To evaluate the antimicrobial efficacy of Clearfil SE Protect (CP) and Clearfil SE Bond (CB) after curing and rinsed against five individual oral microorganisms as well as a mixture of bacterial culture prepared from the selected test organisms. Bacterial suspensions were prepared from single species of Streptococcus mutans, Streptococcus sobrinus, Streptococcus gordonii, Actinomyces viscosus and Lactobacillus lactis, as well as mixed bacterial suspensions from these organisms. Dentin bonding system discs (6 mm×2 mm) were prepared, cured, washed and placed on the bacterial suspension of single species or multispecies bacteria for 15, 30 and 60 min. MTT, Live/Dead bacterial viability (antibacterial effect), and XTT (metabolic activity) assays were used to test the two dentin system's antibacterial effect. All assays were done in triplicates and each experiment repeated at least three times. Data were submitted to ANOVA and Scheffe's f-test (5%). Greater than 40% bacteria killing was seen within 15 min, and the killing progressed with increasing time of incubation with CP discs. However, a longer (60 min) period of incubation was required by CP to achieve similar antimicrobial effect against mixed bacterial suspension. CB had no significant effect on the viability or metabolic activity of the test microorganisms when compared to the control bacterial culture. CP was significantly effective in reducing the viability and metabolic activity of the test organisms. The results demonstrated the antimicrobial efficacy of CP both on single and multispecies bacterial culture. CP may be beneficial in reducing bacterial infections in cavity preparations in clinical dentistry.

The aim of this study was to evaluate the tensile strength of orthodontic wires bonded onto the enamel with cyanoacrylate ester. To obtain the specimens, 120 human premolars (extracted for orthodontic or periodontal reasons) were included in acrylic blocks of rapid polymerization with three teeth each. Four groups were formed with ten specimens each. In the specimens, a dental splint model was made with cyanoacrylate ester and round stainless steel wire. In groups I, II and III, cyanoacrylate ester was used with round steel wires, with variation in diameter: 0.014 inches; 0.016 inches and 0.018 inches, respectively. In group IV, round steel wire 0.018 inches was used with photo polymerizing resin composite with previous acid etching. The adhesive force of the materials was measured in two points under the action of the tensiometer (ETM-USA). The number of loose wires was counted along with those that remained fixed according to the different levels of force applied because of the direction of the tensile force (vertical or horizontal) and the diameter of the wire used. The data obtained were first submitted to a descriptive analysis and then submitted to a statistical analysis (Friedman's Test and Dunn's Test of Multiple Comparison - Epi-info 3.2). Within the limitations of the experimental conditions presented, the cyanoacrylate ester or 'Super Bonder (R)' maintained bonded to enamel and steel wires (0.016 and 0.018 inches) during the tensile strength tests under different levels of applied forces.

Purpose: To assess the effect of the composite surface conditioning on the microtensile bond strength of a resin cement to a composite used for inlay/onlay restorations. Materials and Methods: Forty-two blocks (6 × 6 × 4 mm) of a microfilled composite (Vita VMLC) were produced and divided into 3 groups (N = 14) by composite surface conditioning methods: Gr1 - etching with 37% phosphoric acid, washing, drying, silanization; Gr2 - air abrasion with 50-l̀m Al2O3 particles, silanization; Gr3 - chairside tribochemical silica coating (CoJet System), silanization. Single-Bond (one-step adhesive) was applied on the conditioned surfaces and the two resin blocks treated with the same method were cemented using RelyX ARC (dual-curing resin cement). The specimens were stored for 7 days in water at 37°C and then sectioned to produce nontrimmed beam samples, which were submitted to microtensile bond strength testing (μTBS). For statistical analysis (one-way ANOVA and Tukey's test, · = 0.05), the means of the beam samples from each luted specimen were calculated (n = 7). Results: μTBS values (MPa) of Gr2 (62.0 ± 3.9a) and Gr3 (60.5 ± 7.9a) were statistically similar to each other and higher than Gr1 (38.2 ± 8.9b). The analysis of the fractured surfaces revealed that all failures occurred at the adhesive zone. Conclusion: Conditioning methods with 50-l̀m Al2O3 or tribochemical silica coating allowed bonding between resin and composite that was statistically similar and stronger than conditioning with acid etching.

