Purpose: To investigate the anti-hyperuricemic effect of Dioscorea tokoro Makino extract (DTME) in potassium oxonate-induced hyperuricemic mice. Method: The effect of DTME was investigated in the hyperuricemic mice induced by potassium oxonate. DTME. The extract was administered to the mice daily at doses of 220, 440 and 880 mg/kg for 10 days; allopurinol (5 mg/kg) was given as positive control. Serum and urine levels of uric acid and creatinine were determined by colorimetric method. Simultaneously, protein levels of urate transporter 1 (URAT1) and organic anion transporter 1 (OAT1) in the rat kidney were analyzed by Western blotting. Results: Compared with control, a high dose of DTME inhibited xanthine oxidase (XOD) activity in both serum (18.12 ± 1.33 U/L) and in liver (70.15 ± 5.20 U/g protein) (p < 0.05); decreased levels of serum uric acid (2.04 ± 0.64 mg/L) (p < 0.05), serum creatinine (0.35 ± 0.18 μmol/L) and blood urea nitrogen (BUN) (8.83 ± 0.71 mmol/L) (p < 0.05). Furthermore, the extract increased levels of urine uric acid (38.34 ± 8.23 mg/L), urine creatinine (34.38 ± 1.98 mmol/L), down regulated of URAT1 and up regulated of OAT1 protein expressions (p < 0.05) in the renal tissue of hyperuricemic mice. Conclusion: DTME improves renal dysfunction in rats by regulating renal urate transporters in hyperuricemic rats. This may find therapeutic application in antihypertensive therapy.

The cell suspension cultures, established from the friable callus which was risen from the nodal segments of Dioscorea bulbifera L. in Murashige-Skoog (MS) medium supplemented with indole-3-butryic acid (20 mg L- 1), was examined for cell growth in MS medium fed with cholesterol (50 mg L- 1 and 100 mg L- 1) after 8, 10, 12, 14, 16, and 18 days of culture. The growth index of the cell suspension culture on the 8th day was 1.2 and gradually inclined to 1.9 on the 16th day and remained the same at the 18th day. There is no marked difference in the cell growth of cholesterol-treated and control cell suspension culture. The maximum accumulation of diosgenin was noticed on the 14th day in control and cholesterol-treated cell suspension culture and immobilised cell cultures. The highest concentration of diosgenin, 2.94% and 2.14% dry weight, was obtained in immobilised cell culture and cell suspension culture treated with 100 mg L- 1 cholesterol, respectively.

Se caracterizaron y evaluaron quince accesiones de Ñame (Dioscorea sp) provenientes de ocho países de la cuenca del Caribe. El ensayo se estableció en el programa de Recursos Genéticos Nicaragüenses (REGEN). Del Instituto Superior de Ciencias Agropecuarias (ISCA), Managua. Utilizando un arreglo fr bloques al azar con dos repeticiones. Se observaron 89 descriptores, entre los cuales se seleccionaron para análisis 42 cualitativos y 23 cuantitativos. Los datos se sometieron a análisis de agrupamiento por encadenamiento simple: El fonograma de descriptores cualitativos determino la experiencia de dos especies: D alata y D. Trífida mientras que dos clones que se sospecha eran duplicados por cercanía geográfica, presentan alta diferenciación: Del análisis muy similar al patrón presentado por el total de diez descriptores de color: sugiriendo la importancia de los primeros en la diferenciación de accesiones. El fonograma de descriptores cualitativos resulto diferente en conformación e integrantes de los grupos, que los obtenidos del análisis de descriptores cualitativos: los datos de rendimiento se procesaron de la misma manera, sometidos a un análisis de varianza que arrojo diferentes pruebas de medidas de Duncan se obtuvieron dos niveles de diferenciación a y b. concordante con rendimiento presento estrecha concordancia con el de los descriptores cuantitativos. De 23 descriptores cuantitativos se seleccionaron trece, que sometidos a prueba de t. determinó nueve capaces de encontrar diferencias significativas entre pares de grupos de accesiones. De la relación entre el color y rendimiento, se desprende que los clones con subcuticula morada. Blanco amarillenta y amarillo pálido, con pulpa morada o blanca amarillento. Tienen tendencia a generar altos rendimientos. Se elaboró un catálogo de características morfológicas de las accesiones estudiadas.

