An increased incidence of Clostridium difficile infection (CDI) is associated with the emergence of epidemic strains characterised by high genetic diversity. Among the factors that may have a role in CDI there is a family of 29 paralogs, the cell wall proteins (CWPs), which compose the outer layer of the bacterial cell and are likely to be involved in colonisation. Previous studies have shown that 12 of the29 cwp genes are clustered in the same region, named after slpA (cwp1) the slpA locus, whereas the remaining 17 paralogs are distributed throughout the genome. The variability of 14 of these 17 cwp paralogs was determined in 40 C. difficile clinical isolates belonging to six of the currently prevailing PCR ribotypes. Based on sequence conservation, these cwp genes were divided into two groups, one comprising cwp loci having highly conserved sequences in all isolates, and the other 5 loci showing low genetic conservation between isolates of the same PCR ribotype as well as between different PCR ribotypes. Three conserved CWPs, Cwp16, Cwp18 and Cwp25, and two variable ones, Cwp26 and Cwp27, were characterised further by Western blot analysis of total cell extracts or S-layer preparations of the C. difficile clinical isolates. Expression of genetically invariable CWPs is well conserved in all isolates, while genetically variable CWPs are not always expressed at comparable levels even in strains containing identical sequences but belonging to different PCR ribotypes. In addition, we chose to analyse the immune response obtained in a protection experiment, carried out in hamsters, using a protein microarray approach to study the in vivo expression and the immunoreactivity of several surface proteins, including 18 Cwps.

Antigenic variation of the intestinal protozoan parasite Giardia lamblia is caused by an exchange of the parasite's variant surface protein (VSP) coat. Many investigations on antigenic variation were performed with G. lamblia clone GS/M-83-H7 which produces surface antigen VSP H7. To generate novel information on giardial vsp gene transcription, vsp RNA levels were assessed by quantitative reverse transcription-(RT)-PCR in both axenic VSP H7-type trophozoites and subvariants obtained after negative selection of GS/M-83-H7 trophozoites by treatment with a cytotoxic, VSP H7-specific monoclonal antibody. Our investigation was not restricted to the assessment of the sense vsp transcript levels but also included an approach aimed at the detection of complementary antisense vsp transcripts within the two trophozoite populations. We found that sense vsp H7 RNA predominated in VSP H7-type trophozoites while sense RNA from only one (vsp IVg) of 8 subvariant vsp genes totally analysed predominated in subvariant-type trophozoites. Interestingly, the two trophozoite populations exhibited a similar relative distribution regarding the vsp H7 and vsp IVg antisense RNA molecules. An analogous sense versus antisense RNA pattern was also observed when the transcripts of gene cwp 1 (encoding cyst wall protein 1) were investigated. Here, both types of RNA molecules only appeared after cwp 1 had been induced through in vitro encystation of the parasite. These findings for the first time demonstrated that giardial antisense RNA production did not occur in a constitutive manner but was directly linked to complementary sense RNA production after activation of the respective gene systems.

Cheese whey powder (CWP) is an attractive raw material for ethanol production since it is a dried and concentrated form of CW and contains lactose in addition to nitrogen, phosphate and other essential nutrients. In the present work, deproteinized CWP was utilized as fermentation medium for ethanol production by Kluyveromyces fragilis. The individual and combined effects of initial lactose concentration (50-150 kg m(-3)), temperature (25-35 degrees C) and inoculum concentration (1-3 kg m(-3)) were investigated through a 2(3) full-factorial central composite design, and the optimal conditions for maximizing the ethanol production were determined. According to the statistical analysis, in the studied range of values, only the initial lactose concentration had a significant effect on ethanol production, resulting in higher product formation as the initial substrate concentration was increased. Assays with initial lactose concentration varying from 150 to 250 kg m(-3) were thus performed and revealed that the use of 200 kg m(-3) initial lactose concentration, inoculum concentration of 1 kg m(-3) and temperature of 35 degrees C were the best conditions for maximizing the ethanol production from CWP solution. Under these conditions, 80.95 kg m(-3) of ethanol was obtained after 44 h of fermentation. (C) 2011 Elsevier Ltd. All rights reserved.

