The venom proteome of Daboia russelli siamensis, a snake of medical importance in several Asian countries, was analysed by 2-D electrophoresis, subsequent MS/MS and enzymatic assays. The proteome comprises toxins from six protein families: serine proteinases, metalloproteinases, phospholipases A(2), L-amino acid oxidases, vascular endothelial growth factors and C-type lectin-like proteins. The venom toxin composition correlates with the clinical manifestation of the Russell's viper bite and explains pathological effects of the venom such as coagulopathy, oedema, hypotensive, necrotic and tissue damaging effects. The vast majority of toxins are potentially involved in coagulopathy and neurotoxic effects. The predominant venom components are proteinases capable of activating blood coagulation factors and promoting a rapid clotting of the blood, and neurotoxic phospholipase A(2)s. The analysis of the venom protein composition provides a catalogue of secreted toxins. The proteome of D. r. siamensis exhibits a lower level of toxin diversity than the proteomes of other viperid snakes. In comparison to the venoms of Vipera ammodytes ammodytes and Vipera ammodytes meridionalis, the venom from D. r. siamensis showed quantitative differences in the proteolytic, phospholipase A2, L-amino acid oxidase and alkaline phosphatase activities. 按照Promega 公司的mRNA 提取试剂盒操作手册, 从圆斑蝰蛇( Daboia russellii siamensis ) 的毒腺中提 取mRNA ; 利用RT2PCR 的方法进行体外扩增, 获得C - 型凝集素蛋白的基因, 克隆到pMD182T 载体中。随机挑 选14 个阳性克隆进行核酸测序, 获得7 个编码不同蛇毒C - 型凝集素样蛋白亚基的cDNA , 分别命名为DRS2L1 、 DRS2L2 、DRS2L3 、DRS2L4 、DRS2L5 、DRS2L6 和DRS2L7 。由基因序列推导出的氨基酸序列表明, 克隆到的7 个蛇 毒C - 型凝集素样蛋白的亚基中均有糖识别结构域存在。BLAST 分析显示, 仅有DRS2L1 的蛋白序列与目前已知 的蛇毒C - 型凝集素样蛋白的α亚基相似。序列同源性比较并结合半胱氨酸位点分析, 推测DRS2L1 和DRS2L2 可能分别是圆斑蝰蛇毒Ⅹ因子激活剂的轻链LC2 和LC1 。DRS2L3 和DRS2L4 可能是高分子量的蛇毒C - 型凝集 素样蛋白的β亚基, 而DRS2L5 和DRS2L6 可能是低分子量的蛇毒C - 型凝集素样蛋白的β亚基。DRS2L7 可能是 类似于血小板膜糖蛋白Ib 结合蛋白的β亚基。

One novel Kunitz BPTI-like peptide designated as BBPTI-1, with chymotrypsin inhibitory activity was identified from the venom of Burmese Daboia russelii siamensis. It was purified by three steps of chromatography including gel filtration, cation exchange and reversed phase. A partial N-terminal sequence of BBPTI-1, HDRPKFCYLPADPGECLAHMRSF was obtained by automated Edman degradation and a Ki value of 4.77. nM determined. Cloning of BBPTI-1 including the open reading frame and 3' untranslated region was achieved from cDNA libraries derived from lyophilized venom using a 3' RACE strategy. In addition a cDNA sequence, designated as BBPTI-5, was also obtained. Alignment of cDNA sequences showed that BBPTI-5 exhibited an identical sequence to BBPTI-1 cDNA except for an eight nucleotide deletion in the open reading frame. Gene variations that represented deletions in the BBPTI-5 cDNA resulted in a novel protease inhibitor analog. Amino acid sequence alignment revealed that deduced peptides derived from cloning of their respective precursor cDNAs from libraries showed high similarity and homology with other Kunitz BPTI proteinase inhibitors. BBPTI-1 and BBPTI-5 consist of 60 and 66 amino acid residues respectively, including six conserved cysteine residues. As these peptides have been reported to have influence on the processes of coagulation, fibrinolysis and inflammation, their potential application in biomedical contexts warrants further investigation. © 2013 Elsevier Inc.

