Pollen transport to a receptive stigma can be facilitated through different pollinators, which submits the pollen to different selection pressures. This study aimed to associate pollen and stigma morphology with zoophily in species of the tribe Phaseoleae. Species of the genera Erythrina, Macroptilium and Mucuna with different pollinators were chosen. Pollen grains and stigmas were examined under light microscopy (anatomy), scanning electronic microscopy (surface analyses) and transmission electronic microscopy (ultrastructure). The three genera differ in terms of pollen wall ornamentation, pollen size, pollen aperture, thickness of the pollen wall, amount of pollenkitt, pollen hydration status and dominant reserves within the pollen grain, while species within each genus are very similar in most studied characteristics. Most of these features lack relationships to pollinator type, especially in Erythrina and Mucuna. Pollen reserves are discussed on a broad scale, according to the occurrence of protein in the pollen of invertebrate- or vertebrate-pollinated species. Some pollen characteristics are more associated to semi-dry stigma requirements. This apical, compact, cuticularised and secretory stigma occurs in all species investigated. We conclude that data on pollen and stigma structure should be included together with those on floral morphology and pollinator behaviour for the establishment of functional pollination classes.

Neural mechanisms underlying the onset and maintenance of epileptic seizures involve alterations in inhibitory and/or excitatory neurotransmitter pathways. Thus, the prospecting of novel molecules from natural products that target both inhibition and excitation systems has deserved interest in the rational design of new anticonvulsants. We isolated the alkaloids (+)-erythravine and ( +)-11-alpha-hydroxyerythravine from the flowers of Erythrina mulungu and evaluated the action of these compounds against chemically induced seizures in rats. Our results showed that the administration of different doses of (+)-erythravine inhibited seizures evoked by bicuculline, pentylenetetrazole, and kainic acid at maximum of 80, 100, and 100%, respectively, whereas different doses of (+)-11-alpha-hydroxy-erythravine inhibited seizures at a maximum of 100% when induced by bicuculline, NMDA, and kainic acid, and, to a lesser extent, PTZ (60%). The analysis of mean latency to seizure onset of nonprotected animals, for specific doses of alkaloids, showed that (+)-erythravine increased latencies to seizures induced by bicuculline. Although (+)-erythravine exhibited very weak anticonvulsant action against seizures induced by NMDA, this alkaloid increased the latency in this assay. The increase in latency to onset of seizures promoted by (+)-11-alpha-hydroxy-erythravine reached a maximum of threefold in the bicuculline test. All animals were protected against death when treated with different doses of (+)-11-alpha-hydroxy-erythravine in the tests using the four chemical convulsants. Identical results were obtained when using (+)-erythravine in the tests of bicuculline, NMDA, and VIZ, and, to a lesser extent, kainic acid. Therefore, these data validate the anticonvulsant properties of the tested alkaloids, which is of relevance in consideration of the ethnopharmacological/biotechnological potential of E. mulungu. O cultivo in vitro de embriões zigóticos é uma técnica promissora para se avançar no estudo do desenvolvimento embrionário e da quebra da dormência de sementes. Diante do exposto, objetivou-se, com este trabalho, avaliar o efeito dos reguladores vegetais 6-benzilaminopurina (BAP) e ácido naftalenoacético (ANA) no potencial morfogenético, in vitro, de embriões zigóticos de mulungu. Embriões zigóticos maduros, oriundos de sementes foram utilizados inteiros, ou seccionados em plúmula, região intermediária e radícula, sendo posteriormente inoculados em meio de cultura WPM, suplementado com combinações de BAP (0,0; 2,0; 4,0; 8,0; 12,0 e 16,0 µM) e ANA (0,0; 1,0 e 2,0 µM), acrescido de 87,64 mM de sacarose e solidificado com 0,7% de ágar. Após 30 dias, avaliaram-se a percentagem de regeneração dos embriões e ápice plumular, o número de brotos, o número de folhas, o comprimento da parte aérea dos brotos, o número de raízes e a percentagem de formação de calos oriundos da região intermediária e da radícula. É possível a regeneração in vitro de mulungu, a partir dos explantes plúmula e embriões zigóticos inteiros, cultivados em meio de cultura WPM, suplementado com 4,0 µM de BAP. Regiões intermediárias e da radícula promoveram a formação de calos compactos (96,06%), na combinação de 10,63 µM BAP e 2,0 µM de ANA.

Estudos recentes têm evidenciado a importância de flores como recurso alimentar de diversas espécies de aves neotropicais. Este estudo teve o objetivo de listar espécies e descrever o comportamento das aves que se alimentaram de recursos florais de Erythrina fusca (Fabaceae) no Pantanal Mato-Grossense. Para isso, foram acompanhadas aves que se alimentaram em 14 árvores floridas dessa espécie em um total de cerca de 25 horas de observação. Como resultados, 20 espécies de aves pertencentes a sete famílias taxonômicas foram observadas alimentando-se de recursos florais de E. fusca. Apesar de várias espécies de aves terem apresentado comportamentos destrutivos em maior proporção, algumas espécies, sobretudo das famílias Trochilidae e Icteridae, apresentaram estratégias alimentares compatíveis com o transporte de pólen. O cruzamento dos dados obtidos neste estudo com os da literatura indicam que a família Icteridae pode ter relevante papel na polinização dessa espécie vegetal.

