Solid state compounds of general formula ML2.nH2O [where M is Mg, Ca, Sr or Ba; L is cinnamate (C6H5 -CH=CH-COO-) and n = 2, 4, 0.8, 3 respectively], have been synthetized. Thermogravimetry (TG), derivative thermogravimetry (DTG), differential scanning calorimetry (DSC) and X-ray diffraction powder patterns have been used to characterize and to study the thermal stability and thermal decomposition of these compounds.

Some new compounds of cinnamic acid with lighter trivalent lanthanides were prepared in the solid state. The compounds have general formula ML3·H2O, where L is cinnamate (C6H5-CH=CH-COO-) and M is La, Ce, Pr, Nd or Sm. Thermogravimetry, derivative thermogravimetry, differential scanning calorimetry, infrared absorption spectra and X-ray diffraction powder patterns were used to characterize and to study the thermal stability and thermal decomposition of these compounds.

Some new compounds of cinnamic acid with the latter trivalent lanthanides and yttrium(III) were synthesized in the solid state. The compounds have the general formula LnL3, where Ln represents trivalent Eu to Lu or Y ions and L is the cinnamate anion (C6H5-CH=CH-COO-). Les glucides constituent la classe de molécules organiques la plus abondante et ceux-ci jouent des rôles cruciaux dans divers processus biologiques. De part leur importance médicinale, la préparation des désoxy-sucres, des C-glycosides et des C-disaccharides est devenue un sujet de pointe en synthèse organique. De façon générale, cette thèse décrit une nouvelle synthèse de novo des 4-désoxy hexopyrannoses en plus de la préparation de C-glycosides biologiquement actifs. De plus, une attention particulière a été portée à la préparation de novo de 4-désoxy-C-disaccharides. Dans un premier temps, le catalyseur de Cr(III) de Jacobsen et un complexe binaphtol/titane ont été utilisés pour réaliser des hétéro-Diels-Alder énantiosélectives. Les dihydropyrannes ainsi générés ont été transformés en 4-désoxy hexopyrannoses présents dans la nature. De cette façon, un dérivé de l’acide ézoaminuroïque, un précurseur de la désosamine et de la néosidomycine, a été préparé suivant cette approche de novo. De plus, à titre comparatif, la néosidomycine a également été fabriquée selon une approche chiron, à partir du méthyl alpha-D-mannopyrannoside. Finalement, une évaluation biologique préliminaire de la néosidomycine a été effectuée sur une la concanavaline-A (Chapitre 2). Dans un deuxième temps, une allylation stéréosélective sur un aldéhyde lié via des liens C-C à une unité mannoside a permis de générer un alcool homoallylique. Cette dernière fonctionnalité a été transformée en 4-désoxy hexopyrannose de configuration D ou L. De cette façon, la préparation de pseudo 4-désoxy-C-disaccharides, de 4-désoxy-C-disaccharides et de pseudo 4-désoxy aza-C-disaccharides a facilement été réalisée. Les rapports diastéréoisomériques de la réaction d’allylation ont été déterminés en plus de la configuration absolue des nouveaux centres stéréogéniques formés. La transformation des alcools homoallyliques en pyrannes poly hydroxylés ou en lactames poly hydroxylés a été réalisée, en plus de la déprotection de certains membres de cette famille pour une évaluation biologique préliminaire sur la concanavaline-A (Chapitre 3). Finalement, la synthèse de C-glycosides biologiquement actifs a été réalisée selon deux volets: i) préparation de 3-C-mannopyrannosyl coumarines et ii) synthèse de C-galactosides, inhibiteurs de la lectine PA-IL. Pour ce faire, le couplage de Heck a été utilisé à partir d’un ester alpha,bêta-insaturé, attaché à une unité glycosidique via des liens C-C, pour générer un dérivé glycosyl cinnamate de méthyle. Cependant, lorsque le 2-iodophénol est utilisé comme partenaire de Heck, la coumarine correspondante a été isolée. Les dérivés C-galactopyrannosyl cinnamates de méthyle représentent de bons inhibiteurs monovalents de la PA-IL avec un Kd aussi bas que 37 micro M (Chapitre 4).

