Objective: To test if subpopulations of chondrocytes from different cartilage zones could be used to engineer cartilage constructs with features of normal stratification. Design: Chondrocytes from the superficial and middle zones of immature bovine cartilage were cultured in alginate, released, and seeded either separately or sequentially to form cartilage constructs. Constructs were cultured for 1 or 2 weeks and were assessed for growth, compressive properties, and deposition, and localization of matrix molecules and superficial zone protein (SZP). Results: The cartilaginous constructs formed from superficial zone chondrocytes exhibited less matrix growth and lower compressive properties than constructs from middle zone chondrocytes, with the stratified superficial-middle constructs exhibiting intermediate properties. Expression of SZP was highest at the construct surfaces, with the localization of SZP in superficial-middle constructs being concentrated at the superficial surface. Conclusions: Manipulation of subpopulations of chondrocytes can be useful in engineering cartilage tissue with a biomimetic approach, and in fabricating constructs that exhibit stratified features of normal articular cartilage. Articular cartilage is the load-bearing tissue that consists of proteoglycan macromolecules entrapped between collagen fibrils in a three-dimensional architecture. To date, the drudgery of searching for mathematical models to represent the biomechanics of such a system continues without providing a fitting description of its functional response to load at micro-scale level. We believe that the major complication arose when cartilage was first envisaged as a multiphasic model with distinguishable components and that quantifying those and searching for the laws that govern their interaction is inadequate. To the thesis of this paper, cartilage as a bulk is as much continuum as is the response of its components to the external stimuli. For this reason, we framed the fundamental question as to what would be the mechano-structural functionality of such a system in the total absence of one of its key constituents-proteoglycans. To answer this, hydrated normal and proteoglycan depleted samples were tested under confined compression while finite element models were reproduced, for the first time, based on the structural microarchitecture of the cross-sectional profile of the matrices. These micro-porous in silico models served as virtual transducers to produce an internal noninvasive probing mechanism beyond experimental capabilities to render the matrices micromechanics and several others properties like permeability, orientation etc. The results demonstrated that load transfer was closely related to the microarchitecture of the hyperelastic models that represent solid skeleton stress and fluid response based on the state of the collagen network with and without the swollen proteoglycans. In other words, the stress gradient during deformation was a function of the structural pattern of the network and acted in concert with the position-dependent compositional state of the matrix. This reveals that the interaction between indistinguishable components in real cartilage is superimposed by its microarchitectural state which directly influences macromechanical behavior.

An adhesive elasto-plastic contact model for the discrete element method with three dimensional non-spherical particles is proposed and investigated to achieve quantitative prediction of cohesive powder flowability. Simulations have been performed for uniaxial consolidation followed by unconfined compression to failure using this model. The model has been shown to be capable of predicting the experimental flow function (unconfined compressive strength vs. the prior consolidation stress) for a limestone powder which has been selected as a reference solid in the Europe wide PARDEM research network. Contact plasticity in the model is shown to affect the flowability significantly and is thus essential for producing satisfactory computations of the behaviour of a cohesive granular material. The model predicts a linear relationship between a normalized unconfined compressive strength and the product of coordination number and solid fraction. This linear relationship is in line with the Rumpf model for the tensile strength of particulate agglomerate. Even when the contact adhesion is forced to remain constant, the increasing unconfined strength arising from stress consolidation is still predicted, which has its origin in the contact plasticity leading to microstructural evolution of the coordination number. The filled porosity is predicted to increase as the contact adhesion increases. Under confined compression, the porosity reduces more gradually for the load-dependent adhesion compared to constant adhesion. It was found that the contribution of adhesive force to the limiting friction has a significant effect on the bulk unconfined strength. The results provide new insights and propose a micromechanical based measure for characterising the strength and flowability of cohesive granular materials.

