Die Kernmagnetresonanz (NMR) ist eine vielseitige Technik, die auf spin-tragende Kerne angewiesen ist. Seit ihrer Entdeckung ist die Kernmagnetresonanz zu einem unverzichtbaren Werkzeug in unzähligen Anwendungen der Physik, Chemie, Biologie und Medizin geworden. Das größte Problem der NMR ist ihre geringe Sensitivtät auf Grund der sehr kleinen Energieaufspaltung bei Raumtemperatur. Für Protonenspins, die das größte magnetogyrische Verhältnis besitzen, ist der Polarisationsgrad selbst in den größten verfügbaren Magnetfeldern (24 T) nur ~7*10^(-5).rnDurch die geringe inhärente Polarisation ist folglich eine theoretische Sensitivitätssteigerung von mehr als 10^4 möglich. rnIn dieser Arbeit wurden verschiedene technische Aspekte und unterschiedliche Polarisationsagenzien für Dynamic Nuclear Polarization (DNP) untersucht.rnDie technische Entwicklung des mobilen Aufbaus umfasst die Verwendung eines neuen Halbach Magneten, die Konstruktion neuer Probenköpfe und den automatisierten Ablauf der Experimente mittels eines LabVIEW basierten Programms. Desweiteren wurden zwei neue Polarisationsagenzien mit besonderen Merkmalen für den Overhauser und den Tieftemperatur DNP getestet. Zusätzlich konnte die Durchführbarkeit von NMR Experimenten an Heterokernen (19F und 13C) im mobilen Aufbau bei 0,35 T gezeigt werden. Diese Ergebnisse zeigen die Möglichkeiten der Polarisationstechnik DNP auf, wenn Heterokerne mit einem kleinen magnetogyrischen Verhältnis polarisiert werden müssen.rnDie Sensitivitätssteigerung sollte viele neue Anwendungen, speziell in der Medizin, ermöglichen.

Dynamic nuclear polarisation (DNP) has been used to obtain magic angle spinning (14)N(OT) (nitrogen-14 overtone) solid-state NMR spectra from several model amino acids, with both direct and indirect observation of the (14)N(OT) signal. The crystalline solids were impregnated with biradical solutions of organic liquids that do not dissolve the crystalline phase. The bulk phase was then polarized via(1)H spin diffusion from the highly-polarized surface (1)H nuclei, resulting in (1)H DNP signal enhancements of around two orders of magnitude. Cross polarisation from (1)H nuclei directly to the (14)N overtone transition is demonstrated under magic angle spinning, using a standard pulse sequence with a relatively short contact time (on the order of 100 μs). This method can be used to acquire (14)N overtone MAS powder patterns that match closely with simulated line shapes, allowing isotropic chemical shifts and quadrupolar parameters to be measured. DNP enhancement also allows the rapid acquisition of 2D (14)N(OT) heteronuclear correlation spectra from natural abundance powder samples. (1)H-(14)N(OT) HETCOR and (13)C-(14)N(OT) HMQC pulse sequences were used to observe all single-bond H-N and C-N correlations in histidine hydrochloride monohydrate, with the spectra obtained in a matter of hours. Due to the high natural abundance of the (14)N isotope (99.6%) and the advantages of observing the overtone transition, these methods provide an attractive route to the observation of C-N correlations from samples at natural isotopic abundance and enable the high resolution measurement of (14)N chemical shifts and quadrupolar interaction parameters.

BACKGROUND: Acetate metabolism in skeletal muscle is regulated by acetylCoA synthetase (ACS). The main function of ACS is to provide cells with acetylCoA, a key molecule for numerous metabolic pathways including fatty acid and cholesterol synthesis and the Krebs cycle. METHODS: Hyperpolarized [1-(13)C]acetate prepared via dissolution dynamic nuclear polarization was injected intravenously at different concentrations into rats. The (13)C magnetic resonance signals of [1-(13)C]acetate and [1-(13)C]acetylcarnitine were recorded in vivo for 1min. The kinetic rate constants related to the transformation of acetate into acetylcarnitine were deduced from the 3s time resolution measurements using two approaches, either mathematical modeling or relative metabolite ratios. RESULTS: Although separated by two biochemical transformations, a kinetic analysis of the (13)C label flow from [1-(13)C]acetate to [1-(13)C]acetylcarnitine led to a unique determination of the activity of ACS. The in vivo Michaelis constants for ACS were KM=0.35±0.13mM and Vmax=0.199±0.031μmol/g/min. CONCLUSIONS: The conversion rates from hyperpolarized acetate into acetylcarnitine were quantified in vivo and, although separated by two enzymatic reactions, these rates uniquely defined the activity of ACS. The conversion rates associated with ACS were obtained using two analytical approaches, both methods yielding similar results. GENERAL SIGNIFICANCE: This study demonstrates the feasibility of directly measuring ACS activity in vivo and, since the activity of ACS can be affected by various pathological states such as cancer or diabetes, the proposed method could be used to non-invasively probe metabolic signatures of ACS in diseased tissue.

