Foi avaliado o efeito dos fungos contaminantes Trichoderma sp. e C. olivacearum na produtividade, eficiência biológica e número de cogumelos da produção do A. blazei em composto (mistura de cana-de-açúcar, palha de capim coast-cross, farelo de soja, gesso e calcário calcítico). O delineanamento foi inteiramente casualizado com três tratamentos (Trichoderma sp., C. olivacearum e testemunha) e oito repetições (caixa com 12 kg de composto colonizado com A. blazei). Após a colonização do composto pelo A. blazei, adicionou-se 150g de inóculo à base de triticale de cada um dos fungos contaminantes na superfície do composto seguido da camada de cobertura. O experimento foi conduzido em estufa com cobertura plástica, umidade relativa entre 60-90% e temperatura de 20-34ºC. A produtividade foi determinada pela relação entre a massa fresca de basidiomas e a massa úmida do composto. A eficiência biológica foi determinada pela relação entre a massa fresca de basidiomas e a massa seca do composto ao final do período de colheita. de acordo com os resultados obtidos, os fungos contaminantes C. olivacearum e Trichoderma sp. não afetaram a produtividade, eficiência biológica e número de cogumelos da produção do A. blazei em compostos previamente colonizados.

The effect of the contaminant fungi Trichoderma sp. and Chaetomium olivacearum on the cultivation of the ABL 99/30 and ABL 04/49 isolates of A. blazei in two compost formulations made up with tyfton (Cynodom dactylon) and oat (Avena sativa) was evaluated. The experimental design was a totally randomized 2 x 2 x 3 factorial design with 6 repetitions. The experimental unit consisted of 12-12.5 kg of wet compost. During the spawning, 150 g of Trichoderma sp. and C. olivacearum were added to the compost. The experiment was carried out in a climatized room, with humidity between 75-90% and temperature of 28º C. The productivity averages of the ABL 99/30 isolate of A. blazei were higher than those of ABL 04/49 and Trichoderma sp. and C. olivacearum negatively influenced the production of A. blazei. The different composts (based on tyfton and oat straw) did not influence the production of basidiomata.

Six new azaphilones, 5`-epichaetoviridin A (7), 4`-epichaetoviridin F (9), 12 beta-hydroxychaetoviridin C (10), and chaetoviridins G-I (11-13), and six known azaphilones, chaetoviridins A E (1-5) and 4`-epichaetoviridin A (8), were isolated from the endophytic fungus Chaetomium globosum cultivated in PDB medium for 21 days. The structure elucidation and the assignment of the relative configurations of the new natural products were based on detailed NMR and MS spectroscopic analyses. The structure of compound 1 was confirmed by single-crystal X-ray diffraction analysis. were determined using Mosher`s method. The antibiotic activity of the elegans infection model. The absolute configurations of compounds 4, 7, 8, and 12 compounds was evaluated using an in vivo Caenorhabditis elegans infection model.

Endophytes live in association with host plants during all or part of their life cycle without causing any apparent disease. They are considered outstanding and underexploited sources of novel bioactive compounds. Chaetomium globosum was isolated as an endophytic fungus from the healthy leaves of Viguiera robusta. C.globosum is a remarkable producer of chaetoglobosins, which are typically cytotoxic. In this work, chaetoglobosins B (1), D (2) and E (3) have been produced by the endophytic C. globosum strain. Chaetoglobosin B was evaluated against Jurkat (leukemia) and B16F10 (melanoma) tumoral cells and showed 89.55% and 57.10% of inhibition at 0.1 mg mL-1, respectively. Chaetoglobosin B also showed weak antibacterial activity against Staphylococcus aureus (MIC 120 µg/mL) and Escherichia coli (MIC 189 µg/mL).

O colapso do meloeiro (Cucumis melo), causado por Monosporascus cannonballus, é uma das principais enfermidades que acometem esta olerícola. O presente trabalho objetivou estudar o potencial do controle biológico de M. cannonballus com Chaetomium. Substrato infestado com 0,5; 1; 2; 4 e 8 x 10(5) UFC g-1 de Chaetomium foi colocado em bandejas, nas quais sementes de meloeiro do tipo Pele de Sapo cv. PS 1430 foram semeadas após duas semanas de incubação. As mudas obtidas foram transplantadas para vasos com substrato semelhante, sendo que neste momento o substrato foi infestado com mais 2,5 x 10(4) UFC g-1 de Chaetomium por vaso e 20 UFC g-1 de substrato de M. cannonballus. Três testemunhas foram utilizadas, uma infestada apenas com Chaetomium, outra somente com M. cannonballus e testemunha absoluta. O delineamento estatístico foi inteiramente casualizado com cinco repetições. As avaliações foram efetuadas aos 45 dias após o transplante, sendo calculado o índice geral de doença e o percentual de controle. O valor do índice geral de doença variou conforme o aumento da concentração de Chaetomium, sendo os menores valores encontrados para os tratamentos 4 e 8 x 10(5) UFC g-1de Chaetomium equivalentes a 0,9 e 0,6, respectivamente. A análise estatística demonstrou que os tratamentos correspondentes a 4 e 8 x 10(5) UFC g-1 de Chaetomium foram mais eficientes, diferindo dos demais, com percentual de controle superior a 55 %, evidenciado o potencial de Chaetomium no controle de M. cannonballus.

