The phytoextraction process implies the use of plants to promote the elimination of metal contaminants in the soil. In fact, metal-accumulating plants are planted or transplanted in metal-contaminated soil and cultivated in accordance with established agricultural practices. The objective of the present study was to evaluate the productivity and Cd phytoextraction capacity of white lupine ( Lupinus albus L.) and narrow-leafed lupine ( Lupinus angustifolius L.), as well as the effect on residual Cd concentration in the soil. Both species of lupines were grown at three CdCl2 rates (0, 1, and 2 mg kg-1), under three agroclimatic conditions in Chile in 2013. In the arid zone (Pan de Azúcar, 73 mm precipitation), narrow-leafed lupine production was significantly (P < 0.05) higher than white lupine (4.55 vs. 3.26 Mg DM ha-1, respectively). In locations with higher precipitation (Santa Rosa, 670 mm; Carillanca, 880 mm), narrow-leafed lupine DM production was slightly higher than in Pan de Azúcar, but white lupine was approximately three times higher. Total plant Cd concentrations in white and narrow-leafed lupine increased as Cd rates increased in the three environments, but they were much higher in narrow-leafed lupine than white lupine; 150%, 58%, and 344% higher in Pan de Azúcar, Santa Rosa, and Carillanca, respectively. Cadmium uptake (g Cd ha-1) and apparent recovery were also higher (P < 0.05) in narrow-leafed lupine in two environments (Pan de Azúcar and Carillanca). These results suggest that narrow-leafed lupine present higher potential as phytoremediation species than white lupine.

Obtaining correctly folded proteins from inclusion bodies of recombinant proteins expressed in bacterial hosts requires solubilization with denaturants and a refolding step. Aggregation competes with the second step. Refolding of eight different proteins was carried out by precipitation with smart polymers. These proteins have different molecular weights, different number of disulfide bridges and some of these are known to be highly prone to aggregation. A high throughput refolding screen based upon fluorescence emission maximum around 340 nm (for correctly folded proteins) was developed to identify the suitable smart polymer. The proteins could be dissociated and recovered after the refolding step. The refolding could be scaled up and high refolding yields in the range of 8 mg L-1 (for CD4D12, the first two domains of human CD4) to 58 mg L-1 (for malETrx, thioredoxin fused with signal peptide of maltose binding protein) were obtained. Dynamic light scattering (DLS) showed that polymer if chosen correctly acted as a pseuclochaperonin and bound to the proteins. It also showed that the time for maximum binding was about 50 min which coincided with the time required for incubation (with the polymer) before precipitation for maximum recovery of folded proteins. The refolded proteins were characterized by fluorescence emission spectra, circular dichroism (CD) spectroscopy, melting temperature (T-m), and surface hydrophobicity measurement by ANS (8-anilinol-naphthalene sulfonic acid) fluorescence. Biological activity assay for thioredoxin and fluorescence based assay in case of maltose binding protein (MBP) were also carried out to confirm correct refolding. (C) 2012 Elsevier B.V. Städtische Biomassen der Grünflächen bilden eine potentielle, bisher weitgehend ungenutzte Ressource für Bioenergie. Kommunen pflegen die Grünflächen, lassen das Material aber verrotten oder führen es Deponien oder Müllverbrennungsanlagen zu. Diese Praxis ist kostenintensiv ohne für die Verwaltungen finanziellen Ausgleich bereitzustellen. Stattdessen könnte das Material energetisch verwertet werden. Zwei mögliche Techniken, um Bioenergie zu gewinnen, wurden mit krautigem Material des städtischen Straßenbegleitgrüns untersucht i) direkte anaerobe Fermentation (4 Schnitte im Jahr) und ii) „Integrierte Festbrennstoff- und Biogasproduktion aus Biomasse“ (IFBB), die Biomasse durch Maischen und mechanisches Entwässern in einen Presssaft und einen Presskuchen trennt (2 Schnitte im Jahr). Als Referenz wurde die aktuelle Pflege ohne Verwertungsoption mitgeführt (8faches Mulchen). Zusätzlich wurde die Eignung von Gras-Laub-Mischungen im IFBB-Verfahren untersucht. Der mittlere Biomasseertrag war 3.24, 3.33 und 5.68 t Trockenmasse ha-1 jeweils für die Pflegeintensitäten Mulchen, 4-Schnitt- und 2-Schnittnutzung. Obwohl die Faserkonzentration in der Biomasse der 2-Schnittnutzung höher war als im Material der 4-Schnittnutzung, unterschieden