During the last glacial cycle, Greenland temperature showed many rapid temperature variations, the so-called Dansgaard–Oeschger (DO) events. The past atmospheric methane concentration closely followed these temperature variations, which implies that the warmings recorded in Greenland were probably hemispheric in extent. Here we substantially extend and complete the North Greenland Ice Core Project (NGRIP) methane record from the Preboreal Holocene (PB) back to the end of the last interglacial period with a mean time resolution of 54 yr. We relate the amplitudes of the methane increases associated with DO events to the amplitudes of the local Greenland NGRIP temperature increases derived from stable nitrogen isotope (δ15N) measurements, which have been performed along the same ice core (Kindler et al., 2014). We find the ratio to oscillate between 5 parts per billion (ppb) per °C and 18 ppb °C−1 with the approximate frequency of the precessional cycle. A remarkably high ratio of 25.5 ppb °C−1 is reached during the transition from the Younger Dryas (YD) to the PB. Analysis of the timing of the fast methane and temperature increases reveals significant lags of the methane increases relative to NGRIP temperature for DO events 5, 9, 10, 11, 13, 15, 19, and 20. These events generally have small methane increase rates and we hypothesize that the lag is caused by pronounced northward displacement of the source regions from stadial to interstadial. We further show that the relative interpolar concentration difference (rIPD) of methane is about 4.5% for the stadials between DO events 18 and 20, which is in the same order as in the stadials before and after DO event 2 around the Last Glacial Maximum. The rIPD of methane remains relatively stable throughout the full last glacial, with a tendency for elevated values during interstadial compared to stadial periods.

In order to reconstruct the temperature of the North Greenland Ice Core Project (NGRIP) site, new measurements of δ15N have been performed covering the time period from the beginning of the Holocene to Dansgaard–Oeschger (DO) event 8. Together with previously measured and mostly published δ15N data, we present for the first time a NGRIP temperature reconstruction for the whole last glacial period from 10 to 120 kyr b2k (thousand years before 2000 AD) including every DO event based on δ15N isotope measurements combined with a firn densification and heat diffusion model. The detected temperature rises at the onset of DO events range from 5 °C (DO 25) up to 16.5 °C (DO 11) with an uncertainty of ±3 °C. To bring measured and modelled data into agreement, we had to reduce the accumulation rate given by the NGRIP ss09sea06bm timescale in some periods by 30 to 35%, especially during the last glacial maximum. A comparison between reconstructed temperature and δ18Oice data confirms that the isotopic composition of the stadial was strongly influenced by seasonality. We evidence an anticorrelation between the variations of the δ18Oice sensitivity to temperature (referred to as α) and obliquity in agreement with a simple Rayleigh distillation model. Finally, we suggest that α might be influenced by the Northern Hemisphere ice sheet volume.

S100A8 (also known as CP10 or MRP8) was the first member of the S100 family of calcium-binding proteins shown to be chemotactic for myeloid cells. The gene is expressed together with its dimerization partner S100A9 during myelopoiesis in the fetal liver and in adult bone marrow as well as in mature granulocytes. In this paper we show that S100A8 mRNA is expressed without S100A9 mRNA between 6.5 and 8.5 days postcoitum within fetal cells infiltrating the deciduum in the vicinity of the ectoplacental cone. Targeted disruption of the S100A8 gene caused rapid and synchronous embryo resorption by day 9.5 of development in 100% of homozygous null embryos. Until this point there was no evidence of developmental delay in S100A8(-/-) embryos and decidualization was normal. The results of PCR genotyping around 7.5-8.5 days postcoitum suggest that the null embryos are infiltrated with maternal cells before overt signs of resorption. This work is the first evidence for nonredundant function of a member of the S100 gene family and implies a role in prevention of maternal rejection of the implanting embryo. The S100A8 null provides a new model for studying fetal-maternal interactions during implantation.

The functional importance of members of the S100 Ca2+-binding protein family is recently emerging. A variety of activities, several of which are apparently opposing, are attributed to S100A8, a protein implicated in embryogenesis, growth, differentiation, and immune and inflammatory processes. Murine (m) S100A8 was initially described as a chemoattractant (CP-10) for myeloid cells. It is coordinately expressed with mS100A9 (MRP14) in neutrophils and the non-covalent heterodimer is presumed to be the functional intracellular species. The extracellular chemotactic activity of mS100A8, however, is not dependent on mS100A9 and occurs at concentrations (10(-13)-10(-11) M) at which the non-covalent heterodimer would probably dissociate. This review focuses on the structure and post-translational modifications of mS100A8/A9 and their effects on function, particularly chemotaxis.