Overtone NMR spectroscopy has the potential to provide high-resolution 14N solid-state NMR spectra. The technique was first developed during the 1980s but has only recently been successfully combined with magic angle spinning (MAS), providing improved sensitivity and resolution as well as enabling more advanced approaches such as indirect detection and signal enhancement methods. This report provides a brief background to 14N overtone NMR, describing the ways in which it differs from conventional NMR and the challenges that arise as a result. The effects of MAS on the overtone spectrum are then presented and illustrated with numerous experimental and simulated examples. Finally, several recent developments enabled by MAS are described and some potential future directions are suggested.

In this thesis, the self-assembled functional structure of a broad range of amphiphilic molecular transporters is studied. By employing paramagnetic probe molecules and ions, continuous-wave and pulse electron paramagnetic resonance spectroscopy reveal information about the local structure of these materials from the perspective of incorporated guest molecules. First, the transport function of human serum albumin for fatty acids is in the focus. As suggested by the crystal structure, the anchor points for the fatty acids are distributed asymmetrically in the protein. In contrast to the crystallographic findings, a remarkably symmetric entry point distribution of the fatty acid binding channels is found, which may facilitate the uptake and release of the guest molecules. Further, the metal binding of 1,2,3-triazole modified star-shaped cholic acid oligomers is studied. These biomimetic molecules are able to include and transport molecules in solvents of different polarity. A pre-arrangement of the triazole groups induces a strong chelate-like binding and close contact between guest molecule and metal ion. In absence of a preordering, each triazole moiety acts as a single entity and the binding affinity for metal ions is strongly decreased. Hydrogels based on N-isopropylacrylamide phase separate from water above a certain temperature. The macroscopic thermal collapse of these hydrogels is utilized as a tool for dynamic nuclear polarization. It is shown that a radical-free hyperpolarized solution can be achieved with a spin-labeled gel as separable matrix. On the nanoscale, these hydrogels form static heterogeneities in both structure and function. Collapsed regions protect the spin probes from a chemical decay while open, water-swollen regions act as catalytic centers. Similarly, thermoresponsive dendronized polymers form structural heterogeneities, which are, however, highly dynamic. At the critical temperature, they trigger the aggregation of the polymer into mesoglobules. The dehydration of these aggregates is a molecularly controlled non-equilibrium process that is facilitated by a hydrophobic dendritic core. Further, a slow heating rate results in a kinetically entrapped non-equilibrium state due to the formation of an impermeable dense polymeric layer at the periphery of the mesoglobule.

We report 13C magic angle spinning NMR observation of photochemically induced dynamic nuclear spin polarization (photo- CIDNP) in the reaction center (RC) of photosystem II (PS2). The light-enhanced NMR signals of the natural abundance 13C provide information on the electronic structure of the primary electron donor P680 (chlorophyll a molecules absorbing around 680 nm) and on the pz spin density pattern in its oxidized form, P680⨥. Most centerband signals can be attributed to a single chlorophyll a (Chl a) cofactor that has little interaction with other pigments. The chemical shift anisotropy of the most intense signals is characteristic for aromatic carbon atoms. The data reveal a pronounced asymmetry of the electronic spin density distribution within the P680⨥. PS2 shows only a single broad and intense emissive signal, which is assigned to both the C-10 and C-15 methine carbon atoms. The spin density appears shifted toward ring III. This shift is remarkable, because, for monomeric Chl a radical cations in solution, the region of highest spin density is around ring II. It leads to a first hypothesis as to how the planet can provide itself with the chemical potential to split water and generate an oxygen atmosphere using the Chl a macroaromatic cycle. A local electrostatic field close to ring III can polarize the electronic charge and associated spin density and increase the redox potential of P680 by stabilizing the highest occupied molecular orbital, without a major change of color. This field could be produced, e.g., by protonation of the keto group of ring V. Finally, the radical cation electronic structure in PS2 is different from that in the bacterial RC, which shows at least four emissive centerbands, indicating a symmetric spin density distribution over the entire bacteriochlorophyll macrocycle.

