Phi Beta Delta Presentation - Lawrence Bruner

A year ago I had the privilege of being inducted into Phi Beta Delta, and at that time I was invited to speak at the next induction ceremony, which it is my pleasure to do today. I am delighted to have this opportunity to congratulate our scholarship recipient, our new inductees, Peter Gleick, President of the Pacific Institute for Studies in Development, Environment and Security, and Merlyn Carlson, Director of the Nebraska Department of Agriculture and a member of the Governor's cabinet, on their honorary memberships in the society. I want to acknowledge Harriet Turner, Director of International affairs, for her fine work on behalf of the university and us all, and I want to thank all members of this society of international scholars for the important work you do, not only for Nebraska but, indeed, for our world.

Catharine Waugh McCullouch papers in the Mary Earhart Dillon collection, 1869-1945

Collection primarily documents McCulloch's research on women's legal status, and her work with the Illinois Equal Suffrage Association, the National American Woman Suffrage Association, and the League of Women Voters. There is also documentation of women in the legal profession, of McCulloch's friendships with the other women suffragists and lawyers, and some biographical material. The papers contain little information about her family or social life.

The Penicillium chrysogenum aclA gene encodes a broad-substrate-specificity acyl-coenzyme A ligase involved in activation of adipic acid, a side-chain precursor for cephem antibiotics

Activation of the cephalosporin side-chain precursor to the corresponding CoA-thioester is an essential step for its incorporation into the P-lactam backbone. To identify an acyl-CoA ligase involved in activation of adipate, we searched in the genome database of Penicillium chrysogenum for putative structural genes encoding acyl-CoA ligases. Chemostat-based transcriptome analysis was used to identify the one presenting the highest expression level when cells were grown in the presence of adipate. Deletion of the gene renamed aclA, led to a 32% decreased specific rate of adipate consumption and a threefold reduction of adipoyl-6-aminopenicillanic acid levels, but did not affect penicillin V production. After overexpression in Escherichia coli, the purified protein was shown to have a broad substrate range including adipate. Finally, protein-fusion with cyan-fluorescent protein showed co-localization with microbody-borne acyl-transferase. Identification and functional characterization of aclA may aid in developing future metabolic engineering strategies for improving the production of different cephalosporins. (C) 2009 Elsevier Inc.