Three features of the heat shock response, reorganization of protein expression, intracellular accumulation of trehalose, and alteration in unsaturation degree of fatty acids were investigated in the thermophilic fungus Chaetomium thermophile and compared to the response displayed by a closely related mesophilic species, C. brasiliense. Thermophilic heat shock response paralleled the mesophilic response in many respects like (i) the temperature difference observed between normothermia and the upper limit of translational activity, (ii) the transient nature of the heat shock response at the level of protein expression including both the induction of heat shock proteins (HSPs) as well as the repression of housekeeping proteins, (iii) the presence of representatives of high-molecular-weight {HSPs} families, (iv) intracellular accumulation of trehalose, and finally (v) modifications in fatty acid composition. On the other hand, a great variability between the two organisms was observed for the proteins expressed during stress, in particular a protein of the {HSP60} family that was only observed in C. thermophile. This peptide was also present constitutively at normal temperature and may thus fulfil thermophilic functions. It is shown that accumulation of trehalose does not play a part in thermophily but is only a stress response. C. thermophile contains less polyunsaturated fatty acids at normal temperature than C. brasiliense, a fact that can be directly related to thermophily. When subjected to heat stress, both organisms tended to accumulate shorter and less unsaturated fatty acids.

The purpose of this study was the development and validation of an LC-MS-MS method for simultaneous analysis of ibuprofen (IBP), 2-hydroxyibuprofen (2-OH-IBP) enantiomers, and carboxyibuprofen (COOH-IBP) stereoisomers in fungi culture medium, to investigate the ability of some endophytic fungi to biotransform the chiral drug IBP into its metabolites. Resolution of IBP and the stereoisomers of its main metabolites was achieved by use of a Chiralpak AS-H column (150 x 4.6 mm, 5 mu m particle size), column temperature 8 degrees C, and the mobile phase hexane-isopropanol-trifluoroacetic acid (95: 5: 0.1, v/v) at a flow rate of 1.2 mL min(-1). Post-column infusion with 10 mmol L(-1) ammonium acetate in methanol at a flow rate of 0.3 mL min(-1) was performed to enhance MS detection (positive electrospray ionization). Liquid-liquid extraction was used for sample preparation with hexane-ethyl acetate (1:1, v/v) as extraction solvent. Linearity was obtained in the range 0.1-20 mu g mL(-1) for IBP, 0.05-7.5 mu g mL(-1) for each 2-OH-IBP enantiomer, and 0.025-5.0 mu g mL(-1) for each COOH-IBP stereoisomer (r >= 0.99). The coefficients of variation and relative errors obtained in precision and accuracy studies (within-day and between-day) were below 15%. The stability studies showed that the samples were stable (p > 0.05) during freeze and thaw cycles, short-term exposure to room temperature, storage at -20 degrees C, and biotransformation conditions. Among the six fungi studied, only the strains Nigrospora sphaerica (SS67) and Chaetomium globosum (VR10) biotransformed IBP enantioselectively, with greater formation of the metabolite (+)-(S)-2-OH-IBP. Formation of the COOH-IBP stereoisomers, which involves hydroxylation at C3 and further oxidation to form the carboxyl group, was not observed.

Diketopiperazine (DKP) derivatives, named colletopiperazine, fusaperazine C and E as well as four known DKPs were isolated from cultures of Colletotrichum gloeosporioides, Penicillium crustosum, both endophytic fungi isolated from Viguiera robusta, and a Fusarium spp., an endophyte of Viguiera arenaria, respectively. Their structures were established on the basis of their spectroscopic data. Conformational analysis of two known DKPs showed that folded conformations were as energetically stable as the extended one. (C) 2010 Elsevier Ltd. All rights reserved.

A study was conducted in the subtropical area of Southern Brazil to determine the survival of pathogens in soybean residues under conventional and no-tillage cultivation systems from March to September of 1998 and 1999. The pathogens most frequently isolated were Colletotrichum truncatum, Phomopsis spp., Cercospora kikuchii, Fusarium spp., Macrophomina phaseolina, and Rhizoctonia solani. Other fungi isolated were Myrothecium roridum, Penicillium sp., Chaetomium sp., Epicoccum sp., Corynespora cassiicola and Trichoderma sp. The percent of survival of each pathogen varied according to the month and the year. Survival of C. truncatum, Phomopsis spp. and C. kikuchii were significantly reduced (p<0.05) from the first to the last evaluation either on buried debris or maintained on the soil surface. On the other hand, M. phaseolina and Fusarium spp. were either not affected or favored by burying the debris. The frequency of recovery of Fusarium spp. increased specially in debris kept under the soil. The loss of biomass, measured by debris weight along the period of this study, showed a reduction of 44.4% in the conventional system and 34.9% in the no-tillage system in 1998, when rain was better distributed. In 1999, the reduction was 48.2% and 39.0% for the conventional and no-tillage system, respectively.