Recent developments in dynamic nuclear polarisation now allow significant enhancements to be generated in the cryo solid state and transferred to the liquid state for detection at high resolution. We demonstrate that the Ardenkjaer-Larsen method can be extended by taking advantage of the properties of the trityl radicals used. It is possible to hyperpolarise 13C and 15N simultaneously in the solid state, and to maintain these hyperpolarisations through rapid dissolution into the liquid state. We demonstrate the almost simultaneous measurement of hyperpolarised 13C and hyperpolarised 15N NMR spectra. The prospects for further improvement of the method using contemporary technology are also discussed.

Measurement of heteronuclear spin-lattice relaxation times is hampered by both low natural abundance and low detection sensitivity. Combined with typically long relaxation times, this results in extended acquisition times which often renders the experiment impractical. Recently a variant of dynamic nuclear polarisation has been demonstrated in which enhanced nuclear spin polarisation, generated in the cryo-solid state, is transferred to the liquid state for detection. Combining this approach with small flip angle pulse trains, similar to the FLASH-T(1) imaging sequence, allows the rapid determination of spin-lattice relaxation times. In this paper we explore this method and its application to the measurement of T(1) for both carbon-13 and nitrogen-15 at natural abundance. The effects of RF inhomogeneity and the influence of proton decoupling in the context of this experiment are also investigated.

Trois protéines de la famille TRIM (Motif TRIpartite), TIF1α, β (Transcriptional Intermediary Factor 1) et PML (ProMyelocytic Leukaemia¬), font l’objet de cette étude. TIF1α est connu comme un coactivateur des récepteurs nucléaires et TIF1β comme le corépresseur universel des protéines KRAB-multidoigt de zinc dont le prototype étudié ici est ZNF74. PML possède divers rôles dont le plus caractérisé est celui d’être l’organisateur principal et essentiel des PML-NBs (PML-Nuclear Bodies), des macrostructures nucléaires très dynamiques regroupant et coordonnant plus de 40 protéines. Il est à noter que la fonction de TIF1α, β et PML est régulée par une modification post-traductionnelle, la sumoylation, qui implique le couplage covalent de la petite protéine SUMO (Small Ubiquitin like MOdifier) à des lysines de ces trois protéines cibles. Cette thèse propose de développer des méthodes utilisant le BRET (Bioluminescence Resonance Energy Transfert) afin de détecter dans des cellules vivantes et en temps réel des interactions non-covalentes de protéines nucléaires mais aussi leur couplage covalent à SUMO. En effet, le BRET n’a jamais été exploré jusqu’alors pour étudier les interactions non-covalentes et covalentes de protéines nucléaires. L’étude de l’interaction de protéines transcriptionnellement actives est parfois difficile par des méthodes classiques du fait de leur grande propension à agréger (famille TRIM) ou de leur association à la matrice nucléaire (ZNF74). L’homo et l’hétérodimérisation de TIF1α, β ainsi que leur interaction avec ZNF74 sont ici testées sur des protéines entières dans des cellules vivantes de mammifères répondant aux résultats conflictuels de la littérature et démontrant que le BRET peut être avantageusement utilisé comme alternative aux essais plus classiques basés sur la transcription. Du fait de l’hétérodimérisation confirmée de TIF1α et β, le premier article présenté ouvre la possibilité d’une relation étroite entre les récepteurs nucléaires et les protéines KRAB- multidoigt de zinc. Des études précédentes ont démontré que la sumoylation de PML est impliquée dans sa dégradation induite par l’As2O3 et dépendante de RNF4, une E3 ubiquitine ligase ayant pour substrat des chaînes de SUMO (polySUMO). Dans le second article, grâce au développement d’une nouvelle application du BRET pour la détection d’interactions covalentes et non-covalentes avec SUMO (BRETSUMO), nous établissons un nouveau lien entre la sumoylation de PML et sa dégradation. Nous confirmons que le recrutement de RNF4 dépend de SUMO mais démontrons également l’implication du SBD (Sumo Binding Domain) de PML dans sa dégradation induite par l’As2O3 et/ou RNF4. De plus, nous démontrons que des sérines, au sein du SBD de PML, qui sont connues comme des cibles de phosphorylation par la voie de la kinase CK2, régulent les interactions non-covalentes de ce SBD mettant en évidence, pour la première fois, que les interactions avec un SBD peuvent dépendre d’un évènement de phosphorylation (“SBD phospho-switch”). Nos résultats nous amènent à proposer l’hypothèse que le recrutement de PML sumoylé au niveau des PML-NBs via son SBD, favorise le recrutement d’une autre activité E3 ubiquitine ligase, outre celle de RNF4, PML étant lui-même un potentiel candidat. Ceci suggère l’existence d’une nouvelle relation dynamique entre phosphorylation, sumoylation et ubiquitination de PML. Finalement, il est suggéré que PML est dégradé par deux voies différentes dépendantes de l’ubiquitine et du protéasome; la voie de CK2 et la voie de RNF4. Enfin une étude sur la sumoylation de TIF1β est également présentée en annexe. Cette étude caractérise les 6 lysines cibles de SUMO sur TIF1β et démontre que la sumoylation est nécessaire à l’activité répressive de TIF1β mais n’est pas impliquée dans son homodimérisation ou son interaction avec la boîte KRAB. La sumoylation est cependant nécessaire au recrutement d’histones déacétylases, dépendante de son homodimérisation et de l’intégrité du domaine PHD. Alors que l’on ne connaît pas de régulateur physiologique de la sumoylation outre les enzymes directement impliquées dans la machinerie de sumoylation, nous mettons en évidence que la sumoylation de TIF1β est positivement régulée par son interaction avec le domaine KRAB et suggérons que ces facteurs transcriptionnels recrutent TIF1β à l’ADN au niveau de promoteur et augmentent son activité répressive en favorisant sa sumoylation.

