Background Et aims: To investigate the effect that early weaning associated with the ingestion of either a glutamine-free or supplemented diet has on the functioning of peritoneal. macrophages, hematopoiesis and nutritional status of mice. Methods: Swiss Webster mice were early weaned on their 14th day of life and distributed to two groups, being fed either a glutamine-free diet (-GLN) or a glutamine-supplemented diet (+GLN). Animals belonging to a control group (CON) were weaned on their 21st day of life. Results: The -GLN and +GLN groups had a lower lean body mass, carcass protein and ash content, plasma glutamine concentration and lymphocyte counts both in the peripheral blood and bone marrow when compared to the CON group (P < 0.05). Dietary supplementation with glutamine reversed both the lower concentrations of protein and DNA in the muscle and liver, as well. as the reduced capacity of spreading and synthesizing nitric oxide, hydrogen peroxide, TNF-alpha, IL-1 beta and IL-6 in cultures of peritoneal. macrophages obtained from the -GLN group (P < 0.05). Conclusion: These data indicate that the ingestion of glutamine modulates the function of peritoneal macrophages in early weaned mice. However, a glutamine-supplemented diet cannot substitute maternal milk in respect to immunological and metabolic parameters. Infants who are breast-fed have been shown to have a lower incidence of certain infectious diseases compared with formula-fed infants. Glutamine is one of the most abundant amino acids found in maternal milk and it is essential for the function of immune system cells such as macrophages. The purpose of this study was to investigate the effect of glutamine supplementation on the function of peritoneal macrophages and on hemopoiesis in early-weaned mice inoculated with Mycobacterium bovis bacillus Calmette-Guerin (BCG). Mice were wearied at 14 d of age and distributed to 2 groups and fed either a glutamine-free diet (n = 16) or a glutamine-supplemented diet (+Gln (n = 16). Both diets were isonitrogenous (with addition of a mixture of nonessential amino acids) and isocaloric. At d 21, 2 subgroups of mice (n = 16) were intraperitoneally injected with BCG and all mice were killed at d 28. Plasma, muscle and liver glutamine concentrations and muscle glutamine synthetase activity were not affected by diet or inoculation with BCG. The +GIn diet led to increased leukocyte and lymphocyte counts in the peripheral blood (P < 0.05) and granulocyte and lymphocyte counts in the bone marrow and spleen (P < 0.05). The +GIn diet increased spreading and adhesion capacities, hydrogen peroxide, nitric oxide, and tumor necrosis factor-alpha (TNF alpha) syntheses and the phagocytic and fungicidal activity of peritoneal macrophages (P < 0.05). The interaction between the +GIn diet and BCG inoculation increased the area under the curve of interleukin (IL)-1 beta and TNF alpha syntheses (P < 0.05). In conclusion, the intake of glutamine increases the function of peritoneal macrophages and hemopoiesis in early-weaned and BCG-inoculated mice. These data have important implications for the design of breast milk substitutes for human infants.

Objective: Glutamine is one of the most abundant amino acids found in maternal milk, and its concentration increases throughout lactation. Because glutamine is essential for macrophage functionality, it is hereby suggested that early weaning in conjunction with the absence of glutamine consumption impairs the functioning of macrophages, which could in turn be reversed with an in vitro supplementation with glutamine. Methods: Swiss Webster mice were early weaned at 14 d of age (EW group) or at 21 d of age (control group, n = 8 per group). The EW group was fed a glutamine-free diet from days 14 to 21 of life. Results: Mice in the EW group presented a significant decrease in plasma and muscle concentrations of glutamine and an increase in the activity of glutamine synthetase in the muscle. Peritoneal macrophages obtained from animals in the EW group presented a significant increase in the production of interleukin (IL)-10 and a significant decrease in the synthesis of IL-1 beta, IL-6, tumor necrosis factor-a, nitric oxide, and hydrogen peroxide and in their ability to adhere, spread, phagocytize, and kill fungi. Glutamine in vitro supplementation reversed the decrease in IL-6, nitric oxide, and hydrogen peroxide synthesis and the decrease in the capacity to adhere, spread, and phagocytize in animals of the EW group. Conclusion: These new. data may imply that a lack of glutamine intake in early weaned mice hampers the functioning of peritoneal macrophages. (C) 2008 Elsevier Inc. All rights reserved.

