Intra-host sequence data from RNA viruses have revealed the ubiquity of defective viruses in natural viral populations, sometimes at surprisingly high frequency. Although defective viruses have long been known to laboratory virologists, their relevance in clinical and epidemiological settings has not been established. The discovery of long-term transmission of a defective lineage of dengue virus type 1 (DENV-1) in Myanmar, first seen in 2001, raised important questions about the emergence of transmissible defective viruses and their role in viral epidemiology. By combining phylogenetic analyses and dynamical modelling, we investigate how evolutionary and ecological processes at the intra-host and inter-host scales shaped the emergence and spread of the defective DENV-1 lineage. We show that this lineage of defective viruses emerged between June 1998 and February 2001, and that the defective virus was transmitted primarily through co-transmission with the functional virus to uninfected individuals. We provide evidence that, surprisingly, this co-transmission route has a higher transmission potential than transmission of functional dengue viruses alone. Consequently, we predict that the defective lineage should increase overall incidence of dengue infection, which could account for the historically high dengue incidence reported in Myanmar in 2001-2002. Our results show the unappreciated potential for defective viruses to impact the epidemiology of human pathogens, possibly by modifying the virulence-transmissibility trade-off, or to emerge as circulating infections in their own right. They also demonstrate that interactions between viral variants, such as complementation, can open new pathways to viral emergence.

Background: A single case of paternal co-transmission ofmitochondrial DNA (mtDNA) in humans has been reported so far. Objective: To find potential instances of non-maternal inheritance of mtDNA. Methods: Published medical case studies (of single patients) were searched for irregular mtDNA patterns by comparing the given haplotype information for different clones or tissues with the worldwide mtDNA database as known to date-a method that has proved robust and reliable for the detection of flawed mtDNA sequence data. Results: More than 20 studies were found reporting clear cut instances with mtDNAs of different ancestries in single individuals. As examples, cases are reviewed from recent published reports which, at face value, may be taken as evidence for paternal inheritance of mtDNA or recombination. Conclusions: Multiple types (or recombinant types) of quite dissimilar mitochondrial DNA from different parts of the known mtDNA phylogeny are often reported in single individuals. From re-analyses and corrigenda of forensic mtDNA data, it is apparent that the phenomenon of mixed or mosaic mtDNA can be ascribed solely to contamination and sample mix up.

Des travaux récents démontrent que certains neurones dopaminergiques du mésencéphale ont la capacité de libérer du glutamate en plus de la dopamine (DA). Ce phénomène de « co-transmission » requiert l’expression du transporteur vésiculaire du glutamate de type 2 (VGLUT2) dans les neurones dopaminergiques. Certaines observations montrent que l’expression de VGLUT2 dans les neurones dopaminergiques survient tôt durant leur développement et est essentiellement limitée aux neurones de l’aire tegmentaire ventrale (VTA). De plus, cette libération de glutamate se retrouve principalement au niveau des terminaisons de ces neurones dans le striatum ventral, mais pas dans le striatum dorsal. Ces données suggèrent d’une part un rôle développemental possible du glutamate dans les neurones dopaminergiques, et d’autre part, que les signaux dérivés des neurones cibles puissent réguler le double phénotype des neurones dopaminergiques menant ainsi à une plasticité phénotypique. Par ailleurs, il est toujours inconnu si cette libération de glutamate se fait à partir des terminaisons qui relâchent la DA ou à partir de terminaisons axonales distinctes. De plus, le rôle physiologique de ce surprenant phénomène de co-transmission reste également inconnu. Ainsi, dans cette étude, nous avons d’abord démontré in vitro et in vivo que l’expression de VGLUT2 est nécessaire pour la survie et la croissance d’une sous-population de neurones dopaminergiques. En utilisant une lignée de souris ayant une délétion génique spécifique de VGLUT2 dans les neurones dopaminergiques, nous avons observé une diminution du nombre de terminaisons dopaminergiques et glutamatergiques dans le striatum, une baisse de libération de DA dans le striatum ventral, une diminution de la coordination motrice ainsi qu’une diminution de l’activité locomotrice induite par les drogues d’abus. D’autre part, nous avons démontré in vitro et in vivo que les neurones dopaminergiques au double phénotype établissent des terminaisons distinctes afin de relâcher le glutamate et la DA. De plus, nous démontrons que ce phénomène de ségrégation des sites de libération semble être induit par une interaction avec les neurones du striatum ventral. Ces travaux démontrent le rôle physiologique déterminant de la co-transmission DA-glutamate pour l’homéostasie du système DAergique et dévoile une caractéristique fondamentale de l’établissement des terminaisons axonales de ces neurones. Ces travaux permettent ainsi de mieux comprendre les rôles physiologiques de la co-libération de glutamate par les neurones du système nerveux central et présentent une nouvelle perspective sur les dysfonctions potentielles de ces neurones dans les maladies du cerveau.

