Serum butyrylcholinesterase (BCHE) activity is associated with obesity, blood pressure and biomarkers of cardiovascular and diabetes risk. We have conducted a genome-wide association scan to discover genetic variants affecting BCHE activity, and to clarify whether the associations between BCHE activity and cardiometabolic risk factors are caused by variation in BCHE or whether BCHE variation is secondary to the metabolic abnormalities. We measured serum BCHE in adolescents and adults from three cohorts of Australian twin and family studies. The genotypes from approximately 2.4 million single-nucleotide polymorphisms (SNPs) were available in 8791 participants with BCHE measurements. We detected significant associations with BCHE activity at three independent groups of SNPs at the BCHE locus (P = 5.8 x 10(-262), 7.8 x 10(-47), 2.9 x 10(-12)) and at four other loci: RNPEP (P = 9.4 x 10(-16)), RAPH1-ABI2 (P = 4.1 x 10(-18)), UGT1A1 (P = 4.0 x 10(-8)) and an intergenic region on chromosome 8 (P = 1.4 x 10(-8)). These loci affecting BCHE activity were not associated with metabolic risk factors. On the other hand, SNPs in genes previously associated with metabolic risk had effects on BCHE activity more often than can be explained by chance. In particular, SNPs within FTO and GCKR were associated with BCHE activity, but their effects were partly mediated by body mass index and triglycerides, respectively. We conclude that variation in BCHE activity is due to multiple variants across the spectrum from uncommon/large effect to common/small effect, and partly results from (rather than causes) metabolic abnormalities.

Capillary electrophoresis with electrochemiluminescene detection was used to characterize procaine hydrolysis as a probe for butyrylcholinesterase by in vitro procaine metabolism in plasma with butyrylcholinesterase acting as bioscavenger. Procaine and its metabolite N,N-diethylethanolamine were separated at 16 kV and then detected at 1.25 V in the presence of 5.0 mM Ru(bpy)(3)(2+), with the detection limits of 2.4 x 10(-7) and 2.0 x 10(-8) mol/L (S/N=3), respectively. The Michaelis constant K-m value was 1.73 x 10(-4) mol/L and the maximum velocity V-max was 1.62 x 10(-6) mol/L/min. Acetylcholine bromide and choline chloride presented inhibition effects on the enzymatic cleavage of procaine, with the 50% inhibition concentration (IC50) of 6.24 x 10(-3) and 2.94 x 10(-4) mol/L.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder that has been associated, sometimes controversially, with polymorphisms in a number of genes. Recently the butyrylcholinesterase K variant (BCHE K) allele has been shown to act in synergy with the apolipoprotein E epsilon4 (APOE epsilon4) allele to promote risk for AD. Most subsequent replicative studies have been unable to confirm these findings. We have conducted a case-control association study using a clinically well defined group of late onset AD patients (n=175) and age and sex matched control subjects (n=187) from the relatively genetically homogeneous Northern Ireland population to test this association. The spacer structure of homobivalent quinazolinimes acting as potent acetyl-(AChE)- and butyrylcholinesterase (BChE) inhibitors was chemically modified introducing tertiary amine and acyl-amide moieties, and the activities at both ChEs were evaluated. Molecular docking was applied to explain the data and probe the capacity of the mid-gorge site of both ChEs. The novel spacer structures considerably alter the biological profile of bivalent quinazolinimines with regard to both inhibitory activity and selectivity. Mutual interaction of binding to the various sites of the enzymes was further investigated by applying also different spacer lengths and ring sizes of the alicycle of the tricyclic quinazolinimines. In order to achieve selectivity toward BChE and to improve inhibitory activities, the spacer structure was optimized and identified a highly potent and selective BChE inhibitor. In a previous study, we showed that purified commercial esterase activity can be detected in a chemiluminescent assay based on the hydrolysis of 2-methyl-1-propenylbenzoate (MPB) to 2-methyl-1-propenol, which is subsequently oxidized by the horseradish peroxidase (HRP)-H2O2 system. The purpose of this study was to verify the applicability of this assay to human serum. The existence of an esterase activity capable of hydrolysing MPB is indicated by the fact that the MPB-scruin-HRP-H2O2 System consumes oxygen and emits light. Both signals were abolished by prior serum heat inactivation and were preserved when serum was stored at less than or equal to4 degreesC. Addition of aliesterase inhibitors, such as fluoride ion and trichlorfon or the cholinesterase inhibitor eserine, totally prevents light emission. The butyrylcholinesterase-specific substrate benzoylcholine causes a delay in both O-2 uptake and light emission, while the specific acetylcholinesterase substrate, acetyl-beta -methylcholine, had practically no effect. Purified butyrylcholinesterase, but not acetylcholinesterase, triggered light emission. The finding that butyryleholinesterase is responsible for the hydrolysis of MPB in serum should serve as the basis for the development of a specific chemiluminescent assay for this enzyme. Background: Plasma cholinesterase activity