Cyt b-559是光系统II反应中心的成分之一,它由亚基和亚基组成的。在Cyt b-559中,血红素辅基与两个亚基中的组氨酸连接成有功能的蛋白,并维持PSII的功能稳定性。前人曾将与血红素相连的His突变,导致Cyt b-559功能和PSII稳定性的丧失。基于此研究,本文采用定点突变技术,将亚基中与His23位置最近的上游氨基酸Arg18分别用Gly和Glu取代,下游氨基酸Ser24用Phe取代,获得了衣藻Cyt b-559的突变体。对突变体的分析,有以下新结果:突变体都能进行光合自养,但无论在异养培养基上还是自养培养基上,和对照相比,其生长速度非常缓慢; PSII的活性分析,表明PSII的放氧活性为野生衣藻细胞的50%~80%, Fv/Fm 的荧光参数为40%~70%;对突变体进行强光(1000μE•m-2•s-1)照射,10min后,其放氧活性都降低为0,而野生型衣藻还保持35%的活性;提取类囊体膜蛋白,进行SDS-PAGE电泳和Western-blotting分析,显示突变体的膜蛋白与对照无显著差异。这些结果说明对围绕血红素环境的固有氨基酸的改变,虽然并没有明显影响类囊体膜蛋白的表达和组成,但是却影响了衣藻细胞的生长和PSII的活性,增加了衣藻细胞对强光的敏感性,降低了衣藻细胞自身的光保护能力。这说明靠近血红素配位环境的氨基酸Arg和Ser,尤其是Arg,对Cyt b-559的功能维持不可缺少,对于维持PSII的活性也很重要。

疣螈属的红瘰疣螈(Tylototriton shanjing)和棕黑疣螈(T.verrucosus)的物种界限一直不清楚.测定了来自中国西南地区14个地点的T.shanjing和T.verrucosus共40只标本的线粒体DNA Cyt b基因(753 bp).结果表明:(1)用邻接法、最大简约法和贝叶斯法等3种系统发育分析法分别重建棕黑疣螈种组系统发育树的拓扑结构不支持T.shanjing是单系群;(2)T.shanjing与T.verrucsus的mtDNA Cyt b序列差异平均值仅为1.2%.未达到种级水平.因此,全部T.shanjing样品都属于同一个物种,即T.verrucosus,不支持T.shanjing的物种地位,T.shanjing为T.verrucosus的同物异名,并建议恢复T.verrucosus的中文名红瘰疣螈.根据基于40个样品Cvt b基因序列的系统发育树和遗传变异以及地理分布,这些红瘰疣螈(T.verrucosus)样品聚为3支,即中国西南地区的红瘰疣螈可分为片马、滇中滇西和滇东南3个地理居群.

测定了来自黄河上游和柴达木盆地托索湖的裸裂尻鱼共16个个体的Cytb基因全序列(1141bp),探讨了种群结构和遗传多样性。用MEGA2.1软件分析了碱基组成和序列变异;以青海湖裸鲤、花斑裸鲤和极边扁咽齿鱼为外类群,用PAUP*4.0b10程序构建了单倍型NJ树;用Arlequin Ver.2000程序计算了群体间遗传变异值(Fst)和Nm值以及群体分化概率值。结果显示,来自柴达木水系托索湖的裸裂尻鱼没有形成单系群,Fst=0.204(P<0.05),Nm=1.95。初步判断,黄河和柴达木水系托索湖的裸裂

The genetic diversity and phylogeographical patterns of Trypanosoma species that infect Brazilian bats were evaluated by examining 1043 bats from 63 species of seven families captured in Amazonia, the Pantanal, Cerrado and the Atlantic Forest biomes of Brazil. The prevalence of trypanosonne-infected bats, as estimated by haemoculture, was 12.9%, resulting in 77 Cultures of isolates, most morphologically identified as Trypanosoma cf. cruzi, classified by barcoding using partial sequences from ssrRNA gene into the subgenus Schizotrypanum and identified as T. cruzi (15), T cruzi marinkellei (37) or T. cf. dionisii (25). Phylogenetic analyses using nuclear ssrRNA, glycosomal glyceraldehyde 3-phosphate dehydrogenase (gGAPDH) and mitochondrial cytochrome b (Cyt b) gene sequences generated three clades, which clustered together forming the subgenus Schizotrypanum. In addition to vector association, bat trypanosomes were related by the evolutionary history, ecology and phylogeography of the bats. Tryponosoma cf. dionisii trypanosomes (32.4%) infected 12 species from four bat families captured