O câncer colorretal é um dos tumores humanos mais freqüentes e a terceira causa de morte relacionada ao câncer no mundo. Apesar de importantes progressos terapêuticos, os resultados na doença avançada ainda são muito modestos. Isto deve-se ao fato de que a droga mais utilizada nesta neoplasia, o antimetabólito 5-fluorouracil (5-FU), foi desenvolvido a mais de 40 anos produzindo taxas de respostas de somente 10-15%. Recentemente, o inibidor da topoisomerase I irinotecan (CPT-11) demonstrou, em carcinoma de cólon avançado, respostas comparáveis tanto em pacientes não tratados quanto naqueles que tiveram recaída após terapia com 5-FU. Estes resultados justificam a avaliação da combinação 5-FU/CPT-11 nesta doença. Apesar das respostas dos estudos clínicos serem promissoras, a melhor seqüência de administração destes agentes ainda não foi determinada. Neste estudo, avaliamos a combinação CPT-11/5-FU quanto ao aumento da inibição do crescimento celular quando comparada com os agentes sozinhos nas linhagens celulares de carcinoma de cólon humano SW620, HT-29 e SNU-C4. Para isto, as células foram expostas às drogas sozinhas ou a várias combinações e seqüências de dose baixa e fixa (IC20) de um dos agentes na presença de dose alta e seriada do outro. As células foram avaliadas imediatamente após os tratamentos e/ou cultivadas por mais 2 dias em meio de cultura sem drogas através de coloração com sulforodamina B. As interações entre CPT-11 e 5-FU foram avaliadas por um programa de computador que permite calcular os índices de combinação (CIs) das drogas indicando sinergismo, adição, ou antagonismo (CI < 1, = 1, ou > 1, respectivamente). As respostas celulares foram relacionadas com as atividades das enzimas timidilato sintase, topoisomerase I e carboxil esterase, que foram determinadas através de ensaio que mede os sítios de ligação e atividade catalítica da enzima, ensaio de decatenação do DNA e método espectrofotométrico, respectivamente. Estando a toxicidade no DNA envolvida no mecanismo de ação das duas drogas, também relacionamos as respostas celulares com a introdução de danos ao DNA por método fluorescente. Para melhor entendermos as interações entre as drogas, examinamos os efeitos da exposição à IC20, IC50 e/ou IC80 do CPT-11 ou 5-FU por 2 h ou 24 h em alvos celulares possivelmente relacionados com a citotoxicidade destes agentes. Estes incluem: capacidade de reparo por excisão do 10 DNA, distribuição das células nas fases do ciclo celular, integridade da membrana plasmática e formação de complexos DNA-topoisomerase I. Para isto, utilizamos método de incorporação de [3H-metil]timidina, citometria de fluxo, liberação de lactato desidrogenase (LDH) no meio de cultura e ensaio de precipitação com SDS, respectivamente. Os estudos de inibição do crescimento celular revelaram valores de IC50 do 5-FU nas linhagens SW620, HT-29 e SNU-C4 de aproximadamente 15, 8 e 2 µM, respectivamente, e do CPT-11 próximos a 2, 2 e 4 µM, respectivamente. As diferentes sensibilidades ao 5-FU nas três linhagens foram determinadas principalmente pela diferença na afinidade ao substrato. As respostas comparáveis obtidas pela exposição ao CPT-11 podem ser explicadas pelo equilíbrio entre as diferentes atividades das enzimas topoisomerase I e carboxil esterase, entre as linhagens. O programa de análise da combinação das drogas mostrou adição ou sinergismo após exposição a IC20 do CPT-11 seguido do 5-FU, nas três linhagens celulares. Por outro lado, o pré-tratamento com IC20 do 5-FU antagonizou a inibição do crescimento mediada pelo CPT-11. Nenhum dos tratamentos simultâneos determinou um aumento na inibição do crescimento nas linhagens SW620 e HT-29; mas mostraram adição ou antagonismo na linhagem SNU-C4. Observamos um significativo acréscimo na introdução de danos ao DNA nas linhagens celulares SW620 e HT-29 somente quando a IC20 do CPT-11 precedeu a IC50 do 5-FU. Já na linhagem SNU-C4, não somente este tratamento, mas também a utilização simultânea dos agentes introduziu mais dano ao DNA. Os tratamentos por 2 h ou 24 h com IC20, IC50, e/ou IC80 do 5-FU ou CPT-11 não causaram mudanças significativas na distribuição das células nas fases do ciclo celular, concentração de LDH no meio de cultura, ou formação de complexos DNA-topoisomerse I. Indicando que alterações nestes processos não estão envolvidas com os efeitos modulatórios dos pré-tratamentos na citotoxicidade das drogas. Entretanto, a incorporação de [3H-metil]timidina no DNA de células tratadas com CPT-11, aumentou em função da dose e tempo de exposição à droga. Este resultado sugere que o dano ao DNA introduzido pelo CPT-11 depende da dose e tempo de exposição podendo ser reparado por mecanismos de excisão. Somente foi observada significativa incorporação de [3H-metil]timidina em células tratadas com IC20 do 5-FU por 2 h, sugerindo que o reparo por excisão só ocorre após exposição por curto período e à doses baixas deste agente. Juntos, os resultados deste estudo mostraram que tanto os efeitos anti-proliferativos quanto a introdução de danos ao DNA pela combinação CPT-11/5-FU em dose baixa e fixa de um agente junto com dose alta do outro, nas linhagens celulares de carcinoma de cólon humano SW620, HT-29 e SNU-C4, dependem da seqüência de administração das drogas. Estes achados podem ter sido determinados pelos diferentes efeitos do tratamento com uma dose baixa dos agentes nos mecanismos de reparo por excisão do DNA.

