Estudou-se as alterações clínico-patológicas e laboratoriais em equinos, inoculados experimentalmente com a peçonha de Bothropoides jararaca, Bothrops jararacussu, Bothrops moojeni e Bothropoides neuwiedi, com a finalidade de fornecer subsídios para o diagnóstico do envenenamento pela picada dessas. Os venenos liofilizados foram diluídos em 1ml de solução fisiológica e administrados a seis equinos, por via subcutânea, nas doses de 0,5 e 1mg/kg (B. jararaca), 0,8 e 1,6mg/kg (B. jararacussu), 0,205mg/kg (B. moojeni) e 1mg/kg (B. neuwiedi). Todos os equinos, menos os que receberam o veneno de B. jararacussu, morreram. Os sinais clínicos iniciaram-se entre 8min e 2h10min após a inoculação. O período de evolução variou, nos quatro casos de êxito letal, de 24h41min a 70h41min, e nos dois equinos que se recuperaram foi de 16 dias. O quadro clínico, independente do tipo de veneno e das doses, caracterizou-se por aumento de volume no local da inoculação, arrastar da pinça do membro inoculado no solo, inquietação, apatia, diminuição da resposta aos estímulos externos, mucosas pálidas e hemorragias. Os exames laboratoriais revelaram anemia normocítica normocrômica com progressiva diminuição no número de hemácias, da hemoglobina e do hematócrito, e leucocitose por neutrofilia. Houve aumento de alamina aminotransferase, creatinaquinase, dehidrogenase láctica, ureia e glicose, bem como aumento do tempo de ativação da protrombina e do tempo de tromboplastina parcial ativada. Os achados de necropsia foram extensas hemorragias no tecido subcutâneo, com presença de sangue não coagulado e em boa parte associadas a edema (edema hemorrágico), que se estendia desde o local da inoculação até as regiões cervical, torácica, escapular e membro. Na periferia das áreas hemorrágicas havia predominantemente edema gelatinoso. Havia ainda presença de grande quantidade de líquido sanguinolento nas cavidades torácica, pericárdica e abdominal. Não foram encontradas alterações histológicas significativas.

Throughout the analysis of the collections JJ, CEVAP, IBSP, ZUEC and UEL, a study on the ophiofauna of Botucatu was made. We present a preliminary list of the species of snakes here found, with small comments on their natural history. The number of specimens studied was 943. We registered, for Botucatu, 51 species of snakes, distributed in 31 genera and 6 families. The families here found were Anomalepididae (1 spp.), Boidae (1 spp.), Colubridae (7 spp.), Dipsadidae (32 spp.), Elapidae (3 spp.) and Viperidae (7 spp.). The analysis of the relative abundance shows that Dipsadidae was the most abundant family, with n=425 (44,83%), followed by Viperidae, with n=388 (40,93%), Boidae, n=62 (6,54%), Colubridae, n=57 (6,01%), Elapidae, n=15 (1,6%) and, at last, Anomalepididae, with n=1 (0,1%). The five more representative species were Crotalus durissus (n=135, 14,31%), Oxyrhopus guibei (n=123, 12,8%) Bothropoides jararaca (N=121, 12,6%), Bothropoides neuwiedi (N=95, 9,88%) and Sibynomorphus mikani (N=65, 6,76%). A higher number of individuals collected was registered for the months of january to april that, together, sum up to almost 50% of the total. The months of june to september registered a lower number of individuals. The spacial distribution analysis shows that a higher number of snakes was found on the country areas of Botucatu (n=270, 41,54%), specially on pastures. Due to the lack of studies of Botucatu’s ophiofauna, this list is probably underestimated. It is mandatory that future studies approaching this group and its ecological components on this region are made, using appropriate sampling methodologies, in order to form an accurate list of the species of snakes of Botucatu

The venom gland of viperid snakes has a central lumen where the venom produced by secretory cells is stored. When the venom is lost from the gland, the secretory cells are activated and new venom is produced. The production of new venom is triggered by the action of noradrenaline on both alpha(1)- and beta-adrenoceptors in the venom gland. In this study, we show that venom removal leads to the activation of transcription factors NF kappa B and AP-1 in the venom gland. In dispersed secretory cells, noradrenaline activated both NF kappa B and AP-1. Activation of NF kappa B and AP-1 depended on phospholipase C and protein kinase A. Activation of NF kappa B also depended on protein kinase C. Isoprenaline activated both NF kappa B and AP-1, and phenylephrine activated NF kappa B and later AP-1. We also show that the protein composition of the venom gland changes during the venom production cycle. Striking changes occurred 4 and 7 days after venom removal in female and male snakes, respectively. Reserpine blocks this change, and the administration of alpha(1)- and beta-adrenoceptor agonists to reserpine-treated snakes largely restores the protein composition of the venom gland. However, the protein composition of the venom from reserpinized snakes treated with alpha(1)- or beta-adrenoceptor agonists appears normal, judging from SDS-PAGE electrophoresis. A sexual dimorphism in activating transcription factors and activating venom gland was observed. Our data suggest that the release of noradrenaline after biting is necessary to activate the venom gland by regulating the activation of transcription factors and consequently regulating the synthesis of proteins in the venom gland for venom production.

