Le facteur de transcription BP1 humain est exprimé dans les cellules érythroïdes pendant le développement fœtal mais son niveau d’expression est réduit au stade adulte. Les études antérieures in vitro ont montré que BP1 est un répresseur du gène adulte de β-globine mais sa fonction dans la régulation des gènes ε et γ n’a pas été abordée à ce jour. Dans notre étude, nos analyses de BP1 humain ont été menées in vivo au stade embryonnaire en utilisant une lignée de souris transgénique surexprimant BP1 dans les cellules érythroïdes définitives murines. Au niveau protéique, BP1 humain est exprimé aux âges E12.5 et E13.5 dans les cellules érythroïdes fœtales des embryons transgéniques. Toutefois, les niveaux de BP1 humain ne perturbent pas l’érythropoïèse définitive fœtale: les embryons transgéniques ne sont pas anémiques et ne meurent pas in utero. La surexpression de BP1 humain altère tout de même le niveau endogène des facteurs de transcription Ikaros et SOX6 impliqués dans la régulation des gènes de β-globine durant l’érythropoïèse définitive fœtale murine. Chez les embryons doubles transgéniques exprimant BP1 et les gènes humains de β-globine à E12.5, l’expression du gène adulte β est réduite alors que celle des gènes ε et γ est non réprimée. Les mesures d’expression des gènes humains de β-globine effectuées en absence d’Ikaros à E12.5 précisent le rôle de BP1 humain dans l’activation du gène embryonnaire ε. Dans les cellules érythroïdes fœtales murines dépourvues d’Ikaros à E12.5, BP1 humain augmente grandement l’expression des facteurs de transcription EKLF et BCL11A et semble déréprimer l’expression de SOX6, ce qui conduit à une répression des gènes fœtaux et une activation du gène adulte β au jour embryonnaire murin suivant. Puisque BP1 atténue l’altération de l’expression des gènes fœtaux et adultes causée par l’absence d’Ikaros, nous proposons que BP1 et Ikaros soient liés dans les mécanismes de transcription des gènes humains de β-globine.

Stimulation of phosphatidylinositol 3'-kinase (PI3K) and protein kinase B (PKB) is implicated in the regulation of protein synthesis in various cells. One mechanism involves PI3K/PKB-dependent phosphorylation of 4E-BP1, which dissociates from eIF4E, allowing initiation of translation from the 7-methylGTP cap of mRNAs. We examined the effects of insulin and H(2)O(2) on this pathway in neonatal cardiac myocytes. Cardiac myocyte protein synthesis was increased by insulin, but was inhibited by H(2)O(2). PI3K inhibitors attenuated basal levels of protein synthesis and inhibited the insulin-induced increase in protein synthesis. Insulin or H(2)O(2) increased the phosphorylation (activation) of PKB through PI3K, but, whereas insulin induced a sustained response, the response to H(2)O(2) was transient. 4E-BP1 was phosphorylated in unstimulated cells, and 4E-BP1 phosphorylation was increased by insulin. H(2)O(2) stimulated dephosphorylation of 4E-BP1 by increasing protein phosphatase (PP1/PP2A) activity. This increased the association of 4E-BP1 with eIF4E, consistent with H(2)O(2) inhibition of protein synthesis. The effects of H(2)O(2) were sufficient to override the stimulation of protein synthesis and 4E-BP1 phosphorylation induced by insulin. These results indicate that PI3K and PKB are important regulators of protein synthesis in cardiac myocytes, but other factors, including phosphatase activity, modulate the overall response.

Infection of cells with picornaviruses, such as poliovirus and encephalomyocarditis virus (EMCV), causes a shutoff of host protein synthesis. The molecular mechanism of the shutoff has been partly elucidated for poliovirus but not for EMCV. Translation initiation in eukaryotes is facilitated by the mRNA 5' cap structure to which the multisubunit translation initiation factor eIF4F binds to promote ribosome binding. Picornaviruses use a mechanism for the translation of their RNA that is independent of the cap structure. Poliovirus infection engenders the cleavage of the eIF4G (formerly p220) component of eIF4F and renders this complex inactive for cap-dependent translation. In contrast, EMCV infection does not result in eIF4G cleavage. Here, we report that both EMCV and poliovirus activate a translational repressor, 4E-BP1, that inhibits cap-dependent translation by binding to the cap-binding subunit eIF4E. Binding of eIF4E occurs only to the underphosphorylated form of 4E-BP1, and this interaction is highly regulated in cells. We show that 4E-BP1 becomes dephosphorylated upon infection with both EMCV and poliovirus. Dephosphorylation of 4E-BP1 temporally coincides with the shutoff of protein synthesis by EMCV but lags behind the shutoff and eIF4G cleavage in poliovirus-infected cells. Dephosphorylation of 4E-BP1 by specifically inhibiting cap-dependent translation may be the major cause of the shutoff phenomenon in EMCV-infected cells.

