Ten new cyclic hexadepsipeptides, six isariins and four isaridins, from the fungus Isaria have been identified and characterized by high-performance liquid chromatography, coupled to tandem electrospray ionization mass spectrometry (LC-ESIMS/MS). The isariins possess a beta-hydroxy acid residue and five alpha-amino acids, while isaridins contain a beta-amino acid, an alpha-hydroxy acid, and four alpha-amino acids. One- and two-dimensional NMR spectroscopy confirmed the chemical identity of some of the isariin fractions. Mass spectral fragmentation patterns of [M + H](+) ions reveal clear diagnostic fragment ions for the isariins and isaridins. Previously described cyclic depsipeptides, isarfelins from Isaria felina (Guo, Y. X.; Liu, Q. H.; Ng, T. B.; Wang H. X. Peptides 2005, 26, 2384), are now reassigned as members of the isaridin family. Examination of isaridin sequences revealed significant similarities with cyclic hexadepsipeptides such as destruxins and roseotoxins. The structure of an isariin (isariin A) investigated by NMR spectroscopy indicated the presence of a hybrid alpha beta C-11 turn, formed by the beta-hydroxy acid and glycine residues and a (D)Leu-(L)Ala type II' beta-turn. Additionally, the inhibitory effect of isariins and an isaridin on the intra-erythrocytic growth of Plasmodium falciparum is presented.

Two new cyclohexadepsipeptides have been isolated from the fungus Isaria. Fungal growth in solid media yielded hyphal strands from which peptide fractions were readily isolable by organic-solvent extraction. Two novel cyclodepsipeptides, isaridin A and isaridin B, have been isolated by reverse-phase HPLC, and characterized by ESI-MS and 1H-NMR. Single crystals of both peptides have been obtained, and their 3D structures were elucidated by X-ray diffraction. The isaridins contain several unusual amino acid residues. The sequences are cyclo(β-Gly-HyLeu-Pro-Phe-NMeVal-NMePhe) and cyclo(β-Gly-HyLeu-β-MePro-Phe-NMeVal-NMePhe), where NMeVal is N-methylvaline, NMePhe N-methylphenylalanine, and HyLeu hydroxyleucine (=2-hydroxy-4-methylpentanoic acid). The two peptides differ from one another at residue 3, isaridin A having an (S)-proline at this position, while β-methyl-(S)-proline (=(2S,3S)-2,3,4,5-tetrahydro-3-methyl-1H-pyrrole-2-carboxylic acid) is found in isaridin B. The solid-state conformations of both cyclic depsipeptides are characterized by the presence of two cis peptide bonds at HyLeu(2)-Pro(3)/HyLeu(2)-β-MePro(3) and NMeVal(5)-NMePhe(6), respectively. In isaridin A, a strong intramolecular H-bond is observed between Phe(4)CO⋅⋅⋅HNβ-Gly(1), and a similar, but weaker, interaction is observed between β-Gly(1)CO⋅⋅⋅HNPhe(4). Chemical investigation of the cytotoxic and anti-tuberculosis active butanone extract obtained from the growth media of the marine-derived fungus Beauveria felina led to the isolation of two new destruxins, [β-MePro] destruxin E chlorohydrin (1) and pseudodestruxin C (3), along with five known cyclic depsipeptides. The structures of the new destruxin derivatives were established by analysis of spectroscopic data, while the absolute configuration of the common amino acid residues was established by Marfey's analysis. The absolute configuration of the 2(A),4(5)-5-chloro-2,4-dihydroxypentanoic acid residue in 1 could be established by application of a J-based configuration method followed by derivatization with R-MPA-Cl and NMR analysis. © Japan Antibiotics Research Association.

Two new peptidic proteasome inhibitors were isolated as trace components from a Curacao collection of the marine cyanobacterium Symploca sp. Carmaphycin A (1) and carmaphycin B (2) feature a leucine-derived a,beta-epoxyketone warhead directly connected to either methionine sulfoxide or methionine sulfone. Their structures were elucidated on the basis of extensive NMR and MS analyses and confirmed by total synthesis, which in turn provided more material for further biological evaluations. Pure carmaphycins A and B were found to inhibit the beta 5 subunit (chymotrypsin-like activity) of the S. cerevisiae 20S proteasome in the low nanomolar range. Additionally, they exhibited strong cytotoxicity to lung and colon cancer cell lines, as well as exquisite antiproliferative effects in the NCI60 cell-line panel. These assay results as well as initial structural biology studies suggest a distinctive binding mode for these new inhibitors.