The biological activity of the proline rich decapeptde Bj PRO 10c a processing product of the C type natriuretic peptide precursor protein, expressed in the brain and the venom gland of the pit viper Bothrops jararaca, was originally attributed to the inhibition of the somatic angiotensm converting enzyme activity with subsequent ant hypertensive effect However recent results suggest broader biological activity may also be involved in the cardiovascular effects of this peptide Here we show that Bj PRO 10c enhances and sustains the generation of nitric made (NO) by regulating argininosuccinate synthase activity and thereby velocity of the citrulline NO cycle Bj PRO 10c-mediated effects not restricted to the cardiovascular system since NO production was also induced in cells of astroglial origin Bj PRO 10c was internalized by C6 astroglioma cells where it induces NO production and upregulation of the citrulline NO cycle cells in a dose dependent fashion In view of that, astroglial cells function as L arginine pool for NO production in neighboring neurons, we suggest a regulatory function for Bj PRO-10c on the metabolism of this gaseous neurotransmitter in the CNS Moreover, proliferation of astroglial cells was reduced in the presence of Bj PRO 10c however, cell death was not induced Since NO donors have been studied for the treatment of solid cancers Bj PRO 10c may serve as structural model for developing drugs to improve the effects of cancer therapy based on the peptide`s ability to augment NO production (C) 2010 Elsevier B V All rights reserved

Bothropasin is a 48 kDa hemorrhagic PIII snake venom metalloprotease (SVMP) isolated from Bothrops jararaca, containing disintegrin/cysteine-rich adhesive domains. Here we present the crystal structure of bothropasin complexed with the inhibitor POL647. The catalytic domain consists of a scaffold of two subdomains organized similarly to those described for other SVMPs, including the zinc and calcium-binding sites. The free cysteine residue Cys(189) is located within a hydrophobic core and it is not available for disulfide bonding or other interactions. There is no identifiable secondary structure for the disintegrin domain, but instead it is composed mostly of loops stabilized by seven disulfide bonds and by two calcium ions. The ECD region is in a loop and is structurally related to the RGD region of RGD disintegrins, which are derived from I`ll SVMPs. The ECD motif is stabilized by the Cys(117)_Cys(310) disulfide bond (between the disintegrin and cysteine-rich domains) and by one calcium ion. The side chain of Glu(276) of the ECD motif is exposed to solvent and free to make interactions. In bothropasin, the HVR (hyper-variable region) described for other Pill SVMPs in the cysteine-rich domain, presents a well-conserved sequence with respect to several other Pill members from different species. We propose that this subset be referred to as PIII-HCR (highly conserved region) SVMPs. The differences in the disintegrin-like, cysteine-rich or disintegrin-like cysteine-rich domains may be involved in selecting target binding, which in turn could generate substrate diversity or specificity for the catalytic domain. This research intends to narrate the journey of a simple man who was turned into a saint by a group of believers in Mossoró, west of Rio Grande do Norte. We recognized in the story of José Leite Santana, the "cangaceiro" Jararacá, an appropriated land to build an investigative approach of how characters as different as bandit and saint can be interconnected. From the believers speech, a narrative was created about these detachments of concepts based on the hero myth that is shown here as a part of men´s dealing with death

Este artigo parte de um estudo etnográfico da devoção popular ao cangaceiro Jararaca em Mossoró, Rio Grande do Norte, morto pela polícia local em 1927. Seu objetivo é destacar os ritos que manifestam e suportam tal devoção, sobretudo em seus aspectos verbais: testemunhos, narrações hagiográficas e fatos históricos tidos como reais - principalmente os fatos que cercaram a morte do cangaceiro e suas proezas no cangaço - que dão margem à reelaboração da identidade social do morto, de modo a aproximar sua vida do modelo do Robin Hood, o bom bandido, e sua morte do modelo do martírio cristão. Assim, ele pode tornar-se um santo funerário.