In this study, the behaviour of iron ore fines with varying levels of adhesion was investigated using a confined compression test and a uniaxial test. The uniaxial test was conducted using the semi-automated uniaxial EPT tester in which the cohesive strength of a bulk solid is evaluated from an unconfined compression test following a period of consolidation to a pre-defined vertical stress. The iron ore fines were also tested by measuring both the vertical and circumferential strains on the cylindrical container walls under vertical loading in a separate confined compression tester - the K0 tester, to determine the lateral pressure ratio. Discrete Element Method simulations of both experiments were carried out and the predictions were compared with the experimental observations. A recently developed DEM contact model for cohesive solids, an Elasto-Plastic Adhesive model, was used. This particle contact model uses hysteretic non-linear loading and unloading paths and an adhesion parameter which is a function of the maximum contact overlap. The model parameters for the simulations are phenomenologically based to reproduce the key bulk characteristics exhibited by the solid. The simulation results show a good agreement in capturing the stress history dependent behaviour depicted by the flow function of the cohesive iron ore fines while also providing a reasonably good match for the lateral pressure ratio observed during the confined compression K0 tests. This demonstrates the potential for the DEM model to be used in the simulation of bulk handling applications.

O trabalho consta do estudo do meio físico geotécnico do município de Porto Alegre visando a identificação e caracterização de unidades geotécnias, formadas por associações de solos com características físico-morfológicas e origem semelhantes. As unidades geotécnicas são definidas por um estudo de escritório baseado em levantamentos geológicos, pedológicos, topográficos e geográficos e também com o auxílio de fotografias aéreas, acompanhado de uma investigação de campo com a amostragem de perfis das diferentes unidades estimadas. Uma carta com a ocorrência das unidades geotécnicas e com a representação das estruturas geológicas de maior importância geotécina é construída. Destaque é dado ás unidades geotécnicas de solos subtropicais oriundos dos granitos, gnaisses e migmatitos. E executada uma avaliação das características físicas (mineralogia, granulometria, plasticidade, e índices físicos) e propriedades geotécnicas (compressibilidade, colapsividade e resistência ao cisalhamento) dos solos dos principais horizontes de perfis típicos destas unidades. São utilizados ensaios de caracterização, análises mineralógicas de rochas por lâminas delgadas, análises mineralógicas de argilas por meio de difratograma de raio X, ensaios de compressão confinada e ensaios de cisalhamento direto. Os resultados são apresentados em função dos principais horizontes de solos das unidades geotécnicas , são eles horizontes saprolíticos, formados por solo residual que ainda apresentam minerais primários e vestígios estruturais da rocha de origem e horizontes superficiais muitas das vezes laterizados. A análise foi feita à luz dos processos de formação e dos decorrentes fatores que determinam o comportamento geotécnico destes solos de intemperismo subtropical.

A technological alternative for the correct disposal of tires is the use in the construction of embankment with soil and shredded tires. The use of waste tires in tropical soils requires prior knowledge of the properties and limitations of these materials. In this work, the results of an experimental program was devised to characterize the behavior of mixtures of waste tires and a lateritic soil. The residue used in this study is classified as tire buffings with an average size of 1.4 mm. The laboratory program included testing of particle size analysis, Atterberg limits, compaction, direct shear tests, permeability and confined compression tests with pure soil, pure tire and the mixtures. Proportions of 0% (pure soil), 10%, 20%, 40%, 50 % and 100% (pure tire) by weight were used. For the confining stress levels used in the study, the presence of tire residue provided a considerable increase in shear strength of the mixture. The maximum shear strength was obtained for a residue content of 40% by weight. Permeability tests on samples of waste under a confining stress of 100 kPa showed that the permeability increases significantly with increasing residue content until a residue content of 20%. The increase in permeability after that value showed to be negligible. Confined compression tests showed that the soil mixed with tire residue becomes more compressible than the pure soil. The secant constrained modulus (Msec) for the same vertical stress decreases with increasing percentage of residue.

