Zinc is present in high concentration in many structures of the limbic circuitry, however the role of zinc as a neuromodulator in such synapses is stilt uncertain. In this work, we verified the effects of zinc chelation in an animal model of epileptogenesis induced by amygdala rapid kindling. The basolateral. amygdala was electrically stimulated ten times per day for 2 days. A single stimulus was applied on the third day. Stimulated animals received injections of PBS or the zinc chelator diethildythiocarbamate acid (DEDTC) before each stimulus series. Animals were monitored with video-EEG and were perfused 3 h after the last stimulus for subsequent neo-Timm and Ftuoro-Jade B analysis. Zinc chelation decreased the duration of both behavioral seizures and electrical after-discharges, and also decreased the EEG spikes frequency, without changing the progression of behavioral seizure severity. These results indicate that the zinc ion may have a facilitatory role during kindling progression. (c) 2008 Elsevier B.V. All rights reserved.

A presente dissertação descreve o desenvolvimento e a caracterização de sensores potenciométricos com base em polímeros de impressão molecular e de sensores ópticos com base em membranas de poli(cloreto de vinilo), PVC, para a determinação de cobre em vinhos verdes. Os sensores potenciométricos foram preparados a partir de diferentes solventes (metanol e clorofórmio), tendo o seu crescimento decorrido na presença ou ausência da molécula molde (cobre). Os sistemas sensores selectivos ao cobre continham partículas de polímeros com ou sem impressão molecular como material electroactivo, dispersas em solvente plastificante, PVC e, em alguns casos, aditivo aniónico. A avaliação dos vários sistemas baseou-se na comparação das características operacionais dos diversos eléctrodos onde foram aplicados. Estas características foram obtidas a partir de curvas de calibração, cujos declives e limites de detecção variaram entre -39,9 – 37,0 mV decada-1 e 4,2 – 29,1 μg mL-1, respectivamente. Os sensores não são independentes do pH uma vez que o complexo formado entre o cobre e a difenilcarbazida é favorecido por valores de pH próximos de 5. Assim, obtiveram-se melhores resultados usando água desionizada ou solução tampão de HEPES revelando-se um método rápido e relativamente eficaz nestas condições. Os sensores ópticos basearam-se na reacção colorimétrica entre o cobre e um complexante. Os reagentes complexantes escolhidos foram a neocuproína, a difenilcarbazida e o dietilditiocarbamato de sódio. Avaliou-se o efeito de vários parâmetros experimentais na resposta destes sensores, tais como o pH (avaliado para os valores 3,00 e 5,00), a concentração de cobre (que variou entre 0,06 e 317,7 mg L- 1) e as próprias características da membrana. Os melhores resultados foram obtidos a pH 3, numa gama de concentrações de 0,06 e 31,8 mg L-1 usando a difenilcarbazida como reagente complexante. A aplicação destes sensores a vinhos requer ainda estudos adicionais, especialmente no que diz respeito à necessidade de implementar algum procedimento de pré-tratamento de amostra.

Diethyldithiocarbamate (ditiocarb), a metabolite of the old anti-alcoholic drug disulfiram (Antabuse), forms proteasome-inhibiting metal complexes with copper or zinc that suppress cancer cells both in vitro and in vivo. The drug has been used in a clinical trial (NCT00742911) along with copper gluconate as a dietary supplement in patients with cancer spreading to the liver. In this study, we demonstrate the effect of synthetic complexes of disulfiram with four various metals (Mn, Fe, Cr and Cu) used as food supplements. These complexes may be spontaneously formed in the blood during the use of disulfiram with divalent metals and thus may suppress the growth of cancer in vivo. The cytotoxic effect of the compounds and the compounds' ability to inhibit the cellular proteasome were tested in the osteosarcoma cell line U2OS. After 48 h, copper and manganese complexes exhibited cytotoxic effect on the cell line, in sharp contrast to both iron and chromium complexes. A flow system coupled to a tungsten coil atomizer in an atomic absorption spectrometer (TCA-AAS) was developed for As(III) determination in waters, by extraction with sodium diethyldithiocarbamate (NaDDTC) as complexing agent, and by sorption of the As(III)-DDTC complex in a micro-column filled with 5 mg C18 reversed phase (10 µL dry sorbent), followed by elution with ethanol. A complete pre-concentration/elution cycle took 208 s, with 30 s sample load time (1.7 mL) and 4 s elution time (71 µL). The interface and software for the synchronous control of two peristaltic pumps (RUN/ STOP), an autosampler arm, seven solenoid valves, one injection valve, the electrothermal atomizer and the spectrometer Read function were constructed. The system was characterized and validated by analytical recovery studies performed both in synthetic solutions and in natural waters. Using a 30 s pre-concentration period, the working curve was linear between 0.25 and 6.0 µg L-1 (r = 0.9976), the retention efficiency was 94±1% (6.0 µg L-1), and the pre-concentration coefficient was 28.9. The characteristic mass was 58 pg, the mean repeatability (expressed as the variation coefficient) was 3.4% (n=5), the detection limit was 0.058 µg L-1 (4.1 pg in 71 µL of eluate injected into the coil), and the mean analytical recovery in natural waters was 92.6 ± 9.5 % (n=15). The procedure is simple, economic, less prone to sample loss and contamination and the useful lifetime of the micro-column was between 200-300 pre-concentration cycles.

