Atmospheric aerosol particles serving as cloud condensation nuclei (CCN) are key elements of the hydrological cycle and climate. We have measured and characterized CCN at water vapor supersaturations in the range of S=0.10-0.82% in pristine tropical rainforest air during the AMAZE-08 campaign in central Amazonia. The effective hygroscopicity parameters describing the influence of chemical composition on the CCN activity of aerosol particles varied in the range of kappa approximate to 0.1-0.4 (0.16+/-0.06 arithmetic mean and standard deviation). The overall median value of kappa approximate to 0.15 was by a factor of two lower than the values typically observed for continental aerosols in other regions of the world. Aitken mode particles were less hygroscopic than accumulation mode particles (kappa approximate to 0.1 at D approximate to 50 nm; kappa approximate to 0.2 at D approximate to 200 nm), which is in agreement with earlier hygroscopicity tandem differential mobility analyzer (H-TDMA) studies. The CCN measurement results are consistent with aerosol mass spectrometry (AMS) data, showing that the organic mass fraction (f(org)) was on average as high as similar to 90% in the Aitken mode (D <= 100 nm) and decreased with increasing particle diameter in the accumulation mode (similar to 80% at D approximate to 200 nm). The kappa values exhibited a negative linear correlation with f(org) (R(2)=0.81), and extrapolation yielded the following effective hygroscopicity parameters for organic and inorganic particle components: kappa(org)approximate to 0.1 which can be regarded as the effective hygroscopicity of biogenic secondary organic aerosol (SOA) and kappa(inorg)approximate to 0.6 which is characteristic for ammonium sulfate and related salts. Both the size dependence and the temporal variability of effective particle hygroscopicity could be parameterized as a function of AMS-based organic and inorganic mass fractions (kappa(p)=kappa(org) x f(org)+kappa(inorg) x f(inorg)). The CCN number concentrations predicted with kappa(p) were in fair agreement with the measurement results (similar to 20% average deviation). The median CCN number concentrations at S=0.1-0.82% ranged from N(CCN,0.10)approximate to 35 cm(-3) to N(CCN,0.82)approximate to 160 cm(-3), the median concentration of aerosol particles larger than 30 nm was N(CN,30)approximate to 200 cm(-3), and the corresponding integral CCN efficiencies were in the range of N(CCN,0.10/NCN,30)approximate to 0.1 to N(CCN,0.82/NCN,30)approximate to 0.8. Although the number concentrations and hygroscopicity parameters were much lower in pristine rainforest air, the integral CCN efficiencies observed were similar to those in highly polluted megacity air. Moreover, model calculations of N(CCN,S) assuming an approximate global average value of kappa approximate to 0.3 for continental aerosols led to systematic overpredictions, but the average deviations exceeded similar to 50% only at low water vapor supersaturation (0.1%) and low particle number concentrations (<= 100 cm(-3)). Model calculations assuming aconstant aerosol size distribution led to higher average deviations at all investigated levels of supersaturation: similar to 60% for the campaign average distribution and similar to 1600% for a generic remote continental size distribution. These findings confirm earlier studies suggesting that aerosol particle number and size are the major predictors for the variability of the CCN concentration in continental boundary layer air, followed by particle composition and hygroscopicity as relatively minor modulators. Depending on the required and applicable level of detail, the information and parameterizations presented in this paper should enable efficient description of the CCN properties of pristine tropical rainforest aerosols of Amazonia in detailed process models as well as in large-scale atmospheric and climate models.