sich die Methanausbeuten nicht signifikant. Der Presskuchen aus dem krautigen Material des Straßenbegleitgrüns hatte einen Heizwert von 16 MJ kg-1 Trockenmasse, während der Heizwert des Presskuchens der Gras-Laub-Mischung in Abhängigkeit vom Aschegehalt zwischen 15 und 17 MJ kg-1 Trockenmasse lag. Der Aschegehalt der Mischungen war höher als der Grenzwert nach DIN EN 14961-6:2012 (für nicht-holzige Brennstoffe), was auf erhöhte Bodenanhaftung auf Grund der Erntemethoden zurückzuführen sein könnte. Der Aschegehalt des krautigen Materials vom Straßenrand hielt die Norm jedoch ein. Die Elementkonzentration (Ca, Cl, K, Mg, N, Na, P, S, Al, Cd, Cr, Cu, Mn, Pb, Si, Zn) im krautigen Material war generell ähnlich zu Landwirtschafts- oder Naturschutzgrünland. In den Mischungen nahm die Elementkonzentration (Al, Cl, K, N, Na, P, S, Si) mit zunehmendem Laubanteil ab. Die Konzentration von Ca, Mg und der Neutral-Detergenz-Fasern stieg hingegen an. Die IFBB-Technik reduzierte die Konzentrationen der in der Verbrennung besonders schädlichen Elemente Cl, K und N zuverlässig. Außer den potentiell hohen Aschegehalten, wurde während der Untersuchungen kein technischer Grund entdeckt, der einer energetischen Verwertung des getesteten urbanen Materials entgegenstehen würde. Ökonomische, soziale und ökologische Auswirkungen einer Umsetzung müssen beachtet werden. Eine oberflächliche Betrachtung auf Basis des bisherigen Wissens lässt hoffen, dass eine bioenergetische Verwertung städtischen Materials auf allen Ebenen nachhaltig sein könnte.

Stereotypies in captive animals typically occur under conditions that are stressful for the animals, and there is some anecdotal evidence that stress levels during early stereotypy development predict later stereotypy levels. Based on this and on the involvement of stress in the behavioural sensitization to psychostimulant drugs, it has been hypothesized that stereotypy development might be causally related to stress. To address this question further, we used mice of the commercial outbred stock CD-1 (ICR) and mice of two lines derived from the outbred CD-1 (ICR) strain by selective breeding for high (HR) and low (LR) stress reactivity, respectively, and examined whether genetically driven variation in stress reactivity is associated with variation in the expression of cage-induced stereotypies. From 21 days of age, 10 females of each line were housed in pairs under standard laboratory conditions until they were video recorded for stereotypic behaviour and tested for corticosterone responses in a stress reactivity test (SRT) at 12 weeks of age. As expected, HR females showed a significantly stronger corticosterone response in the SRT than LR females, while ICR females were intermediate. Unexpectedly, however, both HR and LR females showed very low levels of stereotypic behaviour, while ICR females developed the high levels of stereotypies typical for this strain of mouse. Consequently, there was no significant relationship between measures of acute corticosterone reactivity and stereotypy performance, but a trend for reduced recovery of the corticosterone response in the ICR line suggests that variation in recovery rather than the acute response might predict stereotypy levels in these mice. The development of strategies and policies aiming at the reduction of environmental exposure to air pollution requires the assessment of historical emissions. Although anthropogenic emissions from the extended territory of the Soviet Union (SU) considerably influenced concentrations of heavy metals in the Northern Hemisphere, Pb is the only metal with long-term historical emission estimates for this region available, whereas for selected other metals only single values exist. Here we present the first study assessing long-term Cd, Cu, Sb, and Zn emissions in the SU during the period 1935–1991 based on ice-core concentration records from Belukha glacier in the Siberian Altai and emission data from 12 regions in the SU for the year 1980. We show that Zn primarily emitted from the Zn production in Ust-Kamenogorsk (East Kazakhstan) dominated the SU heavy metal emission. Cd, Sb, Zn (Cu) emissions increased between 1935 and the 1970s (1980s) due to expanded non-ferrous metal production. Emissions of the four metals in the beginning of the 1990s were as low as in the 1950s, which we attribute to the economic downturn in industry, changes in technology for an increasing metal recovery from ores, the replacement of coal and oil by gas, and air pollution control.