Objectives: To estimate the prevalence of fibromyalgia (FM) and chronic widespread pain (CWP) in community-dwelling elderly individuals living in Sao Paulo, to assess the spectrum of problems related to these diseases using the Fibromyalgia Impact Questionnaire (FIQ) and to correlate the FIQ with the number of tender points and with pain threshold. Methods: Our sample consisted of 361 individuals (64% women, 36% men, mean age of 73.3 +/- 5.7 years). Individuals were classified into four groups: FM (according to American College of Rheumatology criteria), CWP, regional pain (RP) and no pain (NP). Pain characteristics and dolorimetry for 18 tender points and the FIQ were assessed. Results: The prevalence of FM was 5.5% [95% confidence interval (CI) = 5.4-5.7], and the prevalence of CWP was 14.1% (95% Cl: 10.5-17.7%). The frequency of RP was 52.6% and the prevalence of NP was 27.7%. FIQ scores were higher in people with FM (44.5), followed by CWP (31.4), RP (18.1) and NP (5.5) (p < 0.001). There was a positive correlation between the domains of the FIQ and the number of tender Points (p < 0.05), and a negative correlation between FIQ score and pain threshold (p < 0.05). Conclusion: In our elderly subjects, the prevalence of FM was slightly higher compared to previously reported studies, and CWP was around 14%. The spectrum of problems related to chronic pain was more severe in FM followed by CWP, strongly suggesting that these conditions should be diagnosed and adequately treated in older individuals. A presente Dissertação de Mestrado tem como objetivo o estudo do problema de inversão sísmica baseada em refletores planos para arranjo fonte-comum (FC) e ponto-médiocomum (PMC). O modelo direto é descrito por camadas homogêneas, isotrópicas com interfaces plano-horizontais. O problema é relacionado ao empilhamento NMO baseado na otimização da função semblance, para seções PMC corrigidas de sobretempo normal (NMO). O estudo foi baseado em dois princípios. O primeiro princípio adotado foi de combinar dois grupos de métodos de inversão: um Método Global e um Método Local. O segundo princípio adotado foi o de cascata, segundo a teoria Wichert-Herglotz-Bateman, que estabelece que para conhecer uma camada inferior tem-se que conhecer primeiro a camada superior (dissecação). A aplicação do estudo é voltada à simulação sísmica de Bacia Sedimentar do Solimões e de Bacia Marinha para se obter uma distribuição local 1D de velocidades e espessuras para a subsuperfície em horizontes alvo. Sendo assim, limitamos a inversão entre 4 e 11 refletores, uma vez que na prática a indústria limita uma interpretação realizada apenas em número equivalente de 3 a 4 refletores principais. Ressalta-se que este modelo é aplicável como condição inicial ao imageamento de seções sísmicas em regiões geologicamente complexas com variação horizontal suave de velocidades. Os dados sintéticos foram gerados a partir dos modelos relacionados a informações geológicas, o que corresponde a uma forte informação a priori no modelo de inversão. Para a construção dos modelos relacionados aos projetos da Rede Risco Exploratório (FINEP) e de formação de recursos humanos da ANP em andamento, analisamos os seguintes assuntos relevantes: (1) Geologia de bacias sedimentares terrestre dos Solimões e ma rinha (estratigráfica, estrutural, tectônica e petrolífera); (2) Física da resolução vertical e horizontal; e (3) Discretização temporal-espacial no cubo de multi-cobertura. O processo de inversão é dependente do efeito da discretização tempo-espacial do campo de ondas, dos parâmetros físicos do levantamento sísmico, e da posterior reamostragem no cubo de cobertura múltipla. O modelo direto empregado corresponde ao caso do operador do empilhamento NMO (1D), considerando uma topografia de observação plana. O critério básico tomado como referência para a inversão e o ajuste de curvas é a norma 2 (quadrática). A inversão usando o presente modelo simples é computacionalmente atrativa por ser rápida, e conveniente por permitir que vários outros recursos possam ser incluídos com interpretação física lógica; por exemplo, a Zona de Fresnel Projetada (ZFP), cálculo direto da divergência esférica, inversão Dix, inversão linear por reparametrização, informações a priori, regularização. A ZFP mostra ser um conceito út il para estabelecer a abertura da janela espacial da inversão na seção tempo-distância, e representa a influência dos dados na resolução horizontal. A estimativa da ZFP indica uma abertura mínima com base num modelo adotado, e atualizável. A divergência esférica é uma função suave, e tem base física para ser usada na definição da matriz ponderação dos dados em métodos de inversão tomográfica. A necessidade de robustez na inversão pode ser analisada em seções sísmicas (FC, PMC) submetida a filtragens (freqüências de cantos: 5;15;75;85; banda-passante trapezoidal), onde se pode identificar, comparar e interpretar as informações contidas. A partir das seções, concluímos que os dados são contaminados com pontos isolados, o que propõe métodos na classe dos considerados robustos, tendo-se como referência a norma 2 (quadrados- mínimos) de ajuste de curvas. Os algoritmos foram desenvolvidos na linguagem de programação FORTRAN 90/95, usando o programa MATLAB para apresentação de resultados, e o sistema CWP/SU para modelagem sísmica sintética, marcação de eventos e apresentação de resultados.