It has previously been argued that the repressor of protein synthesis initiation factor 4E, 4E-BP1, is a direct in vivo target of p42mapk. However, the immunosuppressant rapamycin blocks serum-induced 4E-BP1 phosphorylation and, in parallel, p70s6k activation, with no apparent effect on p42mapk activation. Consistent with this finding, the kinetics of serum-induced 4E-BP1 phosphorylation closely follow those of p70s6k activation rather than those of p42mapk. More striking, insulin, which does not induce p42mapk activation in human 293 cells or Swiss mouse 3T3 cells, induces 4E-BP1 phosphorylation and p70s6k activation in both cell types. Anisomycin, which, like insulin, does not activate p42mapk, promotes a small parallel increase in 4E-BP1 phosphorylation and p70s6k activation. The insulin effect on 4E-BP1 phosphorylation and p70s6k activation in both cell types is blocked by SQ20006, wortmannin, and rapamycin. These three inhibitors have no effect on p42mapk activation induced by phorbol 12-tetradecanoate 13-acetate, though wortmannin partially suppresses both the p70s6k response and the 4E-BP1 response. Finally, in porcine aortic endothelial cells stably transfected with either the wild-type platelet-derived growth factor receptor or a mutant receptor bearing the double point mutation 740F/751F, p42mapk activation in response to platelet-derived growth factor is unimpaired, but increased 4E-BP1 phosphorylation is ablated, as previously reported for p70s6k. The data presented here demonstrate that 4E-BP1 phosphorylation is mediated by the FRAP-p70s6k pathway and is independent of mitogen-activated protein kinase.

The 15q11.2-q13 region has been well characterized, being associated with a range of syndromatic copy number variants (CNVs), and comprises five established break points sites (BP1 to BP5). While the clinical effect for BP1-BP3, BP2-BP3 and BP4-BP5 CNVs is well established, the same cannot be said for BP1-BP2 CNVs. Recently the 15q11.2 BP1-BP2 deletion has been reviewed, emerging as a microdeletion syndrome with low penetrance and variable expressivity being the CNV frequently inherited from a healthy parent. This microdeletion is considered to be a risk factor for several neurodevelopment disorders. For the reciprocal duplication the picture has been less conclusive. Aiming for a better understanding of the clinical significance of this CNV, we collected patients with intellectual disability and/or other clinical features, referred for microarray testing, gathering clinical details for the ones with the duplication. Data was collected from two genetic laboratories. With a total of 1545 patients, we identified eleven carrying the duplication at 15q11.2 BP1-BP2. It was possible to assess inheritance in only four cases, all inherited from a healthy parent. All patients presented intellectual disability,and facial dysmorphism was the second most common feature observed. Microcephaly, autism, congenital abnormalities, dystonia and cataplexy where reported individually. The magnitude of the effect of 15q11.2 duplication remains elusive, and the outcome unclear, posing a major challenge to genetic counseling. Nevertheless, we expect the collection of more of these cases will establish this gain, as it happened with the reciprocal deletion, as a microduplication syndrome with low penetrance and variable expressivity.

Leucine is a key amino acid for initiating translation in muscle cells, but the dose-dependent effects of leucine on intracellular signaling are poorly characterized. This study examined the effect that increasing doses of leucine would have on changes in mechanistic target of rapamycin (mTOR)–mediated signaling, rates of protein synthesis, and cell size in C2C12 cells. We hypothesized that a leucine “threshold” exists, which represents the minimum stimulus required to initiate mTOR signaling in muscle cells. Acute exposure to 1.5, 3.2, 5.0, and 16.1 mM leucine increased phosphorylation of mTORSer2448 (~1.4-fold; P < .04), 4E-BP1 Thr37/46 (~1.9-fold; P < .001), and rpS6Ser235/6 (~2.3-fold; P < .001). However, only p70S6kThr389 exhibited a dose-dependent response to leucine with all treatments higher than control (~4-fold; P < .001) and at least 5 mM higher than the 1.5-mM concentration (1.2-fold; P < .02). Rates of protein synthesis were not altered by any treatment. Seven days of exposure to 0.5, 1.5, 5.0, and 16.5 mM leucine resulted in an increase in cell size in at least 5 mM treatments (~1.6-fold, P < .001 vs control). Our findings indicate that even at low leucine concentrations, phosphorylation of proteins regulating translation initiation signaling is enhanced. The phosphorylation of p70S6kThr389 follows a leucine dose-response relationship, although this was not reflected by the acute protein synthetic response. Nevertheless, under the conditions of the present study, it appears that leucine concentrations of at least 5 mM are necessary to enhance cell growth.