in all biomes, from North to South Brazil, and clustered with T. dionisii from Europe despite being separated by some genetic distance. Trypanosoma cruzi marinkellei (49.3%) was restricted to phyllostomid bats from Amazonia to the Pantanal (North to Central). Trypanosoma cruzi (18.2%) was found mainly in vespertilionid and phyllostomid bats from the Pantanal/Cerrado and the Atlantic Forest (Central to Southeast), with a few isolates from Amazonia. 植物已经演化出多种保护其免受强光抑制和破坏的机制,从而使植物体在自然界能够应付复杂多变的光照环境。虽然人们早就确定Cyt b-559存在于PSII反应中心内,但目前对其性质与功能的认识还不充分。本工作的目的就是研究Cyt b-559天然分子特性,探讨其生理功能和存在的意义。取得了一些有新意的结果: 1、依据PSII反应中心分离纯化的原理,应用更有效的层析介质DEAE-Sephacel,我们设计了快速高效的从菠菜和水稻中分离纯化Cyt b-559的方法和流程,获得了高纯度的样品。它们在非变性胶电泳中具有相同的泳动性。蛋白组分的HPLC结果证明,纯化的Cyt b-559的确由两个亚基组成,α亚基和β亚基的分子量用我们设计的适合于分析小蛋白的Tricine—SDS—PAGE方法准确测定为9.4kDa和4.5kDa。 2、利用HPLC技术分析了纯化的Cyt b-559样品的色素组成,结果表明Cytb-559中含有Chl α而不含类胡萝卜素分子,这一结果通过吸收光谱和共振拉曼光谱的分析得到进一步地证明。通过等电聚焦方法分析了Cyt b-559的等电点,发现其亚基的等电点相差很大,全蛋白的等电点与...更多D1、D2蛋白的等电点也不相同,推测在体内生理pH条件下它们具有相反带电性而在PSII组装中发挥作用。 3、低温荧光光谱的检测结果表明,Cyt b-559的荧光发射峰位在563nm和666nm;首次证明Cyt b-559可以发出荧光和将电子传递给结合在其上的辅助叶绿素,但传递能力比较低故而导致其荧光特性与PSII反应中心的不相同。Cytb-559的紫外荧光光谱表明Trp残基位于其内部的疏水区域,证明Cyt b-559中的芳香族氨基酸可能在其功能的发挥中起一定作用。 4、通过MCD的分析,发现Cyt b-559中血红素的MCD信号在540—580nm和400—440nm波段,而且光谱形状和强度与PSII反应中心的相一致,说明PSII反应中心该范围内的MCD信号中有Cyt b-559的贡献。FTIR光谱的测定结果证明Cyt b-559血红素的配体是组氨酸,其二级结构中α-螺旋占了一半。此外,还比较了Cyt b-559和PSII反应中心的膜脂成分,发现两者有很大的相似性。不同植物来源的Cyt b-559在许多性质上都表现出高度一致,从一个侧面证明Cyt b-559在进化中的保守性。 5、PSll反应中心发生光破坏时,原初电子供体P680己受到严重破坏。我们发现,在光抑制的最初一段时间内,Cyt b-559吸收峰值发生变化:在受体侧光抑制的条件下,其吸收峰值先略有增加而后才下降,而在供体侧光抑制条件下则相反,说明 Cyt b-559对光抑制的发生非常敏感,可能在光抑制早期保护PSll反应中心。 6、纯化的Cyt b-559的组氨酸含量在照光前后没有显著的变化,说明 PSll反应中心内被破坏的组氨酸不属于Cyt b-559。PSll反应中心所含的组氨酸中有些可被DEPC修饰,但我们的实验结果表明DEPC不能修饰Cyt b-559的组氨酸。这可能有利于Cyt b-559保护功能的发挥。 7、我们观察到,在两种光抑制条件下,LP Cyt b-559光还原和 HP Cyt b-559光氧化具有对pH值的依赖性,说明Cyt b-559在光保护中的作用不仅与其高低电势态有关,而且与其质子化程度有联系。CCCP促进HP Cyt -559释放质子,从而维持循环电子传递。DCBQ和 DCMU在很低浓度时都抑制 Cyt b-559光还原,前者不影响Cyt b-559光氧化而后者在CCCP存在时也会抑制Cyt b-559光氧化。 8、Cyt b-559有定位PSll反应中心其它蛋白的锚蛋白的作用。黄化苗转绿实验证明在 HP Cyt b-559的含量增加超过 45%以后放氧活性开始逐渐增加。Cytb-559从低电势态到高电势态的转变是放氧复合物组装到PSll反应中心的关键步骤之一。在植物正常生长时,Cyt b-559与 P680的其它电于供体发生竟争,起到安全阀门的作用。 9、在逆境条件下,Cyt b-559具有保护PSll反应中心免受强光破坏而起到“分于开关”的作用。我们的实验表明,在室温条件下存在通过Cyt b-559的环式电子流,存在从氧化态LP Cyt b-559到还原态HP Cyt b-559的一个循环,其中的氧化还原变化与质子化/去质子化反应相连。通过与其它血红素蛋白的比较,我们推测 Cyt b-559“分子开关”的关键是:光抑制情况下,铁原子与远端His之间的疏水空穴被氧自由基占据后使得铁进入叶琳中央孔中,迫使近端HIS向叶琳平面位移,从而引起 Cyt b-559构象改变,使电势态发生转变。