A high performance liquid chromatographic-diode array detection method for the determination of busulfan in plasma was developed and validated. Sample preparation consisted of protein precipitation followed by derivatization with sodium diethyldithiocarbamate and liquid-liquid extraction with methyl-tert-butyl ether. Chromatograms were monitored at 277 nm. Separation was carried out on a Lichrospher RP 18 column (5 µm, 250 x 4 mm). The mobile phase consisted of water and acetonitrile (20:80, v/v). The method presented adequate specificity, linearity, precision and accuracy and allowed reliable determination of busulfan in clinical plasma samples, being applied to three patients submitted to bone marrow transplantation.

A dispersive liquid-liquid microextraction based on solidification of floating organic drop for simultaneous extraction of trace amounts of nickel, cobalt and copper followed by their determination with electrothermal atomic absorption spectrometry was developed. 300 µL of acetone and 1-undecanol was injected into an aqueous sample containing diethyldithiocarbamate complexes of metal ions. For a sample volume of 10 mL, enrichment factors of 277, 270 and 300 and detection limits of 1.2, 1.1 and 1 ng L-1 for nickel, cobalt and copper were obtained, respectively. The method was applied to the extraction and determination of these metals in different water samples.

Zinc salts of ethyl, isopropyl, and butyl xanthates were prepared in the laboratory. The effect of these xanthates in combination with zinc diethyldithiocarbamate (ZDC) on the vulcanization of silica-filled NBR compounds has been studied at different temperatures. The cure times of these compounds were compared with that of NBR compounds containing tetramethylthiuram disulphide/dibenzthiazyl disulphide. The rubber compounds with the xanthates and ZDC were cured at various temperatures from 60 to 150°C. The sheets were molded and properties such as tensile strength, tear strength, crosslink density, elongation at break, compression set, abrasion resistance, flex resistance, heat buildup, etc. were evaluated. The properties showed that zinc salt of xanthate/ZDC combination has a positive synergistic effect on the cure rate and mechanical properties of NBR compounds.

Zinc salts of ethyl, isopropyl and butyl xanthates were prepared in the laboratory. The rubber compounds with the three xanthate accelerators and ZDC were cured at various temperatures from 60°C to 150°C. The sheets were moulded and properties such as tensile strength , tear strength , cross-link density, elongation -at-break, compression set, heat build up, abrasion resistance, flex resistance , etc. were evaluated . The properties showed that zinc xanthate/ZDC accelerator combination has a positive synergistic effect on the mechanical properties of NR compounds. The curing of HAF filled NR compound containing zinc xanthate /ZDC is slightly slower than the curing of the corresponding gum compounds . It is observed that, by gradually increasing the amount of the accelerator, the cure time of black filled NR compound can be made equal to that of the gum compou

This study present a novel NO sensor made of a spin trap (iron(II)-diethyldithiocarbamate complex, FeDETC) incorporated in a latex rubber matrix and works as a trap for NO, which is detectable by Electron Paramagnetic Resonance (EPR). We explored the optimization of our sensors changing systematically two fabrication parameters: the latex rubber matrix temperature of polymerization and FeDETC concentration inside the matrix. The sensor was prepared in four different temperatures: 4, 10, 20 and 40°C. The FeDETC concentration was also varied from 0.975 to 14.8 mM. We observed a variation of the EPR signals from the sensors prepared at different conditions. We found a high stability of the EPR response from our sensor, 40 days at RT. The best sensor was made with a latex rubber matrix polymerized at 10°C and with a FeDETC concentration of 14.8 mM. In vivo tests show good biocompatibility of our sensor. © 2007 Asian Network for Scientific Information.