is known to be correlated with plasma triglycerides, HDL- and LDL-cholesterol, and other features of the metabolic syndrome. A role in triglyceride metabolism has been proposed. Genetic variants that decrease activity have been studied extensively, but the factors contributing to overall variation in the population are poorly understood. We studied plasma cholinesterase activity in a sample of 2200 adult twins to assess covariation with cardiovascular risk factors and components of the metabolic syndrome, to determine the degree of genetic effects on enzyme activity, and to search for quantitative trait loci affecting activity. Methods and Results: Cholinesterase activity was lower in women than in men before the age of 50, but increased to activity values similar to those in males after that age. There were highly significant correlations with variables associated with the metabolic syndrome: plasma triglyceride, HDL- and LDL-cholesterol, apolipoprotein B and E, urate, and insulin concentrations; gamma-glutamyltransferase and aspartate and alanine aminotransferase activities; body mass index; and blood pressure. The heritability of plasma cholinesterase activity was 65%. Linkage analysis with data from the dizygotic twin pairs showed suggestive linkage on chromosome 3 at the location of the cholinesterase WHO gene and also on chromosome 5. Conclusions: Our results confirm and extend the connection between cholinesterase, cardiovascular risk factors, and metabolic syndrome. They establish a substantial heritability for plasma cholinesterase activity that might be attributable to variation near the structural gene and at an independent locus. Rivastigmine is a very important drug prescribed for the treatment of Alzheimer's disease (AD) symptoms. It is a dual inhibitor, in that it inhibits both acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE). For our screening program on the discovery of new rivastigmine analogue hits for human butyrylcholinesterase (hBuChE) inhibition, we investigated the interaction of this inhibitor with BuChE using the complimentary approach of the biophysical method, saturation transfer difference (STD)-NMR and molecular docking. This allowed us to obtain essential information on the key binding interactions between the inhibitor and the enzyme to be used for screening of hit compounds. The main conclusions obtained from this integrated study was that the most dominant interactions were (a) H-bonding between the carbamate carbonyl of the inhibitor and the NH group of the imidazole unit of H434, (b) stacking of the aromatic unit of the inhibitor and the W82 aromatic unit in the choline binding pocket via pi-pi interactions and (c) possible CH/pi interactions between the benzylic methyl group and the N-methyl groups of the inhibitor and W82 of the enzyme.

The first line medication for mild to moderate Alzheimer s disease (AD) is based on cholinesterase inhibitors which prolong the effect of the neurotransmitter acetylcholine in cholinergic nerve synapses which relieves the symptoms of the disease. Implications of cholinesterases involvement in disease modifying processes has increased interest in this research area. The drug discovery and development process is a long and expensive process that takes on average 13.5 years and costs approximately 0.9 billion US dollars. Drug attritions in the clinical phases are common due to several reasons, e.g., poor bioavailability of compounds leading to low efficacy or toxic effects. Thus, improvements in the early drug discovery process are needed to create highly potent non-toxic compounds with predicted drug-like properties. Nature has been a good source for the discovery of new medicines accounting for around half of the new drugs approved to market during the last three decades. These compounds are direct isolates from the nature, their synthetic derivatives or natural mimics. Synthetic chemistry is an alternative way to produce compounds for drug discovery purposes. Both sources have pros and cons. The screening of new bioactive compounds in vitro is based on assaying compound libraries against targets. Assay set-up has to be adapted and validated for each screen to produce high quality data. Depending on the size of the library, miniaturization and automation are often requirements to reduce solvent and compound amounts and fasten the process. In this contribution, natural extract, natural pure compound and synthetic compound libraries were assessed as sources for new bioactive compounds. The libraries were screened primarily for acetylcholinesterase inhibitory effect and secondarily for butyrylcholinesterase inhibitory effect. To be able to screen the libraries, two assays were evaluated as screening tools and adapted to be compatible with special features of each library. The assays were validated to create high quality data. Cholinesterase inhibitors with various potencies and selectivity were found in natural product and synthetic compound libraries which indicates that the two sources complement each other. It is acknowledged that natural compounds differ structurally from compounds in synthetic compound libraries which further support the view of complementation especially if a high diversity of structures is the criterion for selection of compounds in a library.