本文由两部分组成,一部分是关于一组新型除草剂(K-15,K-23)的抑制特性及作用位点的研究;另一部分是关于碳酸氢盐对细胞色素b-559高电势的保护作用的研究。 在第一部分,我们首先研究了抑制剂K-23对Psn放氧活性、DCIP光还原和可变荧光等光合特性的影响。研究发现,K-23在低浓度时刺激放氧活性,而在相对高浓度时抑制放氧活性。但是,K-23在低浓度时却有效地抑制了可变荧光。这些数据表明了新型抑制剂的抑制反应是基于氧化还原作用而不是猝灭作用。此外,通过采用胰蛋白酶消化类囊体膜的方式初步检测了新型抑制剂的作用部位,其结果表明:虽然新型抑制剂抑制受体侧电子传递,但它的抑制部位与DCMU不同。 其次,研究了新型抑制剂对光系统II色素蛋白复合体与多肽组分的影响及抑制剂的键合蛋白。应用SDS-PAGE技术,发现新型抑制剂主要作用于光系统II的反应中心蛋白。用温和的Deriphat-PAGE分析也证实了新型抑制剂作用于核心复合物,导致核心复合物二聚体消失。 进一步用SDS-PAGE分析新型抑制剂对Psn多肽组分的影响,发现新型抑制剂主要影响D.、D2、CP43和CP47。用荧光发射的方法确定了K-15键合在D2蛋白上。 最后,结合荧光动力学和HPLC方法,分别从定性和定量的角度,以核心复合物以及抑制剂存在下从BBY中分离的核心复合物为研究对象,详细研究了抑制剂对QA的取代作用。研究发现,在无去垢剂或低浓度去垢剂存在情况下,由于不能创造出适合于QA存在的疏水环境,我们没有得到QA被K-15取代的实验证据。而在抑制剂K-15存在下,用2.2% HTG从BBY分离的核心复合物的实验中,检测不到正的可变荧光Fv,而是得到了降低的FM,这个结果表明QA已被抑制剂在它的作用位点处所取代。 在第二部分,研究了pH5.0—7.0范围内碳酸氢盐对Cyt b-559氧化还原状态转变的影响。首先研究了pH5.0~7.0条件下碳酸氢盐对PSII Cyt b-559还原减氧化差异吸收光谱的影响,发现铁氰化钾还原的PSII Cyt b-559 HP的含量随介质pH值的降低而减少。然而,碳酸氢盐的加入阻止了由于介质pH降低而引起的Cyt b-559由高电势向低电势的转化。比如,当样品温育在pH5.0的介质中,铁氰化钾还原的Cyt b-559 HP含量占总量的25%-30%,当介质中加入2mmol/L碳酸氢盐后Cyt b-559 HP的含量上升,占总量的50%-56%。碳酸氢盐效应对氢醌还原的Cyt b-559HP含量的影响尤为显著。pH6.5时碳酸氢盐对Cyt b-559的还原氧化状态的影响不显著。其次,分别研究了PSII经Tris处理去除锰簇和三个外周蛋白及NH20H处理去除锰簇和17 kDa和23 kDa后,碳酸氢盐对Cytb一559 HP影响的pH依赖值,发现不论在pH5.0还是pH6.5的介质中碳酸氢盐效应都不存在。 综合以上实验结果,我们认为碳酸氢盐对酸化引起的Cyt b-559氧化还原状态的影响与它和锰的作用有关,但也不能排除钙离子与碳酸氢盐之间的协同作用。

在所研究的个体中,2个蜂猴物种碱基组成具有哺乳动物的共同特点,它们之间转换比(特别是密码子第3位)是颠换比的6倍多,大于其他种间比较;低的Ka/Ks值(<0.1)说明懒猴科cyt-b基因的异义突变位点受到强的选择压力作用。由cyt-b基因构建的系统发育树符合懒猴科化石记录和形态学分类观点。根据化石记录和分化时间有一定线性关系的第3位颠换和同义突变速率,估算蜂猴与倭蜂猴种间、蜂猴与瘠蜂猴属间可能的分化时间分别为300和600万年。

Gao-Yan Li, Xu-Zhen Wang, Ya-Hui Zhao, Jie Zhang, Chun-Guang Zhang, and Shun-Ping He (2009) Speciation and phylogeography of Opsariichthys bidens (Pisces: Cypriniformes: Cyprinidae) in China: analysis of the cytochrome b gene of mtDNA from diverse populations. Zoological Studies 48(4): 569-583. The cyprinid fish Opsariichthys bidens Gunther is distributed in all major river systems of continental East Asia, and represents an attractive model for phylogeographic studies among cyprinid species or within a given species. In this study, we investigated the phylogeographic and demographic history of this species, using partial sequences of the cytochrome (cyt) b gene in mitochondrial (mt)DNA. Fish samples were collected from almost all major river systems where O. bidens is distributed in China. Sequence analysis showed remarkably high polymorphism, with 125 haplotypes in the 234 specimens examined, and with 89.8% of haplotypes occurring in only 1 specimen. A neutrality test