This study evaluated the influence of the dental substrates obtained after the use of different caries removal techniques on bonding of a self-etching system. Forty, extracted, carious, human molars were ground to expose flat surfaces containing caries-infected dentine surrounded by sound dentine. The caries lesions of the specimens were removed or not (control-G1) either by round steel burs and water-cooled, low speed, handpiece (G2), or by irradiation with an erbium, chromium:yttrium scandium gallium garnet (Er,Cr:YSGG) laser (2W, 20 Hz, 35.38 J/cm(2), fiber G4 handpiece with 0.2826 mm(2), non-contact mode at a 2 mm distance, 70% air/20% water-G3) or using a chemo-mechanical method (Carisolv-G4). Caries-infected, caries-affected and sound dentines were submitted to a bonding system followed by construction of a resin-based composite crown. Hour-glass shaped samples were obtained and submitted to a micro-tensile bond test. The bond strength data were compared by analysis of variance (ANOVA), complemented by Tukey`s test (P <= 0.05). The samples of sound dentine presented higher bond strengths than did samples of caries-affected dentine, except for the groups treated with the Er,Cr:YSGG laser. The highest bond strengths were observed with the sound dentine treated with burs and Carisolv. The bond strengths to caries-affected dentine were similar in all groups. Additionally, bonding to caries-affected dentine of the Er,Cr:YSGG laser and Carisolv groups was similar to bonding to caries-infected dentine. Thus, caries-affected dentine is not an adequate substrate for adhesion. Moreover, amongst the caries removal methods tested, the Er,Cr:YSGG laser irradiation was the poorest in providing a substrate for bonding with the tested self-etching system.

OBJECTIVES: To detect the influence of blood contamination (BC) on the bond strength (BS) of a self-etching bonding system (SES) to enamel and dentine. METHODS: 25 human molars were longitudinally sectioned on the mesio-distal axis in order to obtain 50 specimens, which were embedded in acrylic resin. At first, the specimens were ground to expose a flat surface of enamel, and a bond strength test was performed. Afterwards, the samples were ground again in order to obtain a flat surface of dentine. Ten groups (total: n=100) were assigned according to substrate (enamel and dentine), step in the bonding sequence when contamination occurred (before the acidic primer and after the bonding resin), and contamination treatment (dry or rinse and dry procedure). Fresh human blood was introduced either before or after SES application (Clearfil SE Bond) and treated with air drying, or by rinsing and drying following application. Composite resin (Filtek Z-250,3M ESPE) was applied as inverted, truncated cured cones that were debonded in tension. RESULTS: The mean tensile BS values (MPa) for enamel/dentine were 19.4/23.0 and 17.1/10.0 for rinse-and-dry treatment (contamination before and after SES, respectively); while the measurements for the dry treatment, 16.2/23.3 and 0.0/0.0 contamination before and after SES, respectively. CONCLUSIONS: It was determined that blood contamination impaired adhesion to enamel and dentine when it occurred after bond light curing. Among the tested contamination treatments, the rinse-and-dry treatment produced the highest bond strength with BC after SES application, but it was not sufficient to recover the BS in the contamination-free group.

The thesis describes the development and evaluation of epoxy resin as interfacial bonding agent for short Nylon-6 fiber elastomer composites. Epoxy resin is well known for its adhesive property. The potential use of it as interfacial bonding agent in short fiber composite is not explored yet. Three rubbers viz., acrylonitrile butadiene rubber (NBR), Neoprene rubber (CR) and styrene butadiene rubber (SBR) were selected and different fiber loading were tried. The resin concentration was optimized for each fiber loading with respect to cure characteristics and mechanical properties. Rheological characteristics and thermal degradation of the composites containing different fiber loading and different resin concentrations were studied in detail to find the effect of epoxy resin bonding system. The mechanical properties were studied in detail. The short Nylon -6 fiber improved most of the mechanical properties of all the three rubbers. Tensile strength showed a dip at 10 phr fiber loading in the case of CR while it was continuously increased with fiber loading in the case of NBR and SBR. All the composites showed anisotropy in mechanical properties. The epoxy resin is an effective bonding agent for short Nylon -6 fiber reinforced NBR and CR composites. Epoxy resin improved tensile strength, abrasion resistance and modulus of these composites. SEM studies confirmed the improved bonding of fiber and matrix in the presence of epoxy bonding agent. Epoxy resin was not effective as bonding agent in the case of short Nylon fiber- SBR composite. From the rheological studies of the composites with and without bonding agent it was observed that all the composite exhibited pseudoplasticity, which decreased with temperature. At higher shear rates all the mixes showed plug flow. SEM pictures showed that maximum orientation of fibers occured at a shear rate, just before the onset of plug flow. The presence of fiber reduced the temperature sensitivity of the flow at a given shear rate. Die swell was reduced in the presence of fiber. Shear viscosity of the composite was increased in the presence of resin. Die swell was increased in the presence of epoxy resin for composites at all shear rates. The thermal degradation of NBR and SBR composites with and without bonding agent followed single step degradation pattern. Thermal stability of the composites was improved in the presence of bonding agent. The degradation of virgin elastomer and the composites followed first order kinetics.