A autora discute o tratamento jurídico-penal dispensado aos embriões humanos excedentes das técnicas de reprodução assistida, previsto hoje no Brasil principalmente pela Lei de Biossegurança, de 2005, que dispõe sobre os crimes relacionados à proteção do genoma humano. Para ela, a questão tende a se tornar cada vez mais pertinente devido ao vertiginoso crescimento da engenharia genética e a multiplicação dos embriões in vitro mantidos estocados em laboratórios. No trabalho, defende que, embora a existência de vida no embrião extrauterino não tenha sido comprovada, não se pode negar a ele o atributo da dignidade humana, porque seria portador do conjunto de genes que formam o patrimônio genético da humanidade, conferindo especial identidade à espécie. Assim, a autora, se vê com bons olhos o fato de o Direito Penal, como um subsistema do ordenamento jurídico do país, emprestar formas especiais de tutela a este novo bem jurídico e alerta que pode estar faltando, ao mesmo tempo, maior abrangência e focos mais específicos para os riscos de lesão ou destruição dos genomas. Ela também julga indispensável que as novas tecnologias genéticas que vêm surgindo sejam sempre tratadas e refletidas na esfera jurídico-penal

Synthesis and spectroscopic properties of seven new dibutyltin(IV) compounds of 2-{(E)-4-hydroxy-3-[(E)-4-(aryl)iminomethyl]phenyldiazenyl}benzoic acids (L(n)HH'; n=2-8) with general formula {[Bu2Sn(L(n)H)]2O}2 (1-7) are reported. The compounds were characterized by elemental analysis and by UV-Visible, fluorescence, IR, (1)H, (13)C and (119)Sn NMR spectroscopies. Solid state structures of dibutyltin(IV) compounds 1-3, 6 and 7 were accomplished from single crystal X-ray crystallography which reveal the common ladder-type structure with two endo- and two exo-Sn atoms. The redox properties of L(n)HH' (n=2-4, 7 and 8) and their diorganotin(IV) compounds 1-3, 6 and 7 were also investigated by cyclic voltammetry. In general, the dibutyltin(IV) derivatives exhibited significant in vitro cytotoxic potency towards A375 (melanoma) and HCT116 (colon carcinoma) cell lines as determined by several experiments, like Live and Dead assay, MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) cell viability assay, LDH (lactate dehydrogenase), cleavage of caspases and PARP (poly(ADP-ribose)polymerase), and DNA fragmentation. Dibutyltin(IV) compounds increase cell death without cytolysis and decreases membrane fluidity, without interfering with p53. Among the dibutyltin(IV) compounds, compound 6 was found to be the most potent, with an IC50 value of 78nM. A mechanism of action for tumor cell death is proposed.