A double antibody sandwich enzyme linked immunosorbent assay (ELISA) was developed to detect Echis carinatus venom in various organs (brain, heart, lungs, liver, spleen and kidneys) as well as tissue at the site of injection of mice, at various time intervals (1, 6, 12, 18, 24 h and 12 h intervals up to 72 h) after death. The assay could detect E. carinatus venom levels up to 2.5 ng/ml of tissue homogenate and the venom was detected up to 72 h after death. A highly sensitive and species-specific avidin-biotin microtitre ELISA was also developed to detect venoms of four medically important Indian snakes (Bungarus caeruleus, Naja naja, E. carinatus and Daboia russelli russelli) in autopsy specimens of human victims of snake bite. The assay could detect venom levels as low as 100 pg/ml of tissue homogenate. Venoms were detected in brain, heart, lungs, liver, spleen, kidneys, tissue at the bite area and postmortem blood. In all 12 human victim cadavers tested the culprit species were identified. As observed in mice, tissue at the site of bite area showed the highest concentration of venom and the brain showed the least. Moderate amounts of venoms were found in liver, spleen, kidneys, heart and lungs. Development of a simple, rapid and species-specific diagnostic kit based on this ELISA technique useful to clinicians is discussed.

本论文研究了从圆斑蛙蛇泰国亚种(Daboia russellii siamensis)蛇毒中纯化的C一型凝集素样蛋白Dabocetin和L一氨基酸氧化酶DRS一AO的理化性质、生物学活性和分子克隆。Dabocetin是分子量约为28扔。的异二聚体蛋白,它由分子量约为15.0kDa和14.5kDa的两个同源亚基以和p共价结合形成。N-末端氨基酸序列比较显示,Dabocetin与目前已知的蛇毒c一型凝集素样蛋白有很高的同源性。即使在终浓度达50.0。叫而时,Dabocetin也不能直接诱导血小板聚集。此外,在终浓度为40.00μg/ml时,Dabocetin几乎不能抑制由AdP,TMVA和stejnulxin诱导的血小板聚集。但是,Dabocetin呈剂量依赖地抑制瑞斯托霉素诱导的血小板凝集,其半数抑制率ICS。值为10.80ug/ml。流式细胞仪分析表明,Dabocetin显著抑制单克隆抗体522与GPIba的结合,提示Dabocetin很可能是一个GPIb结合蛋白。从圆斑蛙蛇的毒腺中克隆到了7个编码不同蛇毒C一型凝集素样蛋白亚基的七DNA(命名为DRs一1至DRs一7)。其中,DRsLS编码Dabocetin的a亚基,DRS一6编码Dabocetin的p亚基。DRs一1和DRS一2很可能是圆斑蛙蛇毒腺中表达的X因子激活剂的两条轻链LCZ和LCI的山NA。DRS一3,DRS毛4和DRSL7可能是圆斑蛙蛇毒腺中表达的C一型凝集素样蛋白p亚基的。NA。DRsLAO是一个新的L一氨基酸氧化酶,比活力为1.98U加噶。十二烷基磺酸钠一聚丙烯酞氨凝胶电泳(SDs-PAGE)分析显示,该酶在还原和非还原条件下均呈现一条蛋白带,表观分子量约为58kDa。N-末端氨基酸序列比较显示,DRS一AO与目前已知的蛇毒L一氨基酸氧化酶有很高的同源性。该酶的最适底物为L一亮氨酸,最适pH为8.8。DRs一Ao呈剂量依赖地抑制扔P和仆IvA诱导的血小板聚集,其半数抑制率ICS。值分别为32.8μg/ml和32.3μg/ml。DRS-LAO对金黄色葡萄球菌(灯Cc25923)和耐甲氧西林金黄色葡萄球菌有较强的抗菌作用。DRs一AO对金黄色葡萄球菌必Tcc25923)的最低抑菌浓度卿C)和最低杀菌浓度耐甲氧西林金黄色葡萄球菌的孤CS。和呱Cg。值分别为18.。林留时和36.0μg/ml;DRSLAo对耐甲氧西林金黄色葡萄球菌的MBC50和MBCg。值分别为36.0μg/ml和72.0μg/ml。通过对DRS一AO的分子克隆,得到了编码DRS-AO的部分cDNA序列。