Human S100A12 (extracellular newly identified RAGE (receptor for advanced glycosylation end products)binding protein), a new member of the S100 family of EF-hand calcium-binding proteins, was chemically synthesised using highly optimised 2-(1H-benzotriazol-1-yl)-1,1,3,3-tetramethyluronium hexafluorophosphate/tert-butoxycarbonyl in situ neutralisation solid-phase chemistry. Circular dichroism studies indicated that CaCl2 decreased the helical content by 27% whereas helicity was marginally increased by ZnCl2. The propensity of S100A12 to dimerise was examined by electrospray ionisation time-of-flight mass spectrometry which clearly demonstrated the prevalence of the non-covalent homodimer (20 890 Da). Importantly, synthetic human S100A12 in the nanomolar range was chemotactic for neutrophils and macrophages in vitro. (C) 2001 Federation of European Biochemical Societies. o interesse da República de Cabo Verde na utiliza9ao da energia renovável é basicamente económico. O país é muito pobre e todo o custo de energia pesa profundamente na sua economia nacional,visto nao existirem recursos de energia "tradicional". A utiliza~ao da energia renovável para bombagem de água é por isso, de grande importancia porque reduz a importa~ao de petróleo. Tanto a energia eólica como a energia solar oferecem grandes possibilidades em Cabo Verde. O país está sendo atingido pelo pior e mais langa período de seca jamais experimentado na sua história. Pode-se imaginar os problemas enfrentados para abastecimento de água a populac~o, abeberamento de animais e irriga- .., , ,,, , ~ao das poucas areas agrlcolas. O abastecimento de água potável é extremamente importante neste país, é apenas para a sobrevivencia da maior parte da sua popula9ao rural (190.000 habitantes), com pouco poder aquisitivo dado a diminuisao da área agrícola irrigada e a quantidade de gado, nos últimos 15 anos. Na agricultura irrigada, de momento totalizando apenas 2.000 ha, utilizase relativamente grande quantidade de combustível, porque 50% consiste em irrigasao por bombagem. Por esta razao o governo de Cabo Verde estimula fortemente a utilizasao das energias renováveis. As actividades neste campo come~aram em 1977 com a cria~ao da Divisao de Energias Renováveis (DER) dentro do Ministério do Desenvolvimento Rural (MUR). O governo de Cabo Verde e a PNUD (Projecto PNUD CVI/76/X05 - Demonstrasao de Energia Nao Convenciorial) puseram fundos a disposi~ao. Este projecto contribuiu com 3 aerogeradores e um volontário, Sr. Van Meel, que trabalhou anteriormente no Grupo de Energia Eólica de Eindhoven e que faz parte do CWD. Desde o seu início, a Divisao deu prioridade a bombagem de água para o abastecimento de água potável, o que foi e continua a ser urna necessidade urgente nas Ilhas de Cabo Verde. Os Servi~os da Igreja Mundial ofereceram 25 aerobombas Dempster com ventoinha de 8' de diametro e mais tarde a UNICEF distribuiu 10 Dempsters de ]4'. Gradualmente mais organiza90es financiadoras envolveram-se nas actividades da DER, como por exemplo a ClMADE, FAC, Embaixada An~ricana, Embaixada do Canadá, ICCO, etc. O Ministério do Desenvolvimento e Coopera9ao dos Países Baixos (DGIS) encarregou- se, em 1979, da norneasao do voluntárío da PNUD. Também em 1979 a USAID envolveu-se. Foi reconhecido e implementado um projecto de energia renovável (ref. 1). Este projecto comecou em Agosto de 1980. O papel prin cipal do projecto consistia na aquisiq~o de material e equipamento para urna nova oficina da DER. Urna das premissas do projecto era que o DGIS prestaria assistencia él Divisao. A pedido do DGIS a CWD realizou um estudo sobre a viabilidade de aplica9ao de aerobombas durante o primeiro semestre de 1980 (ref. 2). O estudo revelou que 75% das necessidades de água por bombagem em Cabo Verde, pode ria ser desempenhado