The synthesis of methacrylate esters of 4-cyanophenyl-(4-(ω-hydroxyalkyloxy)) cinnamates, with spacer lengths of 2 and 6 methylene units and the synthesis of the corresponding acrylate ester with a spacer of 2 methylene units are described. The methacrylate monomers were polymerized by free radical polymerization, both as homopolymers and as copolymers with the analogous benzoate monomer of spacer length 6. The acrylate ester could not be polymerized successfully under the same reaction conditions. Polymers were characterized by NMR spectroscopy, gel permeation chromatography, differential scanning calorimetry, and thermo-optic observations. Of the monomers prepared, only the cinnamate with a hexamethylene spacer shows a mesophase, seen on supercooling of the melt. All of the polymers prepared were liquid crystalline, with smectic behavior predominating in the polymethacrylates with the longer spacer group. A narrow nematic region is seen just below the clearing temperature with a range of 3–9°C, nematic character is increased in the copolymer series with the degree of incorporation of the cinnamate monomer with the spacer group of length 2.

Leaves from Carpolobia lutea (Polygalaceae) were screened to establish the antiulcer ethnomedicinal claim and to quantitatively isolate, elucidate the active compounds by semi-preparative HPLC. The anti-nociceptive effects of Carpolobia lutea (CL) G. Don (Polygalaceae) organic leaf extracts were tested in experimental models in mice. The anti-nociceptive mechanism was determined using tail-flick test, acetic acid-induced abdominal constrictions, formalin-induced hind paw licking and the hot plate test. The fractions (ethanol, ethyl acetate, chloroform, n-hexane) and crude ethyl acetate extract of CL (770 mg/kg, i.p.) produced significant inhibitions of both phases of the formalin-induced pain in mice, a reduction in acetic acid-induced writhing as well as and an elevation of the pain threshold in the hot plate test in mice. The inhibitions were greater to those produced by indomethacin (5 mg/kg, i.p.). Ethyl acetate fraction revealed cinnamic and coumaric acids derivatives, which are described for the first time in literature. These cinnamalglucosides polyphenols characterised from CL may in part account for the pharmacological activities. These findings confirm its ethnomedical use in anti-inflammatory pain and in pains from gastric ulcer-associated symptoms. © 2011 Springer Basel AG.

Estudos anteriores sugerem que o óleo essencial da Aniba canelilla contendo altos níveis de 1-nitro-2-feniletano apresenta atividade antinociceptiva, antiinflamatória e hipotensora. Um estudo teórico de correlação entre estrutura química versus atividade biológica do 1-nitro-2-feniletano e a prostaglandinaendoperóxido sintase foi realizado. Além disso, foi feita uma análise conformacional do nitrofeniletano, bem como modificações em sua estrutura química, obtendo-se derivados nitroestirenos e cinamatos, para a posterior avaliação da relação estrutura-propriedade. A análise conformacional do nitrofeniletano foi realizada usando o método DFT/B3LYP com o conjunto de base 6-31G(d) através da variação da torção do angulo dihedro entre as ligações dos carbonos C1-C2 ligadas aos grupos fenila e nitro, com variação de intervalos de 5°, na faixa de 0° a 180°, visando a busca dos confôrmeros de menor energia. O confôrmero de 180º do dihedro O2N-C1-C2-C6H5 foi significativamente mais estável e isto pode ser explicado em termos do equilíbrio pela presença de sete geometrias de mais baixas energias. A análise de HOMO e LUMO mostra que os grupamentos fenila e metileno contribuem para a formação deste orbital. O cálculo de energia e determinação de propriedades eletrônicas relacionadas com o mecanismo de interação com a ciclooxigenase demonstrou boa correlação entre os grupos nitro e carboxílico, os quais podem ser classificados como bioisósteros. Os resultados das propriedades físico-químicas calculadas indicam uma maior correlação do nitrofeniletano com o ácido acetilsalicílico, podendo ter seu mecanismo de ação relacionado através de interações com a Arg-120 e Ser-530.Cálculos realizados com a guanidina protonada pelo DFT/B3LYP/6-31G(d,p) parao complexo contendo o grupo carboxilato exibem uma energia de interação mais favorável, com valor de -136,34 kcal/mol, quando comparado com a molécula contendo o grupo nitro, com valor de -12,84 kcal/mol, o que pode explicar a maior efetividade da indometacina, semelhante aos resultados biológicos. Os derivados do nitrofeniletano apresentaram uma relação estrutural com derivados de cinamatos, diferindo nas propriedades redox, o que pode favorecer o desenvolvimento de estruturas protótipo como moléculas bioativas a partir destes esqueletos moleculares.