O objetivo deste trabalho foi comparar a biometria de frutos e sementes, a embebição e germinação de duas populações de Erythrina velutina coletadas na região semiárida do Estado de Pernambuco, nas áreas de caatinga (Alagoinha) e no brejo de altitude (Poção). Os frutos e as sementes foram avaliados quanto ao comprimento, largura, espessura (cm) e peso (g). Com relação às sementes, foram analisados o número por fruto, o volume (cm³) e a embebição, bem como a germinação com e sem escarificação mecânica, na presença constante de luz a 25 ºC. Os frutos e sementes do brejo apresentaram os valores assim discriminados: 9,65 x 1,57 x 1,22 cm, 1,43 g e 1,34 x 0,92 x 0,78 cm, 0,46 g; e na caatinga: 6,85 x 1,19 x 1,03 cm, 0,21 g e 1,06 x 0,63 x 0,63 cm e 0,26 g. O número de sementes e o volume no brejo e caatinga foram de 2,33 e 1,38 cm³ e 1,40 e 0,23 cm³, respectivamente. Todas as sementes escarificadas apresentaram maiores valores de embebição e porcentagem de germinação. Quanto à velocidade de germinação, foi maior na caatinga. Esses resultados indicam que as condições ambientais do brejo de altitude favorecem o desenvolvimento dos frutos e das sementes.

Some environmental factors, including water availability, may influence seed germination. This study investigated the germination of E. velutina seeds submitted to different osmotic potentials and mobilization of reserves during water-stress. Scarified seeds were arranged in paper rolls and soaked in solutions of Polyethylene Glycol (PEG) prepared in osmotic potentials 0.0, -0.2, -0.4, -0.6, and -0.8 MPa and kept into a seed germinator, at 25 °C, and 12/12 h photoperiod (L/D), during 10 days. The percentage, mean time, mean speed, germination speed index; as well as the germination uniformity coefficient were assessed. During germination process the total soluble sugars, reducing sugars, soluble protein, and total amino acids were quantified in the cotyledon, hypocotyl and radicle of soaked seeds and cotyledons of quiescent seeds (control). There was influence of osmotic potential on E. velutina seed germination. The germination percentage remained at high levels until -0.6 MPa and above this osmotic potential there has been no germination. The mobilization of stored reserves of carbon and nitrogen in E. velutina seeds was also influenced by water-stress. There was sensitiveness between -0.2 and -0.6 MPa; however, the degradation and the mobilization of reserves was slower when the osmotic potential decreased.

In this paper of the catalogue of south brazilian arboreal pollen grains, the autor deals with the Papilionatae. The Mimosoideae and Caesalpinioideae are yet in preparation, so that a discussion of the three subfamilies (or families) is not possible. In relation with the systematical subdivision of the Papilionatae, we found a large correspondence with the morphology of the present pollen grains. The group of Phaseoleae contains the genera Mucuna, Erythrina and Dioclea; the grains of the studied species are very different one from another; the first of the genera possesses very volumous grains, with three colpori and a reticulated superficies; the second has three-porated pollen grains with a large reticulated superficies, and the third, Dioclea, is yet different; it possesses oblated grains, each three-colporated, with a thick sexine and a psilated superficies. So, we can say, that Phaseoleae is a erypalynous group. Dalbergieae, with the genera: Andira, Dalbergia, Lonchocarpus, Machaerium, Platymiscium and Pterocarpus (and Dahlstedtia, the only exception), has very uniform pollen grains, and may be considered stenopalynous. It is not possible to include the genus Dahlstedtia into this group. A little exception is represented by Pterocarpus violaceus, because of the reticulated sexine of its grains, while the others, also three-colporated, possess a tectate-reticulated sexine. The genera Myrocarpus and Ormosia, from Sophoreae, are very more similar to the Dalbergieae as to any other genus of the Phaseoleae.