Objectives: Early weaning (EW) increases proliferation of the gastric epithelium in parallel with higher expression of transforming growth factor alpha and its receptor epidermal growth factor receptor (EGFR). The primary objective of the present study was to examine involvement of EGFR signalling in regulating mucosal cell proliferation during the early weaning period. Materials and methods: Fifteen-day-old rats were split into two groups: suckling (control) and EW, in which pups were separated from the dam. Animals were killed daily until the 18th day, 3 days after onset of treatment. To investigate the role of EGFR in proliferation control, EW pups were injected with AG1478, an EGFR inhibitor; signalling molecules, proliferative indices and cell cycle-related proteins were evaluated. Results: EW increased ERK1/2 and Src phosphorylation at 17 days, but p-Akt levels were unchanged. Moreover, at 17 days, AG1478 administration impaired ERK phosphorylation, whereas p-Src and p-Akt were not altered. AG1478 treatment reduced mitotic and DNA synthesis indices, which were determined on HE-stained and BrdU-labelled sections. Finally, AG1478 injection decreased p21 levels in the gastric mucosa at 17 days, while no changes were detected in p27, cyclin E, CDK2, cyclin D1 and CDK4 concentrations. Conclusions: EGFR is part of the mechanism that regulates cell proliferation in rat gastric mucosa during early weaning. We suggest that such responses might depend on activation of MAPK and/or Src signalling pathways and regulation of p21 levels.

Early weaning is a stressful event characterized by a transient period of intestinal atrophy that may be mediated by reduced secretion of glucagon-like peptide (GLP) 2. We tested whether enterally fed bile acids or plant sterols could increase nutrient-dependent GLP-2 secretion and improve intestinal adaptation in weanling pigs. During the first 6 d after weaning, piglets were intragastrically infused once daily with either deionized water -control-, chenodeoxycholic acid -CDC; 60mg/kg body weight-, or b-sitoesterol -BSE; 100 mg/kg body weight-. Infusing CDC increased plasma GLP-2 -P menor que 0.05- but did not affect plasma GLP-1 and feed intake. The intestinal expression of Glp2r -glucagon-like peptide 2 receptor-, Asbt -sodium-dependent bile acid transporter-, Fxr -farnesoid X receptor-, and Tgr5 -guanosine protein?coupled bile acid receptor- genes were not affected by CDC treatment. The intragastric administration of CDC did not alter the weight and length of the intestine, yet increased the activation of caspase-3 in ileal villi -P menor que 0.02- and the expression of Il6 -interleukin 6; P menor que 0.002- in the jejunum. In contrast, infusing BSE did not affect any of the variables that were measured. Our results show that the enteral administration of the bile acid CDC potentiates the nutrient-induced secretion of endogenous GLP-2 in early-weaned pigs. Bile acid?enhanced release of GLP-2, however, did not result in improved intestinal growth, morphology, or inflammation during the postweaning degenerative phase.

The aim of this study was to evaluate the effect of glutamine on the expression of proteins involved in the nuclear factor-kappaB (NF-kappa B) signaling pathway of murine peritoneal macrophages. Since glutamine is essential for the normal functioning of macrophages, it was hypothesized that in vitro glutamine supplementation would increase NF-kappa B activation. Peritoneal macrophages were pretreated with glutamine (0, 0.6, 2 and 10 mM) before incubation with lipopolysaccharide (LPS), and the effects of glutamine on the production of tumor necrosis factor-alpha and on the expression and activity of proteins involved in the NF-kappa B signaling pathway were studied by an enzyme linked immuno-sorbent assay, Western blotting, and an electrophoretic mobility shift assay. Glutamine treatment (2 and 10 mM) increased the activation of NF-kappa B in LPS-stimulated peritoneal macrophages (P < 0.05). In non-stimulated cells, glutamine treatment (2 and 10 mM) significantly reduced I kappa B-alpha protein expression (P < 0.05). Glutamine modulates NF-kappa B signaling pathway by reducing the level of I kappa B-alpha, leading to an increase in NF-kappa B within the nucleus in peritoneal macrophages.

