Introdução – No caso de uma eventual contaminação radioativa esta deve ser quantificada, pelo que é necessário garantir que as condições de funcionamento de um monitor de contaminação de superfícies se encontrem adequadamente caracterizadas, através da determinação da sua eficiência de deteção. Este estudo tem como objetivo determinar a eficiência de deteção de um monitor de contaminação e analisar a influência da distância e da atividade. Metodologia – O monitor de contaminação Thermo Mini 900E e as fontes radioativas planas de referência de Carbono – 14 (C-14), Césio – 137 (Cs-137), Estrôncio – 90 (Sr-90), Cloro – 36 (Cl-36) e Amerício – 241 (Am-241) com débito de emissão superficial rastreável ao Physikalish Technischen Bundesanstalt (PTB) foram utilizadas. Fontes de Sr-90 e C-14 com dois débitos de emissão distintos para cada e distâncias de 1 a 20mm foram usados para estudar a sua influência na eficiência de deteção. Resultados – Verificou-se que as fontes radioativas emissoras de partículas de maior energia apresentam uma maior eficiência de deteção e que esta diminui com o aumento da distância do detetor à fonte, sendo que os radioisótopos de maior energia apresentam um decréscimo na eficiência de 15% ao longo de 20mm, enquanto os radioisótopos de menor energia apresentam um decréscimo de 10%. Não se verificou uma influência expressiva do débito de emissão na eficiência de deteção deste monitor de contaminação. Conclusão – A eficiência de deteção de um monitor de contaminação portátil é dependente da distância deste à contaminação bem como do tipo de radiação emitida e energia dos radioisótopos presentes na contaminação. - ABSTRACT - Introduction – A radioactive contamination needs to be quantified in case it eventually occurs. Therefore it is necessary to ensure that the operating conditions of a surface contamination monitor are characterized by determining its detection efficiency. This experimental study aims to determine the detection efficiency of a contamination monitor and evaluate the distance and surface emission rate influence on the detection efficiency. Methodology – A contamination monitor Thermo Mini 900E was tested with reference radiation sources of Carbon – 14 (C-14), Cesium – 137 (Cs-137), Strontium – 90 (Sr-90), Chlorine – 36 (Cl-36) and Americium – 241 (Am-241) with its emission rate traced to the Physikalish Technischen Bundesanstalt (PTB). Sources of Sr-90 and C-14 with two different emission rates for each one and distances of 1 to 20mm were used to study its influence in the detection efficiency. Results – With the increasing of distance, the detection efficiency decreases. The most energetic radiation sources have higher detection efficiency, boasting a 15% decrease over 20mm whereas the sources bearing a smaller efficiency decrease 10%. No influence of the surface emission rate in the detection efficiency was verified. Conclusion – Thus, it is concluded that the detection efficiency of a contamination monitor is dependent of the distance between it and the contamination as well as the type and energy of the radioisotopes present in the contamination.

In Switzerland, individuals exposed to the risk of activity intake are required to perform regular monitoring. Monitoring consists in a screening measurement and is meant to be performed using commonly available laboratory instruments. More particularly, iodine intake is measured using a surface contamination monitor. The goal of the present paper is to report the calibration method developed for thyroid screening instruments. It consists of measuring the instrument response to a known activity located in the thyroid gland of a standard neck phantom. One issue of this procedure remains that the iodine radioisotopes have a short half-life. Therefore, the adequacy and limitations to simulate the short-lived radionuclides with so-called mock radionuclides of longer half-life were also evaluated. In light of the results, it has been decided to use only the appropriate iodine sources to perform the calibration.