The increasing resistance of Rhipicephalus (Boophilus) microplus tick to commercial insecticides requires alternative methods for the control of this cattle plague. The enthomopathogenic fungus Beauveria feline produces destruxins in culture media, cyclic depsipeptides which display an array of biological activities. The present investigation aimed to evaluate the acaricide action of destruxins isolated from B. felina culture media on R. (B.) microplus engorged females. B. felina was grown in MF medium under 19 different growth conditions. HPLC-PDA analysis of chromatographic fractions obtained from the 19 different growth media extracts indicated the presence of destruxins in all lipophylic fractions. Such fractions were combined and subjected to separation by HPLC. Fractions containing distinct destruxins composition were tested against R. (B.) micro plus. Two fractions, composed of destruxin Ed(1) and pseudodestruxin B and/or pseudodestruxin C (fraction P1) as well as by hydroxyhomodestruxin B and/or destruxin D and/or roseotoxin C (fraction P7), displayed 30% and 28.7% acaricidal efficacy, respectively. This activity profile in such low concentration is adequate to consider destruxins as potential leading compounds to be developed for tick biological control. (C) 2012 Elsevier Inc. Der Suche nach neuen Wirkstoffen für den chemischen Pflanzenschutz kommt insbesondere vor dem Hintergrund der steigenden Weltbevölkerung und weniger zur Verfügung stehenden kulturfähigen Ackerflächen eine stetig wachsende Bedeutung zu. Ziel dieser Arbeit war die Synthese von cyclischen Peptiden und Depsipeptiden, die aufgrund ihrer biologischen Aktivität als potentielle Insektizide für den chemischen Pflanzenschutz in Frage kommen. Darüber hinaus sollten von Kohlenhydraten abgeleitete Katalysatoren zur enantioselektiven Cyanhydrinsynthese entwickelt werden, um einen leichten Zugang zu den Bausteinen der Depsipeptide zu ermöglichen. Als vielversprechender Naturstoff mit insektiziden Eigenschaften gilt das cyclische Pentapeptid Cycloaspeptid E, dessen Totalsynthese in 10 Stufen mit einer Gesamtausbeute von 25% erreicht wurde, sodass die Verbindung für biologische Tests bereitgestellt werden konnte. Zusätzlich gelang die Kristallisation der Verbindung, was eine Röntgenstrukturanalyse ermöglichte. Ein Derivat von Cycloaspeptid E sollte 2-Aminonicotinsäure anstelle von Anthranilsäure enthalten. Die Synthese dieser Verbindung wurde auf drei Wegen versucht. Dabei zeigte sich, dass es bei einer zur Totalsynthese des Naturstoffs analogen Strategie zur quantitativen Bildung eines Diketopiperazins kommt. Auf den anderen Routen ließ sich entweder ein Kupplungsschritt nicht realisieren, oder die Verbindung erwies sich unter den gewählten Bedingungen als instabil. Die Darstellung eines 2-Aminonicotinsäure-Derivats von Cycloaspeptid E bleibt daher weiterhin ein ungelöstes Problem, das weiterer Forschung bedarf. Verticilid A1 ist ein cyclisches Depsipeptid, das aufgrund seiner Bindungsfähigkeit an den Ryanodinrezeptor von Insekten, als Leitstruktur für die Suche nach neuen Insektiziden von Interesse ist. Um zu untersuchen, wie wichtig die Esterbindungen im Molekül für die biologische Aktivität sind, sollte das entsprechende Amid-Derivat und das Cyclodepsipeptid mit nur zwei statt vier Esterbindungen hergestellt werden. Hierbei zeigte sich, dass eine zur Darstellung von Verticilid A1 analoge Syntheseroute zu einer ausgeprägten Epimerisierung führt. Eine lineare Synthese der Derivate endet in der Bildung des Diketopiperazins. Weiterhin wurden zwei neue, zueinander pseudoenantiomere Vanadium(IV)-Katalysatoren auf Basis von D-Glucose einerseits und L-Xylose andererseits dargestellt. Diese lassen sich in fünf bzw. 14 Stufen synthetisieren und liefern in der enantioselektiven Katalyse von Mandelsäurenitril Enantiomerenüberschüsse von 89% bzw. 91% bei hohen Ausbeuten. Zusammenfassend lässt sich feststellen, dass im Rahmen dieser Arbeit die Totalsynthese von Cycloaspeptid E erfolgreich durchgeführt wurde, und die Syntheseversuche von weiteren cyclischen Peptiden wichtige Erkenntnisse für weitere Synthesen lieferten. Mit den beiden hergestellten Vanadium(IV)-Komplexen wurden zwei potente, auf Kohlenhydraten basierende Katalysatoren für die enantioselektive Synthese von Cyanhydrinen entwickelt.

A nematocidal agent present in a southern Australian marine sponge of the genus Echinodictyum has been isolated and identified by detailed spectroscopic analysis and total synthesis as the novel betaine (-)-echinobetaine A (6). Preliminary SAR investigations have been undertaken.