本文以苯并(a)芘为目标污染物,探讨了母体化合物BaP及其次生代谢产物的连续降解的方法、降解过程和微生物的酶蛋白应答,并运用种子(小麦、白菜和萝卜)根伸长生长实验,考查了BaP的不同降解时期次生代谢产物造成的复合污染整体效应,旨在探讨BaP及其次生代谢产物的降解影响因素,减少其环境累积,为BaP污染环境的全面修复提供实验依据和理论基础。 在实验条件下,运用HPLC鉴定出真菌FZSY-1降解BaP的同时生成了三个次生代谢产物BP1,6-quinone, BP7,8-diol 和 3-OHBP;同时鉴定出真菌FZSY-2降解BaP的同时生成了两个代谢产物BP1,6-quinone 和 3-OHBP。 驯化微生物与氧化剂(KMnO4)的耦合降解系统对BaP及其代谢产物的连续降解效果好于单纯微生物降解。三个次生代谢产物中,BP1,6-quinone在环境中最易累积。同时提出了微生物与氧化剂协同的作用可以有效促进环境中持久有机污染物(尤其是高浓度,小面积污染)的连续降解。对于FZSY-1与氧化剂(KMnO4)耦合降解BaP,在TW80存在下,与对照(未加TW80)相比,在降解的前期(3天取样),BaP及其代谢产物的降解相对滞后;而在降解的后期(12天取样),BaP及其代谢产物的降解高于对照。 在不同BaP浓度下,检测了四种酶,C120、C230、CAT和PPO。三株细菌的CAT酶活与BaP的浓度无关;三株细菌的C230酶活都比较高;三株细菌的PPO酶活均较低。加入共代谢底物(琥珀酸钠)后,与对照(未加入共代谢底物)相比,C120、C230酶活明显提高。 以BaP以及FZSY-1(BZSY-2)降解BaP不同时期的复合降解产物(BaP,M6,M12,CK)为目标污染物,它们对小麦种子根伸长的抑制作用顺序为:M6﹥BaP﹥M12﹥CK。BaP,M6,M12,CK对白菜和萝卜种子根伸长的抑制作用顺序和小麦相同;同一目标污染物(M6)对这几种供试种子(小麦、白菜和萝卜)根伸长的抑制作用顺序为小麦﹥白菜﹥萝卜;三种植物种子根伸长抑制作用均表现为:真菌的M6﹥细菌的M6,真菌的M12﹥细菌的M12。

Aims-An increased concentration of insulin-like growth factor 1 (IGF-1) is an independent risk factor for premenopausal breast cancer. Tamoxifen is thought initially to reduce concentrations of IGF-1 and increase concentrations of the IGF binding proteins. The aim of this study was to compare concentrations of IGF-1, IGF binding protein 1 (IGF-BP1), and IGF-BP3 in patients with breast cancer (n = 14) with those seen in control subjects (n = 23) and to assess the effect of tamoxifen on IGF status in these patients.

PRL-3, a metastasis-associated phosphatase, is known to exert its oncogenic functions through activation of PI3K/Akt, which is a key regulator of the rapamycin-sensitive mTOR complex 1 (mTORC1), but a coherent link between PRL-3 and activation of mTOR has not yet been formally demonstrated. We report a positive correlation between PRL-3 expression and mTOR phospho-activation in clinical tumour samples and mouse models of cancer and demonstrate that PRL-3 increased downstream signalling to the mTOR substrates, p70S6K and 4E-BP1, by increasing PI3K/Akt-mediated activation of Rheb-GTP via TSC2 suppression. We also show that PRL-3 increases mTOR translocation to lysosomes via increased mTOR binding affinity to Rag GTPases in an Akt-independent manner, demonstrating a previously undescribed mechanism of action for PRL-3. PRL-3 also enhanced matrix metalloproteinase-2 secretion and cellular invasiveness via activation of mTOR, attributes which were sensitive to rapamycin treatment. The downstream effects of PRL-3 were maintained even under conditions of environmental stress, suggesting that PRL-3 provides a strategic survival advantage to tumour cells via its effects on mTOR.

The mammalian target of rapamycin complex 1 (mTORC1) is a highly conserved protein complex regulating key pathways in cell growth. Hyperactivation of mTORC1 is implicated in numerous cancers, thus making it a potential broad-spectrum chemotherapeutic target. Here, we characterized how mTORC1 responds to cell death induced by various anticancer drugs such rapamycin, etoposide, cisplatin, curcumin, staurosporine and Fas ligand. All treatments induced cleavage in the mTORC1 component, raptor, resulting in decreased raptor-mTOR interaction and subsequent inhibition of the mTORC1-mediated phosphorylation of downstream substrates (S6K and 4E-BP1). The cleavage was primarily mediated by caspase-6 and occurred at two sites. Mutagenesis at one of these sites, conferred resistance to cell death, indicating that raptor cleavage is important in chemotherapeutic apoptosis.