In-situ measurements in convective clouds (up to the freezing level) over the Amazon basin show that smoke from deforestation fires prevents clouds from precipitating until they acquire a vertical development of at least 4 km, compared to only 1-2 km in clean clouds. The average cloud depth required for the onset of warm rain increased by similar to 350 m for each additional 100 cloud condensation nuclei per cm(3) at a super-saturation of 0.5% (CCN0.5%). In polluted clouds, the diameter of modal liquid water content grows much slower with cloud depth (at least by a factor of similar to 2), due to the large number of droplets that compete for available water and to the suppressed coalescence processes. Contrary to what other studies have suggested, we did not observe this effect to reach saturation at 3000 or more accumulation mode particles per cm(3). The CCN0.5% concentration was found to be a very good predictor for the cloud depth required for the onset of warm precipitation and other microphysical factors, leaving only a secondary role for the updraft velocities in determining the cloud drop size distributions. The effective radius of the cloud droplets (r(e)) was found to be a quite robust parameter for a given environment and cloud depth, showing only a small effect of partial droplet evaporation from the cloud's mixing with its drier environment. This supports one of the basic assumptions of satellite analysis of cloud microphysical processes: the ability to look at different cloud top heights in the same region and regard their r(e) as if they had been measured inside one well developed cloud. The dependence of r(e) on the adiabatic fraction decreased higher in the clouds, especially for cleaner conditions, and disappeared at r(e)>=similar to 10 mu m. We propose that droplet coalescence, which is at its peak when warm rain is formed in the cloud at r(e)=similar to 10 mu m, continues to be significant during the cloud's mixing with the entrained air, cancelling out the decrease in r(e) due to evaporation.

Expression of connective tissue growth factor (CTGF), a member of the CCN gene family, is known to be significantly induced by mechanical stress. We have therefore investigated whether other members of the CCN gene family, including Cyr61 and Nov, might reveal a similar stress-dependent regulation. Fibroblasts growing under stressed conditions within a three-dimensional collagen gel showed at least a 15 times higher level of Cyr61 mRNA than cells growing under relaxed conditions. Upon relaxation, the decline of the Cyr61 mRNA to a lower level occurred within 2 h, and was thus quicker than the response of CTGF. The regulation was fully reversible when stress was reapplied. Thus, Cyr61 represents another typical example of a stress-responsive gene. The level of the Nov mRNA was low in the stressed state, but increased in the relaxed state. This CCN gene therefore shows an inverted regulation relative to that of Cyr61 and CTGF. Inhibition of protein kinases by means of staurosporine suppressed the stress-induced expression of Cyr61 and CTGF. Elevated levels of cAMP induced by forskolin mimicked the effects of relaxation on the regulation of Cyr61, CTGF and Nov. Thus, adenylate cyclase as well as one or several protein kinases might be involved in the mechanoregulation of these CCN genes.

Más de 12000 m3 de mucilago de cacao CCN-51 son producidos y abandonados en las fincas de cacao en el Ecuador cada año. El estudio tiene como objetivo caracterizar este residuo en la Zona 6. Las muestras se obtuvieron de 10 lugares dentro de la zona de estudio, las mismas que están geo referenciadas. Para el análisis se usó espectrofotometría UV-Visible para la identificación de azucares reductoras totales, y espectrofotometría de absorción atómica para identificar minerales. Además se determinó parámetros físicos. Los resultados de los análisis fueron los siguientes: pH 4.05±0.004, los sólidos solubles fue de 17.15±0.86 0Brix, la acidez Titulable fue 245.25±21.19 meq/L. Por otra parte las azucares reductoras totales fueron de 1228.82±178.52 g/L y los de calcio, sodio y potasio fueron de 169.21±31.04 mg/L, 161.85±40.41 mg/L, 462.9±49.96 mg/L respectivamente. Se analizó una muestra mediante espectroscopia de infrarrojo para identificar glucosa y sacarosa, los resultados de este análisis fueron 398 g/L, 800g/l, posteriormente se realizó un análisis t student con el resultado obtenido en espectrofotometría UV-Visible de las azucares reductoras totales, como resultado final se estableció que no existe diferencia significativa entre las dos técnicas instrumentales.

