A perturbation of FtsZ assembly dynamics has been shown to inhibit bacterial cytokinesis. In this study, the antibacterial activity of 151 rhodanine compounds was assayed using Bacillus subtilis cells. Of 151 compounds, eight strongly inhibited bacterial proliferation at 2 mu M. Subsequently, we used the elongation of B. subtilis cells as a secondary screen to identify potential FtsZ-targeted antibacterial agents. We found that three compounds significantly increased bacterial cell length. One of the three compounds, namely, CCR-11 (E)-2-thioxo-5-({3-(trifluoromethyl)phenyl]furan-2-yl}methylene) thiazolidin-4-one], inhibited the assembly and GTPase activity of FtsZ in vitro. CCR-11 bound to FtsZ with a dissociation constant of 1.5 +/- 0.3 mu M. A docking analysis indicated that CCR-11 may bind to FtsZ in a cavity adjacent to the T7 loop and that short halogen oxygen, H-bonding, and hydrophobic interactions might be important for the binding of CCR-11 with FtsZ. CCR-11 inhibited the proliferation of B. subtilis cells with a half-maximal inhibitory concentration (IC50) of 1.2 +/- 0.2 mu M and a minimal inhibitory concentration of 3 mu M. It also potently inhibited proliferation of Mycobacterium smegmatis cells. Further, CCR-11 perturbed Z-ring formation in B. subtilis cells; however, it neither visibly affected nucleoid segregation nor altered the membrane integrity of the cells. CCR-11 inhibited HeLa cell proliferation with an IC50 value of 18.1 +/- 0.2,mu M (similar to 15 x IC50 of B. subtilis cell proliferation). The results suggested that CCR-11 inhibits bacterial cytokinesis by inhibiting FtsZ assembly, and it can be used as a lead molecule to develop FtsZ-targeted antibacterial agents.

It is shown how to use non-commutative stopping times in order to stop the CCR flow of arbitrary index and also its isometric cocycles, i.e. left operator Markovian cocycles on Boson Fock space. Stopping the CCR flow yields a homomorphism from the semigroup of stopping times, equipped with the convolution product, into the semigroup of unital endomorphisms of the von Neumann algebra of bounded operators on the ambient Fock space. The operators produced by stopping cocycles themselves satisfy a cocycle relation.

作为植物界广泛存在的一类酚类聚合物,木质素是陆生植物正常生长发育过程中非常重要的生物大分子,而且与人类的生活息息相关。利用分子生物学手段和基因工程方法,从小麦中分离木质素生物合成途径的关键酶-肉桂酰辅酶A还原酶基因(CCR),研究肉桂酰辅酶A还原酶基因在木质素代谢途径中的调控规律,从其催化的限速步骤入手,来调控木质素的合成,有效的改变木质素的组成、含量和结构,是改善木质素在植物生长发育中的作用乃至开发木质素资源的关键所在。本文就小麦肉桂酰辅酶A还原酶基因的分离、表达特征及其在木质素合成途径中的作用开展了研究工作。 首先用RACE方法从小麦中克隆了CCR的两个cDNA的部分序列,序列分析表明它们编码的蛋白具有CCR的典型特点,GC含量高于均60%,两者在核酸水平和蛋白水平的同源性为76%和 69%,证明在小麦中至少存在着两个CCR基因。通过 RT-PCR和Northern 杂交确定W-cr6和W-cr19在小麦的发育中具有不同的表达特征,W-cr6主要在茎中表达,而W-cr19的表达集中在根中。以W-cr6为探针,从cDNA文库中筛选到一个全长1317bp的cDNA,命名为TaCCR1。TaCCR1包括开放阅读框 (ORF) 1047bp、5′端侧翼 72bp和3′端侧翼198bp的非翻译序列。TaCCR1能够编码由349个氨基酸组成的蛋白质,预期的分子量为37.4kD。同源性比较显示TaCCR1基因在核酸水平和蛋白质水平与其他物种的CCR基因的同源性高于60%。 为了分析CCR在木质素合成中的作用,用TaCCR1构建了用于转化烟草的正义和反义表达载体pStCCR和pAtCCR、用于转化小麦的正义和反义表达载体pBSC1和pBAC1。通过农杆菌介导得到了30株反义转基因烟草和12株正义转基因烟草。由于外源基因的抑制作用,转基因烟草在形态、木质素组成和含量、木质部显微结构上都程度不同的发生了变化。正义和反义的转基因株系呈现出株型矮化、木质素含量下降、木质部导管细胞壁受到破坏等现象。同时利用花粉管通道法转化小麦种子5000多粒,部分处理经过初步的PCR和 Southern分子鉴定获得了1株转基因株系,需要对其遗传、生理和形态特征做进一步的研究。 本文还对木质素对小麦茎杆的机械强度的影响做了初步的探讨,得到的结果是小麦茎杆的木质素含量、维管束的数量、茎杆有效的横界面积与其最大弯曲应力存在着正相关,而维管束的结构、密度对茎杆的最大弯曲应力没有明显的影响,从而为通过CCR基因来改善小麦茎杆的抗倒特性建立了生理学基础。

Synechocystis sp. PCC 6803 exposed to chill (5 degrees C)-light (100 mu mol photons m(-2) s(-1)) stress loses its ability to reinitiate growth. From a random insertion mutant library of Synechocystis sp. PCC 6803, a sll1242 mutant showing increased sensitivity to chill plus light was isolated. Mutant reconstruction and complementation with the wild-type gene confirmed the role of sll1242 in maintaining chill-light tolerance. At 15 degrees C, the autotrophic and mixotrophic growth of the mutant were both inhibited, paralleled by decreased photosynthetic activity. The expression of sll1242 was upregulated in Synechocystis sp. PCC 6803 after transfer from 30 to 15 degrees C at a photosynthetic photon flux density of 30 mu mol photons m(-2) S-1. sll1242, named ccr (cyanobacterial cold resistance gene)- 1, may be required for cold acclimation of cyanobacteria in light.