pelos moinhos. - O projecto proposto diz respeito nao apenas a assistencia técnica, mas também ao financiamento da nova oficina e a aquisi9ao de cerca de 30 moi nhos. Este projecto bilateral entre Cabo Verde e os Países Baixos - Energias Renováveis", depois da aprova<;ao de Cabo Verde e do DGIS, veia a comecar no dia 1 de Julho de 1981 e estendeu-se por 3 anos. O período de 1977 a Julho de 1981 classifica-se agora como a primeira fase das actividades. Durante este período o CWD contribuiu com a assist€ncia técnica. O período de Julho de 1981 a Julho de 1984 é classificado como segunda fase e será relatado neste documento. O projecto foi executado pela DER. O Director da DER é, desde Fevereiro de 1983, o Sr. Daniel Livramento, licenciado em Física (Brasil 1982). Outros funcionários do corpo directivo sao o Sr. David Cardoso (desde Novem bro de 1983) e muito recentemente, desde Maio de 1984, o Sr. F. Ferreira7 A colaboracao dos Países Baixos para o projecto tem sido efectuada por En genheiros Consultores do DHV, em nome e como associados do CWD. O Sr. K.Versteegh, consultor do CWD, comecou a trabalhar para o projecto em 1 de Julho de 1981 e o Sr. N.Pieterse,~consultordo CWD, em 15 de Setembro de 1981. O Sr. Van Meel deixou o projecto a 1 de Novembro de 1981. O projecto foi avaliado em Novembro de 1983, por urna missao tripartida (Cabo Verde, USAID, DGIS) - Ref. 3. As conclusoes desta missao de avaliacao foram, em geral, positivas e o prolongamento do projecto foi fortemen te recomendado, com €nfase sobre aprodu~ao local. Entretanto, a DER e o CWD prepararam urna proposta para a extensao do projecto por mais 3 anos (Ref. 4). Esta extensao foi aprovada por Cabo Verde e pelo DGIS. O Sr. Versteegh foi substituído em Julho de 1984 e o Sr. Pieterse será substituido em Setembro de 1984 pelos Srs. J.Diepens e H. van der Spek, respectivamente. O conteúdo deste relatório é o seguinte: No capitulo 2 é apresentada urna exposi~ao do trabalho que incluiu um resumo das actividades realizadas na fase 1. Transition metal alkynyl complexes containing perfluoroaryl groups have been prepared directly from trimethylsilyl-protected mono- and di-ethynyl perfluoroarenes by simple desilylation/metallation reaction sequences. Reactions between Me3SiC CC6F5 and RuCl(dppe)Cp'[Cp' = Cp, Cp*] in the presence of KF in MeOH give the monoruthenium complexes Ru(C CC6F5)(dppe)Cp'[Cp' = Cp (2); Cp* (3)], which are related to the known compound Ru(C CC6F5)(PPh3)(2)Cp (1). Treatment of Me3SiC CC6F5 with Pt-2(mu-dppm)(2)Cl-2 in the presence of NaOMe in MeOH gave the bis(alkynyl) complex Pt-2(mu-dppm)(2)(C CC6F5)(2) (4). The Pd(0)/Cu(I)-catalysed reactions between Au(C CC6F5)(PPh3) and Mo( CBr)(CO)(2) Tp* [Tp* = hydridotris(3.5-dimethylpyrazoyl)borate], Co-3(mu(3)-CBr)(mu-dppm)(CO)(7) or IC CFc [Fc = (eta(5)-C5H4)FeCp] afford Mo( CC CC6F5)(CO)(2)Tp* (5), Co-3(mu 3-CC CC6F5)(mu-dppm)(CO)(7) (6) and FcC CC CC6F5 (7), respectively. The diruthenium complexes 1,4-{Cp'(PP)RuC C}(2)C6F4 [(PP)Cp'=(PPh3)(2)Cp (8); (dppe)Cp (9); (dppe)Cp* (10)] are prepared from 1,4-(Me3SiC C)(2)C6F4 in a manner similar to that described for the monoruthenium complexes 1-3. The non-fluorinated complexes 1,4-{Cp'(PP)RuC C}(2)C6H4 [(PP)Cp' = (PPh3)(2)Cp (11); ( dppe) Cp (12); ( dppe) Cp* (13)], prepared for comparison, are obtained from 1,4-(Me3SiC C)(2)C6H4. Spectro-electrochemical studies of the ruthenium aryl and arylene alkynyl complexes 2-3 and 8-13, together with DFT-based computational studies on suitable model systems, indicate that perfluorination of the aromatic ring has little effect on the electronic structures of these compounds, and that the frontier orbitals have appreciable diethynylphenylene character. Molecular structure determinations are reported for the fluoroaromatic complexes 1, 2, 3, 6 and 10.