ABSTRACT FOR PART I: PHOSPHA-MICHAEL ADDITIONS TO ACTIVATED INTERNAL ALKENES: STERIC AND ELECTRONIC EFFECTS A method for the phospha-Michael addition of bis(4-methyl)phenyl phosphine oxide to activated internal alkenes has been developed. Michael acceptors including cinnamates, crotonates, chalcones, and internal alkenes containing multiple activating groups were all successfully utilized in this reaction. The reaction was fairly tolerant of electron-donating and electron-withdrawing substituents on the Michael acceptor, and moderate to excellent yields (49-96%) of the adducts were isolated. When steric bulk was increased by a second substituent on the -position of the Michael-acceptor the reaction was suppressed. This was usually overcome by adding a second activating substituent to the -position. ABSTRACT FOR PART II: MICROWAVE-ASSISTED ARYLGOLD BOND FORMATION A microwave-assisted method was developed for the formation of arylgold complexes containing (2-Biphenyl)di-tert-butylphosphine (JohnPhos) as the supporting phosphine ligand. Arylboronic acids with increasingly bulky aromatic groups were screened to determine the steric limitations of the reaction. Arylgold complexes (JohnPhos)Au(p-methoxyphenyl), (JohnPhos)Au(2,4,6-trimethylphenyl), and (JohnPhos)Au(4-bromo-10-anthracene) were all synthesized by microwave irradiation at 70ºC in the presence of Cs2CO3 in either THF or iPrOH. Reactions performed with arylboronic acids containing unhindered ortho positions were carried out in THF. Arylboronic acids with substituents on the ortho position required iPrOH as the reaction solvent. Arylboronic acids with extreme steric hindrance on the ortho position of the aryl substituent, 2,4,6-triisopropylpphenylboronic acid, were unreactive. It was determined that increasing the irradiation temperature increased the formation of side products, therefore to promote conversion to the arylgold complex the duration of the reaction time was increased while maintaining a temperature of 70ºC. Arylgold complexes (JohnPhos)Au(p-methoxyphenyl), (JohnPhos)Au(2,4,6-trimethylphenyl), and (JohnPhos)Au(4-bromo-10-anthracene) were synthesized with moderate yields (40-69%).

O estudo químico das folhas e dos frutos de P. richardiaefolium resultou no isolamento de oito lignanas, sendo duas lignanas furofurânicas (sesamina e kobusina), quatro lignanas dibenzilbutirolactônicas (hinokinina, kusunokinina, arctigenina e haplomirfolina), duas lignanas dibenzilbutirolactólicas (cubebina e 3,4- dimetoxi-3,4-desmetilenodioxicubebina), dois cinamatos de bornila (ferulato de bornila e cumarato de bornila) e na identificação de duas amidas (piplartina e diidropiplartina). Das folhas de P. richardiaefolium foi extraído e analisado o óleo volátil. As estruturas das substâncias isoladas foram identificadas através de métodos espectroscópicos (RMN de 1H e de 13C e espectrometria de massas). O estudo de análise de componentes principais (PCA) das espécies Piper (P. truncatum - k 616, P. richardiaefolium - k 290, P. richardiaefolium - k 350, P. richardiaefolium - k 593, P. truncatum - k 597, P. pseudopotifolium - k 598, P. richardiaefolium - k 854, P. richardiaefolium - k 610, P. truncatum - k 112, P. pseudopotifolium - k 211 e P. cernuum - k 137) permitiu agrupar as espécies em dois grandes grupos e quatro subgrupos em relação à similaridade entre elas. Ligninas do caule de seis espécies de Piper foram extraídas utilizando o método de degradação de Klason e método de Bjorkman, e analisadas por métodos espectroscópicos (IV, RMN de 1H e de 13C). O método de degradação por oxidação por nitrobenzeno foi o escolhido para determinar a relação entre os monolignóis siringila e guaiacila. Os principais metabólitos das espécies estudadas foram comparados com os tipos de ligninas das mesmas espécies e os resultados sugeriram uma independência entre as vias biossintéticas de ligninas e lignanas.