Em 2003 e 2004, os fungos Aspergillus spp., Chaetomium spp., Cladosporium sp., Fusarium spp., Paecilomyces sp., Penicillium spp., Pestalotiopsis sp., Phomopsis sp., Aureobasidium pullulans (de Bary) Arnaud e uma levedura não identificada foram isolados de sementes de mangaba (Hancornia speciosa Gomes) coletadas em três localidades do Cerrado do Brasil. Testes de patogenicidade e o subsequente reisolamento de Fusarium sp. (CPAC-1857), Pestalotiopsis sp (CPAC-1838 e CPAC-1844)., Phomopsis sp. (CPAC-1863) e de A. pullulans (CPAC-1845) de mudas de mangabeira inoculadas artificialmente comprovaram os postulados de Koch.

The enantioselective biotransformation of propranolol (Prop) by the endophytic fungi Phomopsis sp., Glomerella cingulata, Penicillium crustosum, Chaetomium globosum and Aspergillus fumigatus was investigated by studying the kinetics of the aromatic hydroxylation reaction with the formation of 4-hydroxypropranolol (4-OH-Prop). Both Prop enantiomers were consumed by the fungi in the biotransformation process, but the 4-hydroxylation reaction yielded preferentially (-)-(S)-4-OH-Prop. The quantity of metabolites biosynthesized varied slightly among the evaluated endophytic fungi. These results show that all investigated endophytic fungi could be used as biosynthetic tools in biotransformation processes to obtain the enantiomers of 4-OH-Prop.

Este trabalho teve como objetivo avaliar a qualidade sanitária de sementes de aveia-preta (Avena strigosa Schreb.) produzidas no Estado do Rio Grande do Sul. Foram avaliados 81 lotes de sementes oriundos de 16 municípios de diferentes regiões fisiográficas. Utilizou-se o método do substrato papel, com 400 sementes por lote. Foram detectados os fungos Alternaria sp., Chaetomium sp.,Cladosporium sp., Colletotrichum sp., Curvularia sp., Epicoccum sp., Nigrospora sp., Colletotrichum sp., Fusarium sp. e os fungos de armazenamento Aspergillus sp.e Penicillium sp.A maior incidência média foi dos fungos Phoma sp.e Bipolaris spp., com 18,4 e 22,4% respectivamente. Pode-se concluir que deve haver um melhor manejo da cultura da aveia-preta, visando diminuir a contaminação das sementes por fungos, principalmente, nas regiões típicas produtoras de cereais de inverno.

In a glasshouse experiment using potted strawberry plants (cv. Cambridge Favourite) as hosts, the effect of selected fungal antagonists grown on 25 or 50 g of mushroom compost containing autoclaved mycelia of Agaricus bisporus, or wheat bran was evaluated against Armillaria mellea. Another glasshouse experiment tested the effect of application time of the antagonists in relation to inoculations with the pathogen. A significant interaction was found between the antagonists, substrates and dose rates. All the plants treated with Chaetomium olivaceum isolate Co on 50 g wheat bran survived until the end of the experiment which lasted 482 days, while none of them survived when this antagonist was added to the roots of the plants on 25 g wheat bran or 25 or 50 g mushroom compost. Dactylium dendroides isolate SP had a similar effect, although with a lower host survival rate of 33.3%. Trichoderma hamatum isolate Tham 1 and T. harzianum isolate Th23 protected 33.3% of the plants when added on 50 g and none when added on 25 g of either substrate, while 66.7% of the plants treated with T. harzianum isolate Th2 on 25 g, or T viride isolate TO on 50 g wheat bran, survived. Application of the antagonists on mushroom compost initially resulted in development of more leaves and healthier plants, but this effect was not sustained. Eventually, plants treated with the antagonists on wheat bran had significantly more leaves and higher health scores. The plants treated with isolate Th2 and inoculated with Armillaria at the same time had a survival rate of 66.7% for the duration of the experiment (475 days), while none of them survived that long when the antagonist and pathogen were applied with an interval of 85 days in either sequence. C. olivaceum isolate Co showed a protective effect only, as 66.7% of the plants survived when they were treated with the antagonist 85 days before inoculation with the pathogen, while none of them survived when the antagonist and pathogen were applied together or the infection preceded protection.