The study was designed to investigate the value of the peels of yam (Dioscorea rotundata) as energy source in the diet of Oreochromis niloticus fry and to investigate the level of inclusion of this peels that will give optimum growth performance. Four diets, three levels of yam peels and a control, was prepared and tested on O. niloticus fry (mean weight of 0.27g) for ten weeks. Fifteen (15) O. niloticus fry were grouped in each of the glass aquaria, measuring 60x30x3Ocm and with a maximum capacity of 52 liters of water. The fry were fed twice daily at 10% biomass. The fry were weighed weekly to determine weight increment or otherwise and the quality of feed was adjusted accordingly. DTl (70% yam peels and 30% yellow maize) in the carbohydrate mixture gave the best performance. The fry fed this diet, gained a mean weight of 1.20g for the period of the experiment. The poorest performance in terms of growth was from fry fed the control diet (100% yellow maize in the carbohydrate mixture). Fry fed this diet gained mean weight of 0.80g for the duration of the experiment. Analysis of the various growth indices like SGR, PER, FCR and NPU shows that DTl was the overall best diet with an SGR value of I. 92 and FCR of 54.10. The difference in weight gain by fry fed the three levels of yam peels diet and the control diet (100% yellow maize) was not statistically significant (P>0.05)

Home gardens are considered as vital units for enhancing food security particularly in developing nations of South Asia, such as Sri Lanka. Although the yam crop Dioscorea spp. constitute a popular but still minor component in Sri Lankan home gardens, they have the potential of producing large quantities of edible material with minimal inputs. However, their real value in South Asian home gardens is not yet reported. Hence, this study was carried out to get insights into home garden characteristics, gardener demography as well as current management practices within 300 Sri Lankan home garden systems that are located along a climatic gradient. By using interviews and field observations, gardeners, who cultivated in particular Dioscorea species, were studied within 10 of the 25 administrative districts distributed in the wet, intermediate and dry climatic zone of Sri Lanka. Furthermore, current management practices of yams cultivation were analyzed on local scale and compared afterwards with management recommendations published in the year 2006 by the Department of Agriculture. Dioscorea species were found in a majority of home gardens, especially in wet and intermediate zones of Sri Lanka. D. alata was the most prominent species and was managed at a subsistence level and not as per recommendations developed by the Department of Agriculture. Our results revealed that Dioscorea alata is an essential component of Sri Lankan home gardens in rural areas and can yield substantial quantities of edible tubers with low input, especially during times of food scarcities, and has therefore the potential to enhance food security and rural development.

White or Guinea yam (Dioscorea rotundata), grown for its underground tubers, is an important food in West Africa. Progress in yam breeding is constrained by variable flowering behaviour, making hybridization difficult. Yam clones may be dioecious, monoecious or hermaphrodite with variable sex ratios. The proportion of plants that flower and the flowering intensity also vary with season and location. The objective of the present work was to investigate whether variation in flowering behaviour was related to factors determining rate of development (photoperiod and temperature through sowing date, location and year) or growth (cumulative solar radiation and temperature). Sex ratios, the proportion of plants that had flower buds and open flowers, and the number of flowers or spikes was recorded in one male (TDr 131) and one female (TDr 99-9) clone of white yam grown in the field in Nigeria at three locations and at different sowing dates. Clone TDr 131 was uniformly male flowering, while clone TDr 99-9 exhibited a number of sex types with gynoecious, monoecious and trimonoecious plants observed. The proportion of flowering plants was low in both clones, averaging 0.34 in clone TDr 131 and 0.13 in clone TDr 99-9. Day of vine emergence had a significant and contrasting effect on the proportion of flowering plants and on flowering intensity in the two clones. In clone TDr 131, the proportion of flowering plants and flowering intensity declined with later vine emergence at all locations (r=0.43-0.53, P<0.05), whereas in clone TDr 99-9 the proportion of flowering plants increased with later emergence (r=0.46, P<0.01). In clone TDr 131, this response was strongly associated with warmer temperatures (r=0.49-0.50; P<0.05) and greater cumulative radiation (r=0.85-0.93; P<0.001) between vine emergence and flowering, rather than photoperiod at vine emergence. This suggests that flowering behaviour in the male clone TDr 131 is strongly influenced by factors that affect growth rather than development. Clone TDr 99-9, on the other hand, exhibited no clear relations between flowering and growth or developmental factors, though the proportion of flowering plants and flowering intensity was greatest at planting dates close to the longest day and at temperatures of 25-26 degrees C. This might suggest that flowering behaviour in clone TDr 99-9 is controlled by photothermal responses.