Pancreatic β-cells are highly sensitive to suboptimal or excess nutrients, as occurs in protein-malnutrition and obesity. Taurine (Tau) improves insulin secretion in response to nutrients and depolarizing agents. Here, we assessed the expression and function of Cav and KATP channels in islets from malnourished mice fed on a high-fat diet (HFD) and supplemented with Tau. Weaned mice received a normal (C) or a low-protein diet (R) for 6 weeks. Half of each group were fed a HFD for 8 weeks without (CH, RH) or with 5% Tau since weaning (CHT, RHT). Isolated islets from R mice showed lower insulin release with glucose and depolarizing stimuli. In CH islets, insulin secretion was increased and this was associated with enhanced KATP inhibition and Cav activity. RH islets secreted less insulin at high K(+) concentration and showed enhanced KATP activity. Tau supplementation normalized K(+)-induced secretion and enhanced glucose-induced Ca(2+) influx in RHT islets. R islets presented lower Ca(2+) influx in response to tolbutamide, and higher protein content and activity of the Kir6.2 subunit of the KATP. Tau increased the protein content of the α1.2 subunit of the Cav channels and the SNARE proteins SNAP-25 and Synt-1 in CHT islets, whereas in RHT, Kir6.2 and Synt-1 proteins were increased. In conclusion, impaired islet function in R islets is related to higher content and activity of the KATP channels. Tau treatment enhanced RHT islet secretory capacity by improving the protein expression and inhibition of the KATP channels and enhancing Synt-1 islet content.

OBJETIVO: Avaliar o efeito do desmame precoce sobre o ganho de peso e a composição corporal de camundongos adultos jovens. MÉTODOS: Camundongos Swiss Webster, machos, foram desmamados precocemente (14º dia de vida) ou amamentados até o 21º dia de vida (grupo controle). Após o desmame, os animais foram alimentados com ração elaborada para roedores em crescimento até o 63º dia de vida, quando então foram sacrificados. RESULTADOS: O peso corporal dos animais do grupo desmamado de forma precoce foi significantemente maior no 28º, 35º e no 63º dias de vida em relação ao grupo controle (p<0,05). Porém, o consumo de ração não diferiu entre os grupos. A concentração sérica de proteínas totais, albumina e ferro, bem como a concentração hepática, muscular e cerebral de proteínas, ácido desoxirribonucléico e a relação proteína/ácido ribonucléico, não diferiram significantemente entre os grupos. O grupo desmamado precocemente apresentou maior quantidade absoluta de massa magra, lipídeos, proteínas e cinzas, em comparação ao grupo controle (p<0,05). A quantidade relativa de umidade, lipídeos, massa magra, proteínas e cinzas não diferiu entre os grupos. CONCLUSÃO: O desmame precoce, associado à ingestão de ração elaborada para roedores em crescimento, resultou em aumento do ganho de peso, porém não afetou a composição corporal de camundongos adultos.

The influence of early weaning and concentrate supplementation strategies on sward characteristics, forage chemical composition and lamb productivity were evaluated in four production systems on Tifton-85 pasture: suckling lambs not supplemented until slaughter; suckling lambs supplemented with concentrate in creep feeding until slaughter; early-weaned lambs not supplemented until slaughter; and early-weaned lambs supplemented with concentrate until slaughter. Structural, morphological and productive characteristics of pasture were measured. The forage was chemically analyzed to estimate its composition. Lambs average daily gain and productivity were calculated. Sward height, forage and morphological components mass were lower in systems without weaning. Forage production was higher in systems with supplementation. Higher levels of neutral and acid detergent fiber were observed in forage ingested by lambs in creep feeding and by weaned and unsupplemented lambs. Average daily gain was higher for lambs in creep feeding (275 g/d) and lower for the weaned and unsupplemented animals (57 g/d). Productivity was higher for weaned and supplemented lambs (21 kg lamb body weight, BW gain/ha/d). Lower productivity was observed in systems without supplementation (5 kg lamb BW gain/ha/d on average). Ewes modify the sward conditions improving the pasture characteristics and the quality of forage produced. Changes in sward conditions affect the chemical composition of forage ingested by lambs. Early weaning may be an alternative to maximize pasture utilization in small areas. Concentrate supplementation may increase lamb performance and productivity in grazing systems. If the objective is to improve lamb individual performance, creep feeding should be used.