Monitoring of internal exposure for nuclear medicine workers requires frequent measurements due to the short physical half-lives of most radionuclides used in this field. The aim of this study was to develop screening measurements performed at the workplace by local staff using standard laboratory instrumentation, to detect whether potential intake has occurred. Such measurements do not enable to determine the committed effective dose, but are adequate to verify that a given threshold is not exceeded. For radioiodine, i.e. (123)I, (124)I, (125)I and (131)I, a calibrated surface contamination monitor is placed in front of the thyroid to detect whether the activity threshold has been exceeded. For radionuclides with very short physical half-lives (≤6 h), such as (99m)Tc and those used in positron emission tomography  imaging, i.e. (11)C, (15)O, (18)F and (68)Ga, screening procedures consist in performing daily measurements of the ambient dose rate in front of the abdomen. Other gamma emitters used for imaging, i.e. (67)Ga, (111)In and (201)Tl, are measured with a scintillation detector located in front of the thorax. For pure beta emitters, i.e. (90)Y and (169)Er, as well as beta emitters with low-intensity gamma rays, i.e. (153)Sm, (177)Lu, (186)Re and (188)Re, the procedure consists in measuring hand contamination immediately after use. In Switzerland, screening procedures have been adopted by most nuclear medicine services since such measurements enable an acceptable monitoring while taking into account practical and economic considerations.

The electrochemical detection of the hazardous pollutant 4-nitrophenol (4-NP) at low potentials, in order to avoid matrix interferences, is an important research challenge. This study describes the development, electrochemical characterization and utilization of a multiwall carbon nanotube (MWCNT) film electrode for the quantitative determination of 4-NP in natural water. Electrochemical impedence spectroscopy measurements showed that the modified surface exhibits a decrease of ca. 13 times in the charge transfer resistance when compared with a bare glassy carbon (GC) surface. Voltammetric experiments showed the possibility to oxidize a hydroxylamine layer (produced by the electrochemical reduction of 4-NP on the GC/MWNCT surface) in a potential region which is approximately 700 mV less positive than that needed to oxidize 4-NP, thus minimizing the interference of matrix components. The limit of detection for 4-NP obtained using square-wave voltammetry (0.12 mu mol L(-1)) was lower than the value advised by EPA. A natural water sample from a dam located in Sao Carlos (Brazil) was spiked with 4-NP and analyzed by the standard addition method using thee GC/MWCNT electrode, without any further purification step. the recovery procedure yielded a value of 96.5% for such sample, thus confirming the suitability of the developed method to determine 4-NP in natural water samples. The electrochemical determination was compared with that obtained by HPLC with UV-vis detection.

The research presented herein aims to investigate the strengths and weaknesses of a relatively new technique called phytoscreening. Parallel to the well-known phytoremediation, it consists of exploiting the absorbing potential of trees to delineate groundwater contamination plumes, especially for chlorinated ethenes (i.e., PCE, TCE, 1,2-cis DCE, and VC). The latter are prevalent contaminants in groundwater but their fate and transport in surface ecosystems, such as trees, are still poorly understood and subjected to high variability. Moreover, the analytical validity of tree-coring is still limited in many countries due to a lack of knowledge of its application opportunities. Tree-cores are extracted from trunks and generally analyzed by gas chromatography/mass spectrometry. A systematic review of former literature on phytoscreening for chlorinated ethenes is presented in this PhD thesis to evaluate the factors influencing the effectiveness of the technique. Besides, we tested the technique by probing eight sites contaminated by chlorinated ethenes in Italy (Emilia-Romagna) in different hydrogeological and seasonal settings. We coupled the technique with the assessment of gaseous-phase concentrations directly on-site, inserting detector tubes or a photoionization detector in the tree-holes left by the coring tool. Finally, we applied rank order statistic analysis on field data along with literature data to assess under which conditions phytoscreening should be applied to either screen or monitor environmental contamination issues. A relatively high correlation exists between tree-core and groundwater concentrations (Spearman’s ρ > 0.6), being higher for compounds with higher sorption, for sites with shallower and thinner aquifers, and when sampling specific tree types with standardized sampling and extraction protocols. These results indicate the opportunities for assessing the occurrence, type, and concentration of solvents directly from the stem of trees. This can reduce the costs of characterization surveys, allowing rapid identification of hotspots and plume direction and thus optimizing the drilling of boreholes.