For the first time, oxygen terminated cellulose carbon nanoparticles (CCN) was synthesised and applied in gene transfection of pIRES plasmid. The CCN was prepared from catalytic of polyaniline by chemical vapour deposition techniques. This plasmid contains one gene that encodes the green fluorescent protein (GFP) in eukaryotic cells, making them fluorescent. This new nanomaterial and pIRES plasmid formed π-stacking when dispersed in water by magnetic stirring. The frequencies shift in zeta potential confirmed the plasmid strongly connects to the nanomaterial. In vitro tests found that this conjugation was phagocytised by NG97, NIH-3T3 and A549 cell lines making them fluorescent, which was visualised by fluorescent microscopy. Before the transfection test, we studied CCN in cell viability. Both MTT and Neutral Red uptake tests were carried out using NG97, NIH-3T3 and A549 cell lines. Further, we use metabolomics to verify if small amounts of nanomaterial would be enough to cause some cellular damage in NG97 cells. We showed two mechanisms of action by CCN-DNA complex, producing an exogenous protein by the transfected cell and metabolomic changes that contributed by better understanding of glioblastoma, being the major finding of this work. Our results suggested that this nanomaterial has great potential as a gene carrier agent in non-viral based therapy, with low cytotoxicity, good transfection efficiency, and low cell damage in small amounts of nanomaterials in metabolomic tests.

As a contribution to the Large-Scale Biosphere-Atmosphere Experiment in Amazonia - Cooperative LBA Airborne Regional Experiment (LBA-CLAIRE-2001) field campaign in the heart of the Amazon Basin, we analyzed the temporal and spatial dynamics of the urban plume of Manaus City during the wet-to-dry season transition period in July 2001. During the flights, we performed vertical stacks of crosswind transects in the urban outflow downwind of Manaus City, measuring a comprehensive set of trace constituents including O(3), NO, NO(2), CO, VOC, CO(2), and H(2)O. Aerosol loads were characterized by concentrations of total aerosol number (CN) and cloud condensation nuclei (CCN), and by light scattering properties. Measurements over pristine rainforest areas during the campaign showed low levels of pollution from biomass burning or industrial emissions, representative of wet season background conditions. The urban plume of Manaus City was found to be joined by plumes from power plants south of the city, all showing evidence of very strong photochemical ozone formation. One episode is discussed in detail, where a threefold increase in ozone mixing ratios within the atmospheric boundary layer occurred within a 100 km travel distance downwind of Manaus. Observation-based estimates of the ozone production rates in the plume reached 15 ppb h(-1). Within the plume core, aerosol concentrations were strongly enhanced, with Delta CN/Delta CO ratios about one order of magnitude higher than observed in Amazon biomass burning plumes. Delta CN/Delta CO ratios tended to decrease with increasing transport time, indicative of a significant reduction in particle number by coagulation, and without substantial new particle nucleation occurring within the time/space observed. While in the background atmosphere a large fraction of the total particle number served as CCN (about 60-80% at 0.6% supersaturation), the CCN/CN ratios within the plume indicated that only a small fraction (16 +/- 12 %) of the plume particles were CCN. The fresh plume aerosols showed relatively weak light scattering efficiency. The CO-normalized CCN concentrations and light scattering coefficients increased with plume age in most cases, suggesting particle growth by condensation of soluble organic or inorganic species. We used a Single Column Chemistry and Transport Model (SCM) to infer the urban pollution emission fluxes of Manaus City, implying observed mixing ratios of CO, NO(x) and VOC. The model can reproduce the temporal/spatial distribution of ozone enhancements in the Manaus plume, both with and without accounting for the distinct (high NO(x)) contribution by the power plants; this way examining the sensitivity of ozone production to changes in the emission rates of NO(x). The VOC reactivity in the Manaus region was dominated by a high burden of biogenic isoprene from the background rainforest atmosphere, and therefore NO(x) control is assumed to be the most effective ozone abatement strategy. Both observations and models show that the agglomeration of NO(x) emission sources, like power plants, in a well-arranged area can decrease the ozone production efficiency in the near field of the urban populated cores. But on the other hand remote areas downwind of the city then bear the brunt, being exposed to increased ozone production and N-deposition. The simulated maximum stomatal ozone uptake fluxes were 4 nmol m(-2) s(-1) close to Manaus, and decreased only to about 2 nmol m(-2) s(-1) within a travel distance >1500 km downwind from Manaus, clearly exceeding the critical threshold level for broadleaf trees. Likewise, the simulated N deposition close to Manaus was similar to 70 kg N ha(-1) a(-1) decreasing only to about 30 kg N ha(-1) a(-1) after three days of simulation.