This thesis describes investigations upon pseudopeptides which were conducted to improve our understanding of the fate of synthetic macromolecules in cells and to develop approaches to influence that fate. The low uptake of molecules across the external cellular membrane is the principal barrier against effective delivery of therapeutic products to within the cell structure. In nature, disruption of this membrane by amphiphilic peptides plays a central role in the pathogenesis by bacterial and toxin infections. These amphiphilic peptides contain both hydrophobic and weakly charged hydrophilic amino acid residues and upon activation they become integrated into the lipid bilayers of the extracellular or endosomal membranes. The architectures of the pseudopeptides described here were designed to display similar pH dependent membrane rupturing activity to that of peptides derived from the influenza virus hemagglutinin HA-2. This HA protein promotes fusion of the influenza virus envelope with the cell endosome membrane due to a change in conformation in response to the acidic pH of the endosome lumen (pH 5.0-6.0). The pseudopeptides were obtained by the copolymerisation of L-lysine and L-lysine ethyl-ester with various dicarboxylic acid moieties. In this way a linear polyamide comprising of alternating pendant carboxylic acids and pendant hydrophobic moieties was made. At physiological pH (pH 7.4), electrostatic repulsion of pendant anionic carboxyl groups along the polymer backbone is sufficient to overcome the intramolecular association of the hydrophobic groups resulting in an extended conformation. At low pH (typically pH 4.8) loss of charge results in increased intramolecular hydrophobic association and the polymer chain collapses to a compact conformation, leading to precipitation of the polymer. Consequently, a conformation dependent functional property could be made to respond to small changes in the environmental pH. Pseudopepides were investigated for their cytoxicity towards a well known cell line, namely C26 (colorectal adenocarcinoma) and were shown through the use of a cell viability assay, MTT (3-(4,5-dimethylthiazol-2-yl)-2,5 diphenyltetrazolium bromide) to be well tolerated by C26 cells over a range of concentrations (2-500,μg/ml) at physiological pH (pH 7.4). A modified version of a shorter 30-minute coupled enzymatic assay, the LDH (lactate dehydrogenase) assay was used to evaluate the ability of the pseudopeptides to disrupt the membrane of two different cell lines (COS-1; African green monkey, kidney and A2780; human ovarian carcinoma) at low pH (pH 5.5). The cell membrane disruption property of the pseudopeptides was successfully demonstrated for COS-I and A2780 cell lines at this pH (pH 5.5). A variety of cell lines were chosen owing to limited availability and to compare the cytotoxic action of these pH responsive psudopeptides towards normal and tumorogenic cell lines. To investigate the intracellular delivery of one of the pseudopeptides, poly (L-lysine iso-phthalamide) and its subcellular location, a Cy3 bisamine fluorophore was conjugated into its backbone, at ratios of dye:lysine of 1:20, 1:30, 1:40, 1:60 and 1:80. Native polyacrylacrylamide gel electrophoresis (PAGE) and high voltage paper electrophoresis (HVPE) studies of the polydyes were conducted and provided evidence that that the Cy3 bisamine fluorophore was conjugated into the backbone of the polymer, poly (L-lysine iso-phthalamide). The subcellular fate of the fluorescentlylabelled "polydye" (hereafter PD20) was monitored by laser scanning confocal microscopy (LSCM) in CHO (Chinese hamster ovary) cells cultured in-vitro at various pH values (pH 7.4 and 5.0). LSCM images depicting time-dependent internalisation of PD20 indicated that PD20 traversed the extracellular membrane of CHO cells cultured in-vitro within ten minutes and migrated towards the endosomal regions where the pH is in the region of 5.0 to 6.0. Nuclear localisation of PD20 was demonstrated in a subpopulation of CHO cells. A further study was completed in CHO and HepG2 (hepatocellular carcinoma) cells cultured in-vitro using a lower molecular weight polymer to demonstrate that the molecular weight of "polydye" could be tailored to attain nuclear trafficking in cells. Prospective use of this technology encompasses a method of delivering a payload into a living cell based upon the hypercoiling nature of the pseudopeptides studied in this thesis and has led to a patent application (GB0228525.2; 20(2).

Compounds derived from fungi has been the subject of many studies in order to broaden the knowledge of their bioactive potential. Polysaccharides from Caripia montagnei have been described to possess anti-inflammatory and antioxidant properties. In this study, glucans extracted from Caripia montagnei mushroom were chemically characterized and their effects evaluated at different doses and intervals of treatment. It was also described their action on colonic injury in the model of colitis induced by 2,4,6-trinitrobenzene sulfonic acid (TNBS), and its action on cells of the human colon carcinoma (HT-29). Compounds extracted of C. montagnei contain high level of carbohydrates (96%), low content of phenolic compounds (1.5%) and low contamination with proteins (2.5%). The (FT-IR) and (NMR) analysis showed that polysaccharides from this species of mushroom are composed of α- and β-glucans. The colonic damage was evaluated by macroscopic, histological, biochemical and immunologic analyses. The results showed a reduction of colonic lesions in all groups treated with the glucans of Caripia montagnei (GCM). GCM significantly reduced the levels of IL-6 (50 and 75 mg/kg, p < 0.05), a major inflammatory cytokine. Biochemical analyses showed that such glucans acted on reducing levels of alkaline phosphatase (75 mg/kg, p < 0.01), nitric oxide (p < 0.001), and myeloperoxidase (p < 0.001). These results were confirmed microscopically by the reduction of cellular infiltration. The increase of catalase activity suggest a protective effect of GCM on colonic tissue, confirming their anti-inflammatory potential. GCM displayed cytostatic activity against HT-29 cells, causing accumulation of cells in G1 phase, blocking the cycle cell progression. Those glucans also showed ability to modulate the adhesion of HT-29 cells to Matrigel® and reduced the oxidative stress. The antiproliferative activity against HT-29 cells displayed by GCM (p <0.001) can be attributed to its cytostatic activity and induction of apoptosis by GCM

