Accurate field data on trophic interactions for suspension feeders are lacking, and new approaches to dietary analysis are necessary. Polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) was integrated with stable isotope analysis to examine dietary patterns in suspension-feeding Mytilus spp. from seven spatially discrete locations within a semi-enclosed marine bay (Strangford Lough, Northern Ireland) during June 2009. Results of the two methods were highly correlated, reflecting dietary variation in a similar manner. Variation in PCR-DGGE data was more strongly correlated with the principal environmental gradient (distance from the opening to the Irish Sea), while values of dC and dN became progressively enriched, suggesting a greater dependence on animal tissue and benthic microalgae. Diatoms and crustaceans were the most frequently observed phylotypes identified by sequencing, but specific DNA results provided little support for the trophic trends observed in the stable isotope data. This combined approach offers an increased level of trophic insight for suspension feeders and could be applied to other organisms. © 2012 Springer-Verlag Berlin Heidelberg.

We report the results of stable carbon and nitrogen isotope analysis of 354 human and faunal samples from five archaeological cultures of the Minusinsk Basin, Southern Siberia – Afanasyevo, Okunevo, Andronovo, Karasuk and Tagar (ca. 2700–1 BC) – a key location in Eurasia due to its position on a northern corridor linking China and central Eurasia. The results indicate that the diet of Eneolithic to Middle Bronze Age (Afanasyevo to Andronovo) populations was primarily C3-based, with C4 plants only becoming an important component of the diet in the Late Bronze Age Karasuk and Early Iron Age Tagar cultures. Freshwater fish seems to have been an important constituent of the diets in all groups. The findings constitute the earliest concrete evidence for the substantial use of millet in the eastern Eurasian steppe. We propose that it was probably introduced from Northwestern China during the Karasuk culture at the start of the Late Bronze Age, ca. 1500 BC. We conclude with a discussion of the implications for the nature of pastoralist economies on the steppes.

The stable isotope values for a range of size classes of Hyporhamphus regularis ardelio from Moreton Bay, south-east Australia were determined. There was a positive linear relationship between 613 C and standard length (L-s) (delta(13)C = 0.034 Ls - 16-23; r(2) = 0.78). delta(13)C ranged from -8.48 to - 17.29 parts per thousand with the smallest size class (50 mm Ls) being on average 1.04 parts per thousand enriched with respect to that of zooplankton (Temora turbinata) and 7.97 parts per thousand depleted compared to Zostera capricorni. delta(13)C was positively correlated with Ls (P 0.0 1) with delta(15) N, ranging from 9.18 to 11.00 parts per thousand. Fish of all size classes were on average 2.32 and 7.63 parts per thousand more enriched than zooplankton and seagrass, respectively. Carbon isotope data indicate that H. r. ardelio commence life as carnivores and change to a diet in which seagrass is the primary carbon source. The dependence on animal matter, however, is always present. Due to the low percentage of nitrogen in Z. capricorni (2.5%) compared to zooplankton (9.1%) it appears that nitrogen from zooplankton is necessary throughout their life history with the carbon requirements for these fish coming chiefly from Z. capricorni. Background: Poor diet is thought to be a risk factor for many diseases, including age-related macular disease (ARMD), which is the leading cause of blind registration in those aged over 60 years in the developed world. The aims of this study were 1) to evaluate the dietary food intake of three subject groups: participants under the age of 50 years without ARMD (U50), participants over the age of 50 years without ARMD (O50), and participants with ARMD (AMD), and 2) to obtain information on nutritional supplement usage. Methods: A prospective cross-sectional study designed in a clinical practice setting. Seventy-four participants were divided into three groups: U50; 20 participants aged < 50 years, from 21 to 40 (mean ± SD, 37.7 ± 10.1 years), O50; 27 participants aged > 50 years, from 52 to 77 (62.7 ± 6.8 years), and ARMD; 27 participants aged > 50 years with ARMD, from 55 to 79 (66.0 ± 5.8 years). Participants were issued with a three-day food diary, and were also asked to provide details of any daily nutritional supplements. The diaries were analysed using FoodBase 2000 software. Data were input by one investigator and statistically analysed using Microsoft Excel for Microsoft Windows XP software, employing unpaired t-tests. Results: Group O50 consumed significantly more vitamin C (t = 3.049, p = 0.005) and significantly more fibre (t = 2.107, p = 0.041) than group U50. Group ARMD consumed significantly more protein (t = 3.487, p = 0.001) and zinc (t = 2.252, p = 0.029) than group O50. The ARMD group consumed the highest percentage of specific ocular health supplements and the U50 group consumed the most multivitamins. Conclusions: We did not detect a deficiency of any specific nutrient in the diets of those with ARMD compared with age- and gender-matched controls. ARMD patients may be aware of research into use of nutritional supplementation to prevent progression of their condition.