indicated that some groups were not at mutation-drift equilibrium, suggesting a past population expansion. These results were supported by a mismatch distribution analysis. Based on our analysis, O. bidens consists of 4 groups belonging to 2 clades. The divergence time of the 2 clades was estimated to be 11.06-8.04 my. This value corresponds to the time of the 2nd uplift of the Qinghai-Tibet Plateau, the emergence of the East Asian monsoon, and the Epoch-6 Event. The sinipercids represent a group of 12 species of freshwater percoid fish, including nine in Siniperca and three species in Coreoperca. Despite several classification attempts and a preliminary molecular phylogeny, the phylogenetic relationships and systematic position of sinipercids remained still unsolved. The complete cytochrome b gene sequences from nine sinipercid species four non-sinipercid fish species were cloned, and a total of 12 cyt b sequences from 10 species of sinipercids and 11 cyt b sequences from 10 species of non-sinipercid fish also in Perciformes were included in the phylogenetic analysis. As expected, the two genera Siniperca and Coreoperca within sinipercids are recovered as monophyletic. However, nine species representing Moronidae, Serranidae, Centropomidae, Acropomatidae, Emmelichtyidae, Siganidae and Centrarchidae included in the present study are all nested between Coreoperca and Siniperca, which provides marked evidence for a non-monophyly of sinipercid fishes. Coreoperca appears to be closest to Centrachus representing the family Centrarchidae. Coreoperca whiteheadi and C. herzi are sibling species, which together are closely related to C. kawamebari. In the Siniperca, the node between S. roulei and the remaining species is the most ancestral, followed by that of S. fortis. S. chuatsi and S. kneri are sibling species, sister to S. obscura. However, the sinipercids do not seem to have a very clear phylogenetic history, for different methods of phylogenetic reconstruction result in different tree topologies, and the only conclusive result in favor of a paraphyletic origin of the two sinipercid genera is the parametric bootstrap test. The paraphyly of Sinipercidae may suggest that the "synapomorphs" such as cycloid scales, upon which this family is based, were independently derived at least twice within sinipercid fishes, and further study should be carried out to include the other two Siniperca species and to incorporate other genes.