磷脂酶AZ(PLA2)是蛇毒中含量较为丰富的一类作用于梭酷键的酶。迄今为止,己有多种形式的PLA2从不同地域、不同种属的蛇毒中得以纯化并进行了较为系统的研究。其中,以VipoXin为代表的异二聚体形式PLA2较为引人注目,原因在于这种形式不同于此类蛋白家族中的诸多其它个体。目前,己经有许多关于此异二聚体PL凡生物学特性的报道,包括对此类形式存在原因、活性变化、结构表现、系统进化等方面的讨论。然而至今,这种以异二聚体形式存在的PLA2仅发现于几种蛙亚科(ViperinaeSubfamily)蛇种的蛇毒中,其中就包括我国台湾岛的圆斑蜂蛇台湾亚种(Doboiarusselliiformosensis),而蝮亚科(CrotaiinaeSubfamil)蛇种的蛇毒至今却没有此类报道。我国大陆西南端接壤东南亚,存在于云南、福建一带的圆斑蛙蛇隶属圆斑蛙蛇泰国亚种(Daboiarusselliisiamensis),那么这种蛇毒中是否也含有异二聚体形式的PLA2呢?本工作就此疑问对云南产圆斑蛙蛇泰国亚种(D.r.siamensis)蛇毒中的PLA2进行了研究,结果得到三个新的PLAZ,分别命名为DRS-PLA2-I、DRS-PLA2-II和DRS-PLA2-III。其中,DRS-PLA2-I的分子量为13864.06Da,理论pI为4.56,PLA2活性为12.35μmol/mg/min;DRS-PLA2-II的分子量为13635.99Da,理论pI为8.74,PLA2活性为8.76μmol/mg/min;DRS-PLA2-III的分子量为13619.80Da,理论厂为4.61,无PLA2活性。这三个蛋白酶N端的30个氨基酸残基恰好和三个阳性克隆的cDNA序列推导的蛋白序列吻合,结合已经报道的PLA2蛋白家族蛋白序列的保守性表现,我们可以断定它们之间存在对应关系。分子系统学分析表明DRS-PLA2-II和DRS-PLA2-III在进化关系上和蛙亚科的异二聚体PLA2关系较近,并且二者酶活性分别与异二聚体PLA2的Normalchain和Inhibitorchain相一致,只是没有发现类似Vipoxin形式的异二聚体结合蛋白。这些分析表明DRS-PLA2-nORS-PLA2-III类似圆斑蛙蛇台湾亚种(D.r.forlnos翻s沽)中的PV-4/RV-7,是PLA2异二聚体的一种特殊形式,在进化上滞后于VinOXin。另夕卜本工作还相继从云南产菜花烙铁头(Trimeresrusjerdonii)蛇毒和湖南产烙铁头(Trimeresurusmucrosquamatus)蛇毒中分离得到Jerdonase和TmF。前者为一个丝氨酸蛋白酶性质的、具有纤维蛋白原水解作用和激肤释放酶原水解作用双重活性表现的、高分子量的份五brinogenase,其活性表现可以被PMSF彻底抑制,而EDTA对此却没有影响。其它的几种抑制剂如大豆胰蛋白酶抑制剂、l-cysteine、DTT对Jerdonase的活性表现也有不同程度的影响。在Jerdonase的这些生化特性上中,分子量的大小和对纤维蛋白酶水解的特性这两方面有别于蛇毒中诸多其它来源的同类蛋白;后者T淤为一个舒缓激肚增强肤(BradykninPQtentiatingPePtide,BPP),电离质谱分析表明其分子量为1110.7Da。此小肚氨基酸序列为促进舒缓激肚(Bradki垃n,BK)诱导的豚鼠回肠纵行肌收缩的活力单位为(1.13±0.3)(m留L),T妊抑制血管紧张素转化酶(ACE)对BK水解的半数抑制剂量IC50为2μg。比较已报道的从Agkistrodon属和Bothrops属中纯化得到的BPP氨基酸序列发现:BPP的N端都是特征性的pGlu,C端为IIe-Pro-Pro,有高度的保守性。另外,TmF是Trimeresurus属中此类小肤的首次纯化。总之,本研究对国产的几种常见蛇毒中的几种常见蛋白多肤进行了一定程度的探讨和分析,和相同类别的其它蛋白、多肤比较可以看到,有许多相同的地方,也有许多不同的表现,研究结果为相应领域的深入研究提供资料和思路。

Siamese mud carp (Henichorynchus siamensis) is a freshwater teleost of high economic importance in the Mekong River Basin. However, genetic data relevant for delineating wild stocks for management purposes currently are limited for this species. Here, we used 454 pyrosequencing to generate a partial genome survey sequence (GSS) dataset to develop simple sequence repeat (SSR) markers from H. siamensis genomic DNA. Data generated included a total of 65,954 sequence reads with average length of 264 nucleotides, of which 2.79% contain SSR motifs. Based on GSS-BLASTx results, 10.5% of contigs and 8.1% singletons possessed significant similarity (E value < 10–5) with the majority matching well to reported fish sequences. KEGG analysis identified several metabolic pathways that provide insights into specific potential roles and functions of sequences involved in molecular processes in H. siamensis. Top protein domains detected included reverse transcriptase and the top putative functional transcript identified was an ORF2-encoded protein. One thousand eight hundred and thirty seven sequences containing SSR motifs were identified, of which 422 qualified for primer design and eight polymorphic loci have been tested with average observed and expected heterozygosity estimated at 0.75 and 0.83, respectively. Regardless of their relative levels of polymorphism and heterozygosity, microsatellite loci developed here are suitable for further population genetic studies in H. siamensis and may also be applicable to other related taxa.
