Effects of plateau zokors (Myospalax fontanierii) on seasonal above- and belowground plant biomass, plant species diversity, and soil moisture and organic matter were examined at an alpine meadow site in Qinghai Province, People's Republic of China. Above- and belowground biomass increased significantly in areas where zokors were removed or burrow systems were abandoned for 5 years compared with areas that zokors had occupied for >10 years. Biomass of monocotyledons was reduced greatly, but biomass of nonpalatable dicotyledons increased significantly, in occupied areas. Diversity of dicotyledons, monocotyledons, and total plants in unoccupied areas was significantly greater than in occupied or abandoned areas. Vegetation cover and height in occupied areas were significantly less than in unoccupied and abandoned areas. No consistent effect by zokors on soil moisture and organic matter was observed.

Isatis capadocica, a brassica collected from Iranian arsenic-contaminated mine spoils and control populations, was examined to determine arsenate tolerance, metabolism and accumulation. I. cappadocica exhibited arsenate hypertolerance in both mine and nonmine populations, actively growing at concentrations of > 1 mm arsenate in hydroponic solution. I. cappadocica had an ability to accumulate high concentrations of arsenic in its shoots, in excess of 100 mg kg(-1) DW, with a shoot : root transfer ratio of > 1. The ability to accumulate arsenic was exhibited in both hydroponics and contaminated soils. Tolerance in this species was not achieved through suppression of high-affinity phosphate/arsenate root transport, in contrast to other monocotyledons and dicotyledons. A high percentage (> 50%) of arsenic in the tissues was phytochelatin complexed; however, it is argued that this is a constitutive, rather than an adaptive, mechanism of tolerance.

Our findings revealed two distinct patterns of substrate preparation: the pattern of leaf-cutting ants foraging on dicotyledons is marked by highly fragmented substrate resulting in a more advanced initial decomposition. The pattern of leaf-cutting ants harvesting grasses is characterized by large pieces of substrate, resulting in little initial decomposition. Ants foraging on both types of plants are apparently intermediary between the two patterns, although more similar to the patterns of those foraging on dicotyledons. Also, the behavior of scraping the substrate was described for the first time, it is very important for the removal of the epicuticular wax layer of the leaves helping the growth of the symbiotic fungus.

The ultrastructural features and the plastid changes caused by sample preparation were studied in sieve elements of Panicum maximum leaves. Samples of expanded leaves, taken near the ligule region, were fixed and processed by common light and transmission electron microscopy methods. In mature sieve-tube elements, the protoplast is electron-translucent and plastids are the most frequent organelles. Mitochondria and smooth endoplasmic reticulum segments are also visible and occupy a parietal position within the cell. The plastids are globular and show electron-dense proteinaceous inclusions in the stroma. The protein crystals are predominantly cuneate, but thin crystalloids and amorphous and/or filamentous proteins also occur. The presence of intact plastids plus others in different phases of plastid envelope rupture were interpreted as evidence that this rupture is a normal event in response to injury. This plastid envelope rupture is possibly activated by the release of pressure in the sieve-tube element. After plastid membrane vesiculation, the stroma and the protein crystals are dispersed within the sieve-element ground cytoplasm. The vesicles originating from the plastid envelope move to one cell pole, while protein crystalloids move to the opposite pole and agglomerate in the sieve-plate region. Our findings indicate that these protein crystalloids, which deposit in the sieve plate, may act in sieve-plate pores occlusion, preventing the release of phloem sap, similar to the role of P-protein in dicotyledons.

Expansins are unusual proteins discovered by virtue of their ability to mediate cell wall extension in plants. We identified cDNA clones for two cucumber expansins on the basis of peptide sequences of proteins purified from cucumber hypocotyls. The expansin cDNAs encode related proteins with signal peptides predicted to direct protein secretion to the cell wall. Northern blot analysis showed moderate transcript abundance in the growing region of the hypocotyl and no detectable transcripts in the nongrowing region. Rice and Arabidopsis expansin cDNAs were identified from collections of anonymous cDNAs (expressed sequence tags). Sequence comparisons indicate at least four distinct expansin cDNAs in rice and at least six in Arabidopsis. Expansins are highly conserved in size and sequence (60-87% amino acid sequence identity and 75-95% similarity between any pairwise comparison), and phylogenetic trees indicate that this multigene family formed before the evolutionary divergence of monocotyledons and dicotyledons. Sequence and motif analyses show no similarities to known functional domains that might account for expansin action on wall extension. A series of highly conserved tryptophans may function in expansin binding to cellulose or other glycans. The high conservation of this multigene family indicates that the mechanism by which expansins promote wall extensin tolerates little variation in protein structure.