Background and Aims: Consumption of antioxidant nutrients can reduce the risk of progression of age-related macular degeneration (AMD) - the leading cause of visual impairment in adults over the age of 50 years in the UK. Lutein and zeaxanthin (L&Z) are of particular interest because they are selectively absorbed by the central retina. The objectives of this study were to analyse the dietary intake of a group of AMD patients, assess their ability to prepare and cook healthy food, and to make comparisons with people not affected by AMD. Methods: 158 participants with AMD were recruited via the UK charity The Macular Society, and fifty participants without AMD were recruited from optometric practice. A telephone interview was conducted by trained workers where participants completed a 24 hour food diary, and answered questions about cooking and shopping capabilities. Results: In the AMD group, the average L&Z intake was low in for both males and females. Those able to cook a hot meal consumed significantly more L&Z than those who were not able. Most participants were not consuming the recommended dietary allowance of fibre, calcium, vitamin D and E, and calorific intake was also lower than recommendations for their age-group. The non-AMD group consumed more kilocalories and more nutrients than the AMD group, but the L&Z intake was similar to those with AMD. The main factor that influenced participant’s food choices was personal preference. Conclusion: For an ‘informed’ population, many AMD participants were under-consuming nutrients considered to be useful for their condition. Participants without AMD were more likely to reach recommended daily allowance values for energy and a range of nutrients. It is therefore essential to design more effective dietary education and dissemination methods for people with, and at risk of, AMD.

It has been proposed that long-term consumption of diets rich in non-digestible carbohydrates (NDCs), such as cereals, fruit and vegetables might protect against several chronic diseases, however, it has been difficult to fully establish their impact on health in epidemiology studies. The wide range properties of the different NDCs may dilution their impact when they are combined in one category for statistical comparisons in correlations or multivariate analysis. Several mechanisms have been suggested to explain the protective effects of NDCs, including increased stool bulk, dilution of carcinogens in the colonic lumen, reduced transit time, lowering pH, and bacterial fermentation to short chain fatty acids (SCFA) in the colon. However, it is very difficult to measure SCFA in humans in vivo with any accuracy, so epidemiological studies on the impact of SCFA are not feasible. Most studies use dietary fibre (DF) or Non-Starch Polysaccharides (NSP) intake to estimate the levels, but not all fibres or NSP are equally fermentable. The first aim of this thesis was the development of the equations used to estimate the amount of FC that reaches the human colon and is fermented fully to SCFA by the colonic bacteria. Therefore, several studies were examined for evidence to determine the different percentages of each type of NDCs that should be included in the final model, based on how much NDCs entered the colon intact and also to what extent they were fermented to SCFA in vivo. Our model equations are FC-DF or NSP$ 1: 100 % Soluble + 10 % insoluble + 100 % NDOs¥ + 5 % TS** FC-DF or NSP 2: 100 % Soluble + 50 % insoluble + 100 % NDOs + 5 % TS FC-DF* or NSP 3: 100 % Soluble + 10 % insoluble + 100 % NDOs + 10 % TS FC-DF or NSP 4: 100 % Soluble + 50 % insoluble + 100 % NDOs + 10 % TS *DF: Dietary fibre; **TS: Total starch; $NSP: non-starch polysaccharide; ¥NDOs: non-digestible oligosaccharide The second study of this thesis aimed to examine all four predicted FC-DF and FC-NSP equations developed, to estimate FC from dietary records against urinary colonic NDCs fermentation biomarkers. The main finding of a cross-sectional comparison of habitual diet with urinary excretion of SCFA products, showed weak but significant correlation between the 24 h urinary excretion of SCFA and acetate with the estimated FC-DF 4 and FC-NSP 4 when considering all of the study participants (n = 122). Similar correlations were observed with the data for valid participants (n = 78). It was also observed that FC-DF and FC-NSP had positive correlations with 24 h urinary acetate and SCFA compared with DF and NSP alone. Hence, it could be hypothesised that using the developed index to estimate FC in the diet form dietary records, might predict SCFA production in the colon in vivo in humans. The next study in this thesis aimed to validate the FC equations developed using in vitro models of small intestinal digestion and human colon fermentation. The main findings in these in vitro studies were that there were several strong agreements between the amounts of SCFA produced after actual in vitro fermentation of single fibre and different mixtures of NDCs, and those predicted by the estimated FC from our developed equation FC-DF 4. These results which demonstrated a strong relationship between SCFA production in vitro from a range of fermentations of single fibres and mixtures of NDCs and that from the predicted FC equation, support the use of the FC equation for estimation of FC from dietary records. Therefore, we can conclude that the newly developed predicted equations have been deemed a valid and practical tool to assess SCFA productions for in vitro fermentation.

