The molecular and ultrastructural profiles of the symbionts found in the midgut and ileum of Cephalotes atratus, Cephalotes clypeatus, and Cephalotes pusillus were determined using the V3 region of the bacterial 16S rDNA gene and transmission electron microscopy (T.E.M.). Two samples of C atratus, three of C clypeatus, and six of C. pusillus were analyzed. The coefficients of similarity ranged from 80% to 94% for the samples of symbionts from C. clypeatus and C. atratus, despite being collected in geographically distant sites. The variability within symbionts found in the samples of C. pusillus varied from 29% to 55%, in samples geographically close as well as distant. PCR-DGGE was effective for the purpose of this study and can be considered a versatile tool to analyze gut microbiota. Details of the ultrastructural aspect of these bacteria are presented. Ants in the tribe Cephalotini are exceptional in that they maintain microorganisms in their digestive tract. To understand what these microorganisms mean to the ants, we observed the feeding habits of Cephalotes pusillus and Cephalotes atratus, finding that in nature they feed on extrafloral nectars, homopteran secretions, and bird droppings. Feeding the antibiotic kanamycin to colonies of C. pusillus in the laboratory kills them. Ants desiccate or starve rather than feed on liquids to which the antibiotics gentamycin and netilmycin have been added, but feed and survive on liquids containing nystatin, penicillin, and ampicillin. We identified over 10 microorganisms from the intestine of C. pusillus with different antibiotic-resistance patterns. The bacteria are from the genera Corynebacterium, Brevibacterium, Sphingobacterium, Ochrobactrum, Myroides, Brevundimonas, Alcaligenes, Stenotrophomonas, Moraxella, and Pseudomonas. We hypothesize that the microorganisms provide nutrients to the ants by synthesizing amino acids from carbohydrates and nitrates. We do not know whether the ants collect the bacteria from the environment, but they transmit them to their young. They culture them in their digestive tract, eventually feeding on them.

The histology (H.E.), histochemistry (carbohydrates, proteins, collagen, and calcium), enzyme profile (ATPase and Acid phosphatase), and ultramorphology of the ileum of Cephalotes atratus, C. clypeatus and C. pusillus were studied in order to obtain some information about the similarities of these species. The relationship among these three species, as well as the results obtained for the wall, and the contents of this portion of the digestive tract were discussed. In C. clypeatus the ileum is somewhat smaller than that of C. atratus, despite the similarities in body size. The ATPase results showed 2 different regions: the anterior region responsible for absorbing material from the haemolymph and the posterior region for absorbtion from the ileum lumen. Other histochemical analyses failed to show any differences.

This study aimed at evaluating the effect of collect time (day and night) on ant fauna attracted to baits in areas of Eucalyptus cloeziana (Myrtaceae) and cerrado. The ants were collected in Fazenda Boa Vista, Mannesmann Fi-El Florestal Ltda, Paineiras, Minas Gerais State. Eigthteen sample units were collected: 12 in E. cloezina and six in cerrado. Each sample unit consisted of three plots (25 x 35 m each). The plot consisted of 34 baits distributed in a grid pattern at 5 m intervals. The sampling was carried out in the diurnal and nocturnal period. The results obtained revealed that both type of vegetation (cerrado x Eucalyptus) and the collect time (day x night) had a significative influence on the epigaeic ant fauna. The ordination (DCA) indicated that collect time effect was more important to fauna structuration than the vegetation effect. Brachymyrmex sp.1, Brachymyrmex sp.2, Camponotus crassus Mayr, Camponotus rufipes (Fabricius), Cephalotes pusillus (Klug) and Ectatomma brunneum Smith were indicator species of nocturnal period, and Camponotus renggeri Emery, Camponotus atriceps (Smith), Camponotus melanoticus Emery and Paratrechina sp.1 were indicators of nocturnal period.

The ultrastructural analysis of the midgut of Cephalotes atratus. C. clypeatus, and C. pusillus reveled that the midgut epithelium lays on a basal lamina and is composed basically of three cell types: digestive cells, regenerative cells, and goblet cells. In these ants, the rough endoplasmic reticulum, in addition to producing digestive enzymes, is involved in the formation of concretions and ion storage in specialized vacuoles present in the midgut. These concretions are spherocrystals and may contribute to stabilize the pH and to maintain symbiotic bacteria found between microvilli. The ultrastructure analysis of these bacteria revealed the presence of a double envelope typical of gram-negative bacteria. For the three species examined, the ultrastructure similarities are conspicuous, suggesting that this may be the pattern for the genus Cephalotes. Studies of histological nature, histochemistry and ultra-morphology of the midgut of three species of ants belonging to the genus Cephalotes (atratus, clypeatus and pusillus) were performed. In histology H-E was employed, and in histochemistry techniques for detection of carbohydrates, proteins in general and collagen and calcium fibers were used. The results obtained from this second technique made possible some discussion about the pH of the midgut of these insects. The species are identical in the characters under study. The digestive cells exhibited a secretion that is typical of ants, the apocrine type.