Produced water is a major problem associated with the crude oil extraction activity. The monitoring of the levels of metals in the waste is constant and requires the use of sensitive analytical techniques. However, the determination of trace elements can often require a pre-concentration step. The objective of this study was to develop a simple and rapid analytical method for the extraction and pre-concentration based on extraction phenomenon cloud point for the determination of Cd, Pb and Tl in produced water samples by spectrometry of high resolution Absorption source continues and atomization graphite furnace. The Box Behnken design was used to obtain the optimal condition of extraction of analytes. The factors were evaluated: concentration of complexing agent (o,o-dietilditilfosfato ammonium, DDTP), the concentration of hydrochloric acid and concentration of surfactant (Triton X -114). The optimal condition obtained through extraction was: 0,6% m v-1 DDTP, HCl 0,3 mol L-1 and 0,2% m v-1 of Triton X - 114 for Pb; 0,7% m v-1 DDTP, HCl 0,8 mol L-1 and 0,2% m v-1 Triton X-114 for Cd. For Tl was evidenced that best extraction condition occurs with no DDTP, the extraction conditions were HCl 1,0 mol L-1 e 1,0% m v-1 de Triton X - 114. The limits of detection for the proposed method were 0,005 µg L-1 , 0,03 µg L-1 and 0,09 µg L-1 to Cd, Pb and Tl, Respectively. Enrichment factors Were greater than 10 times. The method was applied to the water produced in the Potiguar basin, and addition and recovery tests were performed, and values were between 81% and 120%. The precision was expressed with relative standard deviation (RSD) is less than 5%

The determination and monitoring of metallic contaminants in water is a task that must be continuous, leading to the importance of the development, modification and optimization of analytical methodologies capab le of determining the various metal contaminants in natural environments, because, in many cases, the ava ilable instrumentation does not provide enough sensibility for the determination of trace values . In this study, a method of extraction and pre- concentration using a microemulsion system with in the Winsor II equilibrium was tested and optimized for the determination of Co, Cd, P b, Tl, Cu and Ni through the technique of high- resolution atomic absorption spectrometry using a continuum source (HR-CS AAS). The optimization of the temperature program for the graphite furnace (HR-CS AAS GF) was performed through the pyrolysis and atomization curves for the analytes Cd, Pb, Co and Tl with and without the use of different chemical modifiers. Cu and Ni we re analyzed by flame atomization (HR-CS F AAS) after pre-concentr ation, having the sample introduction system optimized for the realization of discrete sampling. Salinity and pH levels were also analyzed as influencing factors in the efficiency of the extraction. As final numbers, 6 g L -1 of Na (as NaCl) and 1% of HNO 3 (v/v) were defined. For the determination of the optimum extraction point, a centroid-simplex statistical plan was a pplied, having chosen as the optimum points of extraction for all of the analytes, the follo wing proportions: 70% aqueous phase, 10% oil phase and 20% co-surfactant/surfactant (C/S = 4). After extraction, the metals were determined and the merit figures obtained for the proposed method were: LOD 0,09, 0,01, 0,06, 0,05, 0,6 and 1,5 μg L -1 for Pb, Cd, Tl, Co, Cu and Ni, re spectively. Line ar ranges of ,1- 2,0 μg L -1 for Pb, 0,01-2,0 μg L -1 for Cd, 1,0 - 20 μg L -1 for Tl, 0,1-5,0 μg L -1 for Co, 2-200 μg L -1 and for Cu e Ni 5-200 μg L -1 were obtained. The enrichment factors obtained ranged between 6 and 19. Recovery testing with the certified sample show ed recovery values (n = 3, certified values) after extraction of 105 and 101, 100 and 104% for Pb, Cd, Cu and Ni respectively. Samples of sweet waters of lake Jiqui, saline water from Potengi river and water produced from the oil industry (PETROBRAS) were spiked and the recovery (n = 3) for the analytes were between 80 and 112% confirming th at the proposed method can be used in the extraction. The proposed method enabled the sepa ration of metals from complex matrices, and with good pre-concentration factor, consistent with the MPV (allowed limits) compared to CONAMA Resolution No. 357/2005 which regulat es the quality of fresh surface water, brackish and saline water in Brazil.