The Eurasian otter (Lutra lutra L.) is a top predator in aquatic systems and plays an important role in ecosystem functioning. However, it has undergone dramatic declines throughout Europe as a result of environmental degradation. We examine the putative role of the otter as a bioindicator in Ireland which remains a stronghold for the species and affords a unique opportunity to examine variation in its ecological niche. We describe diet, using spraint contents, along rivers during 2010 and conduct a review and quantitative meta-analysis of the results of a further 21 studies. We aimed to assess variation in otter diet in relation to river productivity, a proxy for natural nutrification and anthropogenic eutrophication, and availability of salmonid prey (Salmo trutta and Salmo salar), to test the hypothesis that otter diet is related to environmental quality. Otter diet did not vary with levels of productivity or availability of salmonids whilst Compositional Analysis suggested there was no selection of salmonid over non-salmonid fish. There was a distinct niche separation between riverine and lacustrine systems, the latter being dominated by Atlantic eel (Anguilla anguilla). Otters are opportunistic and may take insects, freshwater mussels, birds, mammals and even fruit. Otters living along coasts have a greatest niche breath than those in freshwater systems which encompasses a wide variety of intertidal prey though pelagic fish are rarely taken. It is concluded that the ability of the otter to feed on a wide diversity of prey taxa and the strong influence of habitat type, renders it a poor bioindicator of environmental water quality. It seems likely that the plasticity of the habitat and dietary niche of otters, and the extent of suitable habitat, may have sustained populations in Ireland despite intensification of agriculture during the 20th century.

Aims: Dietary glycemic index (GI) and glycemic load (GL) have been associated with risk of chronic diseases, yet limited research exists on patterns of consumption in Australia. Our aims were to investigate glycemic carbohydrate in a population of older women, identify major contributing food sources, and determine low, moderate and high ranges. Methods: Subjects were 459 Brisbane women aged 42-81 years participating in the Longitudinal Assessment of Ageing in Women. Diet history interviews were used to assess usual diet and results were analysed into energy and macronutrients using the FoodWorks dietary analysis program combined with a customised GI database. Results: Mean±SD dietary GI was 55.6±4.4% and mean dietary GL was 115±25. A low GI in this population was ≤52.0, corresponding to the lowest quintile of dietary GI, and a low GL was ≤95. GI showed a quadratic relationship with age (P=0.01), with a slight decrease observed in women aged in their 60’s relative to younger or older women. GL decreased linearly with age (P<0.001). Bread was the main contributor to carbohydrate and dietary GL (17.1% and 20.8%, respectively), followed by fruit (15.5% and 14.2%), and dairy for carbohydrate (9.0%) or breakfast cereals for GL (8.9%). Conclusions: In this population, dietary GL decreased with increasing age, however this was likely to be a result of higher energy intakes in younger women. Focus on careful selection of lower GI items within bread and breakfast cereal food groups would be an effective strategy for decreasing dietary GL in this population of older women.

Blue (Callinectes sapidus)(Portunidae),lady (Ovalipes ocellatus)(Portunidae), and Atlantic rock (Cancer irroratus) (Cancridae) crabs inhabit estuaries on the northeast United States coast for parts or all of their life cycles. Their distributions overlap or cross during certain seasons. During a 1991–1994 monthly otter trawl survey in the Hudson-Raritan Estuary between New York and New Jersey, blue and lady crabs were collected in warmer months and Atlantic rock crabs in colder months. Sex ratios, male:female, of mature crabs were 1:2.0 for blue crabs, 1:3.1 for lady crabs, and 21.4:1 for Atlantic rock crabs. Crabs, 1286 in total, were subsampled for dietary analysis, and the dominant prey taxa for all crabs, by volume of foregut contents, were mollusks and crustaceans. The proportion of amphipods and shrimp in diets decreased as crab size increased. Trophic niche breadth was widest for blue crabs, narrower for lady crabs, and narrowest for Atlantic rock crabs. Trophic overlap was lowest between lady crabs and Atlantic rock crabs, mainly because of frequent consumption of the dwarf surfclam (Mulinia lateralis) by the former and the blue mussel (Mytilus edulis) by the latter. The result of cluster analysis showed that size class and location of capture of predators in the estuary were more influential on diet than the species or sex of the predators.