It is widely accepted that mitochondrial DNA (mtDNA) control region evolves faster than protein encoding genes with few exceptions. In the present study, we sequenced the mitochondrial cytochrome b gene (cyt b) and control region (CR) and compared their rates in 93 specimens representing 67 species of loaches and some related taxa in the Cobitoidea (Order Cypriniformes). The results showed that sequence divergences of the CR were broadly higher than those of the cyt b (about 1.83 times). However, in considering only closely related species, CR sequence evolution was slower than that of cyt b gene (ratio of CR/cyt b is 0.78), a pattern that is found to be very common in Cypriniformes. Combined data of the cyt b and CR were used to estimate the phylogenetic relationship of the Cobitoidea by maximum parsimony, neighbor-joining, and Bayesian methods. With Cyprinus carpio and Danio rerio as outgroups, three analyses identified the same four lineages representing four subfamilies of loaches, with Botiinae on the basal-most clade. The phylogenctic relationship of the Cobitoidea was ((Catostomidae + Gyrinocheilidae) + (Botiinae + (Balitorinae + (Cobitinae + Nemacheilinae)))), which indicated that Sawada's Cobitidae (including Cobitinae and Botiinae) was not monophyletic. Our molecular phylogenetic analyses are in very close agreement with the phylogenetic results based on the morphological data proposed by Nalbant and Bianco, wherein these four subfamilies were elevated to the family level as Botiidae, Balitoridae, Cobitidae, and Nemacheilidae. Five Mbo I (Mbo-A, Mbo-M, Mbo-C(1), Mbo-C(2) and Mbo-C(3)) and Hinf I (Hinf-1 to Hinf-5) patterns were observed in Apis mellifera samples after restriction of a 485 bp fragment of the mitochondrial cytochrome-b (cyt-b) gene. Associating the cyt-b Restriction fragment length polymorphism (RFLP) pattern of each sample to its respective previously established COI-COII (Dra I sites) pattern, five restriction patterns (Mbo-C(1), Mbo-C(2), Mbo-C(3), Hinf-1 and Hinf-4) were observed in samples of maternal origin associated to the evolutionary branch C. No deletions or insertions were observed and the nucleotide substitution rate was estimated at 5.4%. Higher nucleotide diversity was observed among the branch C-haplotypes when compared with A and M lineages. Further studies are needed to confirm if the cyt-b + COI-COII haplotypes help to assign certain phylogeographic patterns to the branch C and to clarify phylogenetic relationships among A. mellifera subspecies.