Tissue grafts are implanted in orthopedic surgery every day. In order to minimize infection risk, bone allografts are often delipidated with supercritical CO2 and sterilized prior to implantation. This treatment may, however, impair the mechanical behavior of the bone graft tissue. The goal of this study was to determine clinically relevant mechanical properties of treated/sterilized human trabecular bone grafts, e.g. the apparent modulus, strength, and the ability to absorb energy during compaction. They were compared with results of identical experiments performed previously on untreated/fresh frozen human trabecular bone from the same anatomical site (Charlebois, 2008). We tested the hypothesis that the morphology–mechanical property relationships of treated cancellous allografts are similar to those of fresh untreated bone. The morphology of the allografts was determined by μCT. Subsequently, cylindrical samples were tested in unconfined and confined compression. To account for various morphologies, the experimental data was fitted to phenomenological mechanical models for elasticity, strength, and dissipated energy density based on bone volume fraction (BV/TV) and the fabric tensor determined by MIL. The treatment/sterilization process does not appear to influence bone graft stiffness. However, strength and energy dissipation of the bone grafts were found to be significantly reduced by 36% to 47% and 66% to 81%, respectively, for a broad range of volume fraction (0.14 < BV/TV < 0.39) and degree of anisotropy (1.24 < DA < 2.18). Since the latter properties are strongly dominated by BV/TV, the clinical consequences of this reduction can be compensated by using grafts with lower porosity. The data of this study suggests that an increase of 5–10% in BV/TV is sufficient to compensate for the reduced post-yield mechanical properties of treated/sterilized bone in monotonic compression. In applications where graft stiffness needs to be matched and strength is not a concern, treated allograft with the same BV/TV as an appropriate fresh bone graft may be used.

We report the material properties of 26 granular analogue materials used in 14 analogue modelling laboratories. We determined physical characteristics such as bulk density, grain size distribution, and grain shape, and performed ring shear tests to determine friction angles and cohesion, and uniaxial compression tests to evaluate the compaction behaviour. Mean grain size of the materials varied between c. 100 and 400 μm. Analysis of grain shape factors shows that the four different classes of granular materials (14 quartz sands, 5 dyed quartz sands, 4 heavy mineral sands and 3 size fractions of glass beads) can be broadly divided into two groups consisting of 12 angular and 14 rounded materials. Grain shape has an influence on friction angles, with most angular materials having higher internal friction angles (between c. 35° and 40°) than rounded materials, whereas well-rounded glass beads have the lowest internal friction angles (between c. 25° and 30°). We interpret this as an effect of intergranular sliding versus rolling. Most angular materials have also higher basal friction angles (tested for a specific foil) than more rounded materials, suggesting that angular grains scratch and wear the foil. Most materials have an internal cohesion in the order of 20–100 Pa except for well-rounded glass beads, which show a trend towards a quasi-cohesionless (C < 20 Pa) Coulomb-type material. The uniaxial confined compression tests reveal that rounded grains generally show less compaction than angular grains. We interpret this to be related to the initial packing density after sifting, which is higher for rounded grains than for angular grains. Ring-shear test data show that angular grains undergo a longer strain-hardening phase than more rounded materials. This might explain why analogue models consisting of angular grains accommodate deformation in a more distributed manner prior to strain localisation than models consisting of rounded grains.

Effective stress paths for a loessial soil subject to collapse during confined compression have been determined from the results of a testing program consisting of (1) confined compression tests on natural samples of loess with initial water contents ranging from air-dry to saturation, (2) negative pore-water pressure measurements to -300 psi during these tests, and (3) Ko-tests in which the lateral stress ratio was measured for one-dimensional strain. Before collapse, Ko was found to average 0.23, an extremely low value for a loose soil, whereas after collapse, Ko increased to 0.54, which is consistent with values for other soils. Because of the low Ko-values before collapse, the effective stress path for loading in confined compression initially approaches the failure envelope. At collapse the stress path intersects the failure envelope and thereafter it changes direction as a consequence of the higher Ko-value after collapse. From the stress path interpretation of the results, it is demonstrated that the collapse mechanism of loess in confined compression and during wetting is a shear phenomenon and subject of analysis in terms of effective stresses.