The objective of this study was to quantify the effect of photoperiod on the duration from vine (shoot) emergence to flowering in white or Guinea yam (Dioscorea rotundata). The duration from vine emergence to flowering in two clonal varieties of yam (TDr 131 and TDr 99-9) was recorded at 10 different sowing dates/locations in Nigeria. Durations to flowering varied from 40 to > 88 days. Mean daily temperature and photoperiod between vine emergence and flowering varied from 25 to 27 degrees C and 13.1 to 13.4 h day(-1), respectively. Both clones had similar responses to temperature, with base and optimum temperatures of 12 and 25-27 degrees C, respectively. Thermal durations to flowering were strongly related (r(2) > 0.75-0.83) to absolute photoperiod (h) at vine emergence as well as to rate of change of photoperiod (s day(-1)) at vine emergence. The response to absolute photoperiod suggests that white yams are quantitative LDPs, flowering sooner in long than short days. Yams also flowered earlier when the rate of change of photoperiod was positive but small, or was negative. center dot Background and Aims The control of dormancy in yam (Disocorea spp.) tubers is poorly understood and attempts to shorten the long dormant period (i.e. cause tubers to sprout or germinate much earlier) have been unsuccessful. The aim of this study was to identify and define the phases of dormancy in Dioscorea rotundata tubers, and to produce a framework within which dormancy can be more effectively studied. center dot Methods Plants of 'TDr 131' derived from tissue culture were grown in a glasshouse simulating temperature and photoperiod at Ibadan (7 degrees N), Nigeria to produce tubers. Tubers were sampled on four occasions: 30 d before shoot senescence (149 days after planting, DAP), at shoot senescence (179 DAP), and twice during storage at a constant 25 degrees C (269 and 326 DAP). The development of the apical shoot bud was described from tissue sections. In addition, the responsiveness of shoot apical bud development to plant growth regulators (gibberellic acid, 2-chloroethanol and thiourea) applied to excised tuber sections was also examined 6 and 12 d after treatment. center dot Key Results and Conclusions Three phases of tuber dormancy are proposed: Phase I, from tuber initiation to the appearance of the tuber germinating meristem; Phase II, from the tuber germinating meristem to initiation of foliar primordium; and Phase III, from foliar primordium to appearance of the shoot bud on the surface of the tuber. Phase I is the longest phase (approx. 220 d in 'TDr 131'), is not affected by PGRs and is proposed to be an endo-dormant phase. Phases II and III are shorter (< 70 d in total), are influenced by PGRs and environmental conditions, and are therefore endo-/eco-dormant phases. To manipulate dormancy to allow off-season planting and more than one generation per year requires that the duration of Phase I is shortened.

Tuber dormancy enables yams to survive in the ground during the dry season and post-harvest storage. Three clones of Dioscorea rotundata were harvested after five intervals and then stored in a cooler (20.6°C) or at ambient temperature (27.8°C). The time from harvest to sprouting was shorter as harvest was delayed. The period from sowing to sprouting for each clone was similar for tubers harvested from 140 days after planting, but tubers harvested earlier took longer to sprout. The cooler temperature delayed sprouting. Tubers of two clones sprouted after only 70 days of crop growth. If the dormancy period of these young tubers can be broken, the generation time of yam crop improvement programmes could be considerably reduced.

This study had as objective to evaluate the effect of yam starch, modified starch from Cargill-Brasil (Amidomax 4800 (R)) and gelatin from Gelita-Brasil (GEL-LAC) as stabilizers/thickeners in different ratios and combinations in the soy yoghurt fermented with Enterococcus faecium and Lactobacillus helveticus ssp jugurti. Ten soy yoghurt formulations containing these different stabilizers/thickeners, always totalizing 0.5% in relation to the final formulation, were analyzed in sensorial and physical-chemical terms. Based on the observed results, it was concluded in relation to the sensorial point of view that the more appropriate product was processed only with gelatin at 0.5% concentration. This product also presented the best physical-chemical results related to consistency, syneresis and water holding capacity. However, the isolated use of gelatin increased fermentation time of the soy yoghurt.

Starch extraction from roots and tubers uses grating with water and sieves to separate the starch slurry from residual mass. The starch is recovered by decantation or centrifugation. The yam starch extraction is difficult due to high viscosity of the slurry caused by non-starch polysaccharides (NSP). The establishment of an efficient extraction process may turn yam into a competitive raw material. In this paper Dioscorea alata starch extracted by four methods was characterized in order to establish the impact of treatments. When the tubers were digested with an aqueous oxalic acid/ammonium oxalate (OA/AO) 1/1 solution, it was easier to separate the starch slurry from residual mass, because viscosity was reduced. For all the others methods tested, the viscosity remained almost the same. The nitrogen present in yam tubers was removed during the different extractions to a different extent. The largest nitrogen reduction was observed with ONAO followed by the control (water). The spectrum of starch granules sizes obtained also varied according to the treatment. Results proved that NSP carries small starch granules over to the waste water. The smaller starch granules diameter varied from 1.9 mu m (OA/AO extraction) to 13.5 mu m (water and pectinase extractions). The larger diameter varied from 41.0 mu m (NaOH treatment) to 67.7 mu m (ONAO). All starches extracted showed a RVA behavior in agreement with literature for yam starch, but with small differences due to the influence of methods. ONAO extraction showed the best recovery (18 g of starch/100 g tuber yam) and granular variation but it interfered with the rheological behavior of starch.