The study examined diel feeding chronology of six fish species in Anchuthengu backwater, Kerala. Over the 24h period, more or less same items contributed to the diet of Arius arius. Mugil cephalus exhibited substantial diel variation in diet composition, with algae forming the main dietary component in the diurnal diet, and prawns in the crepuscular and nocturnal diets. Algae and rotifers formed the main food items throughout the 24h feeding period in Hyporhamphus xanthopterus. No change was noticed in the diel diet composition in Ambassis commersonii. While Caranx ignobilis showed no diel variation, Gerrus lucidus that feeds on a variety of prey items seemed to exhibit some diel variation in feeding. All the fish examined had diurnal feeding peaks. The results indicate that because of diel variation in diet composition and feeding periodicity for these six species, dietary analysis conducted at only one interval would not provide an accurate representation of the diet of these species.

Numbers of Tufted Ducks Aythya fuligula wintering at Lough Neagh declined dramatically following the winter of 2000/01. The abundance and biomass of benthic macroinvertebrates, their main food source, declined significantly between the winters of 1997/98 and 2010. Therefore, information on recent diet was required to determine if there had been any significant changes before and after the observed declines in numbers of both macroinvertebrates and birds. Here, we used oesophageal content analysis to characterise the contemporary diet of Tufted Ducks at Lough Neagh during 2010-12. Out of 75 shot ducks, only three individuals had prey items in their oesophagi while all four ducks that accidentally drowned in gill nets contained prey items. Oesophageal contents were then compared with data collected during a study conducted in the late 1990s. Contemporary diet of Tufted Ducks was dominated by Asellus aquaticus (48%), but molluscs (14%), grain (13%) and chironomid larvae (11%) were also consumed. Between 1998-99 and 2010-12, the contribution of Asellus aquaticus to the diet significantly decreased while the proportions of chironomid larvae, grain, Gammarus spp. and Mysis spp. increased. Alternative methods of dietary analysis, for example stable isotope analysis, are recommended in future studies of diving duck diet at Lough Neagh.

This work aimed to study the diversity and distribution of marine sea turtles stranded in Potiguar Basin, Rio Grande do Norte, as well as aspects related to feeding behavior associated with human impacts. The study was conducted through the analysis of data from stranded animals, recorded in a daily monitoring in an area bounded on the north by the municipality of Aquiraz, in the state of Ceará, and the east by the municipality of Caicara do Norte, in the state of Rio Grande do Norte. Stranded dead animals were necropsied and for the analysis of the diet of animals, esophagus, stomach and intestines were fixed in 10% formalin and after that, the stomach contents were sorted and stored in 70% alcohol. Representative fragments of these organs were removed for making histological slides, with a view to histological characterization of the digestive tract. 2.046 occurrences of turtles were recorded during the period from 01/01/2010 to 31/12/2012. The Chelonia mydas species showed the highest number of records and it was observed in 66.81 % (N = 1,367) of cases; followed by Eretmochelys imbricata with 4.45 % (N = 91) and by Lepidochelys olivacea with 1.22% (N = 25). The Caretta caretta and Dermochelys coriacea species were, respectively, 0.93 % (N = 19) and 0.05 % (N = 1) records of strandings. In 26.54 % of cases, it was not possible to identify the species. Regarding the spatial distribution, the stretch A was the one that had the highest number of strandings and a larger number of records were registered in the warm months of the year. The dietary analysis showed that C. mydas fed preferentially on algae; C. caretta had a diet with a predominance of the item "coral´s fragments" and E. imbricata species showed preference for an animal origin material. Related to this anthropic interaction, 57.14 % (n = 76) of animals that died at the rehabilitation s base, showed cause of death due to complications from ingesting debris. According to the data presented, the Potiguar Basin presents itself as an area with important diversity and distribution of marine sea turtle as well is characterized as a feeding and nidification area for these species