C,C-Dicyanoketenimines 10a-c were generated by flash vacuum thermolysis of ketene NS-acetals 9a-c or by thermal or photochemical decomposition of alpha-azido-,beta-cyanocinnamonitrile 11. In the latter reaction, 3,3-dicyano-2-phenyl-1-azirine 12 is also formed. IR spectroscopy of the keteniminines isolated in Ar matrixes or as neat films, NMR spectroscopy of 10c, and theoretical calculations (B3LYP/6-31G*) demonstrate that these ketenimines have variable geometry, being essentially linear along the CCN-R framework in polar media (neat films and solution), but in the gas phase or Ar matrix they are bent, as is usual for ketenimines. Experiments and calculations agree that a single CN substituent as in 13 is not enough to enforce linearity, and sulfonyl groups are less effective that cyano groups in causing linearity. C,C-Bis(methylsulfonyl)ketenimines 4-5 and a C-cyano-C-(methylsulfonyl)ketenimine 15 are not linear. The compound p-O2NC6H4N=C= C(COOMe)2 previously reported in the literature is probably somewhat linearized along the CCNR moiety. A computational survey (B3LYP/6-31G*) of the inversion barrier at nitrogen indicates that electronegative C-substituents dramatically lower the barrier; this is also true of N-acyl substituents. Increasing polarity causes lower barriers. Although N-alkylbis(methylsulfonyl)ketenimines are not calculated to be linear, the barriers are so low that crystal lattice forces can induce planarity in N-methylbis(methylsulfonyl)ketenimine 3.

Os aspectos fisiológicos e bioquímicos influenciados pelo silício e que condicionam reações de resistência a pragas no cacaueiro ainda são pouco conhecidos. O objetivo deste experimento foi avaliar os efeitos da aplicação de silício sobre a fotossíntese, sobre o teor foliar de fenóis solúveis totais e a sua influência na preferência de Toxoptera aurantii em genótipos de cacau. O experimento foi conduzido em casa de vegetação, no delineamento de blocos casualizados, com quatro repetições, em arranjo fatorial 3 x 3, constituído por três genótipos (TSH 1188, CCN 51 e Catongo), duas doses de silicato de potássio (3 e 6 mL L-1), aplicadas por via foliar e um controle pulverizado somente com água. Foram realizadas avaliações das trocas gasosas, da fluorescência da clorofila a e do teor de compostos fenólicos foliar, além de um teste de preferência com chance de livre escolha com T. aurantii. Os genótipos TSH 1188 e CCN 51 apresentam maior eficiência fotoquímica e fotossintética, comparados com o Catongo. Independentemente do genótipo, a aplicação de silicato de potássio não alterou o índice de desempenho das plantas (PI ABS e PI TOTAL); no entanto, a dose 3 mL L-1 aumentou a fotossíntese líquida e o teor de fenóis solúveis totais. A dose 6 mL L-1 reduziu a preferência de T. aurantii pelas folhas do genótipo TSH 1188. A aplicação de silicato de potássio é promissora no aumento da resistência do cacaueiro a T. aurantii.

The objective of the present work was to evaluate 27 progenies of cocoa crosses considering the agronomic traits and select F1 plants within superior crosses. The experiment was installed in March 2005, in the Experimental Station Joaquim Bahiana (ESJOB), in Itajuipe, Bahia. The area of the experiment is of approximately 3 ha, with a total of 3240 plants. Thirteen evaluations of vegetative brooms, five of cushion brooms and 15 of number of pods per plant were accomplished. Thirty pollinations were made for each selected plant to test for self-compatibility. The production, based on the number of pods per plant, and resistance to witches´ broom indicated CEPEC 94 x CCN 10, RB 39 x CCN 51 and CCN 10 x VB 1151 as superior progenies. All selections tested were self-compatible. The analyses of progenies and individual tree data, associated to visual field observations, allowed the selection of 17 plants which were included in a network of regional tests to determine the phenotypic stability.