The work presented in this thesis is regarding the development and evaluation of new bonding agents for short polyester fiber - polyurethane elastomer composites. The conventional bonding system based on hexamethylenetetramine, resorcinol and hydrated silica was not effective as a bonding agent for the composite, as the water eliminated during the formation of the RF resin hydrolysed the urethane linkages. Four bonding agents based on MDI/'I‘DI and polypropyleneglycol, propyleneglycol and glycerol were prepared and the composite recipe was optimised with respect to the cure characteristics and mechanical properties. The flow properties, stress relaxation pattern and the thermal degradation characteristics of the composites containing different bonding agents were then studied in detail to evaluate the new bonding systems. The optimum loading of resin was 5 phr and the ratio of the -01 to isocyanate was 1:1. The cure characteristics showed that the optimum combination of cure rate and processability was given by the composite with the resin based on polypropyleneglycol/ glycerol/ 4,4’diphenylmethanediisocynate (PPG/GL/MDI). From the rheological studies of the composites with and without bonding agents it was observed that all the composites showed pseudoplastic nature and the activation energy of flow of the composite was not altered by the presence of bonding agents. Mechanical properties such as tensile strength, modulus, tear resistance and abrasion resistance were improved in the presence of bonding agents and the effect was more pronounced in the case of abrasion resistance. The composites based on MDI/GL showed better initial properties while composites with resins based on MDI/PPG showed better aging resistance. Stress relaxation showed a multistage relaxation behaviour for the composite. Within the-strain levels studied, the initial rate of relaxation was higher and the cross over time was lesser for the composite containing bonding agents. The bonding agent based on MDI/PPG/GL was found to be a better choice for improving stress relaxation characteristics with better interfacial bonding. Thennogravimetirc analysis showed that the presence of fiber and bonding agents improved the thennal stability of the polyurethane elastomer marginally and it was maximum in the case of MDI / GL based bonding agents. The kinetics of degradation was not altered by the presence of bonding agents

Objective. This study evaluated histopathologically the response of pulp and periradicular tissues after pulp capping with an all-in-one self-etching adhesive system in dogs` teeth. Study design. Forty teeth of 4 dogs were assigned to 3 groups according to the pulp capping material: G1 (n = 20): self-etching adhesive system; G2 (n = 10): Ca(OH)(2); G3 (n = 10): zinc oxide-eugenol. The animals were killed 7 and 70 days after pulp capping. The pieces containing the pulp-capped teeth were removed and processed for histologic analysis. Results. At 7 days, no dentin bridge formation was observed; G1 and G3 exhibited inflammatory pulpal alterations, whereas G2 presented only mild inflammatory infiltrate in the pulp tissue adjacent to the capping material, the remainder being intact. At 70 days, no specimen in G1 or G3 presented dentin bridge formation. The remaining pulp tissue exhibited severe inflammatory alterations and areas of necrosis. In G2, all specimens showed dentin bridge formation and absence of inflammation and mineralized tissue resorption. No bacteria were identified using Brown and Brenn staining techniques in all 3 groups at any observation period. Conclusion. According to the conditions of this study, direct pulp capping with the self-etching adhesive system did not allow pulp tissue repair and failed histopathologically in 100% of the cases. OBJECTIVE: The purpose of this study was to assess by means of scanning electron microscopy (SEM) the remaining adhesive interface after debonding orthodontic attachments bonded to bovine teeth with the use of hydrophilic and hydrophobic primers under different dental substrate moisture conditions. MATERIAL AND METHODS: Twenty mandibular incisors were divided into four groups (n=5). In Group I, bracket bonding was performed with Transbond MIP hydrophilic primer and Transbond XT adhesive paste applied to moist substrate, and in Group II a bonding system comprising Transbond XT hydrophobic primer and adhesive paste was applied to moist substrate. Brackets were bonded to the specimens in Groups III and IV using the same adhesive systems, but on dry dental enamel. The images were qualitatively assessed by SEM. RESULTS: The absence of moisture in etched enamel enabled better interaction between bonding materials and the adamantine structure. The hydrophobic primer achieved the worst micromechanical interlocking results when applied to a moist dental structure, whereas the hydrophilic system proved versatile, yielding acceptable results in moist conditions and excellent interaction in the absence of contamination. CONCLUSION: The authors assert that the best condition for the application of primers to dental enamel occurs in the absence of moisture.