The various approximations of vacuum polarization potential and the higher order corrections up to \alpha^3 are reviewed and quantitatively dicussed. The quadrupol part of the vacuum polarization is established. It leads rather straight forward to a small contribution of vacuum polarization to nuclear polarization. These effects are quantitatively investigated.

NMR quantum information processing studies rely on the reconstruction of the density matrix representing the so-called pseudo-pure states (PPS). An initially pure part of a PPS state undergoes unitary and non-unitary (relaxation) transformations during a computation process, causing a ""loss of purity"" until the equilibrium is reached. Besides, upon relaxation, the nuclear polarization varies in time, a fact which must be taken into account when comparing density matrices at different instants. Attempting to use time-fixed normalization procedures when relaxation is present, leads to various anomalies on matrices populations. On this paper we propose a method which takes into account the time-dependence of the normalization factor. From a generic form for the deviation density matrix an expression for the relaxing initial pure state is deduced. The method is exemplified with an experiment of relaxation of the concurrence of a pseudo-entangled state, which exhibits the phenomenon of sudden death, and the relaxation of the Wigner function of a pseudo-cat state.

Zusammenfassung: Michael EbertEntwicklung eines leistungsstarken Polarisators und Kompressorsfür 3-He für medizinische MR Tomographie Durch Optisches Pumpen von metastabilem3-He*--Gas bei einem Druck von 1 mb und Spinübertrag mittels Metastabileraustauschstöße aufden Grundzustand, wird 3-He auf 53 % bei einemmittleren Fluß von f = 58 bar*liter/Tag aufpolarisiert. Bei einem Fluß von f = 122bar*l/Tag wird immer noch eine Polarisation von 30 % erzielt. DurchSteigerung der Laserleistung von derzeit 12 Watt aufwünschenswerte 30 Watt, könnten beigleichem Fluß Kernspinpolarisationen des Heliums von 70 % erreicht werden. Mittels einer eigens entwickelten Ganzmetall--Titan--Kolbenpumpeerfolgt die Kompression in zwei Stufen. Zuerst wird einZwischenvolumen auf Drücke 200 bis 800 mb,je nach Anwendung gefüllt. Mit dem selben Kompressor könnenanschließend abnehmbare, verschließbare Experimentierzellen ausdiesem Zwischenvolumen auf Drücke 1 bis 6 bar gefüllt werden. Auf Grund des großen Hubvolumens von 15,4 Liter können große Gasmengen proKompressionszyklus verdichtet werden. Wegen des großen Verhältnisaus Kompressionshub h = 100 cm undKompressordurchmesser 14 cm kommt es imKompressionsraum zu keinen meßbaren Polarisationsverlusten.Zusammen mit dem großen Kompressionsfaktor von K = 10000sind diese konstruktiven Maßnahmen für den vollständigen Erhaltder Polarisation bei der Kompression des polarizierten 3-He--Gasesverantwortlich. Diese großen Gasmengen an hyperpolarisiertem 3-He haben ersteklinische Studien funktioneller Lungen--MRT ermöglicht. DurchMessung des Sauerstoffpartialdrucks und dessen Abnahmerate, derBestimmung der Ventillation und Messung der DiffusionskonstantenADC in der Lunge, können die Funktionen dieses Organs lokalquantitativ beurteilt werden.