We present and examine a multi-sensor global compilation of mid-Holocene (MH) sea surface temperatures (SST), based on Mg/Ca and alkenone palaeothermometry and reconstructions obtained using planktonic foraminifera and organic-walled dinoflagellate cyst census counts. We assess the uncertainties originating from using different methodologies and evaluate the potential of MH SST reconstructions as a benchmark for climate-model simulations. The comparison between different analytical approaches (time frame, baseline climate) shows the choice of time window for the MH has a negligible effect on the reconstructed SST pattern, but the choice of baseline climate affects both the magnitude and spatial pattern of the reconstructed SSTs. Comparison of the SST reconstructions made using different sensors shows significant discrepancies at a regional scale, with uncertainties often exceeding the reconstructed SST anomaly. Apparent patterns in SST may largely be a reflection of the use of different sensors in different regions. Overall, the uncertainties associated with the SST reconstructions are generally larger than the MH anomalies. Thus, the SST data currently available cannot serve as a target for benchmarking model simulations. Further evaluations of potential subsurface and/or seasonal artifacts that may contribute to obscure the MH SST reconstructions are urgently needed to provide reliable benchmarks for model evaluations.

We analyse the spatial expression of seasonal climates of the Mediterranean and northern Africa in pre-industrial (piControl) and mid-Holocene (midHolocene, 6 yr BP) simulations from the fifth phase of the Coupled Model Intercomparison Project (CMIP5). Modern observations show four distinct precipitation regimes characterized by differences in the seasonal distribution and total amount of precipitation: an equatorial band characterized by a double peak in rainfall, the monsoon zone characterized by summer rainfall, the desert characterized by low seasonality and total precipitation, and the Mediterranean zone characterized by summer drought. Most models correctly simulate the position of the Mediterranean and the equatorial climates in the piControl simulations, but overestimate the extent of monsoon influence and underestimate the extent of desert. However, most models fail to reproduce the amount of precipitation in each zone. Model biases in the simulated magnitude of precipitation are unrelated to whether the models reproduce the correct spatial patterns of each regime. In the midHolocene, the models simulate a reduction in winter rainfall in the equatorial zone, and a northward expansion of the monsoon with a significant increase in summer and autumn rainfall. Precipitation is slightly increased in the desert, mainly in summer and autumn, with northward expansion of the monsoon. Changes in the Mediterranean are small, although there is an increase in spring precipitation consistent with palaeo-observations of increased growing-season rainfall. Comparison with reconstructions shows most models underestimate the mid-Holocene changes in annual precipitation, except in the equatorial zone. Biases in the piControl have only a limited influence on midHolocene anomalies in ocean–atmosphere models; carbon-cycle models show no relationship between piControl bias and midHolocene anomalies. Biases in the prediction of the midHolocene monsoon expansion are unrelated to how well the models simulate changes in Mediterranean climate.

A new online method to analyse water isotopes of speleothem fluid inclusions using a wavelength scanned cavity ring down spectroscopy (WS-CRDS) instrument is presented. This novel technique allows us simultaneously to measure hydrogen and oxygen isotopes for a released aliquot of water. To do so, we designed a new simple line that allows the online water extraction and isotope analysis of speleothem samples. The specificity of the method lies in the fact that fluid inclusions release is made on a standard water background, which mainly improves the δ D robustness. To saturate the line, a peristaltic pump continuously injects standard water into the line that is permanently heated to 140 °C and flushed with dry nitrogen gas. This permits instantaneous and complete vaporisation of the standard water, resulting in an artificial water background with well-known δ D and δ18O values. The speleothem sample is placed in a copper tube, attached to the line, and after system stabilisation it is crushed using a simple hydraulic device to liberate speleothem fluid inclusions water. The released water is carried by the nitrogen/standard water gas stream directly to a Picarro L1102-i for isotope determination. To test the accuracy and reproducibility of the line and to measure standard water during speleothem measurements, a syringe injection unit was added to the line. Peak evaluation is done similarly as in gas chromatography to obtain &delta D; and δ18O isotopic compositions of measured water aliquots. Precision is better than 1.5 ‰ for δ D and 0.4 ‰ for δ18O for water measurements for an extended range (−210 to 0 ‰ for δ D and −27 to 0 ‰ for δ18O) primarily dependent on the amount of water released from speleothem fluid inclusions and secondarily on the isotopic composition of the sample. The results show that WS-CRDS technology is suitable for speleothem fluid inclusion measurements and gives results that are comparable to the isotope ratio mass spectrometry (IRMS) technique.