Oral squamous cell carcinoma (OSCC) is the most prevalent malignancy in the oral cavity and reach a large number of individuals, has become an important public health problem. Studies have demonstrated changes in pathway components BMP in various types of cancers as prostate, colon, breast, gastric and OSCCs. Is the current knowledge that these proteins may exert pro-tumor effect in more advanced stages of neoplastic development coming to favor progression and invasion tumor. The inhibition of the signaling pathway BMP-2 through its antagonists, have shown positive results of antitumor activity and use of Noggin may be a novel therapeutic target for cancer. Given this evidence and the few studies with BMP-2, Noggin and OSCC, the objective of this research was to evaluate the effect of BMP-2 and its antagonist Noggin on proliferation and migration cell in line of cell cultures of human tongue squamous cell carcinoma (SCC25). The study was divided in three groups, a control group, where SCC25 cells suffered no treatment, a BMP-2 group, in which cells were treated with 100ng/ml of BMP-2 and a group of cells that were treated with 100ng/ml of Noggin. For the proliferation assay and cell cycle were established three time intervals (24, 48 and 72 hours). Proliferative activity was investigated by trypan blue and cell cycle analysis by staining with propidium iodide flow cytometry. The potential for migration / invasion of SCC25 cells was performing by a cell invasion assay using Matrigel in a 48-hour interval. The proliferation curve showed a higher proliferation in cells treated with BMP-2 in 72 hours (p < 0.05), and lower overgrowth and cell viability in Noggin group. Recombinant proteins favored a greater percentage of cells in cell cycle phase Go/G1 with a statistically significant difference in the interval of 24 hours (p < 0.05). BMP- 2 produced a greater invasion of cells studied as well as its antagonist Noggin inhibits invasion of cells (p < 0.05). Thus, these results indicate that BMP-2 promotes malignant phenotype, dues stimulates proliferation and invasion of SCC25 cells and, its antagonist Noggin may be an alternative treatment, due to inhibit the tumor progression

We have investigated the role of bone sialoprotein (BSP), a secreted glycoprotein normally found in bone, in breast cancer progression. To explore functions for BSP in human breast cancer invasion and metastasis, the full-length BSP cDNA was transfected into the MDA-MB-231-BAG human breast cancer cell line under the control of the CMV promoter. Clones expressing BSP and vector control clones were isolated. BSP producing clones showed increased monolayer wound healing, a faster rate of stellate outgrowth in Matrigel and increased rate of invasion into a collagen matrix when compared to control clones. Clones were also examined in models of breast cancer growth and metastasis in vivo. BSP transfected clones showed an increased rate of primary tumor growth following mammary fat pad injection of nude mice. BSP transfected clones and vector control clones metastasized to soft organs and bone at a similar rate after intra-cardiac injection as determined by real-time PCR and X-ray analysis. Although these organs were targets for both BSP transfected and non-transfected cells, the size of the metastatic lesion was shown to be significantly larger for BSP expressing clones. This was determined by real-time PCR analysis for soft organs and by X-ray analysis of bone lesions. For bone this was confirmed by intra-tibial injections of cells in nude mice. We conclude that BSP acts to drive primary and secondary tumor growth of breast cancers in vivo.

A series of macrobicyclic dizinc(II) complexes Zn2L1-2B](ClO4)(4) (1-6) have been synthesized and characterized (L1-2 are polyaza macrobicyclic binucleating ligands, and B is the N,N-donor heterocyclic base (viz. 2,2'-bipyridine (bipy) and 1,10-phenanthroline (phen)). The DNA and protein binding, DNA hydrolysis and anticancer activity of these complexes were investigated. The interactions of complexes 1-6 with calf thymus DNA were studied by spectroscopic techniques, including absorption, fluorescence and CD spectroscopy. The DNA binding constant values of the complexes were found to range from 2.80 x 10(5) to 5.25 x 10(5) M-1, and the binding affinities are in the following order: 3 > 6 > 2 > 5 > 1 > 4. All the dizinc(II) complexes 1-6 are found to effectively promote the hydrolytic cleavage of plasmid pBR322 DNA under anaerobic and aerobic conditions. Kinetic data for DNA hydrolysis promoted by 3 and 6 under physiological conditions give observed rate constants (k(obs)) of 5.56 +/- 0.1 and 5.12 +/- 0.2 h(-1), respectively, showing a 10(7)-fold rate acceleration over the uncatalyzed reaction of dsDNA. Remarkably, the macrobicyclic dizinc(II) complexes 1-6 bind and cleave bovine serum albumin (BSA), and effectively promote the caspase-3 and caspase-9 dependent deaths of HeLa and BeWo cancer cells. The cytotoxicity of the complexes was further confirmed by lactate dehydrogenase enzyme levels in cancer cell lysate and content media.