Unreinforced masonry (URM) structures that are in need of repair and rehabilitation constitute a significant portion of building stock worldwide. The successful application of fiber-reinforced polymers (FRP) for repair and retrofitting of reinforced-concrete (RC) structures has opened new avenues for strengthening URM structures with FRP materials. The present study analyzes the behavior of FRP-confined masonry prisms under monotonic axial compression. Masonry comprising of burnt clay bricks and cement-sand mortar (generally adopted in the Indian subcontinent) having E-b/E-m ratio less than one is employed in the study. The parameters considered in the study are, (1) masonry bonding pattern, (2) inclination of loading axis to the bed joint, (3) type of FRP (carbon FRP or glass FRP), and (4) grade of FRP fabric. The performance of FRP-confined masonry prisms is compared with unconfined masonry prisms in terms of compressive strength, modulus of elasticity and stress-strain response. The results showed an enhancement in compressive strength, modulus of elasticity, strain at peak stress, and ultimate strain for FRP-confined masonry prisms. The FRP confinement of masonry resulted in reducing the influence of the inclination of the loading axis to the bed joint on the compressive strength and failure pattern. Various analytical models available in the literature for the prediction of compressive strength of FRP-confined masonry are assessed. New coefficients are generated for the analytical model by appending experimental results of the current study with data available in the literature. (C) 2014 American Society of Civil Engineers.

Commercial purity (99.8%) magnesium single crystals were subjected to plane strain compression (PSC) along the c-axis at 200 and 370 degrees C and a constant strain rate of 10(-3) s(-1). Extension was confined to the < 1 1 (2) over bar 0 > direction and the specimens were strained up to a logarithmic true strain of -1. The initial rapid increase in flow stress was followed by significant work softening at different stresses and comparable strains of about -0.05 related to macroscopic twinning events. The microstructure of the specimen after PSC at 200 degrees C was characterized by a high density of {1 0 (1) over bar 1} and {1 0 (1) over bar 3} compression twins, some of which were recrystallized. After PSC at 370 degrees C, completely recrystallized twin bands were the major feature of the observed microstructure. All new grains in these bands retained the same c-axis orientation of their compression twin hosts. The basal plane in these grains was randomly rotated around the c-axis, forming a fiber texture component. The obtained results are discussed with respect to the mechanism of recrystallization, the specific character of the boundaries between new grains and the initial matrix, and the importance of the dynamically recrystallized bands for strain accommodation in these deformed magnesium single crystals. An approach for producing a large quantity of neutrons is proposed. It involves compression of a fuel foil and confinement of the resulting plasma between two intense laser pulses. It is shown that two circularly polarized laser pulses of amplitude a=7 illuminating a deuterium-tritium foil of areal density 3.3 X 10(18) cm(-2) can produce about 4.2 X 10(6) neutrons per joule of the input laser energy.

The increasing use of Fiber Reinforced methods for strengthening existing brick masonry walls and columns, especially for the rehabilitation of historical buildings, has generated considerable research interest in understanding the failure mechanism in such systems. This dissertation is aimed to provide a basic understanding of the behavior of solid brick masonry walls unwrapped and wrapped with Fiber Reinforced Cementitious Matrix Composites. This is a new type of composite material, commonly known as FRCM, featuring a cementitious inorganic matrix (binder) instead of the more common epoxy one. The influence of the FRCM-reinforcement on the load-carrying capacity and strain distribution during compression test will be investigated using a full-field optical technique known as Digital Image Correlation. Compression test were carried on 6 clay bricks columns and on 7 clay brick walls in three different configuration, casted using bricks scaled respect the first one with a ratio 1:2, in order to determinate the effects of FRCM reinforcement. The goal of the experimental program is to understand how the behavior of brick masonry will be improved by the FRCM-wrapping. The results indicate that there is an arching action zone represented in the form of a parabola with a varying shape according to the used configuration. The area under the parabolas is considered as ineffectively confined. The effectively confined area is assumed to occur within the region where the arching action had been fully developed.