A ação inibitória dos organofosforados sobre as esterases, por ser específica, pode ser empregada como um eficiente biomarcador da exposição de seres vivos aos organofosforados. A inibição da acetilcolinesterase (AChE; EC 3.1.1.7) provoca acúmulo do neurotransmissor acetilcolina nas fendas sinápticas colinérgicas, o que pode resultar na morte do indivíduo. Outra atividade também afetada por organofosforados é a da enzima carboxilesterase (CarbE; EC 3.1.1.1). CarbE estão envolvidas na fase I da biotransformação de xenobióticos e atuam como captadoras (scavengers) de organofosfatos, incluindo os formados pela biotransformação dos organofosforados. As CarbE estudadas até hoje se ligam com maior velocidade aos organofosfatos do que as colinesterases. Por isto se admite que CarbE possam diminuir, por captação estequiométrica, a ligação tóxica de moléculas de organofosfatos às acetilcolinesterases das sinapses colinérgicas e das placas motoras dos músculos. Experimentos realizados em nosso laboratório mostraram que a atividade da CarbE está aproximadamente 50% menor no soro e no fígado de pacus submetidos à hipoxia. Por causa disso, em razão de uma possível diminuição da capacidade captadora da CarbE, decidimos verificar se o pacu em hipoxia seria mais sensível aos agrotóxicos organofosforados. Para este propósito foram colocados seis pacus divididos em dois tanques. No primeiro tanque, os animais foram submetidos a 24 horas de hipoxia seguidos por mais 4 horas de exposição ao organofosforado metilparation em duas concentrações diferentes (0,02 ou 0,01 mg / L). No segundo tanque os animais permaneceram em normoxia durante o mesmo período de 24 horas e depois foram expostos ao metilparation como no primeiro tanque. As atividades da AChE ensaiada com acetiltiocolina, a da butirilcolinesterase (BChE) ensaiada com butiriltiocolina e a da CarbE ensaiada com p-nitrofenilacetato foram avaliadas no soro, fígado, cérebro, músculo e coração dos pacus. Houve redução de aproximadamente 35% da atividade de CarbE no soro dos pacus submetidos a 24 horas de hipoxia. Uma queda de 85% na atividade de CarbE do soro foi observada nos animais que sofreram hipoxia e subsequente exposição a 0,02 mg de metilparation por litro. Com metilparation a 0,01 mg/L a diminuição observada foi de 48,2%. No músculo dos pacus expostos a 0,02 mg/L, as atividades de AChE e BChE cairam pela metade quando os mesmos foram submetidos à hipoxia quando comparados a animais que permaneceram em normoxia. Nos diversos tecidos dos pacus expostos a 0,01 mg/L de metilparation não observamos diferenças significativas nas atividades de AChE, BChE ou CarbE. Concluímos que a duplicação da concentração de metilparation de 0,01 para 0,02 mg/L levou à atividade residual de CarbE do soro de 51,8% para 15%. A ausência de mudanças nas atividades das esterases dos tecidos de animais expostos a 0,01 mg/L entre os grupos hipoxia e normoxia deve ter ocorrido porque a concentração de organofosforado não foi suficiente para superar a primeira barreira de proteção das esterases séricas e atingir os tecidos. Mas, no experimento com 0,02 mg/L de metilparation, as inibições de AChE e de BChE no músculo dos animais em hipoxia podem ser explicadas pela diminuição da atividade de CarbE do soro dos pacus.