El presente trabajo se realizó en el Programa adscrito a la Universidad Nacional Agraria, con el objetivo de estudiar el método de conservación in vitro a tasa mínimas de crecimiento en el cultivo de camote (Ipomoea batatas (L.) Lam.) clon N-1437, se procedió a inocular bajo condiciones de asepsia total, microesquejes entre 3 y 4 mm de longitud aproximadamente, conteniendo una yema axilar. En la evaluación final a las 16 semanas, se observó que en los tratamientos con 10 y 15 g/1 de manitol las variables altura, número de hojas y número de raíces presentaron menores valores de incremento mensual comparados con los alcanzados en los tratamientos testigo y a 5 g/1 de manitol, además se determinó que en el tratamiento testigo hubo mejores resultados en cuanto a la sobrevivencia de los tejidos en el 100%, menor formación de callo en un 5%, vitrificación 10% y en la coloración verde oscuro de las hojas en un 65% de las plántulas. En los tratamientos testigo y dilución de las sales MS, resultó que al 50% de dilución el incremento mensual presento valores intermedios en las variables altura de la plántula y número de hojas, pero el número de raíces fue menor presentando valores de 0.16 cm, 0.95 y 0.56 respectivamente. La sobrevivencia de los tejidos fue del 100%, no se presentó formación de callo ni vitrificación y la coloración verde oscuro de las hojas fue del 90%. Aun cuando el manitol fue efectivo en la reducción de la tasa presentaron morfológico de crecimiento efectos menos y fisiológico, deseables en el comparado con el aspecto efecto producido por las diluciones de las sales Ms.

Se establecieron in vitro yemas apicales de banano (Musa sp.) para su micro propagación durante tres sub-cultivos en un medio nutritivo artificial conteniendo sales minerales (MS), tiamina HCI, sacarosa y mio-inositol; variando con fines de estudio, la consistencia física del medio de cultivo (líquido y semi-sólido) y las concentraciones de reguladores de crecimiento: AIA= O y 1 mg/1 y 6-BAP= 5, 7 y 10 mg/1. Durante el establecimiento y multiplicación de los ex plantes se utilizó un cuarto de crecimiento para la incubación con temperaturas de 25 ± 1 ºc e intensidad lumínica de 2 000 lux. Inicialmente se establecieron in vitro 240 yemas apicales para su adaptación, de las cuales un 85 % (204) se adaptaron satisfactoriamente, el 15 % restante fueron descartados por contaminación, fenolización de las paredes del cormo o muerte de los ex plantes por no adaptación. La mayor contaminación se produjo por hongos y en menor medida por bacterias. La proliferación de hijos fue mayor en los medios de cultivo de consistencia semi-sólido en los tres sub-cultivos, correspondiendo los mejores resultados a la variante semi-sólida de los medios MS + 1mg/1 AIA+ 10 mg/16-BAP con promedio de 3.7 hijos por ex plante y 11 hijos en total, seguido por el medio MS_ + O mg/ AIA + 7 mg/1 6-BAP con 3.6 hijos por ex plante y 10.8 en total al final de los tres sub-cultivos. La menor proliferación se presentó en la variante líquida del medio MS + 1 mg/1 AIA + 10 mg/16-BAP con un promedio de 2.1 hijos por explante y 6.3 hijos en total. No se observó tendencia alguna en la proliferación de hijos con el aumento de los sub-cultivos, siendo evidente la influencia de la consistencia del medio de cultivo y la variación en los niveles de reguladores de crecimiento. El nivel más alto de 6-BAP (10 mg/1) utilizado indujo a la formación de multiyemas en los medios de cultivos líquidos, presentándose éstas a partir del II sub­ cultivo. Los medios de consistencia líquida favorecieron el crecimiento in vitro de los explantes, expresándose en un incremento en la altura y peso. Así mismo los medios líquidos indujeron a un desarrollo y crecimiento de raíces en todos los tratamientos estudiados.
