Techniques of production of enthomopatogenic bacteria are developed aiming to increase the productivity and to reduce the costs of the fermentative process. Like this, it has been using agroindustrial wastes or by-products as nutrient sources in culture medium, having been used, in this study, the manipueira, a by-product of the processing of the cassava flour. Fermentations were performed in flasks of Erlenmeyer of 500 mL containing 250 mL of culture media, conditioned in shaker at 180 r.p.m. and 28°C, and the media were composed by manipueira, in concentrations that varied between 400 and 1000 mL/L. The time of the process varied between 48 and 120 hours. They appraised the following parameters: cellular growth, the production of spores, the reduction of organic matter (COD analysis) and the variation of reduction sugar. Although there was a proportional cellular growth to the manipueira concentration, the production of spores was similar in all the cases, at the end of the process, in spite of the smallest speed of production of the same ones in the highest concentrations. In relation to the variation of COD, it has, also, a percentile minor of reduction in the highest concentrations. In the analysis of variation of reduction sugars, the higher concentrations are the ones that they present larger slowness in the reduction of this.

The effect of the addition of different concentratons of cystine and cysteine on sporulation and parasporal crystal formation in Bacillus thuringiensis var. thuringiensis was studied. The effect was well pronounced when the systine/cysteine additions were made after the stationary phase. Heat stable spores and crystals were formed when the culture was provided with a low concentration of cystine/cysteine (0.05 per cent w/v). At a moderate concentration of cystine or cysteine (0.15%), only heat labile spores were formed without the production of the crystal. When the cystine/cysteine concentration was high (0.25%), spore and crystal formation were completely inhibited. Partial reversal of inhibition of sporulation was brought about by sodium sulphate or zinc sulphate and lead, copper, cadmium or cobalt acetate at 0.2 mM or at 0.2% of sodium or potassium pyruvate, citrate, isaconitate, oxalosuccinate, ∝ -keto-glutarate, succinate, fumarate, malate, or oxalacetate. Glutamate (0.2%) overcame the inhibitory effect of cystine/cysteine completely. The structural changes observed using phase contrast microscopy were dependent upon the concentration of cystine/cysteine.

The antitumour protein from the α-endotoxin of Bacillus thuringiensis var. thuringiensis has been purified, crystallized and partially characterized. The same protein also shows the insecticidal activity. According to amino acid analysis it is an acidic protein with a molecular weight of approx. 13 000.

The proteinaceous crystal of Bacillus thuringiensis Var thuringiensis was found to enhance humoral immune response in rats and guinea pigs immunised with sheep red blood cells. The enhancement was due to the increased levels of both 19S and 7S antibodies in the sera of the treated animals. A novel synthesis of 7S haemolytic antibodies was observed in case of crystal treated animals.

Con el fin de conocer el crecimiento, rendimiento y toxicidad de bacillus thuringiensis var kurstaki 3ª3b producida en diferente medios de cultivo se llevaron a cabo fermentación en frasco Erlenmeyer el medio M-25, el medio base nacional y el medio base balanceado. Además se evaluaron este medio después de ser sometido a un proceso de hidrolisis. Aunque no son optimo, los medios probados en este estudio, se puede considerar adecuados para el crecimiento de bacillus thuringiensis. La hidrolisis aplicada al medio base nacional no produce crecimiento mayor de bacillus thuringiensis en comparación con el medio sin hidrolisis y induciendo a la bacteria la necesidad de un periodo más largo de adaptación durante el cual su crecimiento fue sostenidamente lento. La causa de este fenómeno es aun derminada. El balance nutricional realizado a la media base nacional resulta en un crecimiento inicial rápido de bacillus thuringiensis llegando al máximo punto de crecimiento entre 30 horas comparado con 72 horas en la base nacional. Sin embargo, la producción de esporas en este medio es inferior a la media base nacional bajo condiciones de fermentación utilizada. La cepa nativa de bacillus thuringiensis var kurstaki 3ª3b cultivada en las medias bases nacionales y balanceadas muestra toxicidad para las larvas de plutella xylostella en las concentraciones de 0.000001 a 0.01 mg/ml.