A study was conducted to evaluate early weaning of beef calves at 60-70 days of age on feedlot performance and carcass characteristics. One hundred twenty steer calves sired by either Simmental or Angus sires were weaned at an average age of 67 (early weaned, EW) or 147 (late weaned, LW) days. Calves were allotted to 16 feedlot pens by weaning treatment and sire breed at approximately 750-800 lb. EW calves were heavier (P < .05) in initial feedlot weight. There were no differences due to weaning age on daily gain, dry matter intake, feed efficiency or slaughter weights. Simmental steers required more days on feed than Angus steers (P < .05). Early-weaned calves had a higher percent intramuscular fat (5.7 vs. 5.1%), higher average marbling scores (Small78 vs. Small20, P < .05), a higher percentage of cattle grading average USDA Choice and higher (38% vs. 14%, P < .05) and a higher percentage of USDA Prime (10% vs. 0%, P < .05). These data confirm observations in previous studies that early weaning and placing calves on a higher grain diet improves marbling at slaughter. In this study, the effect was shown in calves weaned at an average of 67 days.

Um experimento foi realizado com o objetivo de verificar os efeitos da indução do consumo de água com edulcorante, na fase de berçário (10 aos 30 dias de idade), sobre o desempenho de 270 leitões (machos e fêmeas) submetidos ao desmame precoce segregado e os efeitos residuais nas fases de pré-creche (30 aos 45 dias) e de creche (45 aos 62 dias de idade). Três tratamentos com dois sexos e três repetições (fatorial 3 x 2) foram utilizados dos 10 aos 30 dias de idade dos leitões, sendo: T1- água sem edulcorante, T2 - água com edulcorante e T3 - água com e sem edulcorante oferecidos simultaneamente em bebedouros distintos. Foram avaliados o ganho diário de peso, o consumo diário de ração e a conversão alimentar. O consumo diário de água somente foi medido na fase de berçário. A incidência de diarréia foi observada até o 10º dia após o desmame. Aos 22 dias de idade, seis leitões por tratamento foram sacrificados para avaliação da altura das vilosidades e profundidade das criptas da mucosa do duodeno e jejuno. Não foram observadas diferenças para as variáveis estudadas, exceto para o ganho diário de peso e consumo diário de ração nas fases compreendidas entre 30 e 45 dias e 45 e 62 dias e para o ganho diário de peso entre 10 e 62 dias, a favor das fêmeas. O uso de edulcorante na água não influenciou o consumo de água, o desempenho dos leitões e a qualidade da mucosa intestinal.

Objetivou-se avaliar a influência do processamento grão de milho (inteiro, tratado com uréia ou moído na forma de quirera) sobre a digestibilidade de rações e no desempenho de bezerros desmamados precocemente e mantidos em regime de confinamento. A dieta foi formulada com silagem de sorgo como volumoso, em uma relação volumoso:concentrado de 40:60, na matéria seca. A digestibilidade in vivo da matéria seca (MS), matéria orgânica, proteína bruta, extrato etéreo, fibra em detergente neutro, fibra em detergente ácido, celulose, energia bruta e amido, o pH das fezes e o consumo de MS foram efetuados em um ensaio com seis bezerros, castrados, com sete meses de idade. A influência das dietas no desempenho dos animais foi avaliada em ensaio com 15 bezerros não-castrados e idades semelhantes aos do ensaio de digestibilidade, com peso vivo inicial médio de 159 kg. O processamento do grão de milho não influenciou as variáveis analisadas, exceto o pH das fezes, que apresentou valor mais baixo nos animais que receberam dieta contendo grão de milho moído.