1 The effects of calcium channel blockers on co-transmission from different populations of autonomic vasomotor neurons were studied on isolated segments of uterine artery and vena cava from guinea-pigs. 2 Sympathetic, noradrenergic contractions of the uterine artery (produced by 200 pulses at 1 or 10 Hz; 600 pulses at 20 Hz) were abolished by the N-type calcium channel blocker omega-conotoxin (CTX) GVIA at 1-10 nM. 3 Biphasic sympathetic contractions of the vena cava (600 pulses at 20 Hz) mediated by noradrenaline and neuropeptide Y were abolished by 10 nM CTX GVIA. 4 Neurogenic relaxations of the uterine artery (200 pulses at 10 Hz) mediated by neuronal nitric oxide and neuropeptides were reduced < 50% by CTX GVIA 10-100 nM. 5 Capsaicin (3 muM) did not affect the CTX GVIA-sensitive or CTX GVIA-resistant neurogenic relaxations of the uterine artery. 6 The novel N-type blocker CTX CVID (100-300 nM), P/Q-type blockers agatoxin IVA (10-100 nM) or CTX CVIB (100 nM), the L-type blocker nifedipine (10 muM) or the 'R-type' blocker SNX-482 (100 nM), all failed to reduce CTX GVIA-resistant relaxations. The T-type channel blocker NiCl2 (100-300 muM) reduced but did not abolish the remaining neurogenic dilations. 7 Release of different neurotransmitters from the same autonomic vasomotor axon depends on similar subtypes of calcium channels. N-type channels are responsible for transmitter release from vasoconstrictor neurons innervating a muscular artery and capacitance vein, but only partly mediate release of nitric oxide and neuropeptides from pelvic vasodilator neurons.

In this paper, a new approach to enhance the transmission system distance relay co-ordination is presented. The approach depends on the apparent impedance loci seen by the distance relay during all possible disturbances. In a distance relay, the impedance loci seen at the relay location is obtained by extensive transient stability studies. Support vector machines (SVMs), a class of patterns classifiers are used in discriminating zone settings (zone-1, zone-2 and zone-3) using the signals to be used by the relay. Studies on a sample 9-bus are presented for illustrating the proposed scheme.

In this paper, knowledge-based approach using Support Vector Machines (SVMs) are used for estimating the coordinated zonal settings of a distance relay. The approach depends on the detailed simulation studies of apparent impedance loci as seen by distance relay during disturbance, considering various operating conditions including fault resistance. In a distance relay, the impedance loci given at the relay location is obtained from extensive transient stability studies. SVMs are used as a pattern classifier for obtaining distance relay co-ordination. The scheme utilizes the apparent impedance values observed during a fault as inputs. An improved performance with the use of SVMs, keeping the reach when faced with different fault conditions as well as system power flow changes, are illustrated with an equivalent 265 bus system of a practical Indian Western Grid.

The design of a sustainable electricity generation and transmission system is based on the established science of anthropogenic climate change and the realization that depending on imported fossil-fuels is becoming a measure of energy insecurity of supply. A model is proposed which integrates generation fuel mix composition, assignment of plants and optimized power flow, using Portugal as a case study. The result of this co-optimized approach is an overall set of generator types/fuels which increases the diversity of Portuguese electricity supply, lowers its dependency on imported fuels by 14.62% and moves the country towards meeting its regional and international obligations of 31% energy from renewables by 2020 and a 27% reduction in greenhouse gas emissions by 2012, respectively. The quantity and composition of power generation at each bus is specified, with particular focus on quantifying the amount of distributed generation. Based on other works, the resultant, overall distributed capacity penetration of 19.02% of total installed generation is expected to yield positive network benefits. Thus, the model demonstrates that national energy policy and technical deployment can be linked through sustainability and, moreover, that the respective goals may be mutually achieved via holistic, integrated design. ©2009 IEEE.
