The organisation of cells of the planctomycete species Pirellula marina, Isosphaera pallida, Gemmata obscuriglobus, Planctomyces mat-is and Candidatus Brocadia anammoxidans was investigated based on ultrastructure derived from thin-sections of cryosubstituted cells, freeze-fracture replicas, and in the case of Gemmata obscuriglobus and Pirellllla marina, computer-aided 3-D reconstructions from serial sections of cryosubstituted cells. All planctomycete cells display a peripheral ribosome-free region, termed here the paryphoplasm, surrounding the perimeter of the cell, and an interior region including any nucleoid regions as well as ribosome-like particles, bounded by a single intracytoplasmic membrane (ICM), and termed the pirellulosome in Pirellula species. Immunogold labelling and RNase-gold cytochemistry indicates that in planctomycetes all the cell DNA is contained wholly within the interior region bounded by the ICM, and the paryphoplasm contains no DNA but at least some of the cell's RNA. The ICM in Isosphaera pallida and Planctomyces mat-is is invaginated such that the paryphoplasm forms a major portion of the cell interior in sections, but in other planctomycetes it remains as a peripheral zone. In the anaerobic ammonium-oxidising (anammox process) chemoautotroph Candidatus Brocadia anammoxidans the interior region bounded by ICM contains a further internal single-membrane-bounded region, the anam-moxosome. In Gemmata obscuriglobus. the interior ICM-bounded region contains the nuclear body, a double-membrane-bounded region containing the cell's nucleoid and all genomic DNA in addition to some RNA. Shared features of cell compartmentalisation in different planctomycetes are consistent with the monophyletic nature of the planctomycetes as a distinct division of the Bacteria. The shared organisational plan for the planctomycete cell constitutes a new type not known in cells of other bacteria.

In vertebrate animals, pleural and peritoneal cavities are repositories of milky spots (MS), which constitute an organised coelom-associated lymphomyeloid tissue that is intensively activated by Schistosoma mansoni infection. This study compared the reactive patterns of peritoneal MS to pleural MS and concluded from histological analysis that they represent independent responsive compartments. Whole omentum, lungs and the entire mediastinum of 54 S. mansoni-infected mice were studied morphologically. The omental MS of infected animals were highly activated, modulating from myeloid-lymphocytic (60 days of infection) to lymphomyeloid (90 days of infection) and lymphocytic or lymphoplasmacytic (160 days of infection) types. The non-lymphoid component predominated in the acute phase of infection and was expressed by monocytopoietic, eosinopoietic and neutropoietic foci, with isolated megakaryocytes and small foci of late normoblasts and mast cells. Nevertheless, pleural or thoracic MS of infected mice were monotonous, consisting of small and medium lymphocytes with few mast and plasma cells and no myeloid component. Our data indicate that compartmentalisation of the MS response is dependent on the lymphatic vascularisation of each coelomic cavity, limiting the effects or consequences of any stimulating or aggressive agents, as is the case with S. mansoni infection.

The gastrointestinal tract is heavily colonized with commensal microbes with the concentration of bacteria increasing longitudinally down the length of the intestine. Bacteria are also spatially distributed transversely from the epithelial surface to the intestinal lumen with the inner mucus layer normally void of bacteria. Maintenance of this equilibrium is extremely important for human health and, as the dominant immunoglobulin at mucosal sites, IgA influences mutualism between the host and its normal microbiota. In this review we focus on the links between immune and microbial geography of the mammalian intestinal tract.