A Internet, conforme a conhecemos, foi projetada com base na pilha de protocolos TCP/IP, que foi desenvolvida nos anos 60 e 70 utilizando um paradigma centrado nos endereços individuais de cada máquina (denominado host-centric). Este paradigma foi extremamente bem-sucedido em interligar máquinas através de encaminhamento baseado no endereço IP. Estudos recentes demonstram que, parte significativa do tráfego atual da Internet centra-se na transferência de conteúdos, em vez das tradicionais aplicações de rede, conforme foi originalmente concebido. Surgiram então novos modelos de comunicação, entre eles, protocolos de rede ponto-a-ponto, onde cada máquina da rede pode efetuar distribuição de conteúdo (denominadas de redes peer-to-peer), para melhorar a distribuição e a troca de conteúdos na Internet. Por conseguinte, nos últimos anos o paradigma host-centric começou a ser posto em causa e apareceu uma nova abordagem de Redes Centradas na Informação (ICN - information-centric networking). Tendo em conta que a Internet, hoje em dia, basicamente é uma rede de transferência de conteúdos e informações, porque não centrar a sua evolução neste sentido, ao invés de comunicações host-to-host? O paradigma de Rede Centrada no Conteúdo (CCN - Content Centric Networking) simplifica a solução de determinados problemas de segurança relacionados com a arquitetura TCP/IP e é uma das principais propostas da nova abordagem de Redes Centradas na Informação. Um dos principais problemas do modelo TCP/IP é a proteção do conteúdo. Atualmente, para garantirmos a autenticidade e a integridade dos dados partilhados na rede, é necessário garantir a segurança do repositório e do caminho que os dados devem percorrer até ao seu destino final. No entanto, a contínua ineficácia perante os ataques de negação de serviço praticados na Internet, sugere a necessidade de que seja a própria infraestrutura da rede a fornecer mecanismos para os mitigar. Um dos principais pilares do paradigma de comunicação da CCN é focalizar-se no próprio conteúdo e não na sua localização física. Desde o seu aparecimento em 2009 e como consequência da evolução e adaptação a sua designação mudou atualmente para Redes de Conteúdos com Nome (NNC – Named Network Content). Nesta dissertação, efetuaremos um estudo de uma visão geral da arquitetura CCN, apresentando as suas principais características, quais os componentes que a compõem e como os seus mecanismos mitigam os tradicionais problemas de comunicação e de segurança. Serão efetuadas experiências com o CCNx, que é um protótipo composto por um conjunto de funcionalidades e ferramentas, que possibilitam a implementação deste paradigma. O objetivo é analisar criticamente algumas das propostas existentes, determinar oportunidades, desafios e perspectivas para investigação futura.

OBJETIVO: Avaliar a acurácia da medida da translucência nucal (TN) entre onze e treze semanas e seis dias como marcador ultra-sonográfico para rastreamento de cardiopatias congênitas (CC). MÉTODOS: Estudo multicêntrico retrospectivo, no qual foram analisadas gestações únicas de fetos euplóides. A medida da TN foi realizada no exame de primeiro trimestre quando os fetos tinham entre 45 e 84 mm de comprimento cabeça-nádega (CCN), segundo os critérios estabelecidos pela Fetal Medicine Foundation. Os casos foram seguidos até um mês após o parto para avaliar a presença de CC. RESULTADOS: Foram analisadas 3.664 gestações, das quais vinte recém-nascidos apresentaram alguma cardiopatia congênita até o primeiro mês de vida (prevalência de 0,55%). A mediana da TN nos fetos com CC foi de 1,70 mm e nos fetos sem CC foi de 1,60 mm; entretanto, não houve diferença significativa entre as duas medianas (Teste de Mann-Whitney, p>0,05). A sensibilidade da TN na detecção de CC variou de 15% a 20%, com probabilidade de falso-positivos de 86,4% a 97,9%, dependendo do ponto de corte utilizado. A razão de chance para CC foi alta, quando comparada com as indicações clássicas de ecocardiografia fetal, variando de 4,7 a 33,7, de acordo com o ponto de corte utilizado. CONCLUSÃO: Apesar da baixa sensibilidade do teste, a TN aumentada é um importante fator de risco para CC, devendo ser incluída na estratégia do seu rastreamento pré-natal.