Le système rénine-angiotensine-aldostérone (SRAA) régule l’homéostasie de la contraction des artères. Or, suivant la liaison de l’angiotensine II (Ang II) à son récepteur AT1, le SRAA est également impliqué dans l’activation de voies de signalisation à l’origine de l’inflammation et de l’hypertrophie des cellules musculaires lisses vasculaires (CMLV), soit deux processus participant au remodelage vasculaire caractéristique de diverses maladies cardiovasculaires, telles l’hypertension et l’athérosclérose. Ces pathologies sont les premières causes de mortalité naturelle en Amérique et les traitements les ciblant ne sont pas optimaux puisqu’ils visent seulement quelques facteurs de risque qui leur sont associés. Ainsi, la détermination des effecteurs intracellulaires régulant ces voies délétères est nécessaire à l'identification de nouvelles cibles thérapeutiques. L’inflammation Ang II-dépendante dans les CMLV est attribuée au facteur de transcription nuclear factor-kappa B (NF-κB). Cependant, les processus moléculaires couplant le récepteur AT1 à son activation sont peu caractérisés. L’étude abordant cette question démontre in vitro que NF-κB est activé par la protéine IκB kinase β (IKKβ) dans les CMLV exposées à l’Ang II et que cette kinase est régulée par deux voies de signalisation indépendantes, mais complémentaires afin d’assurer son activation rigoureuse et soutenue. L’une des voies est précoce et dépend des seconds messagers ainsi que de deux nouveaux effecteurs sous-jacents au récepteur AT1, soit la E3 ligase TNF receptor-associated factor 6 (TRAF6) et la IKK kinase transforming growth factor-beta-activated kinase 1 (TAK1) tandis que la seconde est tardive et résulte de la signalisation mitogen-activated protein kinase kinase 1/2 (MEK1/2) - extracellular signal-regulated kinase 1/2 (ERK1/2) - ribosomal S6 kinase (RSK). L’inhibition conjointe de ces voies abroge complètement la réponse inflammatoire, ce qui indique qu’elles en sont la seule source. Ainsi, l’inhibition d’IKKβ pourrait suffire à contrer l’inflammation impliquée dans le remodelage vasculaire associé à une suractivation du SRAA. Une découverte des plus novatrices découle de cette étude, qui veut que la E3 ligase TRAF6 est un nouvel effecteur des récepteurs couplés aux protéines G et est à l’origine de la formation d’un nouveau type de second messager, soit des chaînes libres de poly-ubiquitines. Les mécanismes moléculaires à la base de l’hypertrophie Ang II-dépendante dans les CMLV sont également peu définis. Or, suivant la parution d’un article démontrant qu’IKKβ dans les cellules cancéreuses participe aux mécanismes d’initiation de la traduction en réponse au facteur de nécrose tumorale α (TNFα) via la phosphorylation de la protéine Tuberous sclerosis 1 (TSC1) et donc l’activation du complexe mammalian target of rapamycin (mTORC1), une hypothèse a été émise selon laquelle cette kinase serait impliquée dans la synthèse protéique Ang II-dépendante dans les CMLV. Les expériences effectuées in vitro dans des CMLV exposées à l’Ang II démontrent qu’IKKβ induit la phosphorylation de TSC1 ainsi que l’activation de mTORC1 et de ses substrats S6 kinase 1 (S6K1) et translational regulators eukaryotic translation initiation factor 4E-binding protein (4E-BP1), deux protéines impliquées directement dans l’hypertrophie. Par ailleurs, la synthèse protéique au niveau des CMLV exposées à l’Ang II est réduite de 75% suivant la diminution de l’expression d’IKKβ et suivant la surexpression d’un mutant de TSC1 dont le site consensus d’IKKβ a été modifié, faisant de cette kinase un médiateur majeur au niveau de ce processus. Ainsi, in vitro IKKβ en réponse à l’Ang II est en amont de deux processus impliqués dans un remodelage vasculaire à l’origine de maladies cardiovasculaires. De plus, plusieurs facteurs de risque de ces pathologies convergent à l’activation d’IKKβ, ce qui en fait une cible thérapeutique particulièrement attrayante. Qui plus est, l’administration d’un inhibiteur d’IKKβ à des rats diminue non seulement la synthèse protéique dépendante de l’Ang II au niveau de l’aorte et des artères mésentériques, mais également la synthèse de la protéine pro-inflammatoire VCAM-1 par les cellules composant l’aorte, ce qui confirme son envergure en tant que cible.