O presente trabalho teve como objetivos a análise e atenuação de múltiplas de superfície livre, o processamento e o imageamento de dados sísmicos marinhos visando obter imagens migradas de utilidade à interpretação geológica voltada à exploração de petróleo. Foi dada uma atenção sistemática ao estudo de múltiplas de superfície livre do ponto de vista do filtro de predição baseado na teoria da comunicação com o objetivo de melhor aplicar o filtro WH de deconvolução preditiva, na etapa de processamento, posterior à correção NMO, embora outros métodos possam ser considerados mais competitivos. A identificação e a atenuação de reflexões múltiplas em dados sísmicos reais continua como um grande desafio no processamento de dados sísmicos, uma vez que elas são consideradas como ruído. No entanto, sendo este ruído classificado como coerente, várias técnicas foram desenvolvidas objetivando sua atenuação para evitar erros em cascata nas etapas posteriores como processamento, marcação de eventos, inversão tomográfica, imageamento, e finalmente na interpretação geológica das imagens obtidas. Outro aspecto do trabalho foi estabelecer um fluxograma de processamento-imageamento sendo a atenuação de múltiplas de superfície livre uma etapa central. Foram obtidas seções migradas em tempo e em profundidade onde se permite a interpretação que se deseja. O desenvolvimento deste trabalho foi realizado com os pacotes CWP/SU e MatLab.

