[Excerpt] Although Acinetobacter baumannii has been the main agent for healthcare infections, recent reports suggest that some Acinetobacter environmental species should be considered as a potential cause of disease. In Angola, there are no previous data on its environmental reservoirs and resistance features. We aimed to unveil the occurrence and diversity of Acinetobacter species and the presence of resistance mechanisms in different non-clinical settings in Angola.

Pseudomonas aeruginosa is a dreaded opportunistic pathogen that causes severe and often intractable infections in immunocompromised and critically ill patients. This bacterium is also the primary cause of fatal lung infections in patients with cystic fibrosis and a leading nosocomial pathogen responsible for nearly 10% of all hospital-acquired infections. P. aeruginosa is intrinsically recalcitrant to most classes of antibiotics and has the ability to acquire additional resistance during treatment. In particular, resistance to the widely used β-lactam antibiotics is frequently mediated by the expression of AmpC, a chromosomally encoded β-lactamase that is ubiquitously found in P. aeruginosa strains. This dissertation delved into the role of a recently reported chromosomal β-lactamase in P. aeruginosa called PoxB. To date, no detailed studies have addressed the regulation of poxB expression and its contribution to β-lactam resistance in P. aeruginosa. In an effort to better understand the role of this β-lactamase, poxB was deleted from the chromosome and expressed in trans from an IPTG-inducible promoter. The loss of poxB did not affect susceptibility. However, expression in trans in the absence of ampC rendered strains more resistant to the carbapenem β-lactams. The carbapenem-hydrolyzing phenotype was enhanced, reaching intermediate and resistant clinical breakpoints, in the absence of the carbapenem-specific outer membrane porin OprD. As observed for most class D β-lactamases, PoxB was only weakly inhibited by the currently available β-lactamase inhibitors. Moreover, poxB was shown to form an operon with the upstream located poxA, whose expression in trans decreased pox promoter (Ppox) activity suggesting autoregulation. The transcriptional regulator AmpR negatively controlled Ppox activity, however no direct interaction could be demonstrated. A mariner transposon library identified genes involved in the transport of polyamines as potential regulators of pox expression. Unexpectedly, polyamines themselves were able induce resistance to carbapenems. In summary, P. aeruginosa carries a chromosomal-encoded β-lactamase PoxB that can provide resistance against the clinically relevant carbapenems despite its narrow spectrum of hydrolysis and whose activity in vivo may be regulated by polyamines.

There are many different designs for audio amplifiers. Class-D, or switching, amplifiers generate their output signal in the form of a high-frequency square wave of variable duty cycle (ratio of on time to off time). The square-wave nature of the output allows a particularly efficient output stage, with minimal losses. The output is ultimately filtered to remove components of the spectrum above the audio range. Mathematical models are derived here for a variety of related class-D amplifier designs that use negative feedback. These models use an asymptotic expansion in powers of a small parameter related to the ratio of typical audio frequencies to the switching frequency to develop a power series for the output component in the audio spectrum. These models confirm that there is a form of distortion intrinsic to such amplifier designs. The models also explain why two approaches used commercially succeed in largely eliminating this distortion; a new means of overcoming the intrinsic distortion is revealed by the analysis. A switch-mode assisted linear amplifier (SMALA) combining a linear (Class B) and a switch-mode (Class D) amplifier is presented. The usual single hysteretic controlled half-bridge current dumping stage is replaced by two parallel buck converter stages, in a parallel voltage controlled topology. These operate independently: one buck converter sources current to assist the upper Class B output device, and a complementary converter sinks current to assist the lower device. This topology lends itself to a novel control approach of a dead-band at low power levels where neither class D amplifier assists, allowing the class B amplifier to supply the load without interference, ensuring high fidelity. A 20 W implementation demonstrates 85% efficiency, with distortion below 0.08% measured across the full audio bandwidth at 15 W. The class D amplifier begins assisting at 2 W, and below this value, the distortion was below 0.03%. Complete circuitry is given, showing the simplicity of the additional class D amplifier and its corresponding control circuitry.

Tässä työssä tutkitaan D-luokan audiovahvistimen pääteastetopologioiden eroja teknisen toiminnan, käytännön toteutuksen ja audiosignaalin laadun näkökulmista. Tutkimuksen perusteella valitaan yksi topologia kitaravahvistinsovellukseen, jossa modulointi on toteutettu Sigma-Delta-modulaattorilla. Pääteastetopologioiden tekninen toiminta selvitetään yksityiskohtaisesti virtojen kulkureittien avulla. Simulaatioilla tutkitaan suoja-ajan ja tulosignaalin variaatioiden vaikutusta lähtösignaalin kokonaisharmoniseen säröön (THD) ja selvitetään signaalin säröytymiseen vaikuttavien tekijöiden syitä D-luokan vahvistimen pääteasteessa.