As formigas do gênero Cephalotes, rapidamente identificadas por suas operárias polimórficas de cutícula resistente e cabeça achatada possuem seu sucesso ecológico creditado à sua dieta predominantemente generalista e nidificação em cavidades pré-existentes de troncos de árvores. Possuem hábitos exclusivamente arborícolas e ocorrem nos trópicos e subtrópicos do Novo Mundo possuindo ampla distribuição geográfica. O grupo apresenta uma interessante associação com microorganismos. No presente trabalho foi feita a caracterização molecular e estudo de relações filogenéticas do gênero através da amplificação e sequenciamento de fragmento do gene 28S do DNA Nuclear de três populações localizadas em Rio Claro-SP e São José do Rio Preto-SP de duas espécies de Cephalotes: C. pusillus e C. clypeatus. Também foi feito o levantamento da ocorrência e frequência do endossimbionte Wolbachia em sete populações de Cephalotes localizadas em São José do Rio Preto-SP, Guaraci-SP, São Carlos-SP, Araraquara-SP, Delfinópolis-MG e Rio Claro-SP; abrangendo três espécies: C. pusillus, C. clypeatus e C. atratus. Esse levantamento foi realizado com a utilização de ferramentas moleculares para a análise do gene codificador da proteína de superfície de membrana do endossimbionte, o wsp. Para analises de filogenia e também do endossimbionte, foi realizada a extração do DNA total de operárias, a amplificação do gene através da técnica de PCR utilizando os primers já estabelecidos e em seguida, as amostras foram sequenciadas pelo método de Sanger. Os resultados obtidos mostraram relações monofiléticas dentro da subfamília Myrmicinae, a qual pertence o gênero Cephalotes. As análises do gene 28S trouxeram resultados otimistas no estudo da caracterização molecular do grupo. Os resultados da analise do endossimbionte corroboraram com estudos anteriores com outras espécies do gênero Cephalotes, onde ocorreu alta...

We developed a suitable diet for mass rearing of Cryptolestes ferrugineus (Stephens) populations under laboratory conditions. Recently, this pest has developed strong level of resistance to phosphine in Australia, and therefore, a significant amount of research has been directed towards its management. In total, nineteen grain-based diets, containing rolled oats, various combinations of cracked grains and flours of wheat, sorghum, maize and barley were tested. Each diet contained a small proportion of wheat germ (4.5% w/w) and torula yeast (0.5% w/w). Experiments were conducted at fixed temperature and relative humidity regimes of 30 ± 2 °C and 70 ± 2%, respectively, and replicated three times. Adults (n = 40) of a laboratory strain of C. ferrugineus were introduced into each diet, removed after 14 days and total numbers of live adult progeny were recorded. The following diets resulted in highest live progeny production: barley flour (95%) (607.67 ± 11.21) = rolled oats (75%) + cracked sorghum (20%) (597.33 ± 33.79) ≥ wheat flour (47.5%) + barley flour (47.5%) (496.67 ± 52.93) > cracked sorghum (95%) (384.00 ± 60.66). The performance of these four diets was then tested with field-collected populations of C. ferrugineus and Cryptolestes pusillus (Schonherr). The diets based on rolled oats + cracked sorghum, wheat flour + barley flour, and barley flour alone consistently produced highest progeny numbers in field-collected populations of both species, with mean progeny numbers ranging from 359.9 to 478.5. The multiplication of C. pusillus was significantly higher than C. ferrugineus on all four diets. Our findings will help in mass rearing of healthy cultures of C. ferrugineus and C. pusillus that will greatly facilitate laboratory and field research and in particular, in developing management tactics for these species.

South African (Cape) fur seals, Arctocephalus pusillus pusillus, interact with the South African trawl fisheries-offshore demersal, inshore demersal, and midwater fisheries. These interactions take thef ollowing forms: Seals take or damage netted fish, on particular vessels they become caught in the propeller, seals drown in the nets, live seals come aboard and may be killed. Except in specific cases of seals damaging particular trawler propellers, interactions result in little cost to the offshore and midwater trawl fisheries. For the inshore fishery, seals damage fish in the net at an estimated cost in excess of R69, 728 (US$18,827) per year, but this is negligible (0.3%) in terms ofthe value of the fishery. Seal mortality is mainly caused by drowning in trawl nets and ranges from 2,524 to 3,636 seals of both sexes per year. Between 312 and 567 seals are deliberately killed annually, but this most likely takes place only when caught and they enter the area below deck, where they are difficult to remove, and pose a potential threat to crew safety. Overall, seal mortality during trawling operations is negligible (0.4-0.6%) in terms of the feeding population of seals in South Africa.