Nanoscale silica was synthesized by precipitation method using sodium silicate and dilute hydrochloric acid under controlled conditions. The synthesized silica was characterized by Scanning Electron Microscopy (SEM), Transmission Electron Microscopy (TEM), BET adsorption and X-Ray Diffraction (XRD). The particle size of silica was calculated to be 13 nm from the XRD results and the surface area was found to be 295 m2/g by BET method. The performance of this synthesized nanosilica as a reinforcing filler in natural rubber (NR) compound was investigated. The commercial silica was used as the reference material. Nanosilica was found to be effective reinforcing filler in natural rubber compound. Filler-matrix interaction was better for nanosilica than the commercial silica. The synthesized nanosilica was used in place of conventional silica in HRH (hexamethylene tetramine, resorcinol and silica) bonding system for natural rubber and styrene butadiene rubber / Nylon 6 short fiber composites. The efficiency of HRH bonding system based on nanosilica was better. Nanosilica was also used as reinforcing filler in rubber / Nylon 6 short fiber hybrid composite. The cure, mechanical, ageing, thermal and dynamic mechanical properties of nanosilica / Nylon 6 short fiber / elastomeric hybrid composites were studied in detail. The matrices used were natural rubber (NR), nitrile rubber (NBR), styrene butadiene rubber (SBR) and chloroprene rubber (CR). Fiber loading was varied from 0 to 30 parts per hundred rubber (phr) and silica loading was varied from 0 to 9 phr. Hexa:Resorcinol:Silica (HRH) ratio was maintained as 2:2:1. HRH loading was adjusted to 16% of the fiber loading. Minimum torque, maximum torque and cure time increased with silica loading. Cure rate increased with fiber loading and decreased with silica content. The hybrid composites showed improved mechanical properties in the presence of nanosilica. Tensile strength showed a dip at 10 phr fiber loading in the case of NR and CR while it continuously increased with fiber loading in the case of NBR and SBR. The nanosilica improved the tensile strength, modulus and tear strength better than the conventional silica. Abrasion resistance and hardness were also better for the nanosilica composites. Resilience and compression set were adversely affected. Hybrid composites showed anisotropy in mechanical properties. Retention in ageing improved with fiber loading and was better for nanosilica-filled hybrid composites. The nanosilica also improved the thermal stability of the hybrid composite better than the commercial silica. All the composites underwent two-step thermal degradation. Kinetic studies showed that the degradation of all the elastomeric composites followed a first-order reaction. Dynamic mechanical analysis revealed that storage modulus (E’) and loss modulus (E”) increased with nanosiica content, fiber loading and frequency for all the composites, independent of the matrix. The highest rate of increase was registered for NBR rubber.

The thesis deals with the development of short nylon fiber-reclaimed rubber/elastomer composites. Three rubbers viz, natural rubber, acrylonitrile butadiene rubber and styrene butadiene rubber were selected and were partially replaced with reclaimed rubber. The blend ratio was optimized with respect to cure characteristics and mechanical properties. Reclaimed rubber replaced 40 parts of NR and SBR and 20 parts of NBR without much affecting the properties. These blends were then reinforced with short nylon fibers. The mechanical properties of the composites were studied in detail. In all the cases the tensile strength, tear strength and the abrasion resistance increased with increase in fiber content. In the case of NRlreclaimed rubber blends, the tensile strength-fiberloading relationship was non-linear where as in the case of NBRlreclaimed rubber blends and SBRlreclaimed rubber blends the tensile strength-fiber loading relationship was linear. All the composites showed anisotropy in mechanical properties. The effect of bonding system on the composite properties was also studied with respect to cure characteristics and mechanical properties. For this, a 20 phr fiber loaded reclaimed rubber/elastomer composites were selected and the effect of MDI/PEG resin system was studied. The resin used was 5 phr and the resin ratios used were 0.67: I, 1:1, 1.5:1 and 2:1. The bonding system improved the tensile strength, tear strength and abrasion resistance. The best results are with SBRlreclaimed rubber-short nylon fiber composites. The optimized resin ratio was 1:1 MDI/PEG for all the composites.

The purpose of this study was to evaluate the effectiveness of different light-curing units on the bond strength (push-out) of glass fiber posts in the different thirds of the root (cervical, middle and apical) with different adhesive luting resin systems (dual-cure total-etch; dual-cured and self-etch bonding system; and dual-cure self-adhesive cements), Disks of the samples (n = 144) were used, with approximately 1 mm of thickness of 48 bovine roots restored with glass fiber posts, that were luted with resin cements photo-activated by halogen LCU (QTH, Optilux 501) and blue LED (Ultraled), with power densities of 600 and 550 mW/cm(2), respectively. A universal testing machine (MTS 810 Material Test System) was used with a 1 mm diameter steel rod at cross-head speed of 0.5 mm/min until post extrusion, with load cell of 50 kg, for evaluation of the push-out strength in the different thirds of each sample. The push-out strength values in kgf were converted to MPa and analyzed through Analysis of Variance and Tukey`s test, at significance level of 5%. The results showed that there were no statistical differences between the QTH and LED LCUs. The self-adhesive resin cement had lower values of retention. The total-etch and self-adhesive system resin cements seem to be a possible alternative for glass fiber posts cementation into the radicular canal and the LED LCU can be applied as an alternative to halogen light on photo-activation of dual-cured resin cements.