A descriptive study was developed to monitor air fungal contamination in two elementary schools in Lisbon, Portugal. Eight air samples of 250 litres through impaction method were collected in canteen, library, classrooms and also, outside premises as reference place. Simultaneously, were also monitored environmental parameters, including temperature, and humidity through the equipment Babouc, LSI Sistems and according to the International Standard ISO 7726 - 1998. Considering both schools, sixteen different species of fungi in air were identified, being the 2 most commonly isolated Cladosporium sp. (51,1%) and Penicillium sp. (27,5%). Besides these genera Trichoderma, Aspergillus, Alternaria, Chrysonilia, Botritys, Ulocladium, Athrium, Aureobasidium, Phoma, Scedosporium e Geotrichum were also isolated. Regarding yeasts, Candida sp., Cryptococcus sp. and Rhodotorula sp. were isolated. The youngest school, as well canteens in each school, presented the worst results concerning the air fungal contamination, maybe due to the higher number of occupants. There was no significant relationship (p>0,05) between fungal contamination and temperature and humidity.

When decommissioning a nuclear facility it is important to be able to estimate activity levels of potentially radioactive samples and compare with clearance values defined by regulatory authorities. This paper presents a method of calibrating a clearance box monitor based on practical experimental measurements and Monte Carlo simulations. Adjusting the simulation for experimental data obtained using a simple point source permits the computation of absolute calibration factors for more complex geometries with an accuracy of a bit more than 20%. The uncertainty of the calibration factor can be improved to about 10% when the simulation is used relatively, in direct comparison with a measurement performed in the same geometry but with another nuclide. The simulation can also be used to validate the experimental calibration procedure when the sample is supposed to be homogeneous but the calibration factor is derived from a plate phantom. For more realistic geometries, like a small gravel dumpster, Monte Carlo simulation shows that the calibration factor obtained with a larger homogeneous phantom is correct within about 20%, if sample density is taken as the influencing parameter. Finally, simulation can be used to estimate the effect of a contamination hotspot. The research supporting this paper shows that activity could be largely underestimated in the event of a centrally-located hotspot and overestimated for a peripherally-located hotspot if the sample is assumed to be homogeneously contaminated. This demonstrates the usefulness of being able to complement experimental methods with Monte Carlo simulations in order to estimate calibration factors that cannot be directly measured because of a lack of available material or specific geometries.

A descriptive study was developed to monitor air fungal contamination in one Portuguese maternity. Sixty air samples were collected through impaction method. Air sampling was performed in food storage facilities, kitchen, food plating, canteen, pharmacy, sterilization areas, genecology wards, intensive care unit, operating rooms, urgency and also, outside premises, since this was the place regarded as reference. Besides air samples, forty three samples were collected by swabbing the surfaces using a 10 by 10 cm square stencil. Simultaneously, temperature, relative humidity and particles counting (PM10) were registered. Twenty three species of fungi were identified in air, being the two most commonly isolated the genera Penicillium (41,5%) and Cladosporium (28,4%). Regarding yeasts, only Rhodotorula sp. (45,2%), Trichosporon mucoides (51,6%) and Cryptococcus neoformans (3,2%) were found. Thirteen species of fungi were identified in surfaces, being the most frequent the Penicillium genus (91,6%). Concerning yeasts found in surfaces, four species were identified being Rhodotorula sp. (29,1%) the most frequent. There was no coincidence between prevailing genera indoors and outside premises. Moreover, some places presented fungal species different from the ones isolated outside. In the inside environment, Aspergillus species were isolated in air and surfaces. There was no significant relationship (p>0,05) between fungal contamination and the studied environmental variables. This thesis collects several ecotoxicological studies focused on the quali- quantitative analysis of several classes of chemical compounds. Our studies have been conducted on different aquatic species occupying different food chain trophic levels and characterized by differences in biology, ethology, and nutrition, but all considered excellent bioindicators. This choice allowed us to have a broad overview of the contamination of aquatic environments. Detrimental effects of several chemical compounds on the species investigated have been discussed, considering the economic and public health implications linked to the pollution of the environment and the exposure to old and emerging xenobiotics. Our studies underline the importance of a multidisciplinary and integrated approach that includes the application of the one health concept to ensure the protection of public health and respect for natural environments. Studies collected in this thesis also aim to overcome some critical limitations of the branch of ecotoxicology, such as the lack of standardization in laboratory methods. Our data also underline the importance of expanding research to a greater number of various biological matrices than those indicated by the literature as target tissues for specific pollutants. This condition enables more detailed information on the kinetics of xenobiotics in animal organisms. Our studies also allow us to expand the knowledge related to the mechanisms of synergy and antagonism of mixtures of pollutants that can simultaneously accumulate in wildlife.