Pelo uso de técnicas mais baratas, como a do SDDC, é possível determinar níveis traços de arsênio em cabelo; entretanto esta técnica apresenta alguns inconvenientes como baixa estabilidade e o odor desagradável da piridina. A piridina foi substituída por trietanolamina/CHCl3 e as características analíticas do complexo foram estudadas. O complexo foi estável por 270 minutos, a faixa de aplicação da lei de Beer foi de 0,0 a 25,0 mg As, a repetibilidade foi de 0,028 mg As, o limite de detecção foi de 18,6 mg de As/L e a sensibilidade (e)? de 1,12 104 L.mol-1.cm-1. O método foi aplicado à amostras de cabelo. A lavagem das amostras foi feita com extran e água desionizada e seca em estufa (40-60ºC). 0,1000 g de amostra foi submetida à 11 métodos de digestão. O melhor método foi o que usou uma solução 1:1 de HNO3 e H2SO4 concentrados em temperatura de 100-110ºC com evaporação até fumos de SO3. O tempo de abertura é um inconveniente neste tipo de digestão.

Várias toneladas de rejeito de manganês contendo arsênio, gerado por uma empresa de mineração, foram utilizadas como aterro de ruas da cidade de Santana-AP. A possibilidade de exposição das pessoas residentes nessas localidades levou ao estudo de quantificação de arsênio total nos solos. Após a digestão, os teores de arsênio foram quantificados por espectrofotometria de absorção molecular usando um sistema automático de geração de hidretos (HG-MAS), diciclohexilamina/CHCl3 como solvente do dietilditiocarbamato de prata (SDDC) e KBH4 como redutor. O método apresentou bons resultados com sensibilidade (ε) de 1,10 104 L.mol-1.cm-1, estabilidade de 2,96% e outras vantagens em relação ao método oficial. O método foi aplicado em amostras de referência de solo com recuperação de 98,82 % (N=10). As análises de solos mostraram que do total de amostras analisadas 94,74 % apresentaram concentração de arsênio acima do valor editado pela CETESB para solo residencial (50 mg.kg-1) com valor médio de 682,96 mg.kg-1, variando de 48,08 mg.kg-1 a 1.713,00 mg.kg-1 que comprova a contaminação do solo pelo arsênio.

Urines from patients administered mutagenic antineoplastic drugs were significantly mutagenic in the Ames assay, and hence may pose a genotoxic hazard to hospital personnel or family members caring for the patient. The urines were tested for mutagenicity in several different strains of Salmonella typhimurium that were uvr positive or negative (TA98, TA100, TA102, UTH8413, UTH8414). The urines were fractionated by high pressure liquid chromatography (HPLC) and the fractions assayed for mutagenicity in the strains in which the whole urine was mutagenic. Only fractions of urines containing the parent compound (cisplatin, doxorubicin, or mitomycin) were mutagenic; no other fraction showed significant mutagenicity. However, urine containing cyclophosphamide had two fractions that were mutagenic. One fraction, the fraction containing cyclophosphamide, required metabolic activation for mutagenicity. The other fraction did not require activation for mutagenicity.^ The chemical and mutagenic stability of these urines at room temperature was assayed over a 14 day period. The parent compound degraded within the first seven days, but the urines remained mutagenic. Cis-platinum was chemically stable in the urine; however, the urine decreased in mutagenicity. The decrease was probably the result of stable ligands binding to the platinum.^ Inactivation methods were developed to reduce the genotoxic hazard. Urine containing cisplatin was inactivated by complexing the cisplatin with diethyldithiocarbamate (DDTC). Oxidation with NaOCl of urines containing mitomycin and doxorubicin (sodium thiosulfate must be added to the doxorubicin urine) results in mutagenic inactivation. Inactivation of urine containing cyclophosphamide requires oxidation with alkaline potassium permaganate and trapping of active degradation products with sodium thiosulfate. Urines containing these drugs can be inactivated, but not always by the same method that inactivates the drug alone in solution. Therefore, in the future development of inactivation methods, both chemical and mutagenic assays are necessary to determine effectiveness. Methods of inactivation of mutagenic excreta developed in this study are both effective and practical. ^