En la etapa embrionaria temprana de los vertebrados, las células de la cresta neural (CCN) se segregan del tubo neural y se distribuyen con patrones temporales y espaciales muy precisos, contribuyendo a la formación de muchos derivados: neuronas y glía del sistema nervioso periférico, parte del sistema endócrino, sistema pigmentario y la mayor parte de los tejidos cráneo-faciales. Las bases morfogenéticas de esta movilización de las CCN se asocian con la disponibilidad de componentes de la matriz extracelular, con fenómenos de apoptosis selectiva, y con la expresión de genes homeóticos, de proteínas transportadoras de retinoides y de receptores de ácido retinoico (AR). En consecuencia, el mecanismo que define el comportamiento de las CCN migratorias asume una especial importancia, desde que cualquier fallo producido en estos "reguladores topográficos" podrá alterar la ordenada traslocación de CN, induciendo una dismorfogénesis. (...) Objetivos: 1) Analizar el comportamiento dinámico de la etapa migratoria temprana de las CCN de niveles cefálico y troncal expuestas a etanol o AR in vitro. 2) Evaluar la posible reversibilidad de los efectos del etanol y del AR sobre la morfología y dinámica migratoria de las CCN de niveles cefálico y troncal in vitro. 3) Analizar los componentes del citoesqueleto asociados con los cambios de forma y motilidad celular en CCN de niveles cefálico y troncal expuestas a etanol y AR in vitro. Los resultados del proyecto permitirán aportar al conocimiento de los mecanismos básicos que regulan la movilidad de poblaciones celulares de gran importancia para el desarrollo humano. Los datos obtenidos servirán de base para futuros enfoques de biología molecular tendientes a innovar aspectos del diagnóstico, pronóstico y prevención de patologías humanas de creciente prevalencia provocadas por el etanol (FAS) y los retinoides (RAE).

La motilidad celular orientada (o mecanismo quimiotáctico de orientación), es una respuesta celular a señales moleculares de su micro-ambiente necesaria para modular la distribución celular en sitios específicos y con elevada precisión. Nuestra hipótesis establece que la migración orientada de células neurales es regulada por gradientes de concentración de moléculas solubles liberadas por sus regiones "blanco". Este proyecto es continuación del estudio de la quimiotaxis de células de cresta neural (CCN) y de neuronas ventriculares (NV) inducida respectivamente por factores difusibles de la región del futuro ganglio ciliar y del bulbo olfatorio.En el sistema de CCN, hemos caracterizado como moléculas quimiotácticas a la quimioquina Stromal Cell-Derived Factor-1, y los factores tróficos Stem Cell Factor y Neurotrophic Factor-3, habiendo determinado la expresión de sus respectivos receptores CXCR4, TrkC y p75 en la población de CCN mesencefálicas de ambrión de pollo. Actualmente, estamos desarrollando experimentos con el Ciliary Neurotrophic Factor y factores de la familia Bone Morphogenetic Proteins. Además de la estrategia experimental in vitro, hemos determinado en el embrión entero la expresión de las moléculas quimioatractantes mediante hibridación in situ del ARNm y la presencia de las respectivas proteínas mediante inmunocitoquímica. En el sistema de NV, estamos analizando la motilidad celular en relación con moléculas liberadas por el bulbo olfatorio. En los dos sistemas biológicos, estamos analizando elementos de la transducción de señales y cambios en el citoesqueleto, en ambos casos asociados con la respuesta temprana en la orientación quimiotáctica de la célula. Asimismo, en ambos sistemas biológicos, evaluamos los efectos del etanol sobre la migración y distribución celular, en condiciones equivalentes a las que inducen el Sindrome Fetal Alcohólico en mamíferos.En base a resultados ya obtenidos en experimentos in vitro, en la presente etapa intentaremos su caracterización in vivo mediante el bloqueo funcional de las moléculas quimiotácticas (y/o sus receptores) sobre embriones enteros, mediante silenciamiento con ARNsi (o morfolinos específicos) mediante electroporación, y posterior determinación de la distribución celular mediante marcadores específicos anti-CCN (o lipofílicos de tipo DiI).Los resultados permitirán mejorar el conocimiento del mecanismo de la migración celular orientada y aportar al diseño de recursos diagnósticos, terapéuticos o de control de anomalías embrionarias o patologías tumorales por mala distribución celular como las Neurocristopatías, o inducidas por tóxicos exógenos como el Sindrome Fetal Alcohólico.