Quando as esterases acetilcolinesterase (AChE), butirilcolinesterase (BChE) e carboxilesterase (CarbE) hidrolisam ésteres de fosfato seus sítios ativos sofrem fosfatação inibitória. Por isto, tal fosfatação pode proteger seres vivos contra o espalhamento de xenobióticos organofosforados dentro de seus corpos, já que estas enzimas têm a capacidade de captar moléculas de pesticidas organofosforados estequiometricamente. Os organismos terrestres vivem em um ambiente com mais oxigênio do que os organismos aquáticos. Na água, quando o nível de oxigênio atinge aproximadamente 2,6 mg/L o ambiente está em hipoxia. Este fenômeno afeta ecossistemas aquáticos, uma vez que muitos organismos não conseguem se adaptar à baixa do oxigênio. Estudamos peixes em hipoxia e hiperoxia para entender melhor a bioquímica do funcionamento de suas enzimas captadoras de organofosforados quando eles estão expostos às variações físico-químicas de seus habitats. Dois grupos de no mínimo seis pacus (Piaractus mesopotamicus), seis peixes dourados (Carassius auratus auratus), seis tilápias (Oreochromis niloticus niloticus), seis piavussus (Leporinus macrocephalus), seis apaiaris (Astronotus ocellatus), ou seis carpas (Cyprinus carpio carpio) foram aclimatados à temperatura ambiente em dois aquários de 250 L. No primeiro aquário, pelo menos três animais ensaio de cada espécie sofreram hipoxia por diminuição da concentração de oxigênio até 0,5 mg/L através de borbulhamento de nitrogênio na água. Quando estes animais atingiram a hipoxia foram mantidos a 0,5 mg/L de oxigênio por 6, 8, 24 ou, no máximo, por 42 horas. Três peixes controle de cada espécie foram mantidos em normoxia (4,5 até 7,0 mg/L de oxigênio). Após estes tempos houve a retirada de cerca de 3,5 mL de sangue e dos fígados. Depois de coagular, o sangue foi centrifugado para retirada do soro sobrenadante, que foi usado como amostra para ensaios das esterases. Os fígados foram armazenados em freezer a -70 C e, no momento do ensaio, homogeneizados e centrifugados para obter as frações citosólica e microssomal. As atividades das esterases foram ensaiadas em espectrofotômetro com os substratos acetiltiocolina, butiriltiocolina ou p-nitrofenilacetato. As atividades sobre p-nitrofenilacetato (CarbE) do soro e do fígado sofreram queda em todos os exemplares das espécies submetidos à hipoxia. Tipicamente, esta atividade caiu cerca de 50% nos soros de pacus mantidos por 42 h sob concentrações de oxigênio abaixo de 1,0 mg/L. O tempo para que ocorresse a queda desta atividade enzimática variou de espécie para espécie.

A discussion of the most interesting results obtained in our laboratories, during the supercritical CO(2) extraction of bioactive compounds from microalgae and volatile oils from aromatic plants, was carried out. Concerning the microalgae, the studies on Botryococcus braunii and Chlorella vulgaris were selected. Hydrocarbons from the first microalgae, which are mainly linear alkadienes (C(23)-C(31)) with an odd number of carbon atoms, were selectively extracted at 313 K increasing the pressure up to 30.0 MPa. These hydrocarbons are easily extracted at this pressure, since they are located outside the cellular walls. The extraction of carotenoids, mainly canthaxanthin and astaxanthin, from C. vulgaris is more difficult. The extraction yield of these components at 313 K and 35.0 MPa increased with the degree of crushing of the microalga, since they are not extracellular. On the other hand, for the extraction of volatile oils from aromatic plants, studies on Mentha pulegium and Satureja montana L were chosen. For the first aromatic plant, the composition of the volatile and essential oils was similar, the main components being the pulegone and menthone. However, this volatile oil contained small amounts of waxes, which content decreased with decreasing particle size of the plant matrix. For S. montana L it was also observed that both oils have a similar composition, the main components being carvacrol and thymol. The main difference is the relative amount of thymoquinone, which content can be 15 times higher in volatile oil. This oxygenated monoterpene has important biological activities. Moreover, experimental studies on anticholinesterase activity of supercritical extracts of S. montana were also carried out. The supercritical nonvolatile fraction, which presented the highest content of the protocatechuic, vanilic, chlorogenic and (+)-catechin acids, is the most promising inhibitor of the enzyme butyrylcholinesterase. In contrast, the Soxhlet acetone extract did not affect the activity of this enzyme at the concentrations tested. (C) 2011 Elsevier B.V. All rights reserved.