High-resolution pollen and dinoflagellate cyst records from sediment core M72/5-25-GC1 were used to reconstruct vegetation dynamics in northern Anatolia and surface conditions of the Black Sea between 64 and 20 ka BP. During this period, the dominance of Artemisia in the pollen record indicates a steppe landscape and arid climate conditions. However, the concomitant presence of temperate arboreal pollen suggests the existence of glacial refugia in northern Anatolia. Long-term glacial vegetation dynamics reveal two major arid phases ~64–55 and 40–32 ka BP, and two major humid phases ~54–45 and 28–20 ka BP, correlating with higher and lower summer insolation, respectively. Dansgaard–Oeschger (D–O) cycles are clearly indicated by the 25-GC1 pollen record. Greenland interstadials are characterized by a marked increase in temperate tree pollen, indicating a spread of forests due to warm/wet conditions in northern Anatolia, whereas Greenland stadials reveal cold and arid conditions as indicated by spread of xerophytic biomes. There is evidence for a phase lag of ~500 to 1500 yr between initial warming and forest expansion, possibly due to successive changes in atmospheric circulation in the North Atlantic sector. The dominance of Pyxidinopsis psilata and Spiniferites cruciformis in the dinocyst record indicates brackish Black Sea conditions during the entire glacial period. The decrease of marine indicators (marine dinocysts, acritarchs) at ~54 ka BP and increase of freshwater algae (Pediastrum, Botryococcus) from 32 to 25 ka BP reveals freshening of the Black Sea surface water. This freshening is possibly related to humid phases in the region, to connection between Caspian Sea and Black Sea, to seasonal freshening by floating ice, and/or to closer position of river mouths due to low sea level. In the southern Black Sea, Greenland interstadials are clearly indicated by high dinocyst concentrations and calcium carbonate content, as a result of an increase in primary productivity. Heinrich events show a similar impact on the environment in the northern Anatolia/Black Sea region as Greenland stadials.

We present a selection of methodologies for using the palaeo-climate model component of the Coupled Model Intercomparison Project (Phase 5) (CMIP5) to attempt to constrain future climate projections using the same models. The constraints arise from measures of skill in hindcasting palaeo-climate changes from the present over three periods: the Last Glacial Maximum (LGM) (21 000 yr before present, ka), the mid-Holocene (MH) (6 ka) and the Last Millennium (LM) (850–1850 CE). The skill measures may be used to validate robust patterns of climate change across scenarios or to distinguish between models that have differing outcomes in future scenarios. We find that the multi-model ensemble of palaeo-simulations is adequate for addressing at least some of these issues. For example, selected benchmarks for the LGM and MH are correlated to the rank of future projections of precipitation/temperature or sea ice extent to indicate that models that produce the best agreement with palaeo-climate information give demonstrably different future results than the rest of the models. We also explore cases where comparisons are strongly dependent on uncertain forcing time series or show important non-stationarity, making direct inferences for the future problematic. Overall, we demonstrate that there is a strong potential for the palaeo-climate simulations to help inform the future projections and urge all the modelling groups to complete this subset of the CMIP5 runs.

Objetivou-se, com este trabalho, avaliar os efeitos da adição de diferentes níveis de enzima mais levedura na dieta de bovinos sobre a digestibilidade aparente dos nutrientes e o desempenho em confinamento, utilizando-se 18 animais machos, mestiços, de origem leiteira, com peso vivo (PV) médio de 190±10 kg e 12 meses de idade, e 27 animais da raça Guzerá com PV médio de 325 kg e idade média de 27 meses. Os níveis de enzima mais levedura foram zero (controle), 5 g de enzima (E) mais 5 g de levedura (L) e 10 g de E mais 5 g de L por animal por dia. Todos os animais receberam a mesma dieta, constituída de 65% de silagem (50% de milho:50% de sorgo) e 35% de concentrado composto de milho (24,6%), farelo de algodão (7,4%), uréia (1,2%), sal mineralizado (0,54%) e calcário (0,86%). Esta dieta apresentava 12,4% de PB; 10,1 MJEM/kg MS e 41,1% de FDN. A adição de enzima mais levedura não alterou o consumo de MS (3,31% do PV ou 126,6 g/kg PV0,75), e CDA da MS (63,0%), PB (68,2%), EE (83,3%), CHOT (61,3%), FDN (47,2%), FDA (43,8%) e valor de NDT (62,8%). O GMD e o CMS diminuíram de forma quadrática e a CA alimentar piorou linearmente, em função dos dias de confinamento. O fornecimento de enzima mais levedura para bovinos de corte não traz benefícios nutricionais ou de desempenho.