Snakehead fishes in the family Channidae are obligate freshwater fishes represented by two extant genera, the African Parachannna and the Asian Channa. These species prefer still or slow flowing water bodies, where they are top predators that exercise high levels of parental care, have the ability to breathe air, can tolerate poor water quality, and interestingly, can aestivate or traverse terrestrial habitat in response to seasonal changes in freshwater habitat availability. These attributes suggest that snakehead fishes may possess high dispersal potential, irrespective of the terrestrial barriers that would otherwise constrain the distribution of most freshwater fishes. A number of biogeographical hypotheses have been developed to account for the modern distributions of snakehead fishes across two continents, including ancient vicariance during Gondwanan break-up, or recent colonisation tracking the formation of suitable climatic conditions. Taxonomic uncertainty also surrounds some members of the Channa genus, as geographical distributions for some taxa across southern and Southeast (SE) Asia are very large, and in one case is highly disjunct. The current study adopted a molecular genetics approach to gain an understanding of the evolution of this group of fishes, and in particular how the phylogeography of two Asian species may have been influenced by contemporary versus historical levels of dispersal and vicariance. First, a molecular phylogeny was constructed based on multiple DNA loci and calibrated with fossil evidence to provide a dated chronology of divergence events among extant species, and also within species with widespread geographical distributions. The data provide strong evidence that trans-continental distribution of the Channidae arose as a result of dispersal out of Asia and into Africa in the mid–Eocene. Among Asian Channa, deep divergence among lineages indicates that the Oligocene-Miocene boundary was a time of significant species radiation, potentially associated with historical changes in climate and drainage geomorphology. Mid-Miocene divergence among lineages suggests that a taxonomic revision is warranted for two taxa. Deep intra-specific divergence (~8Mya) was also detected between C. striata lineages that occur sympatrically in the Mekong River Basin. The study then examined the phylogeography and population structure of two major taxa, Channa striata (the chevron snakehead) and the C. micropeltes (the giant snakehead), across SE Asia. Species specific microsatellite loci were developed and used in addition to a mitochondrial DNA marker (Cyt b) to screen neutral genetic variation within and among wild populations. C. striata individuals were sampled across SE Asia (n=988), with the major focus being the Mekong Basin, which is the largest drainage basin in the region. The distributions of two divergent lineages were identified and admixture analysis showed that where they co-occur they are interbreeding, indicating that after long periods of evolution in isolation, divergence has not resulted in reproductive isolation. One lineage is predominantly confined to upland areas of northern Lao PDR to the north of the Khorat Plateau, while the other, which is more closely related to individuals from southern India, has a widespread distribution across mainland SE Asian and Sumatra. The phylogeographical pattern recovered is associated with past river networks, and high diversity and divergence among all populations sampled reveal that contemporary dispersal is very low for this taxon, even where populations occur in contiguous freshwater habitats. C. micropeltes (n=280) were also sampled from across the Mekong River Basin, focusing on the lower basin where it constitutes an important wild fishery resource. In comparison with C. striata, allelic diversity and genetic divergence among populations were extremely low, suggesting very recent colonisation of the greater Mekong region. Populations were significantly structured into at least three discrete populations in the lower Mekong. Results of this study have implications for establishing effective conservation plans for managing both species, that represent economically important wild fishery resources for the region. For C. micropeltes, it is likely that a single fisheries stock in the Tonle Sap Great Lake is being exploited by multiple fisheries operations, and future management initiatives for this species in this region will need to account for this. For C. striata, conservation of natural levels of genetic variation will require management initiatives designed to promote population persistence at very localised spatial scales, as the high level of population structuring uncovered for this species indicates that significant unique diversity is present at this fine spatial scale.

Abstract: Nanostructured titanium dioxide (TiO2) electrodes, prepared by anodization of titanium, are employed to probe the electron-transfer process of cytochrome b5 (cyt b5) by surface-enhanced resonance Raman (SERR) spectroscopy. Concomitant with the increased nanoscopic surface roughness of TiO2, achieved by raising the anodization voltage from 10 to 20 V, the enhancement factor increases from 2.4 to 8.6, which is rationalized by calculations of the electric field enhancement. Cyt b 5 is immobilized on TiO2 under preservation of its native structure but it displays a non-ideal redox behavior due to the limited conductivity of the electrode material. The electron-transfer efficiency which depends on the crystalline phase of TiO2 has to be improved by appropriate doping for applications in bioelectrochemistry. Nanostructured TiO2 electrodes are employed to probe the electron-transfer process of cytochrome b5 by surface-enhanced resonance Raman spectroscopy. Concomitant with the increased nanoscopic surface roughness of TiO2, the enhancement factor increases, which can be attributed to the electric field enhancement.