Smooth muscle cells (SMC) exhibit a functional plasticity, modulating from the mature phenotype in which the primary function is contraction, to a less differentiated state with increased capacities for motility, protein synthesis, and proliferation. The present study determined, using Western analysis, double-label immunofluorescence and confocal microscopy, whether changes in phenotypic expression of rabbit aortic SMC in culture could be correlated with alterations in expression and distribution of structural proteins. Contractile state SMC (days 1 and 3 of primary culture) showed distinct sorting of proteins into subcellular domains, consistent with the theory that the SMC structural machinery is compartmentalised within the cell. Proteins specialised for contraction (alpha -SM actin, SM-MHC, and calponin) were highly expressed in these cells and concentrated in the upper central region of the cell. Vimentin was confined to the body of the cell, providing support for the contractile apparatus but not co-localising with it. In line with its role in cell attachment and motility, beta -NM actin was localised to the cell periphery and basal cortex. The dense body protein alpha -actinin was concentrated at the cell periphery, possibly stabilising both contractile and motile apparatus. Vinculin-containing focal adhesions were well developed, indicating the cells' strong adhesion to substrate. In synthetic state SMC (passages 2-3 of culture), there was decreased expression of contractile and adhesion (vinculin) proteins with a concomitant increase in cytoskeletal proteins (beta -non-muscle [NM] actin and vimentin). These quantitative changes in structural proteins were associated with dramatic chan-es in their distribution. The distinct compartmentalisation of structural proteins observed in contractile state SMC was no longer obvious, with proteins more evenly distributed throughout die cytoplasm to accommodate altered cell function. Thus, SMC phenotypic modulation involves not only quantitative changes in contractile and cytoskeletal proteins, but also reorganisation of these proteins. Since the cytoskeleton acts as a spatial regulator of intracellular signalling, reorganisation of the cytoskeleton may lead to realignment of signalling molecules, which, in turn, may mediate the changes in function associated with SMC phenotypic modulation. El projecte de recerca “Església, poder i societat als comtats catalans d'origen carolingi (877-1010)” contempla l'estudi de l'Església catalana del segle X en tots els seus vessants: com a estructura formada per diferents institucions (seus episcopals, cenobis, parròquies i esglésies-oratoris); com a conjunt de persones (clergues i laics que participen de diferent manera i amb diferent implicació dels afers eclesiàstics); en relació al poder polític-administratiu dels comtats catalans (comtes, vescomtes, senyors i altres delegats del poder i la justícia); com en relació amb els aspectes socioeconòmics i d'ordenació territorial (la gran massa de camperols, l'acaparament de terres per part dels grans propietaris, l'articulació d'un sistema de rendes, els vincles entre persones de diferents rangs, la compartimentació d'un espai creixent). Observem, doncs, que l'Església catalana altmedieval resta present en tots els àmbits de la vida i que el seu estudi permet, de retruc, un coneixement més profund i rigorós de la societat. Tot això, contextualitzat durant el segle X, a cavall entre dos períodes ben estudiats i que en ocasions l'han ennuvolat. El segle X és un període privilegiat per a l'estudi d'un bon nombre de dinàmiques polítiques, socials, econòmiques i culturals que demostren que aquesta centúria tenia una personalitat pròpia i que la posen en rellevància respecte als temps anteriors i posteriors. La historiografia tradicional altmedieval catalana tendeix a buscar l'origen dels canvis socials, institucionals i polítics, de la Catalunya medieval, en dos moments concrets: el món carolingi o els anys immediatament posteriors al canvi de mil•lenni. No obstant això, l'augment de fonts històriques, el millor coneixement dels conceptes i de les institucions i l'apropament a la història mitjançant nous enfocaments i paradigmes, permet visualitzar el segle X com a moment de continuació, però també de naixement de situacions prou importants com per a no ser negligides.

Ce mémoire s’intéresse aux expériences de femmes en processus de sortie de la prostitution. Il vise à comprendre les obstacles auxquels ces femmes sont confrontées pour pouvoir bénéficier d’interventions sociales accessibles et facilitant leur sortie de la prostitution. Cette recherche qualitative prend appui sur 11 entretiens individuels réalisés auprès de femmes âgées de 26 à 55 ans et habitant Montréal, les Laurentides et l’Abitibi. Bien que nombre de femmes aux prises avec la prostitution souhaitent en sortir, on compte peu d’interventions sociales pour les aider en ce sens. Les services publics sont largement insuffisants à la fois du point de vue de leur accessibilité et de leur réponse aux besoins de ces femmes. Peu d’études s’intéressent aux services d’aide à la sortie de la prostitution, notamment au Québec. Ce mémoire privilégie une perspective féministe abolitionniste et un cadre épistémologique de la théorie standpoint. Les résultats mettent en lumière les obstacles à l’accessibilité des interventions sociales, dont le cloisonnement des services et le refus manifeste d’offrir de l’aide aux femmes. Cette recherche rend compte également de l’expérience de pratiques d’intervention entravant le processus de sortie : 1) les pratiques punitives, 2) celles proposant une aide limitée aux femmes ou 3) leur adaptation à la prostitution. La conclusion de ce mémoire propose la mise en œuvre de pratiques sociales novatrices qui prennent en compte les contraintes sociales qui mènent les femmes à l’industrie du sexe et les y maintiennent ainsi que les conséquences de l’expérience même de la prostitution sur elles.

Over the last decade issues related to the financial viability of development have become increasingly important to the English planning system. As part of a wider shift towards the compartmentalisation of planning tasks, expert consultants are required to quantify, in an attempt to rationalise, planning decisions in terms of economic ‘viability’. Often with a particular focus on planning obligations, the results of development viability modelling have emerged as a key part of the evidence base used in site-specific negotiations and in planning policy formation. Focussing on the role of clients and other stakeholders, this paper investigates how development viability is tested in practice. It draws together literature on the role of calculative practices in policy formation, client feedback and influence in real estate appraisals and stakeholder engagement and consultation in the planning literature to critically evaluate the role of clients and other interest groups in influencing the production and use of development viability appraisal models. The paper draws upon semi-structured interviews with the main producers of development viability appraisals to conclude that, whilst appraisals have the potential to be biased by client and stakeholder interests, there are important controlling influences on potential opportunistic behaviour. One such control is local authorities’ weak understanding of development viability appraisal techniques which limits their capacity to question the outputs of appraisal models. However, this also is of concern given that viability is now a central feature of the town planning system.