Fibromyalgia (FM) is characterized by chronic non-inflammatory widespread pain (CWP) and changes in sympathetic function. In attempt to elucidate the pathophysiological mechanisms of FM we used a well-established CWP animal model. We aimed to evaluate changes in cardiac autonomic balance and baroreflex function in response to CWP induction in rats. CWP was induced by two injections of acidic saline (pH 4.0, n = 8) five days apart into the left gastrocnemius muscle. Control animals were injected twice with normal saline (pH 7.2, n = 6). One day after the second injection of acidic saline or normal saline, the animals had pulse interval (PI) and systolic arterial pressure (SAP) variability, and spontaneous baroreflex sensitivity (BRS) evaluated. After induction of CWP, there was an increase of power in the low frequency (LF) band of PI spectrum (12.75 +/- 1.04 nu), a decrease in the high frequency (HF) band (87.25 +/- 1.04 nu) and an increase of LF/HF ratio (0.16 +/- 0.01), when compared to control animals (7.83 +/- 1.13 nu LF; 92.16 +/- 1.13 nu HF; 0.08 +/- 0.01 LF/HF). In addition, there was an increase of power in the LF band of SAP spectrum (7.93 +/- 1.39 mmHg(2)) when compared to control animals (2.97 +/- 0.61 mmHg(2)). BRS was lower in acidic saline injected rats (0.59 +/- 0.06 ms/mmHg) when compared to control animals (0.71 +/- 0.03 ms/mmHg). Our results showed that induction of CWP in rats shifts cardiac sympathovagal balance towards sympathetic predominance and decreases BRS. These data corroborate findings in humans with FM. Background Chronic localized pain syndromes, especially chronic low back pain (CLBP), are common reasons for consultation in general practice. In some cases chronic localized pain syndromes can appear in combination with chronic widespread pain (CWP). Numerous studies have shown a strong association between CWP and several physical and psychological factors. These studies are population-based cross-sectional and do not allow for assessing chronology. There are very few prospective studies that explore the predictors for the onset of CWP, where the main focus is identifying risk factors for the CWP incidence. Until now there have been no studies focusing on preventive factors keeping patients from developing CWP. Our aim is to perform a cross sectional study on the epidemiology of CLBP and CWP in general practice and to look for distinctive features regarding resources like resilience, self-efficacy and coping strategies. A subsequent cohort study is designed to identify the risk and protective factors of pain generalization (development of CWP) in primary care for CLBP patients. Methods/Design Fifty-nine general practitioners recruit consecutively, during a 5 month period, all patients who are consulting their family doctor because of chronic low back pain (where the pain is lasted for 3 months). Patients are asked to fill out a questionnaire on pain anamnesis, pain-perception, co-morbidities, therapy course, medication, socio demographic data and psychosomatic symptoms. We assess resilience, coping resources, stress management and self-efficacy as potential protective factors for pain generalization. Furthermore, we raise risk factors for pain generalization like anxiety, depression, trauma and critical life events. During a twelve months follow up period a cohort of CLBP patients without CWP will be screened on a regular basis (3 monthly) for pain generalization (outcome: incident CWP). Discussion This cohort study will be the largest study which prospectively analyzes predictors for transition from CLBP to CWP in primary care setting. In contrast to the typically researched risk factors, which increase the probability of pain generalization, this study also focus intensively on protective factors, which decrease the probability of pain generalization.

The Federal Coal Mine Health and Safety Act of 1969 required that periodic chest radiographs be offered to underground coal miners to protect the miners from the development of Coal Workers' Pneumoconiosis (CWP) and progression of the disease to progressive massive fibrosis (PMF). These examinations are administered by the National Institute for Occupational Safety and Health (NIOSH) through the Coal Workers' Health Surveillance Program (CWHSP). The mine operator is required to provide each miner with the opportunity to have the chest radiograph at no cost to the miner.^ Three rounds of examinations have been conducted since 1969 and the fourth is underway. The decrease in participation over rounds is of great concern if the incidence and progression of CWP are to be understood and controlled.^ This study developed rates of participation for each of 558 West Virginia underground coal mines who submitted or had NIOSH assigned plans for making chest radiographs available during the third round, July 1978 through December 1980. These rates were analyzed in relation to desired levels of participation and to reinforcing, predisposing and enabling factors presumed to affect rates of participation in disease prevention and surveillance programs.^ Two reinforcing factors, size of mine and inclusion of the mine in the National Coal Study (NCS) epidemiology research program, and the enabling factor, use of an on-site radiograph facility, demonstrated highly significant relationships to participation rates.^ The major findings of the study were: (1) Participation in the CWHSP is even lower than previously estimated; (2) CWHSP program evaluation is not systematic and program data base is not complete and comprehensive; and (3) NIOSH program policy is not clear and administration of the CWHSP is fragmented and lacks adequate fiscal and personnel resources. ^

