluctuated significantly in sediment environments. The activities of catalase (CAT), glutathione peroxidase (GPx) and malondialdehyde (NIDA) reached the highest on the 11(th) day, 6(th) day and 6(th) day. respectively, and then dropped down remarkably in the following days. The ratios of Fv/Fm and the maximal electron transfer rate (ETRm) declined during the initial days (1 similar to 11(th) day), but rebounded on the 16(th) day, which were consistent with the variations of total protein. In the end of incubation. gas vacuoles were hard]), observed and the gelatinous sheath was partly disappeared in the population of Microcystis. Nevertheless, the remaining populations. upon transferred to culture medium, were able to grow though experiencing a longer lag phase of nine days. The results indicated that the sediment environments were able to cause negative effects on M. aeruginosa cells. The cells, however, responded to against the possible damage afterwards. It is thus proposed the acute responses in the population during the early stage of sedimentation could be of importance in aiding the long-term survivor of Microcystis and recruitment in lake sediments. The present study also demonstrated the utility of the device in simulating the sediment environments for further investigation.

In order to gain insight into the bloom sustainment of colonial Microcystis aeruginosa Katz., physiological characterizations were undertaken in this study. Compared with unicellular Microcystis, colonial Microcystis phenotypes exhibited a higher maximum photosynthetic rate (Pm), a higher maximum electron transfer rate (ETRmax), higher phycocyanin content, and a higher affinity for inorganic carbon (K-0.5 DIC <= 8.4 +/- 0.7 mu M) during the growth period monitored in this study. This suggests that photosynthetic efficiency is a dominant physiological adaptation found in colonial Microcystis, thus promoting bloom sustainment. In addition, the high content of soluble and total carbohydrates in colonial Microcystis suggests that this phenotype may possess a higher ability to tolerate enhanced stress conditions when compared to unicellular (noncolonial) phenotypes. Therefore, high photosynthetic activities and high tolerance abilities may explain the bloom sustainment of colonial Microcystis in eutrophic lakes.

Theoretical calculations of the mode characteristics of an equilateral-triangle resonator (ETR) with a 10 mu m cavity side length show that the fundamental mode, with longitudinal mode index of 25, has a wavelength of 2.185 mu m and a longitudinal mode separation of 100 nm. This mode has a quality factor (similar to 2x10(5)) that is much larger than the first (similar to 5x10(4)) and second (similar to 3x10(4)) order modes, indicating that single fundamental mode lasing should be accessible over a broad wavelength tuning range. An electrically injected ETR based on this design is fabricated from an InGaAsSb/AlGaAsSb/GaSb, graded-index separate-confinement heterostructure, laser diode wafer with a 2.1 mu m emission wavelength. This device achieved single mode, continuous wave operation at 77 K with a threshold current of 0.5 mA and a single mode wavelength tuning range of 3.25 nm, which is accomplished by varying the injection current from 0.5 to 6.0 mA.