In dieser Arbeit wird zum Erreichen hoher Kernspinpolarisationen die Methode des metastabilen optischen Pumpens benutzt. Das Verfahren beruht auf dem "Ubertrag des Drehimpulses absorbierter Photonen auf das hierbei angeregte Valenzelektron, welches durch Hyperfeinkopplung den Drehimpuls weiter auf den $^3$He-Kern transferiert. Da der Polarisationsprozess nur bei Dr"ucken von ca. 1~mbar effizient funktioniert, f"ur die meisten Anwendungen aber polarisiertes $^3$He bei einem Druck von $geq 1$~bar ben"otigt wird, muss das Gas nach der Aufpolarisation komprimiert werden. In unserer Arbeitsgruppe steht eine Maschine ("`Polarisator"') zur Verf"ugung, die das Gas polarisiert und anschlie"send komprimiert. Ziel dieser Dissertation ist, einerseits die Leistungsf"ahigkeit des Polarisators bez"uglich Maximalpolarisation und Gasfluss zu verbessern und andererseits den metastabilen Pumpprozess selbst genauer zu untersuchen.\ noindent Durch die Verwendung neuer Laser auf Basis der Fasertechnologie sowie einer systematischen Optimierung der optischen Komponenten konnten in abgeschlossenen Pumpzellen Rekord-Polarisationsgrade von $91pm 2$% erzielt werden.\ noindent Mit der Implementierung neuartiger Optiken und Laser am Mainzer Polarisator konnte die Leistungscharakteristik entscheidend verbessert werden. So wurde die erreichbare Polarisation bei identischer Produktionsrate um 20 Prozentpunkte gesteigert. Zurzeit sind maximale Polarisationsgrade von mehr als 75% im optischen Pumpvolumen erreichbar. Eine am Mainzer Triga-Reaktor durchgef"uhrte Polarisationsbestimmung ergab einen Wert von $72.7pm 0.7$%. Dies veranschaulicht die geringen Polarisationsverluste infolge der Gaskompression, des Transports und einer Lagerung "uber mehrere Stunden.\ noindent Zur Dynamik der geschwindigkeitsver"andernden St"o"se sowie zur Bestimmung der mittleren Photonen-Absorptionsrate wurde ein Modell entwickelt, welches auch experimentell best"atigt wurde. Damit konnte erstmalig das gemessene Absorptionsverhalten einer spektral schmalbandigen Laserdiode korrekt beschrieben werden.\ noindent Zudem stimmen die an so genannten abgeschlossenen Pumpzellen gemessenen extrem hohen Polarisationswerte mit theoretischen Vorhersagen "uberein, sofern der Druck im optischen Pumpvolumen geringer als 1~mbar ist und das $^3$He nicht durch Fremdgase verunreinigt ist. Bei derartigen Pumpzellen ist die gemessene Abh"angigkeit der Polarisation von Laserleistung, Metastabilendichte und falscher Zirkularkomponente mit der Theorie kompatibel.\

Collinear laser spectroscopy has been used as a tool for nuclear physics for more than 30 years. The unique possibility to extract nuclear properties like spins, radii and nuclear moments in a model-independent manner leads to important physics results to test the predictive power of existing nuclear models. rnThis work presents the construction and the commissioning of a new collinear laser spectroscopy experiment TRIGA-LASER as a part of the TRIGA-SPEC facility at the TRIGA research reactor at the University of Mainz. The goal of the experiment is to study the nuclear structure of radioactive isotopes which will be produced by neutron-induced fission near the reactor core and transported to an ion source by a gas jet system. rnThe versatility of the collinear laser spectroscopy technique will be exploited in the second part of this thesis. The nuclear spin and the magnetic moment of the neutron-deficient isotope Mg-21 will be presented, which were measured by the detection of the beta-decay asymmetry induced by nuclear polarization after optical pumping. A combination of this detection method with the classical fluorescence detection is then used to determine the isotope shifts of the neutron-rich magnesium isotopes from Mg-24 through Mg-32 to study the transition to the ''island of inversion''.

Electrical property derivative expressions are presented for the nuclear relaxation contribution to static and dynamic (infinite frequency approximation) nonlinear optical properties. For CF4 and SF6, as opposed to HF and CH4, a term that is quadratic in the vibrational anharmonicity (and not previously evaluated for any molecule) makes an important contribution to the static second vibrational hyperpolarizability of CF4 and SF6. A comparison between calculated and experimental values for the difference between the (anisotropic) Kerr effect and electric field induced second-harmonic generation shows that, at the Hartree-Fock level, the nuclear relaxation/infinite frequency approximation gives the correct trend (in the series CH4, CF4, SF6) but is of the order of 50% too small