O chumbo é utilizado em muitos produtos, tais como baterias, gasolina, tintas e corantes, resultando na sua libertação no meio ambiente. Neste trabalho, foi examinado o papel da parede celular da levedura Saccharomyces cerevisiae como uma barreira ou como alvo da toxicidade do chumbo. A biodisponibilidade do Pb é muito reduzida pelos componentes do meio de cultura YEPD, o que dificulta a avaliação da toxicidade deste elemento em concentrações ambientalmente realistas. Para avaliar a toxicidade de Pb em S. cerevisiae, em condições de crescimento, foram efetuadas diferentes diluições (10-100 vezes) do meio YEPD, as quais foram misturadas com várias concentrações de Pb (0,1-1,0 mmol/l). Observou-se que o YEPD diluído 25 vezes constituía a melhor condição de compromisso entre o crescimento celular e a precipitação de Pb. Os genes CWP1 e CWP2 codificam para duas grandes manoproteínas da parede celular da levedura S. cerevisiae; a deleção destes genes CWP aumenta a permeabilidade da parede celular. A suscetibilidade de células de levedura interrompidas no gene CWP1 (estirpe cwp1Δ) ou CWP2 (estirpe cwp2Δ) foi comparada com a da estirpe, isogénica, selvagem (WT). Verificou-se que o crescimento das estirpes cwp1Δ e cwp2Δ, no meio de cultura YEPD 25 vezes diluído, na presença de Pb, não diferiu do crescimento da estirpe WT. Este resultado sugere que a alteração da permeabilidade da parede celular não altera a sensibilidade de células de levedura ao Pb. Foi investigada o impacto do Pb na parede celular de levedura. Para este efeito, comparou-se a suscetibilidade ao dodecil sulfato de sódio (SDS), ao calcofluor (CFW) e a uma enzima que degrada a parede da célula (liticase), em células da estirpe WT não expostas ou expostas a Pb durante 4, 8 ou 24 h. Além disso, o conteúdo de quitina da parede celular de levedura foi investigada por coloração das células com CFW. Os resultados não mostraram uma alteração da suscetibilidade ao SDS e ao CFW, nas células tratadas com Pb; contudo, nas células tratadas durante 24 h com Pb, observou-se um aumento da sensibilidade à liticase e um aumento da coloração com CFW. Estes resultados sugerem que o chumbo interage com a parede celular da levedura e influencia a sua composição. Deve ser levado a cabo trabalho adicional a fim de confirmar estes resultados.

La relación entre el liderazgo y emprendimiento es un tema de estudio y análisis relativamente nuevo. El término de liderazgo ha sido estudiado y enfocado a otras áreas de estudio, pero en cuanto a la relación existente con emprendimiento se presentan aún carencias conceptuales. A lo largo del presente estudio, se hace una revisión de la literatura la cual permite identificar y analizar estos conceptos y su relación en la última década en Colombia, consultando literatura comprendida en bases de datos en línea tales como, Redalyc, Scielo y Dialnet. En este documento, se destacan los atributos de un emprendedor y de un líder, igualmente se recogen los modelos de emprendimiento, las teorías de liderazgo, y los elementos que caracterizan cada concepto. Finalmente, se evidencia una dicotomía entre el líder y el emprendedor y se concluye que el liderazgo es una característica intrínseca del emprendimiento; sin embargo, el liderazgo no es por regla utilizado para emprender.
Cloud parameter retrievals from Meteosat and their effects on the shortwave radiation at the surface

A method based on Spinning Enhanced Visible and Infrared Imager (SEVIRI) measured reflectance at 0.6 and 3.9 µm is used to retrieve the cloud optical thickness (COT) and cloud effective radius (re) over the Iberian Peninsula. A sensitivity analysis of simulated retrievals to the input parameters demonstrates that the cloud top height is an important factor in satellite retrievals of COT and re with uncertainties around 10% for small values of COT and re; for water clouds these uncertainties can be greater than 10% for small values of re. The uncertainties found related with geometries are around 3%. The COT and re are assessed using well-known satellite cloud products, showing that the method used characterize the cloud field with more than 80% (82%) of the absolute differences between COT (re) mean values of all clouds (water plus ice clouds) centred in the range from ±10 (±10 µm), with absolute bias lower than 2 (2 μm) for COT (re) and root mean square error values lower than 10 (8 μm) for COT (re). The cloud water path (CWP), derived from satellite retrievals, and the shortwave cloud radiative effect at the surface (CRESW) are related for high fractional sky covers (Fsc >0.8), showing that water clouds produce more negative CRESW than ice clouds. The COT retrieved was also related to the cloud modification factor, which exhibits reductions and enhancements of the surface SW radiation of the order of 80% and 30%, respectively, for COT values lower than 10. A selected case study shows, using a ground-based sky camera that some situations classified by the satellite with high Fsc values correspond to situations of broken clouds where the enhancements actually occur. For this case study, a closure between the liquid water path (LWP) obtained from the satellite retrievals and the same cloud quantity obtained from ground-based microwave measurements was performed showing a good agreement between both LWP data set values.