Tämän kandidaatintyön aiheena on D-luokan audiovahvistimen särö ja kohina. Tarkoituksena on selvittää vahvistinluokan merkittävin särö- ja kohinamekanismi, sekä arvioida, voidaanko häiriöitä vähentää lähdön suodattimella. Tutkimusmenetelminä on kirjallisuus ja simulointi. Aineistona on käytetty IEEE:ssä julkaistuja tieteelisiä artikkeleita, eri valmistajien laatimia ohjeita, sekä aihetta käsitteleviä kirjoja. Keskeisimmät tulokset olivat, että merkittävin särömekanismi on transistoreiden suoja-ajan aiheuttama vääristymä, sekä että merkittävin kohina syntyy modulaatiossa käytetystä kantoaallosta. Kantoaallon näkyvyyteen kuormassa voidaan vaikuttaa ulostulon alipäästösuodattimella. Suoja-ajan aiheuttama harmoninen kokonaissärö asettuu musiikin kaistanleveydelle, joten sitä ei voida poistaa suodattamalla.

D-luokan vahvistimien etu perinteisiin A- tai AB-luokan vahvistimiin nähden on niitten korkea hyötysuhde ja pieni koko. Lisäksi ne ovat edullisia ja niihin voidaan asentaa pienemmät jäähdytyslevyt kuin perinteisiin vahvistimiin korkean hyötysuhteen ansiosta. Tässä tutkimuksessa asennetaan Peavey Solo –kitaravahvistimen pääteasteena toimivan AB-luokan päätevahvistimen tilalle tehokkaampi ja uudempaa vahvistinluokkaa edustava D-luokan vahvistin. Tutkimuksessa selvitetään, miten D-luokan vahvistimella toteutettu kytkentä toimii verrattuna alkuperäiseen kytkentään tutkimalla kitaravahvistimelle tyypillisiä sähköisiä ominaisuuksia. Työ toteutetaan mittauksin. Koska työssä olevissa kytkennöissä on jännitteisiä osia, on siinä perehdyttävä sähköturvallisuuteen. D-luokan vahvistin asettaa tietyt sähkötekniset kestävyysvaatimukset kytkennän teholähteelle ja kaiutinelementille, jolloin työssä paneudutaan myös niiden valintaan. D-luokan vahvistimeen päivitetyn kytkennän tuottamat äänenpainetasot olivat valtaosassa mittauksia suuremmat kuin alkuperäisellä kytkennällä. Äänenpainetasojen mittaustilanteissa myös kuormalle syötetyt tehot olivat suuremmat päivitetyllä kytkennällä verrattuna alkuperäiseen. Suurin päivitetyn kytkennän kuorman teho 72,72 W saavutettiin, kun kaiutinelementin yli oli 24,12 V:n jännitteen tehollisarvo ja kuorman teho oli noin 36 kertaa suurempi kuin alkuperäisellä kytkennällä. D-luokan vahvistimen mitatut harmonisen särön arvot ovat huomattavasti pienempiä kuin alkuperäisellä vahvistimella. Muokkaamattoman vahvistimen mitatusta sähköisestä taajuusvasteeesta nähdään, että alkuperäinen vahvistin korostaa tiettyjä taajuuksia, kun D-luokan vahvistimen taajuusvaste on sen sijaan likimain tasainen. Akustinen taajuusvaste ei ole niin tasainen D-luokan vahvistimeen päivitetyllä kytkennällä kuin alkuperäisellä kytkennällä ja sillä on myös kapeampi taajuusalue. Jos päivitetyn kytkennän sointia haluttaisiin parantaa, pitäisi tutkia lisää akustiseen taajuusvasteeseen vaikuttavia asioita.

The convergence of information technology and consumer electronics towards battery powered portable devices has increased the interest in high efficiency, low dissipation amplifiers. Class D amplifiers are the state of the art in low power consumption and high performance amplification. In this thesis we explore the possibility of exploiting nonlinearities introduced by the PWM modulation, by designing an optimized modulation law which scales its carrier frequency adaptively with the input signal's average power while preserving the SNR, thus reducing power consumption. This is achieved by means of a novel analytical model of the PWM output spectrum, which shows how interfering harmonics and their bandwidth affect the spectrum. This allows for frequency scaling with negligible aliasing between the baseband spectrum and its harmonics. We performed low noise power spectrum measurements on PWM modulations generated by comparing variable bandwidth, random test signals with a variable frequency triangular wave carrier. The experimental results show that power-optimized frequency scaling is both feasible and effective. The new analytical model also suggests a new PWM architecture that can be applied to digitally encoded input signals which are predistorted and compared with a cosine carrier, which is accurately synthesized by a digital oscillator. This approach has been simulated in a realistic noisy model and tested in our measurement setup. A zero crossing search on the obtained PWM modulation law proves that this approach yields an equivalent signal quality with respect to traditional PWM schemes, while entailing the use of signals whose bandwidth is remarkably smaller due to the use of a cosine instead of a triangular carrier.