The tourist use of Santana cave provides a row of environmental damages, compromising partially the pristine conditions. Among the measures adopted to avoid this situation, this issue presents a contribution for the physical limits to tourist visitation and the indications for speleotouristic management. It was used the Cifuentes Carrying Capacity method and some basic principles of Visitors Impact Management Framework - VIM. The results show a Real Carrying Capacity (CCR) about 120 visits daily on the cave. The discussions raise a few hypotheses about the alteration in the method used and the temporal frequency between visitors groups, suggesting a provisional CCR of 117 and 135 visitors daily, respectively, during the week and in the weekends and holidays. The conclusions appoint the necessity of a conceptual revision in the Carrying Capacity methods, mainly about its adaptation for tourist management in caves. In the case studied, it is suggested the immediate beginning of environmental monitoring of the cave, to verify the plausibility from proposed visitation limits and the possible contributions for to mitigate the environmental impacts from speleotourism.

PURPOSE: To compare gene expression of the chemokines RANTES and eotaxin-2, its receptor, CCR-3, adhesion molecule ICAM-1 and its receptor LFA-1 in eosinophilic polyps and in control normal nasal mucosa. METHODS: Gene expression was quantified by Real Time PCR in polyps (n=35) and in healthy nasal mucosa (n=15). RESULTS: Eosinophilic polyps showed a higher expression of eotaxin-2 and RANTES, but not of CCR-3, ICAM-1 or LFA-1 compared to control nasal mucosa. CONCLUSION: Eosinophilic polyps present greater expression of eotaxin-2 and RANTES, but not of CCR-3, ICAM-1 or LFA-1 compared to control nasal mucosa.

Although infection by primary HIV type 1 (HIV-1) isolates normally requires the functional interaction of the viral envelope protein with both CD4 and the CCR-5 coreceptor, a subset of such isolates also are able to use the distinct CCR-3 receptor. By analyzing the ability of a series of wild-type and chimeric HIV-1 envelope proteins to mediate CCR-3-dependent infection, we have determined that CCR-3 tropism maps to the V1 and V2 variable region of envelope. Although substitution of the V1/V2 region of a CCR-3 tropic envelope into the context of a CCR-5 tropic envelope is both necessary and sufficient to confer CCR-3 tropism, this same substitution has no phenotypic effect when inserted into a CXCR-4 tropic HIV-1 envelope context. However, this latter chimera acquires both CCR-3 and CCR-5 tropism when a CCR-5 tropic V3 loop sequence also is introduced. These data demonstrate that the V1/2 region of envelope can, like the V3 loop region, encode a particular coreceptor requirement and suggest that a functional envelope:CCR-3 interaction may depend on the cooperative interaction of CCR-3 with both the V1/V2 and the V3 region of envelope.

The β-chemokine receptor CCR-5 is essential for the efficient entry of primary macrophage-tropic HIV-1 isolates into CD4+ target cells. To study CCR-5-dependent cell-to-cell fusion, we have developed an assay system based on the infection of CD4+ CCR-5+ HeLa cells with a Semliki Forest virus recombinant expressing the gp120/gp41 envelope (Env) from a primary clade B HIV-1 isolate (BX08), or from a laboratory T cell line-adapted strain (LAI). In this system, gp120/gp41 of the “nonsyncytium-inducing,” primary, macrophage-tropic HIV-1BX08 isolate, was at least as fusogenic as that of the “syncytium-inducing” HIV-1LAI strain. BX08 Env-mediated fusion was inhibited by the β-chemokines RANTES (regulated upon activation, normal T cell expressed and secreted) and macrophage inflammatory proteins 1β (MIP-1β) and by antibodies to CD4, whereas LAI Env-mediated fusion was insensitive to these β-chemokines. In contrast soluble CD4 significantly reduced LAI, but not BX08 Env-mediated fusion, suggesting that the primary isolate Env glycoprotein has a reduced affinity for CD4. The domains in gp120/gp41 involved in the interaction with the CD4 and CCR-5 molecules were probed using monoclonal antibodies. For the antibodies tested here, the greatest inhibition of fusion was observed with those directed to conformation-dependent, rather than linear epitopes. Efficient inhibition of fusion was not restricted to epitopes in any one domain of gp120/gp41. The assay was sufficiently sensitive to distinguish between antibody- and β-chemokine-mediated fusion inhibition using serum samples from patient BX08, suggesting that the system may be useful for screening human sera for the presence of biologically significant antibodies.

A genetic defect in a CC-chemokine receptor (CCR)-5, the principal coreceptor for the macrophage-tropic HIV type 1 (HIV-1), recently was found to naturally protect CCR-5-defective, but healthy, individuals from HIV-1 infection. In this study, we mimic the natural resistance of the CCR-5-defective individuals by designing a strategy to phenotypically knock out CCR-5. The inactivation of the CCR-5 coreceptor is accomplished by targeting a modified CC-chemokine to the endoplasmic reticulum to block the surface expression of newly synthesized CCR-5. The lymphocytes transduced to express the intracellular chemokine, termed “intrakine,” were found to be viable and resistant to macrophage-tropic HIV-1 infection. Thus, this gene-based intrakine strategy targeted at the conserved cellular receptor for the prevention of HIV-1 entry should have significant advantages over currently described approaches for HIV-1 therapy.