We investigated the effect of acute oral treatment with a water-alcohol extract of the inflorescence of Erythrina mulungu (EM, Leguminosae-Papilionaceae) (100, 200 and 400 mg/kg) on rats submitted to different anxiety models: the elevated T-maze (for inhibitory avoidance and escape measurements), the light/dark transition, and the cat odor test. These models were selected for their presumed capacity to demonstrate specific subtypes of anxiety disorders as recognized in clinical practice. Treatment with 200 mg/kg EM impaired avoidance latencies (avoidance 1 - 200 mg/kg EM: 18 ± 7 s, control group: 40 ± 9 s; avoidance 2 - 200 mg/kg EM: 15 ± 4 s, control group: 110.33 ± 38 s) in a way similar to the reference drug diazepam (avoidance 1: 3 ± 0.79 s; avoidance 2: 3 ± 0.76 s), without altering escape. Additionally, the same treatments increased the number of transitions (200 mg/kg EM: 6.33 ± 0.90, diazepam: 10 ± 1.54, control group: 2.78 ± 0.60) between the two compartments and the time spent in the lighted compartment in the light/dark transition model (200 mg/kg EM: 39 ± 7 s; diazepam: 61 ± 9 s; control group: 14 ± 4 s). The dose of 400 mg/kg EM also increased this last measurement (38 ± 8 s). These results were not due to motor alterations since no significant effects were detected in the number of crossings or rearings in the arena. Furthermore, neither EM nor diazepam altered the behavioral responses of rats to a cloth impregnated with cat odor. These observations suggest that EM exerts anxiolytic-like effects on a specific subset of defensive behaviors, particularly those that have been shown to be sensitive to low doses of benzodiazepines.

Prosopis rubriflora and Prosopis ruscifolia are important species in the Chaquenian regions of Brazil. Because of the restriction and frequency of their physiognomy, they are excellent models for conservation genetics studies. The use of microsatellite markers (Simple Sequence Repeats, SSRs) has become increasingly important in recent years and has proven to be a powerful tool for both ecological and molecular studies. In this study, we present the development and characterization of 10 new markers for P. rubriflora and 13 new markers for P. ruscifolia. The genotyping was performed using 40 P. rubriflora samples and 48 P. ruscifolia samples from the Chaquenian remnants in Brazil. The polymorphism information content (PIC) of the P. rubriflora markers ranged from 0.073 to 0.791, and no null alleles or deviation from Hardy-Weinberg equilibrium (HW) were detected. The PIC values for the P. ruscifolia markers ranged from 0.289 to 0.883, but a departure from HW and null alleles were detected for certain loci; however, this departure may have resulted from anthropic activities, such as the presence of livestock, which is very common in the remnant areas. In this study, we describe novel SSR polymorphic markers that may be helpful in future genetic studies of P. rubriflora and P. ruscifolia.

Aeschynomene falcata is an important forage species; however, because of low seed production, it is underutilized as forage species. Aeschynomene is a polyphyletic genus with a challenging taxonomic position. Two subgenera have been proposed, and it is suggested that Aeschynomene can be split in 2 genera. Thus, new markers, such as microsatellite sequences, are desirable for improving breeding programs for A. falcata. Based on transferability and in situ localization, these microsatellite sequences can be applied as chromosome markers in the genus Aeschynomene and closely related genera. Here, we report the first microsatellite library developed for this genus; 11 microsatellites were characterized, with observed and expected heterozygosities ranging from 0.0000 to 0.7143 and from 0.1287 to 0.8360, respectively. Polymorphic information content varied from 0.1167 to 0.7786. The departure from Hardy-Weinberg equilibrium may have resulted from frequent autogamy, which is characteristic of A. falcata. Of the 11 microsatellites, 9 loci were cross-amplified in A. brevipes and A. paniculata and 7 in Dalbergia nigra and Machaerium vestitum. Five of these 7 cross-amplified microsatellites were applied as probes during the in situ hybridization assay and 2 showed clear signals on A. falcata chromosomes, ensuring their viability as chromosome markers.

Multidrug-resistant microbial infections represent an exponentially growing problem affecting communities worldwide. Photodynamic therapy is a promising treatment based on the combination of light, oxygen, and a photosensitizer that leads to reactive oxygen species production, such as superoxide (type I mechanism) and singlet oxygen (type II mechanism) that cause massive oxidative damage and consequently the host cell death. Indigofera genus has gained considerable interest due its mutagenic, cytotoxic, and genotoxic activity. Therefore, this study was undertaken to investigate the effect of crude extracts, alkaloidal fraction, and isolated substance derived from Indigofera truxillensis in photodynamic antimicrobial chemotherapy on the viability of bacteria and yeast and evaluation of mechanisms involved. Our results showed that all samples resulted in microbial photoactivation in subinhibitory concentration, with indigo alkaloid presenting a predominant photodynamic action through type I mechanism. The use of CaCl2 and MgCl2 as cell permeabilizing additives also increased gram-negative bacteria susceptibility to indigo.

Chromosome numbers of 11 South-Brazilian species of Adesmia were determined. The cytological preparations were obtained by squashing cells of root tips, using the acetic-orcein method. The chromosome number for all the species studied was 2n=20, excepting A. incana var. incana with 2n=ca.40. The counts are new for nine species, and the other two agree with the literature. It is suggested x=10 as the basic number for the genus. Up to the present only four species were cited as polyploid.

This work is a floristic survey of arboreous and climbing shrub taxa of Papilionoideae in a semideciduos forest at Parque Estadual do Rio Doce, eastern Minas Gerais State. Field work was carried out from April/1998 to May/1999, when montly visits were conducted to collect botanical material along trails and of the main road. The floristic survey resulted in 22 taxa belonging to eight genera. An identification key, descriptions, illustrations and comments on the analyzed taxa are presented.
